Establishment of Chlorantraniliprole-Resistant Drosophila Strains and Identification of Their Resistant Characteristics (Chlorantraniliprole 저항성 초파리 계통 확립과 저항성 특성 구명)

  • Ryanodine receptors (RyRs) regulate the contractions of insect muscles by altering intracellular $Ca^{2+}$ concentration and are the targets of chlorantraniliprole. Recently, a chlorantraniliprole-resistant strain was reported in the diamondback moth Plutella xylostella by obtaining point mutations on the RyRs. In the present study, we established two resistant strains from Drosophila melanogaster, which were treated with low or high concentrations of chlorantraniliprole, and their resistance levels were determined on the basis of contact and ingestion toxicities. Compared with the control strain, the two resistant strains did not show any significant differences in contact toxicity. However, they showed significantly increased resistance ratios in ingestion toxicity than that by the control strain. The low and high concentration resistant strains exhibited 2.1- and 8.1-fold increased resistance ratios, respectively, compared with that by the control strain. Moreover, we found that the resistant strains had altered expression levels of RyRs and more enhanced Acetylcholinesterase and Glutathione-S-transferase activities than that by the non-selected strain. These results suggested that the resistance development of chlorantraniliprole in the two strains might be mediated by the activation of detoxification pathways in D. melanogaster.

Effect of Cyclohexane Treatment on Serum Level of Glutathione S-Transferase Activity in Liver Damaged Rats ($CCl_4$ 에 의한 간손상 모델 실험동물에 있어서 cyclohexane 투여가 혈청 glutathione S-transferase 활성에 미치는 영향)

  • To evaluate the effect of cyclohexane(CH) treatment on the serum levels of glutathion S-transferase(GST) activity in liver damaged animals, damaged liver was induced with pretreatment of 50% $CCl_4$ dissolved in olive oil (0.1 m1/100g body weight) intraperitoneally 17 times every other day. To $CCl_4$-treated rats, CH (1.56 g/kg body weight, i.p) was injected once and then the animals were sacrificed at 4 hours after injection of CH. The $CCl_4$-treated animals were identified as severe liver damage on the basis of liver functional findings, 1,e, increased serum levels of alanine aminotransferase(ALT), alkaline phosphate(ALP) and xanthine oxidase(XO) activities. On the other hand, $CCl_4$-treated animals injected with CH once($CCl_4$-pretreated animals) showed more decreased serum levels of ALT and XO, and more increased those of ALP rather than $CCl_4$-treated animals. In case of comparing the GST with ALT activity in liver, both $CCl_4$-treated and pretreated animals showed similar changing pattern of enzyme actvity. Especially $CCl_4$-pretreated animals showed significantly increased serum level of GST actvity compared with the $CCl_4$-treated those, whereas those of ALT showed reversed tendency. In aspects of GST enzyme kinetics, $CCl_4$-pretreated animals showed higher Vmax of liver GST enzyme than $CCl_4$-treated animals. In conclusion, injection of CH to the liver damaged rats led to enhanced liver damage and more increased activity of serum GST which may be chiefly caused by the enzyme induction.

Effect of Tota1 Saponin from Red Ginseng on Acvtivities of Antioxidant Enzymes in Pregnant Rats (홍삼 사포닌이 수태중인 흰쥐의 항산화 효소활성에 미치는 영향)

  • Pregnancy is a physiological state accompained by a high energy demand of many bodily functions and an increased oxygen requirement. Because of the increased intake and utilization of oxygen, increased levels of oxidative stress would be expected. So we observed the activities of the hepatic antioxidant enzymes from rat treated with total saponin from the red ginseng against free raicals produced in pregnant rats. The activity of superoxide dismutase (SOD) in the control group was slightly decreased during pregnancy, and SOD activity in total saponin treated group was not observed any siginificant change compared with the control group. The activities of glutathione peroxidase (GPX), glutathione reductase (GRD) and catalase in the control group have shown the decreasing tendency during pregnancy, whereas the activities of GRD and catalase in total saponin treated group showed significant increased tendency compared with the control group. GPX activity in total saponin treated group was slightly decreased tendnency compared with the control group. The activity of glutathione-S-transferase (GST) in the control group was increased to keep the state of homaeostasis tendency in pregnant rats. On the other hand, the activity of GST after total saponin treatment was increased than control group. Activity of all enzymes in the control group and total saponin treated group recovered the normal level after delivery of rats. In spite of the physiological changes in vivo, the inflaunce of total saponin on activaties of hepatic antioxidant enzyme in pregnant rats seems to be regulated the biological homeostatic adaptation mechanism which protects the maternal liver aganist oxygen induced toxicity

