A Clinical study of Atopic Dermatitis treated by external application with Gynostemma Pentaphyllum MAKINO in cosmetics (아토피 피부염 환자에서 칠엽담(七葉膽)을 함유한 화장품의 임상적 연구)

  • Objectives : This study was conducted to test patients with Atopic Dermatitis by external application with Gynostemma Pentaphyllum MAKINO in cosmetics. Methods : We gave scores to 59 patients who visited the Dept. of Oriental Medical Opthalmology & Otolaryngology & Dermatology of Daegu Hanny Oriental Medical Center from July 1st, 2005 to December 31th. We checked the clinic index score(extent. intensity : the degree of erythema, induration or papulation, puritus, excorination, lichenification, scaling or dryness, erosion or oozing or weeping. the degree of itching), Eosinophil count and IgE. Results : There are 46 patients we could check by clinic index score, and 44 patients by laboratory. 1. In the 46 cases clinic index score was significantly decreased between the before- and afterexternal application treatment periods(P<0.01). 2. The 46 cases was devided into two types (moist heat type and blood dryness type) by intensity. External application of Gynostemma Pentaphyllum MAKINO in cosmetics did not significant change by two types. 3. In the 44 laboratory cases, the number of eosinophil counts was decreased but the number of IgE was significantly increased. Conclusions : Considering the above results, we have concluded that cosmetics with Gynostemma Pentaphyllum MAKINO have the remedical effect for Atopic Dermatitis.

  • This study was carried out in order to analyse the amino acid constituents of Gynostemma pentaphyllum Makino (G. P. M. ) produced in Korea and Japan. Total and free amino acids of those were analysed by HPLC. Total amino acid contents of G. P. M. produced in Korea were similar to that of produced in Japan. The Histidine content of G. P. M. produced in Korea was more rich than that of produced in Japan. The content of glutamic acid was the most rich among in kinds of amino acids in both hind of G. P. M. G. P. M. in Korea contained the least methionine and G. P. M. from Japan contained the least cystine.

Antioxidative Activity of Gynostemma pentaphyllum Makino Extracts (덩굴차(Gynostemma pentaphyllum Makino) 추출물의 항산화 효과)

  • This study was carried out to examine the antioxidant activity of Gynostemma pentaphyllum Makino. Using the DPPH method, we found that free radical scavenging activity was strong in all the fractions except the water fraction of the water extract (GPW) and ethanol extract (GPE) of G pentaphyllum Makino. Pseudo-SOD activity was highest in the diethyl ether fraction of the ethanol extract, while the other fractions of the ethanol and water extracts were lower. For xanthine oxidase inlribition activity, the diethyl ether fraction and butanol fraction of GPW, and the diethyl ether fraction and butanol fraction of GPE, showed activities over 80%. Nitrite scavenging ability was strong (over 60%) in all the GPW and GPE fractions. The diethyl ether fraction and butanol fraction of GPW had more effective nitrite scavenging abilities than the other extract fractions. These results suggest that the extracts of G pentaphyllum Makino can be used as a functional material in a tea or drink.

Antioxidative and Cellular Protective Effects of Dolwoe (Gynostemma pentaphyllum Makino) Extracts against Oxidative Stress (돌외(Gynostemma pentaphyllum Makino) 추출물의 산화적 스트레스에 대한 항산화 및 세포보호효과)

  • In this study, we investigated the total phenolic and flavonoid contents, component analysis, antioxidative activity and cellular protective effects against oxidative stress on human skin cells in 50% ethanol extract, ethyl acetate fraction and aglycone fraction obtained from Gynostemma pentaphyllum (G. pentaphyllum) Makino. The DPPH (1,1-diphenyl-2-picrylhydrazyl) scavenging activites ($FSC_{50}$) of the 50% ethanol extracts, ethyl acetate fraction and aglycone fraction were 246.8, 147.2, $128.9{\mu}g/mL$, respectively. Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of the 50% ethanol extract, ethyl acetate fraction and aglycone fraction on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system using the luminol-dependent chemiluminescence assay were 37.15, 10.74, $7.19{\mu}g/mL$, respectively. We investigated the cellular protective activity and the results showed that treatment of aglycone fraction ($0.05-0.39{\mu}g/mL$) protect human skin cells in a concentration-dependent manner when the skin cell damages were induced by treating them with $H_2O_2$. These results suggest that extract/fractions of G. pentaphyllum Makino may be applicable as natural antioxidants in cosmetics.

