Bactericidal Activity of Grapefruit (Citrus paradisl) Seed Extract-Based Disinfectant

  • Bactericidal activity of grapefruit seed extract (GSE)-based disinfectant, as a safe disinfectant, was measured against five bacteria by Korean Food & Drug Administration (KFDA) dilution-neutralization method. GSE-based disinfectant showed a 99.9999% bactericidal activity against Escherichia coli ATCC 10536, Salmonella typhi ATCC 29629, Staphylococcus aureus ATCC 6538, Bacillus cereus ATCC 11778, and Listeria monocytogenes ATCC 1911 at the concentration of 2.15% GSE. It showed better bactericidal activity against Gram-negative bacteria of E. coli ATCC 10536 and S. typhi ATCC 29629 at lower concentration of GSE (0.43%). Based on the results, it was suggested that a possible bactericidal mechanism of GSE active ingredients was due to the abrupt osmotic shift during the bactericidal activity test by KFDA method.

Grapefruit Seed Extract (DF-100) Treatment of Poultry to Reduce Attached Salmonella

  • Chicken skins or carcasses inoculated with Salmonella typhimurium were exposed to 0.1 or 0.5% grapefruit seed extracts (DF-100) for 1 or 3 min to evaluate antibacterial activity of DF-100 and its possible application in proultry processing. The numbers of live salmonellae on chicken skins were reduced by 0.8-1.2 logs/cm2 with 0.5% DF-100. Dipping chicken carcasses into 0.5% DF-100 for 3 min reduced salmonelae by 4.3 logs/carcass. Scanning electron microscopy showed that DF-100 killed the cells attached but did not detach cells from the skin. No odor or changes in the color of chicken skin were detected after DF-100 treatment.

  • To investigate the antimicrobial activities and effect of grapefruit seed extract(GFSE) on the physiological function of microorganism, antimicrobial activity, fatty acids of bacterial cell lipid and amino acids of bacterial cell protein were measured. The change of cell morphotype was observed by transmission electron microscope. GFSE was very stable on the wide range temperture and pH. The growth rate of E. coli and B. suvtilis were decreased above 40ppm GFSE There fore, minimum inhibitory concentration (MIC) of the E. coli and B. subtilis to GFSE were determined around 40ppm. In the change of fatty acids quantities, hexadecanoate was significantly decreased on the treatment compared with control in case of E. coli, whereas tridecanoate was not detected in case of B. subtilis. In the change of amino acids quantities, alanine, glutamic acid, glycine, lysine were decreased on the treatment compared with control in case of E. coli and B. subtilis Transmission electron microsgraphs(TEM) showed the microbial cells were destroyed by GFSE.

콘택트렌즈 보존제 H2O2와 자몽씨 추출물의 세포 독성 비교 연구 (The comparative study on Cell Cytotoxicity of H2O2 and Grapefruit Seed extract)

  • 콘택트렌즈 관리용액으로 사용되는 합성보존제로서 대표적인 $H_2O_2$와 천연보존제인 자몽씨 추출물(DF-100)이 결막세포에 미치는 저해효과를 비교 조사하기 위하여 본 연구를 시행하였다. 자몽씨 추출물의 주성분은 Narigin이며, 이는 항산화를 일으키는 Flavonoid의 구성성분중 하나이다. $H_2O_2$와 자몽씨 추출물(DF-100)의 세포는 MTT assay에 의해 측정하였고, DNA 손상은 Comet assay 측정으로 분석하였다. 5%의 자몽씨 추출물은 과산화수소에 의해 손상된 세포를 회복시켜주는 효과를 나타내었으며, 또한 배양 결막세포주에 과산화수소 및 자몽씨 추출물을 농도별로 동시 처리하여 24시간 뒤 LDH leakage assay용 이용한 세포독성을 측정하여 분석하였다. 자몽씨 추출물은 과산화수소의 세포증식 저해와 apoptosis를 현저하게 억제해 주었다. 또한 자몽씨 추출물은 bioflavonoids의 구성성분이며 이는 음식으로써도 제공된다. 이 연구는 자몽씨 추출물이 anti-oxidant와 anti-apoptopic activity로 천연보존제로 유용하게 사용될 수 있는 것을 보여준다.

