항균성 펩타이드인 Apidaecin Ib의 살조효과 (The Algicidal Effect of Antimicrobial Peptide, Apidaecin Ib)

  • 꿀벌 (Apis mellifera)의 lymph 액으로부터 정제된 항균성 펩타이드인, Apidaecin Ⅰb는 18개의 아미노산 잔기로 구성된 염기성 non-helical 펩타이드이다. 본 연구에서는 우리나라 연안의 적조 (HABs, harmful algal blooms)를 일으키는 4종의 적조생물 (Alexandrium tamarense, Chattonella marina, Cochlodinium polykrikoides 및 Gymnodinium catenatum)에 대한 Apidaecin Ⅰb의 살조효과를 조사하였다. Apidaecin Ⅰb의 4종의 적조생물에 대한 살조효과는 12.5$\mu{g}/mL$부터 50$\mu{g}/mL$에서 세포의 lysis 또는 ecdysis와 같은 형태로 현미경으로 관찰할 수 있었다. 또한 Apidaecin Ⅰb는 C. marina에 대해서 A. tamarense, C. polykrikorides 및 G. catenatum보다 더욱 강한 살조효과를 나타내었다. 이러한 HABs에 대한 Apidaecin Ⅰb의 살조효과 연구는 새로운 살조물질을 개발하기 위한 자료가 될 것으로 생각된다.

남해안 연안에서 적조생물, Cochlodinium polykikoides, Gyrodinium impudicum, Gymnodinium catenatum의 출현상황과 온도, 염분, 조도 및 영양염류에 따른 성장특성 (Abundance of Harmful Algae, Cochlodinium polykrikoides, Gyrodinium impudicum and Gymnodinium catenatum in the Coastal Area of South Sea of Korea and Their Effects of Temperature, Salinity, Irradiance and Nutrient on the Growth in Culture)

  • 적조생물 Cochlodinium polykrikoides, Gyrodinium impudicum, Gymnodinium catenatum은 독성을 지니거나, 적조를 일으킴으로써 수산피해 및 보건위생상의 문제를 야기시키는 종이다. 이 종들의 적조발생 환경과 기작을 이해하기 위해서는 종별 생태생리 (eco-physiology) 특성 등을 파악할 필요가 있다. 본 실험에서는 한국 남해안 연안에서 이들 3종의 출현상황과 성장특성을 파악하기 위해 이 해역에서 분리한 종을 대상으로 온도, 염분, 조도 및 영양염류에 따른 성장도를 조사하였다. 1999년도 남해안 남해도, 나로도, 완도 연안에서 이들 3종의 최초출현시기는 수온이 $22.8\sim26.5^{\circ}C$인 7월 중순에서 8월 중순으로써 서로 비슷한 시기에 동반 출현하였다. 유영세포의 소멸시기는 G. catenatum의 경우 8월 중, 하순이었고, C. polykrikoides와 G. impudicum은 수온이 $23^{\circ}C$ 이하로 하강하는 9월 하순이었다. 출현기간 중의 최대밀도는 C. polykrikoides의 경우 $40\times10^6$cells/L 이상으로써 고밀도 증식을 하였으나, G. impudicum과 C. catenatum은 각각 3,460ce11s/L 및 440ce11s/L로써 매우 낮은 밀도로 존재하였다. 배양실험에서 C. polykrikoides, G. impudicum, G. catenatum는 $22\sim28^{\circ}C$에서 양호한 성장을 보였고, 최적수온은 $25^{\circ}C$ 내외로 판단되었는데, 이러한 결과는 적조발생시의 수온과 대체로 일치하였다. 염분에 따른 성장률은 3종 모두 $30\sim35\%$에서 양호한 성장률을 보였다. 3종 중 G. impudicum은 비교적 광염성의 특징을 보였고, G. cstenatum은 $35\%$ 이상의 고염분에서 특히, 저조한 성장률을 보였다. 조도에 따른 성장은 C. polykrikoides와 G. impudicum의 경우 특히 7,5001ux 이상의 고조도에서 성장률이 현저히 높은 것으로 나타났다. 이러한 결과는 C. polykrikoides의 경우 조도가 높은 하계에 표층에서 강한 집적현상을 보이면서도 광저해현상을 밟지 않고 양호한 증식을 할 수 있는 특성과 관련이 있을 것으로 추정되었다. C. polykrikoides와 G. impudicum의 질산 및 암모니아 질소 농도에 따른 성장은 $40{\mu}M$까지는 농도가 높을수록 성장률도 증가하였으나 그 이상에서는 큰 차이를 보이지 않아, 두 종의 질소 임계농도는 $13.5\~40{\mu}M$로 판단되었다. 또한, 인산인은 $4.05{\mu}M$ 까지는 농도가 높을수록 성장률도 증가하였으나 그 이상에서는 큰 성장차를 보이지 않아, 두 종 모두 인산인의 임계농도는 $1.35\sim4.05{\mu}M$로 판단되었다. 한편, C. polyklikoides는 DIN과 DIP 농도가 각각 $1.2{\mu}M$$0.3{\mu}M$ 이하로 낮았던 나로도와 남해도 외측해역에서도 적조를 형성하였다. 이와 같이 낮은 영양염류 하에서 왕성하게 증식할 수 있었던 이유는 이 종의 경우 일간 수직이동을 통해 야간에 저층에서 풍부한 영양염류를 흡수할 수 있었기 때문으로 해석되었다.

