Chloroanilines are aromatic amines used as intermediate products in the synthesis of herbicides, azo-dyes, and pharmaceuticals. 3,4-dichloroaniline (DCA) is the degradation product of some herbicides (diuron, propanil, and linuron) and of trichlorocarbanilide, a chemical used as an active agent in the cosmetic industry. The compound, however, is considered a potential pollutant due to its toxicity and recalcitrant property to humans and other species. With the increasing necessity for bioremediation, we sought to isolate bacteria that degraded 3,4-DCA. A bacterium capable of growth on 3,4-DCA as the sole carbon source was isolated from seaside sediment using a dilution method with a culture enriched in 3,4-DCA. The isolated strain, YM-7 was identified to be Pseudomonas sp. The isolated strain was also able to degrade other chloroaniline compounds. The isolated strain showed a high level of catechol 2,3-dioxygenase activity on exposure to 3,4-DCA, suggesting that this enzyme is an important factor in 3,4-DCA degradation. The activity toward 4-methylcatechol was 53.1% that of catechol, while the activity toward 3-methylcatechol, 4-chlorocatechol and 4,5-chlorocatechol was 18.1, 33.1, and 6.9%, respectively.
The sensitivity of a mehtanogen and sulfate-reducing bacterium isolated from a sea-based landfill site Cd$^{2+}$ and CU$^{2+}$ was studied. Methanogens and sulfate-reducing bacteria in leachates of the waste disposal site were enumerated using the MPN method. Methanobacterium thermoautotrophicum KHT, isolated from the leachate, could not grow at 0.5 mM Cd$^{2+}$ or 1.0 mM CU$^{2+}$. Desulfotomaculum sp. RHT, isolated from the same leachate, was able to insolubilization 3.0 mM Cd$^{2+}$ or 2.0 mM CU$^{2+}$ by production of hydrogen sulfide. When strains KHT and RHT were cultured together in the presence of the heavy metals, strain KHT could grow at high heavy metal concentrations after insolubilization of the metals by strain RHT. strain RHT.

We investigated annealing effects of seed layers on the properties of ZnO nanorods grown on the seed layers. ZnO nanorods were grown by a hydrothermal method. ~100 nm-thick ZnO films were sputtered on oxidized Si wafers and quartz as seed layers. The ZnO films were annealed at $400^{\circ}C$, $600^{\circ}C$, and $800^{\circ}C$, respectively. ZnO nanorods were grown at $90^{\circ}C$ for 3 hours in the mixed solution of zinc nitrate hexahydrate and hexamethylenetetramine. X-ray diffraction was carried out to estimate the crystallinity and strain of ZnO films and nanorods. A field emission scanning electron microscope was employed to observe the morphology of the films and nanorods. PL(photoluminescence) measurements were conducted with 266 nm light. It was found that the annealing of seed layers increase the growth rate of nanorods, and change compressive strain of nanorods to tensile strain. The intensity of PL in the UV region reduced by using the annealed seed layers.

In this talk, I will present two research works in progress, which are: i) mapping of piezoelectric polarization and associated charge density distribution in the heteroepitaxial InGaN/GaN multi-quantum well (MQW) structure of a light emitting diode (LED) by using inline electron holography and ii) in-situ observation of the polarization switching process of an ferroelectric Pb(Zr1-x,Tix)O3 (PZT) thin film capacitor under an applied electric field in transmission electron microscope (TEM). In the first part, I will show that strain as well as total charge density distributions can be mapped quantitatively across all the functional layers constituting a LED, including n-type GaN, InGaN/GaN MQWs, and p-type GaN with sub-nm spatial resolution (~0.8 nm) by using inline electron holography. The experimentally obtained strain maps were verified by comparison with finite element method simulations and confirmed that not only InGaN QWs (2.5 nm in thickness) but also GaN QBs (10 nm in thickness) in the MQW structure are strained complementary to accommodate the lattice misfit strain. Because of this complementary strain of GaN QBs, the strain gradient and also (piezoelectric) polarization gradient across the MQW changes more steeply than expected, resulting in more polarization charge density at the MQW interfaces than the typically expected value from the spontaneous polarization mismatch alone. By quantitative and comparative analysis of the total charge density map with the polarization charge map, we can clarify what extent of the polarization charges are compensated by the electrons supplied from the n-doped GaN QBs. Comparison with the simulated energy band diagrams with various screening parameters show that only 60% of the net polarization charges are compensated by the electrons from the GaN QBs, which results in the internal field of ~2.0 MV cm-1 across each pair of GaN/InGaN of the MQW structure. In the second part of my talk, I will present in-situ observations of the polarization switching process of a planar Ni/PZT/SrRuO3 capacitor using TEM. We observed the preferential, but asymmetric, nucleation and forward growth of switched c-domains at the PZT/electrode interfaces arising from the built-in electric field beneath each interface. The subsequent sideways growth was inhibited by the depolarization field due to the imperfect charge compensation at the counter electrode and preexisting a-domain walls, leading to asymmetric switching. It was found that the preexisting a-domains split into fine a- and c-domains constituting a $90^{\circ}$ stripe domain pattern during the $180^{\circ}$ polarization switching process, revealing that these domains also actively participated in the out-of-plane polarization switching. The real-time observations uncovered the origin of the switching asymmetry and further clarified the importance of charged domain walls and the interfaces with electrodes in the ferroelectric switching processes.

