Catechol and 4-chlorobenzoate (4CBA) which are produced from the biodegradation of a variety of aromatic and chloroaromatics have been recognized as toxic to living organisms. In this study, the toxic effects of catechol and 4-chlorobenzoate on gram-positive and -negative bacteria were examined in terms of survival, morphology, change in fatty acids and membrane protein composition. The survival rate of the organisms during treatment for 6 h was decreased, as the concentration of each aromatic was increased. Escherichia coli and Pseudomonas cells treated with catechol and 4CBA at concentrations causing a significant decrease in their viability, showed destructive openings in their cell envelopes. Bacills subtilis treated with the aromatics were reduced in cell size and Staphylococcus aureus cells displayed irregular rod shapes with wrinkled surfaces. The bacterial cells treated with 20 mM catechol showed increases in unsaturated fatty acids, but several saturated fatty acids were decreased. In the E. coli cells treated with 20 mM catechol, inner membrane proteins of 150 kDa and 105 kDa were decreased. But several kinds of the inner and outer membrane proteins were increased. In B. subtilis treated with 20 mM catechol, several kinds of proteins were increased or decreased in membrane proteins.
A new rod-shaped endospore-forming bacterium isolated from the hindgut flora of the termite, Reticulitermes speratus kyushuenesis Morimoto is described. The isolate stained Gram positive, but the KOH test and the test for L-alanine aminopeptidase were negative. The length of a single cell varies from 2.5-9.0 $\mu $m, and the cell is about 0.5-0.7$\mu $m thick. The isolate had a high cellulolytic ability and was identified as Bacillus amyloliquefaciens.
Yogurt is produced by bacterial fermentation of milk and contains lactic acid bacteria (LAB), which produce various metabolites such as organic acid, hydrogen peroxide, and bacteriocin. This study aimed to investigate cell-free supernatants (CFS) of LAB isolated from Tibetan yogurt. CFS (TY1, TY2, TY3, TY4, TY5, TY6, and TY7) from selected strains of LAB were co-incubated with four different foodborne pathogenic bacteria, namely E. coli O157:H7, Listeria monocytogenes, Salmonella typhimurium, and Staphylococcus aureus. Inhibition of foodborne pathogenic bacterial growth was not affected in the presence of CFS (pH 6.5). In contrast, CFS without neutralization completely inhibited the growth of the bacteria. Furthermore, when the concentration of CFS (without neutralization) was changed to 1:4 and 1:8, a difference in inhibition was observed between Gram-positive and Gram-negative bacteria. CFS more effectively inhibited the growth of Gram-negative E. coli O157:H7 and S. Typhimurium than Gram-positive L. monocytogenes and S. aureus. These results suggest that organic acids in LAB may inhibit the growth of foodborne pathogenic bacteria, particularly Gram-negative bacteria.
The suspension culture of an anchorage-dependent cell line (Bok-l) from Bombina orientalis was successful in respects of cost and efficiency. The amount of cells obtained from the suspension culture was almost equivalent to that from the anchorage-dependent culture. This result shows the possibility of suspension culture for scale-up. The cells in suspension produced an antibacterial peptide as much as anchorage-dependent cells did. The cell growth ($6.0\times10^6cells/m\ell$) and viability (>80%) at 10 rpm were higher than that at 0 rpm ($1.9\times10^6cells/m\ell$, 65~80%) and 30 rpm ($1.8\times10^6cells/m\ell$ 40~76%). The size of cells became smaller at the agitation rate of 30 rpm. The antibacterial activities of cell extracts from suspension cultured cells were confirmed against gram-negative and gram-positive bacteria by the inhibition zone assay and the liquid growth inhibition assay.