진동만의 살조세균의 동태와 살조 특성 (The Activities and Characteristics of Algicidal Bacteria in Chindong Bay)

  • 적조 다발지역인 진동만에서 살조세균의 동태를 파악하기 위하여, 1998년 5월 19일부터 7월 7일까지, chlorophyll-a 및 살조세균의 계수$\cdot$분리하였고, 분리한 살조세균의 살조특성을 조사하였다. 본 연구기간동안 살조세균의 밀도는 $6.0\times10^1\~6.4\times10^5$cell $\ell^{-1}$의 범위에서 변동하였으며, chlorophyll-a의 농도의 증가 및 감소에 대하여 각각 1주일의 lag time을 두고 이와 동일한 변동양상을 나타내었다. 현장시료에서 분리된 3종의 살조세균 H519S5-4, H605S5-15 및 H605S5-22 균주의 살조범위를 침편모조류 Heterosigma akashiwo와 Chattonelia sp., 와편모조류 Gymnodinium catenatum, Gyrodinium impudicum 및 Cochlodinium polyklikoides, 규조류 Chaetoceros sp., Cescinodiscus granii, Ditylum brightwellii 및 Thalassiosira rotula를 대상으로 조사하였다. H60585-22 균주는 실험에 사용된 9종의 미세조류를 전부 사멸시켜 광범위한 살조범위를 나타내었다. 또한, 균주 H605S5-15는 H. akashiwo, G. catenatum, T. rotula 및 Chattonella sp.에 대하여, H519S5-4는 H. akashiwo, Chattonella sp., Chaetoceros sp. 및 G. catenatum에 대하여 살조력을 나타내었다. 살조유형은 3종 모두 세포 외 물질을 분비하여 살조하는 살조인자분비형 세균으로 나타났다. 세균이 나타내는 살조력은 식물성 플랑크톤의 군집동태에 영향을 미치는 중요한 요인이며, 연안에서 적조의 급격한 소멸에 관여하는 것으로 사료된다.

한국 연안에 출현하는 무각와편모조류 5종에 대한 형태 및 분자계통학적 특성 (Morphological Characteristics and Molecular Phylogeny of Five Unarmored Dinoflagellates in Korean Coastal Waters)

  • Many species in Gymnodiniales, which are unarmored dinoflagellates, are responsible for marine algal blooms and some of them have potent toxin in the cell. Their taxonomy has so far been well-defined, and several genera (e.g. Akashiwo, Gymnodinium, Karenia) have recently been re-described. In Korea, few works have been carried out on their taxonomical and molecular studies. This study focused on comparison of both morphological and molecular characteristics of five unarmored dinoflagellates on Korean coastal water: Akashiwo sanguinea, Cochlodinium polykrikoides, Gymnodinium catenatum, Gymnodinium impudicum and Karenia aureolum (=K. mikimotoi). Morphological characteristics observed here was in good accordance with the original descriptions of individual species. In addition, none of difference was found in morphological comparisons between the Korean and foreign strains. Furthermore, molecular analysis showed that the SSU rDNA sequences were generally identical according to each species. In some distinct features, A. sanguinea, which has generally the same morphological features, were divided into two groups: one was Korean isolates including European isolates, the other was American isolates. In the two groups, the nucleus was positioned differently: middle of the cells in the Korean isolates (GnSg02, GnSg03), near the epicone in American isolates (CCMP1593, CCMP1837). In addition, this was strongly supported by phylogenetic analysis, inferred from the SSU rDNA sequences. K. aureolum (GrAr01) was corresponded to European G. aureolum (=K. mikimotoi) in shape and position of nucleus, chloroplast, however, which is similar to K. digitata in view of having a finger-like sulcus. This was in good agreement with phylogenetic study of these species. G. catenatum have identical morphology except the ridge location, and their genotype of SSU rDNA was also identical to GenBank data of the same species. From this study, we found that the five Korean unarmored dinoflagellates are identical morphological characteristics and genotype to each species of foreign isolates.

