Tomato (Lycopersicon esculentum) seedlings exposed to various concentrations of $CdC1_2$ (0∼100 $\mu$M) in the nutrient solution for up to 9 days were analyzed with the seedling growth, $H_2O_2$ production, glutathione levels and activity changes of enzymes related to $H_2O_2$ removal. The growth of seedlings was inhibited with over 50 $\mu$M Cd, whereas the levels of $H_2O_2$ and glutathione were enhanced with Cd exposure level and time. Meanwhile, Cd exposure increased the activities of catalase (CAT) and glutathione reductase (GR) but decreased the activities of dehydroascorbate acid reductase (DHAR) and ascorbate peroxidase (APX) in both leaves and roots. These results suggest that the altered activities of antioxidant enzymes particularly involved in the $H_2O_2$ removal and the subsequent $H_2O$$_2$ accumulation could induce the Cd-induced phytotoxicity.

Background: Cigarette smoking is an important risk factor for chronic bronchitis and COPD. Airway epithelial cells exposed to cigarette smoke components such as nicotine, cotinine and benzopyrene can generate reactive oxygen species (ROS) and be subject to oxidative stress. This oxidative stress can induce the inflammatory response in the lung by the oxidant itself or by the release of proinflammatory cytokines. It has been reported that nicotine stimulates ROS, which are associated with NF-${\kappa}B$. Methods: Beas2B cells were treated with nicotine, cotinine and benzopyrene. RT PCR was used to measure the expression of several antioxidant factors using the total RNA from the Beas2B cells. The level of superoxide dismutase(CuZnSOD), thioredoxin, glutathione reductase expression was examined. Results: 0.5 to 4 hours after the benzopyrene, nicotine and cotinine theatments, the level of thioredoxin and glutathione reductase expression decreased. Longer exposure to these compounds for 24 to 72 hours inhibited the expression of most of these antioxidant factors. Conclusion: During exposure to smoke compounds, thioredoxin and glutathione reductase are the key antioxidant factors induced sensitively between 0.5 and 4 hours but the levels these antioxidants decrease between 24 hour and 72hours.

Dissolved oxygen is one of the most important factors controlling growth in aquatic organisms. Hypoxia is generally defined as dissolved oxygen less than 2.8 mg $O_2/L$ (equivalent to 2 mL $O_2/L$ or 91.4 mM). Therefore, hypoxia zone can cause a serious problem in marine ecosystem. In this study, to investigate embryotoxic (fertilization and embryo development rates) effects of hypoxia on sea urchin Strongylocentrotus nudus were exposed to dissolved oxygen levels of 7.6 mg $O_2/L$ (normoxia) and 1.8 mg $O_2/L$ (hypoxia) for 2 days at $15^{\circ}C$ and 33 ‰. Also, Expression levels of stress related gene (HSP70) and antioxidant related gene (glutathione reductase) in the sea urchins exposed to hypoxia were confirmed by Immunoblotting and RT-PCR analysis. In results, we showed that developmental rates were dramatically reduced in hypoxia condition. Molecular analysis demonstrated that higher HSP70 (5.5 fold) and glutathione reductase gene (2.79 fold) were present in the sea urchin exposed to hypoxia. Our results suggested that hypoxia can cause the abnormal development and elicits a stress and antioxidant response on sea urchin.