Papyriflavonol A (PapA), a prenylated flavonoid [5,7,3',4'-tetrahydroxy-6,5'-di-(${\gamma},{\gamma}$-dimethylallyl)-flavonol], was isolated from the root barks of Broussonetia papyrifera. Our previous study showed that PapA has a broad-spectrum antimicrobial activity against pathogenic bacteria and fungi. In this study, the mode of action of PapA against Candida albicans was investigated to evaluate PapA as an antifungal agent. The minimal inhibitory concentration (MIC) values were 10~25 ${\mu}g/ml$ for C. albicans and Saccharomyces cerevisiae, Gram-negative bacteria (Escherichia coli and Salmonella typhimurium), and Gram-positive bacteria (Staphylococcus epidermidis and Staphylococcus aureus). The kinetics of cell growth inhibition, scanning electron microscopy, and measurement of plasma membrane florescence anisotrophy revealed that the antifungal activity of PapA against C. albicans and S. cerevisiae is mediated by its ability to disrupt the cell membrane integrity. Compared with amphotericin B, a cell-membrane-disrupting polyene antibiotic, the hemolytic toxicity of PapA was negligible. At 10~25 ${\mu}g/ml$ of MIC levels for the tested strains, the hemolysis ratio of human erythrocytes was less than 5%. Our results suggest that PapA could be a therapeutic fungicidal agent having potential as a broad spectrum antimicrobial agent.
This study was carried out to evaluate cytotoxic effects of Poncirus trifoliata Raf. extract on lymphocytic leukemia tumor (L1210) cell lines. Disruptions in cell organelles were determined by 3-(4,5-dimethyl thiazol-2-yl)-2,5-diphenyl-2H-tetrazoliumbromide (MTT) assay The comparison of Ic50 Values of Poncirus trifoliata Raf. extract in L1210 cell lines showed that their susceptibility to these fractons decreased in the following order: adriamycin > Fr.4> Fr. 6> Fr. 5> Fr. 3> Fr. 1> Fr. 2 by the MTT assay. In order to develop an antumicrobial agent, Poncirus trifoliata Raf. was extracted wit ethanol, and then it was fractionated with several mobile phase. The antitumor activities of fractions of the ethanol soluble extract was investigated. The minimal inhibitory concentrations (MIC) of fractions of the ethanol soluble extract of Poncirus trifoliata Raf. against microorganisms were also examined. Antimicrobial activities of ampicillin and ketoconazole as references were compared to those of fractions of the ethanol soluble extract of Poncirus trifoliata Raf. The antimicrobial activities of all fractions from the extract had growth inhibition activities against gram-positive bacteria, gram-negative bacteria and fungi $(MIC\;>\;200{\mu}g/ml)$. These results suggest that fraction 4 of the ethanol soluble extract of Poncirus trifoliata Raf. possessed the most antitumorous agent.
Lactobacillus sp. GM7311이 생산하는 박테리오신의 Gram 양성균에 대한 작용 형태를 알아보기 위하여, 세가지 시험균주의 TSB 배양액에 일정한 시간 간격별로 $100\;Bu/m{\ell}$ 되도록 박테리오신을 첨가한 후 그 균수 변화와 작용 특성을 살펴보았다. 우선, 시험 균주의 증식 시기별로 박테리오신을 첨가한 경우 Listeria monocytogenes는 대수증식기 후기 이후에 억제작용을 나타낸 반면, Bacillus subtilis의 경우는 대수증식기 중기 이전까지의 시험균에 대해서 강한 억제작용을 보였으며, Staphylococcus aureus는 정지기 이전의 시험균에 대해 큰 항균 효과를 나타냈다 또한 박테리오신을 처리한 세 가지 시험 균주를 전자 현미경으로 관찰했을 때, L. monocpogenes와 B. subtilis는 세포벽 파괴에 의한 세포내 물질의 유출이 관찰되었으나, S. aureus는 다른 두 균주와 달리 박테리오신에 의한 세포벽의 파괴 현상은 없었으며 다만 세포벽이 대조구에 비해 엷어진 것이 관찰되었다. 아미노산 및 지방산 조성의 변화를 조사하여 박테리오신을 처리하지 않은 대조구와 비교하였을 때, 아미노산의 경우는 세 균주 모두에서 대부분의 성분이 감소하는 경향을 보였고 지방산은 큰 조성상의 변화나 공통된 특정지방산 성분의 감소 등은 나타나지 않았으며 각 성분들의 함량이 증감하는 경향을 보였다.