광합성 측정기를 이용한 미세조류의 광합성 효율 측정 (Evaluation of the ETRmax in Microalgae Using the PHYTO-PAM Fluorometer)

  • In this study, the PHYTO-PAM-fluorometric method was used to evaluate the ETR$_{max}$ in terms of sensitivity to DIN/DIP against 14 microalgae: Prorocentrum micans, Heterocapsa triquetra, Gymnodinium impudicum, Cymnodinium catenatum, Amphidinium caterae, Chlorella vulgaris, Chroococcus minutus, Microcystis aeruginosa, Chlorella ellipsoidea, Nannochloris oculata, Oocystis lacustris, Chroomonas salina, Gloeocystis gigas, and Prymnessium parvum. We found that P. micans, H. triquetra, and A. caterae exposed to the maximum level of DIN/DIP were significantly smaller in the ETR$_{max}$ than that of the minimum and moderate mixture. Unlikely the ETR$_{max}$, the initial slope alpha was not significantly different at the level of 60 DIN/DIP. In G. catenatum, the moderate levels of 15 and 20 in DIN/DIP were found to be significantly different from the ETR$_{max}$ at Chl-Ch4. Gymnodinium impudicum had a higher value than that of the ETR$_{max}$ than that of dinoflagellates used in this study, ranging from 306.1 (Ch4, DIN/DIP: 10) to 520.1 (Ch4, DIN/DIP: 30). The ETR$_{max}$ value obtained from other microalgae was similar to C. impudicum at any of the ratios of DIN/DIP and channels. Consequently, the influence of offshore water current assures us of the suppression of photosynthesis and electron transport rate in dinoflagellates. Gymnodinium impudicum has not been researched in the area of red tides in Korea, but it will be enough to creat the massive algal blooms in the future because of higher potential photochemical availability.

Identification of Cochlodinium polykrikoides against Gyrodinium impudicum and Gymnodinium catenatum in Field Samples using FITC Lectin Probes

  • We have investigated lectin binding patterns in order to apply binding records of previous laboratory experiments to field settings before the first ourbreaks of harmful algal bloom (HAB). Although cells were grown under different conditions, the binding patterns were the same as in the control. In addition, culture days was not associated with the binding patterns, when compared with the control. In nature, this results suggest that ECA, HPA and WGA lectin are able to discriminate between C. polykrikoides and G. impudicum, as well as ECA and SBA have a capability as a tool for differentiating between C. polyrikoides and G. catenatum, although these species are closely similar under the light microscope fiexed with Lugol solution.

2018년 이른 여름 남해 연안해역 식물플랑크톤 군집의 공간분포 특성 (Spatial distributions of phytoplankton community in the coastal waters of South Sea, Korea during the early summer of 2018)

  • 남해 연안해역에서 이른 여름 식물플랑크톤 군집의 분포특성을 파악하기 위해 2018년 6월말에서 7월 중순까지 11개 해역을 대상으로 실시하였다. 결과 출현한 식물플랑크톤 출현 종은 56속 105종으로 규조류가 52.4%, 와편모조류가 40.0%, 기타 편모조류가 7.6%였다. 현존량은 표층에서 5.5~593.2 cells mL-1로 변화하여, 출현종과 현존량 모두 동부해역에서 높고, 서부 해역에서 낮은 특정을 나타내었다. 식물플랑크톤 군집은 남해 금포, 여수 오천동 및 고흥 외나로도 해역을 제외하면 규조류에 지배되는 특성을 보였으며, 우점종은 여수 오천동과 완도 충도를 제외하면 중심규조 Skeletonema costatum-like species (ls)에 의해 우점되었다. 그러나 오천동 해역은 유독와편모조류 Gymnodinium catenatum에 의해 극우점되었고, 남해 금포 및 외나로도 해역은 와편모조류 Tripos fusus에 의해 12% 이상의 우점율을 나타내었다. 이른 여름 남해 연안해역의 식물플랑크톤 군집의 공간분포는 강수량 등 영양염류 공급에 크게 지배되고 있는 것으로 판단할 수 있었다.

