Effect of EM-fermented Orange in Commercial Diet on Growth of Juvenile Flounder, Paralichthys olivaceus (상품 사료에 첨가한 감귤발효액이 치어기 넙치, Paralichthys olivaceus의 성장에 미치는 영향)

  • Diets containing 0.02 to 10.0% EM-fermented orange (EFO) were fed to Paralichthys olivaceus(8.5 g), which were reared in flow-through system for 16 weeks. Groups fed on diets containing 0.1 and 0.2% EFO grew significantly faster; their feed coefficient and daily feeding rate were also higher. The number of goblet cells present in the mid-intestine of the fish receiving dietary EFO was significantly more. Total cholesterol level in plasma of the fish fed with 0.02% EFO was lower. However, there was no significant difference in GOT and GPT among the groups receiving different levels of EFO.

  • As a series of studies to investigate the effect of immunosuppression on Ascaris suum infection in undefinitive hosts, a delicate relationship between host and parasite, rabbits were divided into experiment 1(control group) and experiment 2(immnunosuppressive group treated with prednisolone acetate) and inoculated with a single dose of 5,000 embryonated A suum eggs. The recovery rates, sizes and morphology of the larvae and immunological responses in the rabbits were chronologically monitored according to somatic migration. In both experiments, the larvae failed to develop into the adults, but young adults in the experiment 2 grew somewhat faster and survied later than those in the experiment 1. The mast cells of small intestinal mucosa and mesenteric lymph nodes and the goblet cells of small intestinal mucosa in the worm detected cases of experiment 2 decreased remarkably in number comparing with those of experiment 1. Considering the experimental results. the expulsion mechanism of somatic migrant larvae may he related to the temporary increasing tendency of the mast cells, the goblet cells, T-cells of mesenteric lymph nodes and spleens, eosinophils in peripheral blood, degranulation rates of peritoneal mast cells and the migration inhibition rates of leucocytes. In addition, patent infection of A suum in the rabbits was not obviously observed despite of immunosuppression by prednisolone acetate.

STAT6 Expression and IL-13 Production in Association with Goblet Cell Hyperplasia and Worm Expulsion of Gymnophalloides seoi from C57BL/6 Mice

  • In intestinal helminth infections, Th2 immune respones are generally associated with mucin secretion for worm expulsion from the host intestine. In particular, IL-4 and IL-13 are the important cytokines related with intestinal mucus production via STAT6 signalling in nematode infections. However, this perspective has never been studied in Gymnophalloides seoi infection. The present study aimed to observe the STAT6 signalling and cytokine responses in C57BL/6 mice, a mouse strain resistant to infection with this trematode. The results showed that worm expulsion occurred actively during days 1-2 post-infection (PI), when goblet cells began to proliferate in the small intestine. The STAT6 gene expression in the mouse spleen became remarkable from day 2 PI. Moreover, G. seoi infection induced a significant increase of IL-13 from day 4 PI in the spleen of infected mice. Our results suggested that goblet cell hyperplasia and worm expulsion in G. seoi-infected mice should be induced by STAT6 signalling, in which IL-13 may be involved as a dominant triggering cytokine.

  • The integumental secretory structure is exocrine unicellular gland located in the epidermis of goldfish, Carassius auratus, and divided into two groups, mucous and granular cells. By the histochemical studies of integumental secretions the mucos cells reacted for acidic polysaccharides, and the granular cells for neutral glycoprotein. According to concentration of the secretion the integumental mucous are gradually sulphated. The mucous cells are typical form of goblet cell located in the upper region of the epidermis, and membrane bounded vesicles of the mucous are observed several size and electron densities by the cellular differentiation. The granular cells in middle and lower epidermis are present syncitial forms occasionally, and contain electron dense granules sized $1.0{\mu}m$ which are accumulated in cytoplasmic process held the cells to the basal lamina. The precursors of the integumental secretory materials are originated from the rough endoplasmic reticulum and next transported through the Golgi apparatus as a form of membrane bounded vesicles. After accomplish this process mature secretions are extruded to integumental surface by the mechanism of merocrine secretion in response to nerve stimulations respectively.

