운동강도와 지속시간에 따른 트레드밀 운동이 비만 쥐의 심장근 내 항산화에 미치는 영향 (The Effect of Antioxidative Change in Cardiac Muscle of Obesity Rat by Treadmill Exercise with Intensity and Time)

  • 트레드밀 운동이 비만 쥐의 심장근 내 항산화에 어떠한 영향을 주는지 알아보고자 흰쥐에 6주 동안 고지방 식이로 비만을 유발시킨 뒤 운동강도와 적용시간을 달리하여 트레드밀을 3주 동안 적용하였으며, 체중변화, 혈중지질성분, 조직학적 변화, 심장근 내 항산화 효소와 MCP-1(monocyte chemoattractant protein-1) 등의 변화를 연구하였다. 4주령 Sprague Dawley계 수컷 흰쥐 32마리를 사용하여 각각 일반사료를 섭취한 정상군 (normal), 비만 대조군 (control), 비만 쥐에게 고강도 분할운동을 적용한 실험군 I(Experimental I), 비만 쥐에게 중강도 지구력 운동을 적용한 실험군 II (Experimental II)로 나누었다. 실험 결과 첫째, 체중변화는 고강도 분할 운동을 적용한 실험군 I과 중강도 지구력 운동을 적용한 실험군 II는 대조군에 비해 유의한 감소가 있었다 (p<0.001). 특히, 운동을 적용한 1주에서 3주까지 유의한 차이가 있었다. 둘째, 혈중지질성분 수치는 고강도 분할 운동을 적용한 실험군 I과 중강도 지구력 운동을 적용한 실험군 II는 대조군에 비해 총콜레스테롤, 중성지방, 저밀도 지단백질의 유의한 감소, 고밀도 지단백질에서는 유의한 증가가 있었다 (p<0.001). 셋째, 심장근의 조직학적 소견은 심장근에서 아교질 형성과 평활근 섬유의 정렬 상태와 밀집도, 심장근 세포크기가 중강도 지구력 운동 적용한 실험군 II에서 정상군과 비슷한 형태인 근육들이 잘 밀집해 있는 것을 볼 수 있었다. 또한, 섬유아세포, 염증세포들의 침윤 등이 고지방 식이 유발 쥐인 대조군과 고강도 분할 운동을 적용한 실험군 I에서 가장 심하였으며, 중강도 지구력 운동을 적용한 실험군 II는 대조군에 비해 섬유아세포, 염증세포들의 침윤 등이 현저히 감소하였다. 넷째, 심장근의 항산화 효소 SOD(superoxide dismutase), CAT(catalase), GPx(glutathione peroxidase) 수치는 중강도 지구력 운동을 적용한 실험군 II와 고강도 분할 운동을 적용한 실험군 I에서 모두 유의한 증가가 있었다 (p<0.001). 다섯째, 심장근의 항산화 단백질 MCP-1 발현 수치는 고강도 분할 운동을 적용한 실험군 I과 중강도 지구력 운동을 적용한 실험군 II 모두에서 감소가 있었으며 (p<0.01), 특히, 중강도 지구력 운동을 적용한 실험군 II에서는 유의한 감소가 있었다 (p<0.001). 따라서, 중강도 지구력 운동은 고지방 식이로 유발된 비만 쥐의 심장근 내 항산화에 영향을 주어 비만 개선과 산화적 스트레스에 의한 심장근육 손상 감소 등에 효과적인 것으로 나타났다.