Evaluation of the inhibitory effect of Gynostemma pentaphyllum extracts on CYP450 enzyme activities using LC-MS/MS

  • Gynostemma pentaphyllum (Thunb.) Makino extract, a natural product with a history of traditional use, has gained attention for its potential health benefits. This study aimed to investigate its effects on key cytochrome P450 (CYP) enzymes using LC-MS/MS. Human liver microsomes and cDNA-expressed CYP2C8, CYP2C9, CYP2C19, and CYP3A4 supersomes were employed. Enzyme activity was assessed based on the formation of CYP-specific marker metabolites. The resulting data showed that the extract exhibited inhibitory effects on CYP2C8, CYP2C9, CYP2C19, and CYP3A4. Thus, G. pentaphyllum extract may influence the pharmacokinetics of drugs metabolized by CYP2C8, CYP2C9, CYP2C19, and CYP3A4. These findings emphasize the importance of considering potential herb-drug interactions when incorporating this extract into therapeutic regimens or dietary supplements.

  • Gynostemma pentaphyllum Makino is a kind of perennial liana plant belonging to the Cucurbitaceae family. Since it was first discovered and reported to the Japanese academy in 1977, it came to be widely known to China and Southeast Asia areas as well as Japan. In Korea its name began to appear in the first half of 1980's, and it is now being nationwidely cultivated and popularized as tea stuffs. Gynostemma pentaphyllum Makino has remarkably strong propagation power. In addition. since it contains a great quantity of beneficial components, especially saponin it is believed to have excellent medical effects, too. This study was undertaken to examine the value of Gynostemma pentaphyllum Makino as a health-promotint tea, by investigating the components creating the flaver of tea and the saponin ingredient making medical effects as well as by inspecting whether Gynostemma pentaphyllum Makino products show different component arrangements according to their growing districts. As raw materials of the experiment, three kinds of Gynostemma pentaphyllum Makino samples produced in Suwon, Geochang and Uleungdo were taken. To accomplish the formerly stated goals, the contained quantity of the proximate composition, free sugar, reducing sugar, free amino acid, minerals, tannin, caffeine and vitamin C were measured respectively and compared between the samples, while the saponin content was compared with heat extracted from panax ginseng. The results of the experiment are as follows: 1. In the case of the proximate composition, the crude fat content was the highest in the Gymostemma pentaphyllum Makino material from Geochang (1.62%), the second in that from Suwon (1.56%), and the lowest in that from Uleungdo (1.0%). In addition the Geochang: produced sample had the greatest quantity of the crude protein and ash contents: the order of the crude Protein content was the Geochang produced (17.83%), the Suwon-produced (15.87%), and the Uleungdo-produced(12.28%), while that of the ash content was the Geochang-produced (14.80%), the Uleungdo-produced(10.17%), and the Suwon-produced(9.34%). 2. As for the reducing sugar contents, the Suwon-produced scored the highest of the three (3.3%), while the Geochang-produced was 1.3% and the Uleungdo-produced 0.5%. The total content of free sugar was 1.07% (the Suwon-produced), and 0.49% (the Geochang-produced) respectively but the sample from Uleungdo showed almost no free sugar content. The contained quantity of fructose and glucose was the highest among the kinds of free sugar both in the Suwon-produced and in the Geochang-produced. 3. The content of amino acid was the highest in the Suwon-produced(1.41%), the second in the Geochang-produced(1.37%), and the lowest in the Uleungdo-produced(0.53%). In the experiment, sixteen kinds of amino acid were extracted-Asp. Thr. Ser. Glu. Gly. Ala. Val. Het. Ileu. Leu. Tyr. Phe. Lys. His. Arg. Try. All of them except glutamic acid and methionine showed the highest quantity score in the Suwon-produced, while the glutamic acid content was the higest in the Uleungdo-produced and the methionine content in the Geochang-produced. The sequential arrangement of the sixteen contents according to their magnitude ranged from glycine, aspartic acid, and glutamic acid (the highest) to tryptophan, serine and lysine (the lowest). 4. Ten kinds of mineral were detected-Ca, Mn, Cd, K, Na, Pb, Mg, Fe, Zn, Cu. Among them, the content of Cd, Na, Mg, Zn and Fe was the highest in the Geochang-produced and that of K in the Suwon-produced. 5. The Geochang-produced materials contained much a larger quantity of tannin (6.3%) than the Suwon-produced (2.6%). Neither caffeine nor vitamin C was detected in the three kinds of materials. 6. In the case of the saponin content the Geochang-produced showd 2.39%, the Uleungdo-produced 1.77% and the Suwon-produced 1.49% respectively. However, it was found also that the saponin content of Gymostemma pentaphyllum Makino was the same kind as that of panax ginseng.