The potential of seven flavonoid glycosides to induce quinone reductase (QR), an anticarcinogenic marker enzyme, in murine hepatoma cells (hepalc1c7) and its mutant cells (BPRc1) was evaluated. Among test compounds, kaempferol-3-O-glucoside, luteolin-6-c-glucoside, and quercetin-3-O-glucoside (Q-3-G) induced QR in hepalc1c7 cells in a dose-dependent manner. However, in BPRc1 cells lacking arylhydrocarbon receptor nuclear translocator (ARNT), only Q-3-G caused a significant induction of quinone reductase at the concentration range of 0.5 to 8 ug/mL, suggesting that it is a monofunctional inducer. Q-3-G induced not only phase 2 enzymes, including QR and glutathione-S-transferase, but also nitroblue tetrazolium reduction activity in HL-60 cells, a biochemical marker for cell differentiation promoting agents. In conclusion, Q-3-G merits further study to evaluate its cancer chemopreventive potential.
The effects of waterborne arsenic (As) exposure on antioxidant defense were studied in liver and gills of tilapia, Oreochromis niloticus under thermal stress. Tilapia were exposed to different As concentrations (0, 200 and $400{\mu}g/L$) at three water-temperatures (WT; 20, 25 and $30^{\circ}C$) for 10 days. In antioxidant response, glutathione (GSH) levels, glutathione reductase (GR), glutathione peroxidase (GPx) and glutathione S-treansferase (GST) activities were significantly decreased depend on WT in the gills after As exposure. Also, the range of fluctuation in these enzymes activities was most significantly increased at $30^{\circ}C$ in the liver of tilapia exposed to As. The present findings suggest that a simultaneous stress by temperature change and As exposure could accelerate the alteration in antioxidant enzymes activities of tilapia.
Serum alanine aminotransferase(ALT), aspartate aminotransferase (AST), hepatic glutathione, glutathione S-transferase(GST), cytochrome P450 and cytochrome P450 reductase activity were measured to investigate the effects of hepatic detoxication system and metabolic activities of carbendazim in Sprague Dawley(S.D.) male rat at dose levels of 375, 750 or 1,500 mg/kg body weight. Serum alanine aminotransferase(ALT) and aspartate aminotransferase(AST) activities were slightly increased in all test groups after 120 minutes of administration. Glutathione was increased about 20% at high and medium dose level within 120 minutes after administration, while activity of glutathione S-transferase was decreased $36{\sim}50%$. However, the enzyme activity was recovered from all test groups after 240 minutes of administration. Cytochrome P450 and activity of cytochrome P450 reductase were decreased $25{\sim}50%$ until 120 minutes after administration, but recovered after 240 minutes.
Cnidii Rhizoma water extract (CRW) was tested for liver cancer chemopreventive potential by measuring the inhibition of phase I enzyme and benzo[a]pyrene-DNA adduct formation and induction of phase II detoxification enzymes. There was 17.0% inhibition in the activity of cytochrome P450 1A1 enzyme with the treatment of 150 mg/ml CRW. At concentration of 30 mg/ml CRW, the binding of $[^3H]B[a]P$ metablites to DNA of NCTC-clone 1469 cell was inhibited by 33.3%. CRW was potent inducer of quinone reductase (QR) and glutathione S-transferase (GST) activities in cultured murine hepatoma Hepalc1c7 cells. However, hepatic glutathione (GSH) level was not influenced by CRW. These findings suggest that CRW has chemopreventive potential of liver cancer by inhibiting cytochrome P450 1A1 activity and benzo[a]pyrene-DNA adduct formation and inducing QR and GST activities.
Scenedesmus spp. were cultured for 51 days in newly developed medium, KEP I (Kim and Ecopeace: initials of corresponding author and environmental company) made with Bacterio-Mineral-Water (3%, v/v) that had been bio-reacted with swine urine medium to 10% (v/v) Bold's Basal medium, and investigated for cancer chemopreventive potential by measuring the induction of quinone reductase (QR), glutathione S-transferase (GST), and reduced glutathione (GSH), and inhibition of cytochrome P450 (CYP) 1A1 activity. The activitives of QR and GST of Scenedesmus spp. cultured in KEP I medium were increased by 3.0-fold and 1.5-fold, respectively. However, Scenedesmus spp. cultured in control medium (CT) increased the activitives of QR and GST by 1.8-fold and 1.3-fold, respectively. Scenedesmus spp. in KEP I medium strongly inhibited CYP 1Al activity. These results show that Scenedesmus spp. in KEP I medium has cancer chemopreventive potential and may be a candidate for further development as a chemopreventive agent.

Water extract from Prunella vulgaris L. (PVW) was tested for colon cancer chemopreventive activity by measuring the activities of cytochrome P450 1A1, phase Ⅱ detoxification enzyme [quinone reductase (QR) and glutathione S-transferase (GST)] and ornithine decarboxylase (ODC) and glutathione (GSH) levels in cultured human colorectal adenocarcinoma HT-29 cells. PVW significantly inhibited 7,12-dimethylbenz[a]anthracene (DMBA)-induced cytochrome P450 1A1 activity at 10 and 50 ${\mu}g/ml$. PVW induced QR activity in a dose-dependent manner over a concentration range of $1{\sim}50\;{\mu}g/ml$. GST activity was also induced with the treatment of PVW in HT-29 cells. In addition GSH levels were increased with PVW. PVW inhibited ODC activity, a key enzyme of polyamine biosynthesis, which is enhanced in tumor promotion. These results suggest that Prunella vulgaris L. has colon cancer chemopreventive activity by inhibiting cytochrome P450 1A1 and ODC activities and by increasing phase Ⅱ enzyme activity and GSH levels.