Alkaline Phosphatase Activity and Phosphatase Hydrolyzable Phosphorus for Phytoplankton in Hiroshima Bay, Japan

  • We investigated the seasonal variability of tree alkaline phosphatase activity in seawater and alkaline phosphatase hydrolysable phosphorus (APHP) at 3 stations in Hiroshima Bay using alkaline phosphatase extracted from the dinoflagellates Alexandrium tamarense and Gymnodinium catenatum. The dissolved inorganic phosphorus (DIP) was lower than $1\;{\mu}M$ all samples; the lowest values were in May. The amount of APHP was high at the surface and bottom waters of all stations in May, showing DIP-depleted conditions. In August and November, the amount of APHP was much less than the amount of APHP in May, indicating that the availability of dissolved organic phosphorus (DOP) for these species was low and/or uptake during the dinoflagellate blooming might have occurred in the area. The results obtained from short-term variations of AP activity might suggest that the growth of dinoflagellates in this season may be partly supported by the AP produced by other diatoms.

Phylogenetic Analysis of Harmful Algal Bloom (HAB)-Causing Dinoflagellates Along the Korean Coasts, Based on SSU rRNA Gene

  • Twenty-three cultures of harmful algal bloom (HAB)-(causing dinoflagellates were isolated from the coastal waters of Korea. For each of the 14 morphospecies, the nuclearencoded small subunit (SSU) rDNA was analyzed to determine the phylogenetic relatedness of the species. Despite temporal and spatial isolation, 3-4 clonal cultures of Alexandrium catenella, Cochlodinium polykrikoides, and Gymnodinium catenatum had 100% identical SSU rDNA sequences. In contrast, heterogeneities in the SSU rDNA sequences were observed in Akashiwo sanguinea and Lingulodinium polyedrum strains. Extreme sequence polymorphism was shown within the SSU rRNA genes of an Al. tamarense clonal culture. A homology search in GenBank revealed that 11 dinoflagellate species were located in clusters corresponding to their morphological classification. The SSU rDNA sequences of C. polykrikoides, Gyrodinium instriatum, and Pheopolykrikos hartmannii, which were determined for the first time in this study, showed the following phylogenetic relationships: C. polykrikoides formed an independent branch separated from other dinoflagellates; Gyr. instriatum was placed in a monophyletic group with Gyr. dorsum and Gyr. uncatenum; and Ph. hartmanii, which forms a distinct two-celled pseudocolony, belonged to Gymnodinium sensu Hansen and Moestrup.

Modern Dinoflagellate Cysts Distribution off the Eastern Part of Geoje Island, Korea

  • Distributional characteristics of dinoflagellate cysts in surface sediments were investigated in relation to environmental factors in the eastern part of Geoje Island, Korea. Samples were collected from 10 stations in February of 2004 and water temperature and salinity were measured in February, May, September and November of 2004. Total 30 taxa of dinoflagellate cysts were identified representing 19 genera, 28 species and 2 unidentified species. Among these dinoflagellate cysts, Brigantedinium spp. of which relative proportion in th e total dinoflagellate cysts was 23.5%, was the most abundant at all stations except St. 1, and was followed by Spiniferites bulloideus (8.6%), Lingulodinium machaerophorum (8.2%) and Diplopsalis lenticula (6.7%). In addition, ellipsoidal cysts of the genus Alexandrium (Alexandrium catenella - tamarense type) and Gymnodinium catenatum, known to be causative organisms for PSP, occurred with high concentrations. Scrippsiella trochoidea was also found; however, its cyst concentration was low. Generally, species composition in the study area was similar to these reported from Jinhae Bay and Busan Harbor and several dinoflagellate cysts reflected the eutrophic condition. Cyst distribution in th e eastern part of Geoje Island seems to be influenced by the Tsushima Warm Current flowing from the southwest. The mean water temperature was $12.0^{\circ}C$ in February, $14.7^{\circ}C$ in May, $20.9^{\circ}C$ in September and $17.2^{\circ}C$ in November, which was most favorable for Alexandrium spp. growth. The abundances of dinoflagellate cysts rang e d from 528 to 2,834 cysts/g dry sediment. Higher concentrations were recognized in sediments of west area of the Jisimdo than at other stations. The cyst composition of this area was closely related to these of Jinhae Bay and Busan Harbor from which currents flow into this area. Higher cyst concentration in the west area of Jisimdo might be due to formation of the gyre.