Soft contact lens에 있어서 천연보존제와 합성보존제의 살균효과 : 주사 전자현미경적 관찰 (Effects of natural and chemical disinfectants on soft contact lens infected with Pseudomonas aeruginosa : Scanning Electron Microscopic study)

  • 본 연구는 천연보존제인 자몽씨 추출물과 합성보존제인 $H_2O_2$를 사용하여 soft contact lens 착용시 각막염의 원인중 하나인 Gram negative bacteria인 Pseudomonas aeruginosa 대한 살균효과를 알아보았다. Pseudomonas aeruginosa는 4주간 배양하여 포낭 형태를 이루게 한 다음 $7{\times}10^6CFU/m{\ell}$의 현탁액에 soft contact lens를 오염시킨 후 천연보존제와 합성보존제의 살균 효과를 비교하였다. 현탁액에 담군 120분후 균의 생존률은 천연보존제는 $1{\times}10^3CFU/m{\ell}$ 합성보존제는 $0.5{\times}10^2CFU/m{\ell}$으로 나타나 유의한 차이는 없었다. 하지만 합성보존제의 경우 눈에 자극감이나 화학적 안 외상을 줄 수 있는 점을 고려하면 자몽씨 추출액과 같은 인체에 무독한 천연 보존제의 개발이 필요한 것으로 나타났다.

Production of an Acidic Polygalacturonase from Aspergillus kawachii by Solid State Fermentation and Their Application for Pectin Extraction

  • An acidic polygalacturonase (PG) from Aspergillus kawachii was produced by solid state fermentation employing a polyurethane foam support. The conditions used for the production of acidic PG were particle size of support (0.6 or 500 $mm^3$) and fermentation time. From the factors studied, the particle size had important influence on enzyme production. The best conditions for acidic PG production were $0.6\;mm^3$ particle size, 18 hr at $30^{\circ}C$ and initial pH of 5.0. In addition, pectin was extracted from citrus pomaces (grapefruit, lime, and tangerine) by acidic PG at $50^{\circ}C$ for 24 hr with citric acid solution. Infrared spectroscopy showed that lime pomace had more high-methoxylated (65%) endogenous pectin than was obtained than from grapefruit or tangerine pomaces. The enzymatically extracted pectin yield in dry basis (d.b.) for grapefruit and lime pectins were 6.95 and 4.25%, respectively. The citric acid solution alone also contributed to pectin extraction from citrus pomaces (7-9%, d.b.). Limited pectin extraction by acidic PG from tangerine pomace was most likely due to the presence of low-methoxylated endogenous pectin. The enzymatic method for pectin extraction using acidic PG from A. kawachii is a promising technique for releasing highly polymerized pectic substances from high-methoxylated lime or grapefruit pomaces.

자몽껍질 유래 플라바논의 최적 추출 및 기능성 소재 캡슐화 (Optimum Conditions for Extracting Flavanones from Grapefruit Peels and Encapsulation of Extracts)

  • 버려지는 자몽 껍질에 다량 함유되어있는 플라바논을 추출하여 기능성 성분을 재이용하였다. 친환경용매인 아임계수 추출기술을 이용하여 $170^{\circ}C$, 10 min의 최적 조건에서 추출함으로써 무독성 용매로 빠르고 경제적으로 추출할 수 있었다. 자몽껍질 추출물을 ${\beta}$-cyclodextrin을 이용하여 처리함으로써 플라바논과 같은 비극성 물질을 캡슐화하여 소재의 가용화를 용이하게 하였다. 이것은 항산화 기능이 향상된 대체 소재의 개발 및 건강지향식품에 이용할 수 있으며, 이를 통하여 세계 기능성 식품 시장으로의 진출을 위한 발판으로도 삼을 수 있을 것으로 기대된다.