To examine the growth-phase dependent control of Escherichia coli rnpB gene we used a combination of Northern analysis for RNA determination and Southern analysis for plasmid DNA determination. The relative amounts of metabolically unstable transcript derived from the internally deleted rnpB gene harbored on a multicopy plasmid as well as the relative plasmid contents were measured by Northern analysis and Southern analysis, respectively, of total nucleic acids from E coli cells containing the plasmid. The relative transcription activity of the rnB was represented by a ratio of the relative amount of the transcript to that of the plasmid DNA during bacterial growth. The rnpB transcription increased rapidly with time during exponential growth, but started to decrease before the transition period of an exponential growing cell culture into the stationary phase. Although the expression pattern was similar to the changes of ${\beta}-galactosidase$ activity expressed from the lysogenic strain carrying the chromosomal rnpB-lacZ fusion which were shown in a previous work, the present data appears to represent a more actual growth-phase control of the rnpB transcription than the previous data by the ${\beta}-galactosidase$ assay. In addition the present method described for a direct analysis of both RNA and plasmid DNA provides a rapid and efficient method that can applied to an examination of transcription control by using a multicopy plasmid.
To develop a new cultivar of king oyster mushroom(Pleurotus eryngii), G09-21 as parental strain was selected by the method of Di-mon crossing between monokaryotic strain ASI 2824-21 derived from ASI 2824(Keunneutari No. 2) and dikaryotic strain ASI 2887(Aeryni 3). The Pe21-53(G09-21-10 x ASI 2844-9) was shown the best cultural characteristics, selected to be a new cultivar and designated as 'Seolsong'. The 'Seolsong' was distinctly formed incompatibility line in the confrontation growth of parental strains Keunneutari No. 2, Aeryni 3 and ASI 2844. Analysis of the genetic characteristics of the new cultivar 'Seolsong' showed a different DNA profile as that of the control strains, Keunneutari No. 2, Aeryni 3 and ASI 2844, when RAPD(Random Amplified Polymorphic DNA) primer URP4 was used. The optimum temperature and pH arrange for mycelial growth were $25{\sim}30^{\circ}C$ and pH 5~8, respectively. This new cultivar 'Seolsong' of fruiting body production per bottle was about $131.4{{\pm}}43.1$ g which is about 102% quantity compared to that of other cultivar Keunneutari No. 2. And also the stipe is thick and long, but the number of available stipe is few. Particularly, it was tolerant of high moisture above 90% during the growth period after primodia formation. We therefore expect that this new strain will save of labor and cost of cultivation by without culling work.