미역첨가 식이가 당뇨 유발쥐의 혈청지질과 혈당 및 항산화 효소에 미치는 영향 (Effects of Dietary Seaweed on Blood Glucose, Lipid and Glutathione Enzymes in Streptozotocin-Induced Diabetic Rats)

  • 미역 첨가식이가 당뇨쥐의 당질과 지질대사 및 항산화효소계에 미치는 영향을 규명하기 위하여 SD계 흰쥐에 식이 무게량의 20% 미역 분말을 급여하여 7주간 실험 사육한 후 혈청의 포도당 및 지질농도와 주요 장기의 항산화효소의 활성도를 관찰하였다. 미역 당뇨군에서 미역의 섭취는 당뇨에 의한 체중감소 현상을 보였다. 당뇨군의 혈당 농도는 미역 섭취에 의한 혈당강하 효과는 2주 째에 나타났으나 7주 째는 보이지 않았다. 혈청지질중 총 콜레스테롤과 LDL-콜레스테롤은 변화가 없었으나 중성지방은 미역 당뇨군에서 감소하는 경향이고 HDL-콜레스테롤은 증가하는 경향으로 나타났다. 일반적으로 당뇨시 증가하는 신장의 G6Pase 활성도는 MD군에서 미역의 섭취에 의해 유의적(p<0.05)으로 감소했다. 대조군에서 증가된 간장의 GST 활성도는 미역 섭취에 의해 유의적(p<0.05)으로 감소했다. 간장의 GPX 활성도의 경우, 미역 섭취에 의해 증가하는 경향이었다. 간장의 GR 활성도는 미역 당뇨쥐에서 미역 섭취에 의해 감소하는 경향이나 신장에서는 상반되는 경향으로 나타났다. 간장의 TB-ARS는 미역 당뇨군에서 증가하는 경향이었다. 따라서 7주간의 미역첨가 섭취로 당뇨쥐의 고혈당이 강하되는 것은 2주에 나타났고 7주에는 관찰하지 못하였으나, 당뇨에 의한 산화적 스트레스로 인해 증가한 간장의 항산화 효소 활성도(GST)가 미역 섭취로 인해 저하됨으로써 미역의 섭취가 당뇨시 항산화적 반응을 제공할 수 있음을 유추할 수 있었다. 또한 중성지방이 감소하고 HDL-콜레스테롤의 증가하는 경향으로 심혈관계와 관련되는 당뇨합병증의 억제 가능성을 제시하였다.n)에서 Fe과 Mn 간의 길항작용으로 현저히 낮아졌다. 본 시험재배 조건별 Fe-함량의 변화는 white clover가 orchardgrass보다 상대적으로 더 큰 차이를 보였다. Mn-함량은 초종 간 큰 차이를 보였고 두 목초 공히 Mn을 함유한 조합시비로 크게 증가하였지만 이들 조합시비 간에는 차이가 경미하였다. 3. Mo-함량은 모든 경우 $T_6$ 6및 $T_7$ 에서 다소 높은 수준 이였다. $T_7$$T_6$ 에 비해서 Mo-함량이 다소 낮아졌다. Mo-과다피해는 B/Mo 비율의 조화 또는 B-시비를 통해서 경감되거나 방지할 수 있는 것으로 보였다. 모든 경우 white clover가 orchardgrass보다 B-함량이 높았고 $T_7$ 에서 B-함량이 크게 증가하였다. 혼파보다 단파재배에서 더 높았다. 처리구보다 더 높았으며 반면에 white clover는 가장 낮았다. 큰 요인을 형성하는 1차적 환경이 가정이라는 점에서 2000 가족과 함께 하는 창의성 경진대회는 창의성 축제의 한마당이었다고 감히 결론짓고자 한다.계시비 수준은 3회 예취구에서 558.9 kg $ha^{-1}\;yr^{-1}$, 4회 예취구에서 531.4 kg $ha^{-1}\;yr^{-1}$, 5회 예취구에서 546.3 kg $ha^{-1}\;yr^{-1}$ 이었으며, 이 때 얻어지는 최대 건물수량은 각각 18.4 ton, 12.7 ton, 10.6 ton $ha^{-1}$

시호추출물의 ICR 발암생쥐의 생존율 및 J774A.1 세포와 L1210 세포의 증식에 미치는 영향 (Effects of Bupleurum falcatum Extract on the Survival of Cancered ICR Mouse and the Growth of Cancer Cells such as J774A.1 Cells and L1210 Cells)

