Effects of 4-nonylphenol (4-NP) on mixed function oxygenase (MFO) in the kidneies of rockfish (Sebastes schlegeli) were investigated. The cytochrome P45O (CYP) contents and NADPH cytochrome P450 reductase, NADH cytochrome b5 reductase and 7-ethoxyredorufin-O-deethylase (EROD) activities of microsome, glutathione S-transferase (GST) activity of cytosol in rockfish exposed to 4-NP for 7 days using an intraperitoneal injection (25 mg/kg) were quantitatively determined. The GST activity of rockfish exposed to 4-NP were higher, up to 5.2 times higher, than those in the control fish. The activities of NADPH-and NADH-dependent reductases were inhibited. On the other hand, CYP contents and EROD activity of the 4-NP exposed fish demonstrated neither an increasing or decreasing trend.
To find new inhibitory effects from oriental drugs, Albizziae root was extracted in methanol and the extracted was stepwisely fractionated by hexane, chloroform, ethylacetate, butanol and water. In cytotoxic effect of Albizziae root fractions against cancer cell lines including human hepatoma cells(HepG2) were investigated. Expecially the butanol fraction exhibited a inhibition effects on the growth of human hepatoma cells(HepG2). It inhibited of HepG2 cells with the value of IC50. The activities of qutathione after B(a)P treatment were markedly decreased than control, but those levels were increased by the treatment of Albizziae root methanol fraction. The activity of glutathione-S-transferase after B(a)P treatment were markedly decreased than control, but those levels were increased by the treatment of Albizziae root methanol traction. Induction of phase II enzymes is a major mechanism of chemoprevention. The induction levels of quinone reductase(QR) activity in cultured murine hepatoma(Hepa IcIc7)cell by methanol extract of Albizziae root were measured. Among the tested tractions, the extracts of butanol were found to induce QR activities over 2.8 fold than control. These results suggest that Albizziae root has chemopreventive Potential by inducing QR activities and GST levels and increasing GSH
암(暗) 조건(條件)에서 3일간(日間) 생육(生育)시킨 수수에 있어서 CPMI, 식물(植物) 생장조절물질(生長調節物質) 및 akylating agent 의한 GSH 함량(含量) 및 GST 활성(活性)의 변화(變化)를 조사(調査)하였다. 무처리(無處理) 수수의 지상부(地上部) 추출물(抽出物)에서 GST의 활성(活性)과 GSH의 함량(含量)이 근부(根部)에 비하여 높게 나타났다. CPMI로 처리(處理)한 수수에서 나타난 GSH의 분포(分布) 양상(樣相)은 전반적으로 무처리(無處理)와 차이(差異)를 보이지 않았으나, 초엽부(葉部)에서 GST[metolachlor]의 활성(活性)이 무처리(無處理)에 비하여 약 2.3배 증대되었다. 이러한 GST 활성(活性)의 증대(增大)는 metolachlor에 특이적(特異的)으로 작용하는 GST isozyme의 발현(發現)에 기인하는 것으로 나타났다. 이어서 metolachlor 부활성화(不活性化) 효소(酵素)인 GST의 활성(活性) 변화(變化)를 가져오는 관연기작(關聯機作), 즉 plant hormone regulation 및 substrate induction에 근거(根據)한 일련의 실험(實驗)을 실시(實施)하였다. GST [metolachlor]의 활성(活性)은 2,4-D 처리시(處理時)에 약 2.1배 증가(增加) 하였으나, ethylene과 ABA에 의한 변화(變化)는 나타나지 않았다. 또한 metolachlor는 이 제초제(除草劑)에 특이성(特異性)을 갖는 GST isozymes의 발현(發現)을 통하여 3.4배의 GST 활성(活性) 증가(增加)를 가져온 반면에, 기타 alkylating agent로 사용(使用)된 2-iodoacetamide와 2-bromo-N-phenylacetamide에 있어서는 GST의 활성(活性)을 억제(抑制) 또는 영향(影響)을 주지 않는 것으로 나타났다. 이상의 결과(結果)로 부터 수수에 있어서 CPMI에 의한 GST[metolachlor] 활성(活性)의 증대(增大)가 auxin regulation 또는 substrate induction에 부분적(部分的)으로 관련(關聯)되어 있는 것으로 나타났다.