This study investigated crystal structures, microstructures, and electric-field-induced strain (EFIS) properties of Bi-based lead-free ferroelectric/relaxor composites. Bi1/2Na0.82K0.18)1/2TiO3 (BNKT) as a ferroelectric material and 0.78Bi1/2(Na0.78K0.22)1/2TiO3-0.02LaFeO3 (BNKT2LF) as a relaxor material were synthesized using a conventional solid-state reaction method, and the resulting BNKT2LF powders were subjected to high-energy ball milling (HEBM) after calcination. As a result, HEBM proved a larger average grain size of sintered samples compared to conventional ball milling (CBM). In addition, the increased sintering time led to grain growth. Furthermore, HEBM treatment and sintering time demonstrated a significant effect on EFIS of BNKT/BNKT2LF composites. At 6 kV/mm, 0.35% of the maximum strain (Smax) was observed in the HEBM sample sintered for 12 h. The unipolar strain curves of CBM samples were almost linear, indicating almost no phase transitions, while HEBM samples displayed phase transitions at 5~6 kV/mm for all sintering time levels, showing the highest Smax/Emax value of 700 pm/V. These results indicated that HEBM treatment with a long sintering time might significantly enhance the electromechanical strain properties of BNT-based ceramics.
Strain SA72 was isolated from Jeot-gal and identified as producer of a bacteriocin, which showed some bactericidal activity against Lactobacillus delbrueckii ATCC 4797. Strain SA72 was tentatively identified as Lactococcus lactis according to the AOI test. Lactococcus lactis SA72 showed a broad spectrum of microorganisms, tested by the modified deferred method. The activity of lacticion SA72, named tentatively as a bacteriocin produced by Lactococcus lactis SA72, was detected during the mid-lon growth phase, reached a maximum during the early stationary phase, and then declined after the late stationary phase. Lacticin SA72 also showed a relatively broad spectrum of activity against non-pathogenic and pathogenic microorganisms when assessed by the spot-on-lawn method. Its anitimicrobial activity on sensitive indicator cells disappeared completely by protease XIV treatment. The inhibitory activity of lacticin SA72 remained after treatment for 15 min at $121^{\circ}C$, 문 was stable in a pH range of 2.0 to 9.0 and all organic solvents examined. It demonstrated a typical bactericidal mode of inhibition against Lactobacillus delbrueckii ATCC 4797. The apparent molecular mass of lacticin SA72 was in the region of 3-3.5 kDa, determined by SDS-PAGE.
The work has been carried out for the development of antifungal antibiotics possessing curative effect in the control of sheath blight disease of rice plant. Soil samples were collected from over 1600 spots throughout the country. More than 1300 specimens which seem to be the genus Streptomyces were isolated from the soil samples. Screening procedures consist of respective processes by four steps. Those are growth inhibition test in liquid culture, paper disk method, dendroid test and green house test. 102 isolates appeared to be active against Pellicularia sasakii when all specimens isolated were examined by the first growth inhibition test. Finally a strain of Streptomyces forming strong antifungal substances against P. sasakii was selected from a soil sample of Mt. Soyo, Kyeongi Province. Antifungal substances formed by the strain were isolated and purified from the culture broth and examined for antimicrobial activities as to be specific against fungi but not active on bacterial growth.

Microalgae are receiving an increasing attention because of their potential use as $CO_2$ capture method and/or as feedstock for biofuels production. On the other hand the current microalgae-based technology is still not widespread since it is characterized by technical and economic constraints that hinder its full scale-up. In such contest the growth kinetics of Nannochloris eucaryotum (a relatively unknown marine strain) in batch and semi-batch photobioreactors is quantitatively investigated with the aim of obtaining the corresponding kinetic parameters suitable for process engineering and its optimization. In particular the maximum growth rate was evaluated to be 1.99 $10^{-3}\;h^{-1}$. Half saturation concentrations for nitrates ($K_N$) and phosphates uptake ($K_P$) were evaluated as 5.4 $10^{-4}\;g_N\;L^{-1}$ and 2.5 $10^{-5}\;g_P\;L^{-1}$, respectively. Yield factors for nitrogen ($Y_N$) and phosphorus ($Y_P$) resulted to be 5.9 $10^{-2}\;g_N\;g^{-1}$ biomass and 6.0 $10^{-3}\;g_P\;g^{-1}{_{biomass}}$, respectively. The possibility of using 100% (v/v) $CO_2$ gas as carbon source is also evaluated for the first time in the literature as far as N. eucaryotum is concerned. The strain showed a good adaptability to high concentrations of dissolved $CO_2$ as well as to low pH. The lipid content under 100% $CO_2$ is about 16.16 %wt $wt^{-1}$ and the fatty acid methyl esters composition of the extracted oil is in compliance with the European regulation for quality biodiesel.