The antioxidant, antimicrobial, and cytotoxic activities of ovotransferrin were investigated in vitro. The antioxidant capacity of ovotransferrin was evaluated using the 2,2-Diphenyl-1-picryl hydrazyl (DPPH) radical scavenging method, antimicrobial effects using the agar well diffusion method, and cytotoxicity using the 3-(4,5-dimethylthizol-2-yl)-2,5-diphenylatetetrazolium bromide (MTT) assay. The DPPH radical-scavenging capacity of ovotransferrin at 1 mg/mL level reached approximately 60% after 48 h of reaction. The antimicrobial effects of ovotransferrin against common food-borne pathogens, Staphylococcus aureus KCCM 32395, Bacillus cereus KCCM 40935, Listeria monocytogenes ATCC 15313, Escherichia coli O157:H7 ATCC 43895, and Helicobacter pylori HpKCTC 26695 were dose dependant. Gram-positive bacteria was more sensitive to ovotransferrin than gram-negative bacteria. Ovotransferrin showed stronger antimicrobial effect against L. monocytogenes than other gram-positive bacteria tested. The cytotoxicity of ovotransferrin was evaluated in human cancer cell lines, various tissue origins, including the larynx (Hep-2), stomach (AGS), lung (SK-MES-1), liver (HepG2), breast (MCF-7), cervix (HeLa), and colon (HT-29). Ovotransferrin displayed relatively high cytotoxicity (${\leq}60%$ inhibition effects) at 40 mg/mL. At lower concentrations (${\leq}10mg/mL$), however, ovotransferrin cytotoxic effects were not significant in all cancer cell lines tested. These results indicated that ovotransferrin has potential to be used as an antioxidant or antimicrobial agent in foods or a pharmaceutical agent against cancers.
본 연구에서는 커피(Coffea arabica)의 에탄올 추출물을 이용하여 기능성 화장품소재로서의 가능성을 검토하였다. 미백활성으로는 tyrosinase 저해 활성과 멜라닌 함량을 측정하였으며, 항산화 활성으로는 SOD-like activity를 측정하였고, 그 결과 $500{\mu}g/mL$의 농도에서 $94.8{\pm}6.2%$의 활성을 나타내었다. CAE는 3개의 그람양성균과 6개의 그람음성균을 대상으로 항균활성을 조사하였으며, 그 결과 그람음성균에 대해서는 폭넓은 활성을 나타내었지만, 그람양성균에 대한 활성은 나타내지 않았다. 하지만 항생제내성균주인 MRSA CCARM3561에는 강한 활성을 나타내었다. 반면에 Candida albicanse에 대한 항곰팡이 활성은 나타내지 않았다. Tyrosinase 저해활성과 DOPA 산화능은 대조군인 arbutin 보다는 낮게 나타내었고, B16-F10 melanoma cell에 대한 멜라닌 생합성 저해능은 농도 의존적으로 감소하는 경향을 보였으며, $100{\mu}g/mL$의 농도에서 89.2%를 나타내었다. 이러한 결과들은 커피 추출물이 천연물, 무독성 항균제 또한 천연 화장품 소재로써의 가치가 있음을 나타내는 것이다.
Xylanase를 생산하는 호알칼리성 균주를 토양으로부터 분리하여 동정하였다. 토양으로부터 분리한 호알칼리성 균주 3000여종 중 xylanase 활성이 가장 높은 균주인 strain DK-2386 균주를 선별하였다. Gram 염색과 SEM을 통한 형태학적 특성을 관찰하였으며, Methods for General and Molecular Bacteriology와 Bergey's manual of systematic bacteriology, 그리고 Biochemical tests for identification of medical bacteria에 준하여 생화학적 특성을 확인한 결과 배양액의 색은 붉은 빛을 나타내었으며, Gram양성 간균, 내생포자형성, catalase 양성, oxidase 양성, 혐기성 양성, glucose로부터 gas 형성 양성, casein 가수분해 양성, starch 가수분해 양성, CMC 가수분해 양성, xylan 가수분해 양성, nitrate 환원능 양성이며 10% NaCl 농도에서도 생육하는 것을 확인하였다. 균체의 지방산 조성 분석 결과 $C_{15:0\;ISO}$ 27.35%와 $C_{15:0\;ANTEISO}$ 31.54%, $C_{16:0}$ 9.70%로 이루어져 Bacillus속으로 동정되었으며, 16S rDNA 염기서열 분석결과 B. agaradhaerens와 100%의 유사성을 갖는 것을 확인하였다. (사)한국종균협회로부터 분양받은 Bacillus agaradhaerens 40952, B. alcalophilus와 strain DK-2386 균주사이의 생화학적 특성을 비교한 결과 B. agaradhaerens와 속과 종이 같으나 미세하게 특성이 다른 균주로 판단되어 B. agarad haerens DK-2386이라 명명하였다.