미세캡슐화한 녹차 및 자몽종자 추출물이 Murine RAW 264.7 대식세포주의 항염증에 미치는 영향 비교 (A comparison of anti-inflammatory activities of green tea and grapefruit seed extract with those of microencapsulated extracts)

  • We compared the effects of grapefruit seed extract (GFSE), green tea extract (GT) and their microencapsulated extract on anti-inflammatory activities in murine RAW 264.7 macrophages cell line. In order to protect the bioactive compounds in the extracts, they were microencapsulated with maltodextrin and $H_2O$. Nitric oxide (NO), prostaglandin E2 (PGE2), tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), inducible nitric oxide synthase (iNOS) protein expression and thiobarbiturate reactive substances (TBARS) were analyzed in LPS activated RAW 264.7 macrophages. The green tea extract at the range of $100-600{\mu}g/mL$ inhibited NO, PGE2 production and iNOS protein expression without cytotoxicity in a dose-dependent manner. Grapefruit seed extract had strong inhibitory effects on NO and PGE production and iNOS protein expression at the range of $5-20{\mu}g/mL$ without cytotoxicity. Microencapsulation of green tea extract had further inhibitory effects on NO and PGE2 production and on iNOS protein expression, whereas microencapsulated GFSE did not show any further inhibitory effects on these parameters. Taken together, our results suggest that GSFE might be a promising candidate for preventing inflammation related diseases, such as cardiovascular disease, cancer or diabetes, and the microencapsulation of green tea extract could improve its bioactivity.

천연 추출물을 이용한 무 피클의 제조 및 품질 평가 (Quality Characteristics of Radish Pickle with Natural Preservatives)

  • This study examined the physicochemical properties of radish pickle containing different natural preservatives (grapefruit seed extract, green tea extract, rosemary, or olive) stored for 0, 1, 2, 3, and 4 weeks. The hardness and color of the radish pickles with the grapefruit seed extract was higher than the other radish pickles during storage from week 0 to week 4. A 14.52% and 13.80% decrease in hardness and color were observed in the radish pickles with grapefruit seed extract (GFE), respectively. In addition, the total phenolic content was highest in the GFE in natural preservatives. Based on the results, GFE was selected as the optimal natural preservatives, and the growth of total viable bacteria and yeast were evaluated. The total viable bacteria and yeast showed similar patterns to the control. These results are expected to be useful in producing radish pickles with optimal quality and contribute to the development of various foods in the food industry.

Grapefruit Juice Suppresses Azoxymethane-induced Colon Aberrant Crypt Formation and Induces Antioxidant Capacity in Mice

  • In the present report we determined the protective capacity of grapefruit juice (GJ) against molecular and cellular damage in azoxymethane (AOM) treated mice. Animals were daily administered GJ orally (0.8, 4.1, and 8.2 ${\mu}l/g$) for seven weeks, as well as intraperitoneally (ip) injected with AOM twice (weeks 2 and 3 of the assay). Control groups administered with water, with the high dose of GJ, and with AOM injected in weeks 2 and 3 were also included. The results showed a significant, dose-dependent protection of GJ on the number of colon aberrant crypts (AC) induced by AOM. The highest inhibitory effect was reached with the highest tested dose of GJ, decreasing ACF by 51% and 43% at weeks 4 and 7 of the assay. Regarding protein and lipid oxidation we also found a dose-dependent decrease caused with GJ in comparison with the increased levels produced by AOM. Therefore, our results established chemopreventive potential for GJ, and suggested effects related to its antioxidant capacity. Finally, we found that the tested agents induced neither micronuclei increase nor alteration in bone marrow cytotoxicity.