  • The current investigation was carried out to find out the anticancer activity of the methanol extract from Buplerum falcatum against cancered ICR mouse and cancer cell lines such as J774A.1 and L1210 cells. Extract of Buplerum falcatum displayed the considerable augmentation(134%) of the survival of ICR mouse bearing Sarcoma 180 cancer. In addition, the cytotoxic effects of methanol extract of Buplerum falcatum against J774A.1 cells and L1210 cells were found to show $IC_{50}$ values of $57.3\;{\mu}g/ml$ and $54.6\;{\mu}g/ml$, respectively. In contrast to such cytotoxicity against cancer cells, the extract exerted only meagre toxicity against normal lymphocytes. The increased generation of $O_2^-$ and the considerably increased activities of super-oxide dismutase(SOD) and glutathione peroxidase(GPx) of both J774A.1 cells and L1210 cells in the presence of Buplerum falcatum extract implied that the observed cytotoxicities may have resulted from the detrimental effect of reactive oxygen species(ROS) evoked by Buplerum falcatum extract on the cancer cells.

Oxidative Stress Is Decreased in Off-pump Versus On-pump Coronary Artery Surgery

  • Oxidative stress occurs in patients undergoing coronary artery bypass operation. The aim of this study was to investigate the difference in oxidative stress in off-pump versus on-pump coronary artery bypass surgery. In the present study, in serial blood samples, plasma malondialdehyde (MDA) as index of lipid peroxidation, red blood cells glutathione peroxidase (GPx) and superoxide dismutase (SOD) were measured to compare the extent of oxidative stress in 30 patients undergoing OPCAB (off-pump coronary artery bypass grafting), 12 patients undergoing CABG (on-pump coronary artery bypass grafting) and 18 healthy controls. In CABG group, MDA levels increased significantly from $2.87{\pm}0.62\;nmol/mL$ before anesthesia and $2.87{\pm}0.65\;nmol/mL$ after anesthesia to $3.05{\pm}0.66\;nmol/mL$ after ischemia (p < 0.05). Similarly, SOD levels also elevated significantly from $661.58{\pm}78.70\;U/g$ Hb before anesthesia and $659.42{\pm}81.21\;U/g$ Hb anesthesia induction to $678.08{\pm}75.80\;U/g$ Hb after ischemia (p < 0.01, p < 0.01, respectively). In OPCAB group, only SOD levels increased from $581.73{\pm}86.24\;U/g$ Hb anesthesia induction to $590.90{\pm}88.90\;U/g$ Hb after reperfusion (p < 0.05). Glutathione peroxidase levels were not changed according to blood collection times in both of CABG group or OPCAB group (p > 0.05). Our results show that only mild signs of oxidative stress is found after reperfusion in OPCAB operation compared with CABG operation. Further studies are needed in order to confirm this hypothesis.

발효강화쑥의 간장해 보호효과 (Hepatoprotective Effect of Fermented Artemisia princeps PAMPANINI by Lactic Acid Bacteria)

  • Artemisia princeps PAMPANINI has been used in traditional medicine of the treatment of inflammatory, liver dysfunction and order disorder in the far east countries including Korea. The present study was carried out to investigate the hepatoprotective effects of ethanol extract of Artemisia princeps (AP) and its fermented agents (AP-F) by lactic acid bacteria derived from human intestinal bacteria on liver injured rat induced by $CCl_4$ and d-galactosamine. Hepatoprotective activity was monitored by estimating serum ALT, AST, ALP, LDH, superoxide dismutase (SOD), glutathione redeuctase (GR) and glutathione peroxidase (Gpx) activities in the liver injured by hepatotoxin. Pretreating rats with AP or AP-F at the same dosage regimen significantly suppressed the acute elevation of serum transaminase, ALP, LDH and GR activities, and significantly increased the lowering of blood SOD and GR activites induced by hepatoxin. Based on these findings, it is presumed that AP and APF may have the hepatoprotective effect on $CCl_4$ and d-galactosamine-induced hepatotoxicity rat.