브롬(Br-)이온을 포함하는 지표수 또는 해수를 상수원 또는 어류양식용 물로 사용할 경우 소독, 살균을 위하여 오존(O$_3$)을 많이 사용한다. 이때 물속에 포함되어 있는 브름(Br-) 이온과 오존과의 반응에 의하여 독성물질인 Bromate(BrO$_3$-)가 산화 부산물로서 생성된다. 이 bromate의 생성반응에 대한 용액의 pH와 반응온도의 영향 및 bromate를 음용수중에 함유시키고 실험동물에 섭취시켰을 때의 생체독성에 미치는 기전을 관찰하였다. Bromate의 생성은 반응온도가 증가할수록 증가되지만, 낮은온도(15$^{\circ}C$)에서는 용액의 초기 pH가 3 인 경우는 초기 pH가 7, 10 인 용액의 경우보다 훨씬 적은 생성량을 나타내었다. Bromate를 음료수중에 0, 0.1, 0.2, 0.4g/L로하여 4, 12, 16, 20, 24주 투여하고서 신조직중의 지질과산화의 함량이 증가되었으며, 혈중 뇨소질소의 활성 및 뇨중 ${\gamma}$-glutamy-ltransferase의 활성은 대조군에 비하여 bromate의 투여로 현저히 증가되었으며, 뇨중 lactate dehydrogenase의 활성에는 별다른 영향이 없었다. Bromate의 투여로 xanthine oxidase 및 aldehyde oxidase의 활성은 bromate의 투여로 현저히 증가되었으며 glutathione의 농도 및 glutathione S-transferase의 활성도 대조군 보다 현저히 억제되었다. Glutathione의 생성계에 미치는 ${\gamma}$-giutarnylcystein synthetase의 활성은 대조군에 비해 bromate의 투여로 억제되었으며 glutathione redurtase의 활성은 별다른 영향이 없었다.
Objectives: Most of human cancers may result from exposure to environmental carcinogens, and individual effectiveness in the detoxification of these chemicals will influence susceptibility to malignant disease. Glutathione S-Transferases(GSTs) enzymes are involved in the detoxification of active metabolites of many carcinogens from tobacco smoke and may be important in modulating susceptibility to smoke-related cancer. The purpose of this study is to determine the polymorphism of GSTM1, GSTT1, and GSTP1 in control group and head and neck squamous cell carcinoma group of Korean, and to investigate the effect of GSTs polymorphism on the risk of head and neck cancer. Materials and Methods: A hospital-based case-control study was performed with a group of 133 control individual and 136 head and neck squamous cell carcinoma patients. The polymorphisms of GSTs were analysed using polymerase chain reaction in GSTM1 and GSTTl, and polymerase chain reaction-restriction fragment length polymorphism in GSTP1. Results: The relative risk (odds ratio) of GSTM(-) genotype was 1.14(95% CI, 0.70-1.85) compared to GSTM1(+). The odds ratio of GSTTl(-) genotype was 0.91(95% CI, 0.55-1.50). In old age($65$) group, the odds ratio of GSTT1(-) genotype was 5.2(95% CI, 1.53-17.89). The GSTP1 Val/Val genotype conferred a 1.7-fold risk(95% CI, 0.40-7.34) of head and neck cancer compared with GSTP1 Ile/Ile genotype. Among the combined genotypes of GSTs, GSTM1(-)/GSTT1(+)/GSTP1 Val/Val and GSTM1(-)/GSTTl(-)/GSTP1 Ile/Val genotypes conferred a 2.6-fold and 1.3-fold risk(95% CI, 0.24-14.15 and 0.43-3.14) compared with the GSTM1(+)/GSTTl(+)/GSTP1 Ile/Ile genotype, respectively. Conclusion: Polymorphism of GSTs might modulate susceptibility to head and neck cancer in Korean population. The genotype of GSTP1 Val/Val and combined genotypes of GSTM1(-)/GSTT1(+)/GSTP1 Val/Val, and GSTM1(-)/GSTT1(-)/GSTP1 Ile/Val might be important risk factors to determine the individual susceptibility to head and neck squamous cell carcinoma.