  • Asthma is characterized by a chronic inflammatory disorder of the airways that leads to tissue injury and subsequent structural changes collectively called airway remodelling. Characteristic changes of airway remodelling in asthma include goblet cell hyperplasia, deposition of collagens in the basement membrane, increased number and size of microvessels, hypertrophy and hyperplasia of airway smooth muscle, and hypertrophy of submucosal glands. Apart from inflammatory cells, such as eosinophils, activated T cells, mast cells and macrophages, structural tissue cells such as epithelial cells, fibroblasts and smooth muscle cells can also play an important effector role through the release of a variety of mediators, cytokines, chemokines, and growth factors. Through a variety of inflammatory mediators, epithelial and mesenchymal cells cause persistence of the inflammatory infiltrate and induce airway structural remodelling. The end result of chronic airway inflammation and remodelling is an increased thickness of the airway wall, leading to a increased the bronchial hyperresponsiveness and fixed declined lung function.

  • Histological changes and ontogeny, distribution and relative frequency of bovine SP-1/chromogranin(BCG)-, serotonin-, human pancreatic polypeptide(HPP)- and glucagon-immunoreactive cells were investigated in the colo-rectum of the chicken embryos. The pseudostratified columnar-like epithelium was observed in 10 days of incubation to 15 days of incubation, thereafter these epithelium were differentiated to simple columnar epithelium and goblet cells were detected for the first time on 19 days of incubation. BCG and serotonin-immunoreactive cells were observed for the first time on 15 days of incubation respectively, thereafter these cells were increased with ages. However no HPP and glucagon-immunoreactive cells were found in this study.

  • The tentacular epithelial cells of Cipangopaludina chinensis were studied with electron microscope. The tentacular epithelium consists of columnar supporting cells, numerous glandular cells and ciliated cells. Glandular cells are classified into three types; type I (mucous metachromasia cell), type II(mucous goblet cell) and type III. Ciliated cells are subdivided into two types; type I ciliated cell has cilia with typical axoneme(9+2), and type II ciliated cell has cilia with unusual axoneme.

  • Chronic rhinosinusitis (CRS) is a multifactorial, heterogeneous disease characterized by persistent inflammation of the sinonasal mucosa and tissue remodeling, which can include basal/progenitor cell hyperplasia, goblet cell hyperplasia, squamous cell metaplasia, loss or dysfunction of ciliated cells, and increased matrix deposition. Repeated injuries can stimulate airway epithelial cells to produce inflammatory mediators that activate epithelial cells, immune cells, or the epithelial-mesenchymal trophic unit. This persistent inflammation can consequently induce aberrant tissue remodeling. However, the molecular mechanisms driving disease within the different molecular CRS subtypes remain inadequately characterized. Numerous secreted and cell surface proteins relevant to airway inflammation and remodeling are initially synthesized as inactive precursor proteins, including growth/differentiation factors and their associated receptors, enzymes, adhesion molecules, neuropeptides, and peptide hormones. Therefore, these precursor proteins require post-translational cleavage by proprotein convertases (PCs) to become fully functional. In this review, we summarize the roles of PCs in CRS-associated tissue remodeling and discuss the therapeutic potential of targeting PCs for CRS treatment.

Histochemical Properties Study on the Mucosubstances of the Intestinal Mucosa in the Raja kenojei (홍어 장점막 점액의 조직화학적 성상에 관한 연구)

  • This study is performed in order to clarify the histochemical structure, the distribution of mucous cell and goblet cell, and the histochemical properties of the mucosubstances in the middle region of intestinal mucosa and rectum of Raja kenojei. In the H&E stain, distribution of the mucous cells and acidophilic cells were a more compacted middle portion than other regions of intestine, but the former was more than the latter in the number of mucous cells to rectum. The mucosubstances were stained with aldehyde fuchsin pH 1.7-alcian blue(pH 2.5) stain and then compared to distribution of the mucosubstances used in image and microscope technology(IMT-Size5). The middle intestine of Raja kenojei was composed of mucous cells having only large amounts of mucosubstances in the distal region was much more than that of proximal region. It was two types of mucous cells to rectum, one type was the same as proximal intestine while the other had small amounts of weakly sulfated and large amounts of carboxylated mucins.

Effects of Histidine Polymers on Mucin Release from Primary Cultured Airway Epithelial Cells (히스티딘 중합체가 일차배양 기도 상피세포에서의 뮤신 유리에 미치는 영향)

  • We investigated whether poly-L-histidine (PLH) significantly affect mucin release from cultured airway epithelial cells. Confluent primary hamster tracheal surface epithelial (HTSE) cells were metabolically radiolabeled with $^$^3H$$glucosamine for 24 hr and chased for 30 min in the presence of PLH to assess the effect on $^3H$-mucin release. PLH 9,850 and PLH 6,700 specifically inhibit mucin release from airway goblet cells without significant cytotoxicity. This finding suggests that poly-L-histidine might function as an airway mucoregulative agent.