HPV 16 E6/E7 Transgenic Mice have Hyperkeratosis and Elevated ROS Related Enzyme Activities

  • Human papillomavirus type 16(HPV16) has been known to the major factor for the development of uterine cervical carcinomas. We have extended these studies to investigate the in vivo activities of HPV-16 E6/E7 when expressed in squamous epithelia of transgenic mice. Grossly, hK14HPV16E6/E7 transgenic mice had multiple phenotypes, including wrinkled skin that was apparent prior to the appearance of hair on neonates, thickened ears, and loss of hair in adults. In the transgenic mice, the wrinkled skin phenotype on the body and legs died at the age of 3∼4 weeks. Histological analysis of demonstrated that E6/E7 causes epidermal hyperplasia in multiple transgenic lineages with high penetrance. This epithelial hyperplasia was characterized by an expansion of the proliferating compartment and an expansion of the keratinocyte and was associated with hyperkeratosis. These transgenic mice expressed E6/E7 transgene mainly in skin, heart, pancreas and kidney. Hyperplasia was found at the skin. The enzyme activities of GR, GPx and CuZnSOD were measured from the transgene cause keratinocyte at the skin. The specific enzyme activities were significantly higher in transgenic mice skin compared to the normal mice skin. Thus these transgenic mice may be useful for the develpment of antioxidant enzymes or other therapies for HPV-associated hyperkeratosis.

Streptozotocin-유발 당뇨쥐에서 다시마 추출물 첨가식이의 항당뇨 및 항산화 효과 (Hypoglycemic and Antioxidative Effects of Dietary Sea-Tangle Extracts Supplementation in Streptozotocin-Induced Diabetic Rats)

  • The purpose of this study was to investigate the effect of dietary sea-tangle extracts on blood glucose levels, serum lipid levels, thiobarbituric acid reactive substance (TBARS) and glutathione enzymes in diabetic rats treated with streptozotocin (STZ) Four groups of rats (Sprague-Dawley male rats, 180 - 200g) were consisted of normal rats fed control diet (C), diabetic rats fed control diet (CD), normal rats fed sea-tangl extracts diet (E), and diabetic rats fed sea-tangle extracts diet (ED). Diabetes was induced by single injection of streptozotocin (60 mg/kg B.W.). After 7 weeks, rats were sacrificed, serum glucose, serum total cholesterol, triglyceride levels and glutathione enzymes were measured. Urine was significantly higher in CD and ED groups than those of others (p < 0.05). Levels of amylase, calcium, uric acid, hemoglobin, cholesterol and low density lipoprotein (LDL)-cholesterol were different among four groups. But high density cholesterol (HDL)-cholesterol of ED group was significantly higher (p < 0.05) than other groups (C and E group) And the weekly change of serum glucose was decreased in the 3th,4th and 5th weeks. But serum triglyceride (TG) of diabetic rats fed sea-tangle extracts diet (ED) was lower than diabetic rats fed control diet (CD). Activity of hepatic microsomal G6Pase was significantly increased CD and ED groups higher than C and E group, but kidney was decreased ED group. Hepateic glutathione S-transferase (GST) of CD and ED group were significantly lower than C and E group (p<0.05), glutathione peroxidase (GPX) of E and ED group were significantly higher than C and CD group (p<0.05), glutathione reductase (GR) activities of ED group was significantly lower than other groups, malondialdehyde (MDA) of ED was lower than E and CD group, but kidney was increased significant in ED group compared to liver. These results suggested that dietary sea-tangle extracts reduce .hepatic disorders such as oxidant than kidney. In conclusion, dietary sea-tangle extracts groups reduced blood TG and hepatic MDA levels in STZ-induced diabetic rats.