  • This study was conducted to obtain basic information on growth characteristics and major components of three Korean endemic resource plant species, Artemisia sp., Gynostemma pentaphyllum Makino and Humulus japonicus S. et Z. , growing in different areas in Korea. Three geographical types of artemisia, namely Pangssuk, Ongjinssuk and Yakssuk which were collect in Suwon, Ongjin and Eumsong, respectively were compared in yield, and Pangssuk artemisia was higher in yield than the others. However, essential oil content was the highest in Yakssuk artemisia. Dry weight of areal parts of G. pentaphyllum was higher in the cultural method with props for tendril growth than in the conventional cultural method without props. Rutin was detected in areal parts of G. pentaphyllum. Dry weight of areal parts of H. japonicus collected at the Kyeryong mountain in Chungnam province was higher than that collected in Eumsong. A phenolic compound isoquercitrin was detected in areal parts of H. japonicus.

A Study on the Sugar Contents of Dolwoe Tea(Gynostemma pentaphyllum Makino) (돌외차(Gynostemma pentaphyllum Makino)의 당(糖)에 관한 연구)

  • The sugar contents of Dolwoe, which were prepared in the laboratory and marketed Korean products of Dolwoe, Japanese product of Dolwoe and Korean-roasted products of Dolwoe were analyzed and compared. The contents of total sugar and reducing sugar of stem in prepared Dolwoe tea were 2 times higher than those of leaf. The sugar content of Korean product was similar to that of Japanese, but the contents of total sugar and reducing sugar in Korean-roasted sample decreased to 17.2% and 40.9%, respectively. It was confirmed that free sugar in Dolwoe tea was composed of rhamnose, fructose, glucose and sucrose. The ratio of free sugar content was quite different between Dolwoe-leaf tea and Dolwoe-stem tea. Comparing the free sugar content in prepared Dolwoe tea, glucose was the highest followed by fructose, sucrose and rhamnose. On the other hand, in marketed Dolwoe tea, glucose was the highest substance and followed by fructose, rhamnose and sucrose. It was found that rhamnose content in Japanese product was approximately 6 times more than that of Korean.

  • The mineral content of Dolowe, which were prepared in the laboratory and marketed Korean products of Dolwoe, Japanese product of Dolwoe and Korean-roasted products of Dolwoe were analysed and compared. Dolwoe-leaf tea had high mineral content of Ca, Mg, Mn and Cu, while Dolwone-stem tea had high mineral content of K, Na and Fe. Especially in the ash of Dolwoe-stem tea showed the highest mineral content of K (33.4%) Dolwoe teas were extracted for 5 minutes as ratio of $1,000m{\ell}$ boiling water to 4g Dolwoe tea (250: 1, v/w). Comparing the ratio of extraction of minerals in Dolwoe tea made by us and marketed Dolwoe products, K was the most abundant element of all extractions with the rate of $83.2%{\sim}96.3%$ followed by Mg with $44.8%{\sim}62.2%$, Ca with $23.1%{\sim}64.4%$ and Na with $6.3%{\sim}55.6%$. On the other hand Mn, Fe and Cu were extracted in extremely small quantities.

Screening of the Antibacterial Effects by Ethanol Extracts from Natural Plant in Jeju against Propionibacterium acnes (Propionibacterium acnes에 대한 제주 자생식물 에탄올 추출물의 항균효과 검색)

  • Propionibacterium acnes, a major pathogens bacterium of acne, is one of the microflora associated with the inflammation. In this study, the antibacterial effects of 70% ethanol extracts from the 61 natural plants in Jeju against P. acnes were investigated using the disc diffusion method. The 45 natural plant extracts showed antibacterial effects and the 16 natural plants extracts didn't show antibacterial effects. The Phyllanthus urinaria L. (stem and leaves) extract showed thr highest antimicrobial activity with $18.96{\pm}0.69mm$ clear zone of the growth of P. acnes, followed by Angelica gigas Nakai (roots), Gynostemma pentaphyllum (Thunb.) Makino, Angelica gigas Nakai (stem and leaves), Isodon inflexus (Thunb.) Kudo, camellia sinensis L. (flowers), Rosmarinus officinalis, etc. These results suggest that the natural plant in Jeju can be used as functional cosmetic biomaterials or antimicrobial agents against P. acnes.