Antioxidative Effect of Ethanol Extract on Arctium lappa root in Streptozotocin Induced Diabetic Rats (우엉 뿌리 에탄올 추출물이 Streptozotocin으로 유발된 당뇨 흰쥐의 항산화에 미치는 영향)

  • This study was carried to investigate the antioxidative effect of ethanol extract of Arctium lappa(Al) root in Streptozotocin(STZ)-induced diabetic rats. Diabetes was induced by intravenous injection of STZ at a dose of 45mg/kg.body wight(b.w) dissolved in citrate buffer. The ethanol extract of Al root was orally administrated once a day for 7 days at a dose of 1,000mg/kg.b.w. The contents of malondialdehyde(MDA) and activities of catalase (CAT), glutathione peroxidase(GSH-Px) were significantly decreased(p<0.05) in Al treated group compared to the those of STZ-control group. The content of glutathione(GSH) and activity of glutathione-s-transferase(GST) was significantly increased(p<0.05). These results indicated that ethanol extract of Al root would have antioxidative effect in STZ-induced diabetic rats.

Effects of Extract of Pueraria radix on Lipid Peroxidation in Ethanol-Administered Rats (갈근추출물이 에탄올을 투여한 흰쥐의 지질과산화에 미치는 영향)

  • This study was designed to investigate the effect of Pueraria radix extract on lipid peroxidation in ethanol administered rats. Male sprague dawley rats were given 25% ethanol(2.5g per Kg body weight; E), 10% pueraria radix extract(CP), 25% ethanol and 10% pueraria radix extract(EP). The activity of hepatic superoxide dismutase was increased by ethanol and was lower in the EP group than in the E group. Hepatic catalase activity was increased by ethanol, but decreased by Pueraria radix extract. E group rats had significantly higher liver glutathione peroxidase activity. Activity of hepatic glutathione S transferase was higher in the CP group than in the other groups. No significant dif ferences was found in liver glutathione and lipid peroxide contents between control and EP group. These data indicate that the peroxidative damage associated with chronic ethanol consumption might be decreased by Pueraria radix extract.

  • The treatment with 5ng/ml of mercuric chloride caused time-dependent decreases, and in the activities of GSH S-transferase and GSH-peroxidase, and in the concentration of GSH in CHO cells. Three hours after treatment of $Hg^{2+}$, the activity of GSH S-transferase was decreased to almost half value of control group and the activity of GSH-peroxidase was reduced significantly at 6 hr after treatment. The concentration of GSH was decreased 2 hr after treatment of $Hg^{2+}$ and was decreased to nearly half value of control group 3 hr after treatment.

Effect of Ethanol Pretreatment on the Bromobenzene Metabolism in Rats (흰쥐에 있어서 주정중독이 Bromobenzene 대사에 미치는 영향)

  • To investigate an effect of ethanol pretreatment on the bromobenzene metabolism, the brornobenzene (400 mg/kg body wt. i. p.) was given 3 times at intervals of one day to the male rats orally pretreated with 5% ethanol throughout 2 months. In the ethanol pretreated rats, liver injuries were not demonstrated on the basis of the liver weight per body weight, serum levels of alanine aminotransferase (ALT) activity and histopathologic findings. By the bromobenzene treatment, ethanol pretreated rats showed the more decreased levels of serum ALT and liver weight/body weight(%), and decreased degree of liver damage on histopathological observation than the control group. The ethanol pretreated rats showed the more increased activities of hepatic aniline hydroxylase, glutathione Stransf erase (GST) and the more decreased contents of glutathione than the control. Concomitantly the ethanol pretreated rats showed the more decreased contents of hepatic glutathione and increased activities of GST by the bromobenzene treatment. Above results indicate that ethanol pretreatment enhance the metabolizing ability of bromobenzene in rats.