일부 폐경 후 여성에서 골밀도와 항산화효소 활성에 대한 대두 이소플라본 보충 효과 (Effect of Soy Isoflavone Supplementation on the Bone Mineral Density and Antioxidant Enzyme Activity in Postmenopausal Women)

  • To elucidate the effect of soy isoflavone supplementation on bone mineral density and antioxidant enzyme activity in 60 postmenopausal women residing in Iksan area were recruited. There were 31 participants in the treatment group and 29 in the control group. The treatment group consumed isoflavone extract capsules daily (which contained 90 mg of soy isoflavones) for 12 weeks. The study compared before and after isoflavone intake in the following areas. Physical examination, diet survey, bone mineral density (BMD) and antioxidant enzyme activity (superoxide dismutase (SOD), glutathione peroxidase (GPx), catalase (CAT) and total antioxidant capacity (TA)). The average age of the treatment group was 64.6 years and that of the control group was 66.5 years. There were no significant differences between the two groups in terms of height, weight and body mass index. Both groups maintained a regular diet pattern in terms of their average daily nutrient intake. There were no significant differences between the treatment group (23.3 mg) and the control group (24.0 mg) in terms of daily isoflavone intake based on diet. Isoflavone supplementation of 12 weeks did not resulted in any significant changes in BMD or parameters of antioxidant enzyme activity, implying the necessity of more intensive intervention for a substantial change. In conclusion, this study revealed that antioxidant enzyme activity holds a significant relationship with the bone mineral density in postmenopausal women and further systematic research on dose and period of isoflavone supplementation is needed to clearify the positive effect of isoflavone on BMD and/or blood antioxidant capacity in postmenopausal women.

저온에 대한 식물의 양분흡수, 탄수화물 및 항산화 반응 특성 (Responses of nutrient uptake, carbohydrates and antioxidants against low temperature in plants)

  • Recently, a quick drop of air temperature in plastic film houses by adverse weather conditions leads to the occurrence of low temperature damages to growing crops. Chilling injury, defined as a variety of growth restriction occurring below the optimal temperature, is one of environmental factors strongly affecting crop growth and yield. Low temperature causes the restricted evapotranspiration, reduced mineral uptake (P > K > $NO_3{^-}$), and an increase in electrolyte leakage such as K. Despite being different with plant species, an accumulation of soluble carbohydrates such as glucose, fructose, sucrose and starch under chilling condition is well known. A variety of environmental stresses are known to cause oxidative damage to plants either directly or indirectly by triggering an increased level of production of reactive oxygen species (ROS), and, to combat the oxidative damage, plants have the antioxidant defense systems comprising of enzymes, SOD, POD, CAT, GPX and APX, and non-enzymes, ascorbate, gluthathione, ${\alpha}$-tocopherol, phenolic compounds, carotenoid and flavonoids. The aim of this review is to provide basic information to build chilling-indicators and optimal nutrition management under adverse temperature conditions as broadly considering mineral uptake, carbohydrate metabolism and antioxidative defense system.

Anti-oxidative and Anti-inflammatory Effects of Protulaca Oleracea on the LPS-stimulated AGS Cells

  • Protulaca oleracea, a widely distributed weed, has been reported to exhibit different health promoting effects. The objective of this study was to evaluate the anti-oxidative and anti-inflammatory effects of P. oleracea on LPS-stimulated AGS cells. The cytotoxicity of P. oleracea in AGS cells was examined by MTT assay. The anti-oxidative effects of P. oleracea were examined by DPPH assay. RT-PCR was carried out to examine the effect of P. oleracea in the mRNA expression of different inflammatory mediators. MTT assay revealed that P. oleracea have almost no cytotoxity in AGS cells. DPPH radical scavenging activities were better than butylated hydroxyl toluene (BHT). The mRNA expression of different endogenous anti-oxidative enzymes (SOD2, GPx3 and catalase) were preserved by P. oleracea in AGS cells. The nitric oxide production and expression of iNOS in LPS stimulated RAW264.7 were suppressed in P. oleracea treated groups. Based on these findings, P. oleracea has protective anti-oxidant and anti-inflammatory effects.