Glutathione S-transferase (GST) is a multigene family of phase II detoxifying enzymes that metabolize a wide range of exogenous and endogenous electrophilic compounds. GSTM1 and GSTT1 gene polymorphisms may account for inter-individual variability in coping with oxidative stress. We investigated the relationships between the level of lymphocyte DNA and antioxidative parameters and the effect on GST genotypes. GSTM1 and GSTT1 were characterized in 301 young healthy Korean adults and compared with oxidative stress parameters such as the level of lymphocyte DNA, plasma antioxidant vitamins, and erythrocyte antioxidant enzymes in smokers and non smokers. GST genotype, degree of DNA damage in lymphocytes, erythrocyte activities of superoxide dismutase, catalase, and glutathione peroxidase (GSH-Px), and plasma concentrations of total radical-trapping antioxidant potential (TRAP), vitamin C, ${\alpha}$- and ${\gamma}$-tocopherol, ${\alpha}$- and ${\beta}$-carotene, and cryptoxanthin were analyzed. Lymphocyte DNA damage assessed by the comet assay was higher in smokers than that in non-smokers, but the levels of plasma vitamin C, ${\beta}$-carotene, TRAP, erythrocyte catalase, and GSH-Px were lower than those of non-smokers (p < 0.05). Lymphocyte DNA damage was higher in subjects with the GSTM1- or GSTT1-present genotype than those with the GSTM1-present or GSTT1- genotype. No difference in erythrocyte antioxidant enzyme activities, plasma TRAP, or vitamin levels was observed in subjects with the GSTM1 or GSTT1 genotypes, except ${\beta}$-carotene. Significant negative correlations were observed between lymphocyte DNA damage and plasma levels of TRAP and erythrocyte activities of catalase and GSH-Px after adjusting for smoking pack-years. Negative correlations were observed between plasma vitamin C and lymphocyte DNA damage only in individuals with the GSTM1-present or GSTT1- genotype. The interesting finding was the significant positive correlations between lymphocyte DNA damage and plasma levels of ${\alpha}$-carotene, ${\beta}$-carotene, and cryptoxanthin. In conclusion, the GSTM1- and GSTT1-present genotypes as well as smoking aggravated antioxidant status through lymphocyte DNA damage. This finding confirms that GST polymorphisms could be important determinants of antioxidant status in young smoking and non-smoking adults. Consequently, the protective effect of supplemental antioxidants on DNA damage in individuals carrying the GSTM1- or GSTT1-present genotypes might show significantly higher values than expected.
댑싸리하고초 약침액을 이용하여 phase II detoxification 효소인 quinone reductase (QR 및 GST) 유도, GSH 생성량, phase I 효소 cytochrome P4501A1 활성억제, 발암물질 B[a]P-DNA adduct 형성의 저해효과 등을 측정하였다. QR 생성 유도를 Hepa1c1c7로 실험한 결과, 댑싸리하고초 약침액및 열수추출액 모두에서 유도 되었으며, 농도가 높아질수록 유도율이 더 높게 나타났으며, GSH와 GST의 유도율도 약침액에서 나타났다. 댑싸리하고초 약침액 $5{\times}$에서 45.2%의 cytochrome P4501A1 효소활성 저해효과를 측정할 수 있었다. 또한 B[a]P-DNA binding 저해효과를 실험한 결과, 약침액 $3{\times}$ 농도에서 45.0%의 저해효과가 있었다. 이상의 결과에 의하면 댑싸리하고초 약침액은 암억제 물질로서의 기능을 충분히 발휘할 것으로 사료된다.