  • Objectives: In traditional medicine, Wolgukwhan has been used for the treatment of digestive system disease, such as indigestion, brash, ructation, nausea and vomiting. This study was purposed to investigate the effects of Wolgukwhan methnol extract (WGWM) on oxidative liver cell injury. Methods: In vivo assay, we administerated acetaminophen(500mg/kg, i.p.) to starved mice 24hrs after pretreatment of WGWM for 6days. In the liver homogenates, lipid peroxide and glutathione(GSH) levels were measured. In addition, activities of hepatic enzyme, such as catalase, glutathione peroxidase(GPX), glutathione S-transferase(GST) were measured in the hepatic mitochondrial and cytosolic fractions. Results: In vivo administeration of WGWM showed effective inhibition of acetaminophen induced lipid peroxidation and elevations of glutathione level. The acetaminophen treatment resulted in a decrease of catalase, GPX and GST activities. By contrast, WGWM pretreatment increased compare to those of untreated groups. Conclusions: These results suggested that WGWM might protect against lipid peroxidation by free radicals, destruction of hepatic cell membranes.

Effect of Brief Treatment of Bromobenzene on the Liver $N /

  • Bromobenzene 투여에 의한 간조직중 ATPase 활성을 관찰할 목적으로 흰쥐에 bromobenzene을 체중 kg당 400 mg을 복강으로 투여한 다음 4시간 후에 처치하여 다음과 같은 결과를 얻었다. Bromobenzene 투여로 인한 체중당 간무게는 유의하게 증가되었으나 간조직중 단백질 함량은 감소되었다. 혈청중 alanine aminotransferase 활성은 대조군과 별다른 차이를 볼수 없었다. 따라서 본 실험조건에서 Bromobenzene 처치 실험동물에서 간조직은 가역적상해로 생각되며 이러한 실험동물모델에 $Na^+/K^+$-ATPase 활성은 유의하게 (p<0.05)증가되었으며 이때 V$_{max}$ 치역시 대조군에 비하여 증가 되었다. 이때 간조직중 glutathione 함량은 감소되었으며 glutathione S-transferase 활성 및 cytochrome P-450 함량치는 증가되는 경향을 보였다.

Molecular Characterization and Expression Analysis of a Glutathione S-Transferase cDNA from Abalone (Haliotis discus hannai) (북방전복 (Haliotis discus hannai)에서 분리한 Glutathione S-transferase 유전자의 분자생물학적 고찰 및 발현분석)

  • Glutathione S-transferases (GSTs) are a superfamily of detoxification enzymes that primarily catalyze the nucleophilic addition of reduced glutathione to both endogenous and exogenous electrophiles. In this study, we isolated and characterized a full-length of alpha class GST cDNA from the abalone (Haliotis discus hannai). The abalone GST cDNA encodes a 223-amino acid polypeptide with a calculated molecular mass of 25.8 kDa and isoelectric point of 5.69. Multiple alignments and phylogenetic analysis with the deduced abalone GST protein revealed that it belongs to the alpha class GSTs and showed strong homology with disk abalone (Haliotis discus discus) putative alpha class GST. Abalone GST mRNA was ubiquitously detected in all tested tissues. GST mRNA expression was comparatively high in the mantle, gill, liver, and digestive duct, however, lowest in the hemocytes. Expression level of abalone GST mRNA in the mantle, gill, liver, and digestive duct was 182.7-fold, 114.8-fold, 4675.8-fold, 406.1-fold higher than in the hemocytes, respectively. Expression level of abalone GST mRNA in the liver was peaked at 6 h post-infection with Vibrio parahemolyticus and decreased at 12 h post-infection. While the expression level of abalone GST mRNA in the hemocytes was drastically increased at 3 h post-infection with Vibrio parahemolyticus. These results suggest that abalone GST is conserved through evolution and may play roles similar to its mammalian counterparts.