Recent genome annotation revealed that Populus trichocarpa contains 81 glutathione S-transferase (GST) genes. GST genes play important and varying roles in plants, including conferring tolerance to various abiotic stresses. Little information is available on the relationship - if any - between drought/salt stresses and GSTs in woody plants. In this study, we screened the PatgGST genes in hybrid poplar (Populus alba ${\times}$ Populus tremula var. glandulosa) that were predicted to confer drought tolerance based on our expression analysis of all members of the poplar GST superfamily following exposure to salt (NaCl) and drought (PEG) stresses, respectively. Exposure to the salt stress resulted in the induction of eight PatgGST genes and down-regulation of one PatgGST gene, and the level of induction/repression was different in leaf and stem tissues. In contrast, 16 PatgGST genes were induced following exposure to the drought (PEG) stress, and two were down-regulated. Taken together, we identified seven PatgGSTs (PatgGSTU15, PatgGSTU18, PatgGSTU22, PatgGSTU27, PatgGSTU46, PatgGSTU51 and PatgGSTU52) as putative drought tolerance genes based on their induction by both salt and drought stresses.
Phase II 효소계는 자연계에 존재하는 다양한 화학물질과 천연소재들에 의해 유도되며, 이들의 유도는 화학적 발암물질과 그 밖의 여러 가지 독성물질들로부터 생체를 보호하는데 중요한 역할을 한다. 특히, phase II 효소 중 QR은 quinone류 자체에 대한 보호효과가 있고 다른 암예방 효소계와 공통으로 유도되며, 항암 작용이 있는 많은 화합물에 의해 유도되어지기 때문에 암예방 물질 탐색의 지표가 되는 대표적인 효소로 많이 이용된다. 본 연구에서는 제주도에서 자생하는 귤류 중 지각을 이용하여 메탄올추출물을 제조한 후 대표적인 암예방계 효소로 알려진 quinone reductase 및 glutathion S-transferase의 유도 활성을 조사하였다. 우선 QR 및 GST 유도활성 측정에 앞서 각 시료의 Hepa 1c1c7 세포에 대한 독성을 조사하여 시료자체의 세포독성을 관찰하였으며, 이 결과를 바탕으로 QR, GST 유도 활성을 측정할 시 세포독성을 거의 나타내지 않는 각각 시료의 최대 처리 농도를 결정하였다. 총 6개의 지각 분획물 중 mouse 유래 Hepa 1c1c7 세포주에서는 hexane 충과 chloroform 충에서 QR, GST의 활성이 $200{\mu}g/mL$에서 각각 1.8배, 1.5배 이상으로 증가되어 높은 QR 및 GST의 유도활성을 보였다. 따라서 지각의 hexane 층이나 chloroform층에 존재하는 높은 QR및 GST의 유도활성이 나타나는 물질을 분리 정제하여 앞으로 계속적인 연구가 진행되어야 할 것이다.
We investigated the effect of propolis on the activity of antioxidant enzymes in rat liver exposed by X-ray irradiation. The dosage of propolis showed the effect of lowering the concentration of superoxide anion in irradiated rat liver, suggesting that propolis has a significant role to remove superoxide anion as an antioxidant and/or by activating the antioxidant enzyme. The activities of superoxide dismutase (SOD) and glutathione reductase (GR), disturbed by X-ray irradiation, were restored in 30 days to normal status in the group which dosed propolis before X-ray irradiation. Interestingly, catalase (CAT) and glutathione peroxidase (GPOX) activities were highly increased with feeding propolis to rat compared to untreated group, whereas glutathione s-transferase (GST) activity was little affected. Taken together, it suggests that the propolis has a protective role in the rat liver cells against X-ray irradiation by increasing and recovering the activities of antioxidant enzymes.
