  • A series of S-(2,2-diethoxycarbonyl-1-phenylethyl)-L-glutathione derivatives (11a-e) were synthesized from the reaction of ${\beta},\;{\beta}$-diethoxycarbonylstyrene with L-glutathione in 9 : 1 aqueous methanol. Thus, S-(2,2-diethoxycarbonyl-1-phenylethyl)-L-glutathione (11a), S-2,2-diethoxycarbonyl-1-(3',4'-methylenedioxy)phenylethyl-L-glutathione (11b), S-2,2-diethoxycarbonyl-1-(3',4',5'-trimethoxy)phenylethyl-L-glutathione (11c), S-2,2-diethoxycarbonyl-1-(4'-hydroxy)phenylethyl-L-glutathione (11d), S-2,2-diethoxycarbonyl-1-(4'-methoxy)phenylethyl-L-glutathione (11e) were obtained in good yields. The structure of the adducts was characterized by analytical and spectral data. The effects of pH and solvents upon the yields were also briefly examined. In the range of pH from 4.0 to 8.0, the aqueous methanol were found to be the best solvent for the addition reaction and the antibacterial activities of the adducts to Gram(+) bacteria were found to be weak.

Effects of Bromate on the Glutathione Synthesis in Various Organs of Rats (Bromate가 흰쥐의 장기 Glutathione 함량에 미치는 영향)

  • The effects of bromate administration on glutathione were studied in rats. The contents of glutathione in the liver and kidney were significantly decreased but the alteration was not significant in lung and blood by bromate adminstration. The decrease occurred without concomitant increases in oxidized glutathione (GSSG) or in the GSSG/GSH+GSSG ratio. The activities of $\gamma-glutamyl$ cysteine synthetase in the liver and kidney were decreased by bromate administration. $\gamma-Glutamyl$ transpeptidase activities was significantly decreased in the kidney and not significantly decreased in the lung of bromate treated-rats. These results suggest that the decreased synthesis of glutathione by bromate may be an important reason for the decreased level of glutathione in the liver and kidney, thus the decreased glutathione transport would be a factor on the changes of glutathione contents in bromate-treated rats.

  • Glutathione and ascorbic acid have been shown to fulfill many essential functions in animal and plant growth, development, defence and protection against oxidative damage. Effects of glutathione and ascorbic acid were examined in transgenic N. tabacum cells producing hGM-CSF to determine the effects of the vitamins on growth and cell viability. In lag phase, cell viability was preserved by glutathione and ascorbic acid. Therefore, recombinant protein productivity was increased. The purpose of present study is to investigate the role of antioxidants in cold stress-induced apoptosis in plant suspension cells. Cold stress lowered cell viability and increased total genomic DNA fragmentation. Supplementing the cell cultures with glutathione and ascorbic acid inhibited cold stress-induced decrease in cell viability and increase in total genomic DNA fragmentation.

Influence of Gami-oryungsan on bromobenzene-induced liver injury in experimental animal (Bromobenzene독성(毒性)에 의한 간기능손상(肝機能損傷)에 미치는 가미오령산의 영향(影響))

  • Objective : To investigate the hepatoprotective effects of Gami-oryungsan on the liver damage induced by bromobenzene. Method : The development of fibrosis and acute liver injury was examined by the chemical analysis of AST, AL T, ${\gamma}$-GTP . and epoxide hydrolase glutathione S-transferase glutathione peroxidase enzyme activity, lipidoperoxide levels, glutathione levels were measured and oberved. Results : The increasing levels of lipidoperoxide was decreased proportionally according to dose of extract GO. Epoxide hydrolase glutathioneS-transferase glutathione peroxidase enzyme activity highly increased in GO pre-acupunctured group compared with the group treated with only bromobenzene. The increase of serum AST, AL T, ${\gamma}$-GTP enzyme activity of mice by bromobenzene was inhibited by the administration of GO. Lipidoperoxide levels in rat's liver decreased compared to the case of bromobenzene-treated group. The levels of Glutathione decreased by bromo benzene were increased highly in GO pre-acupunctured group. Conclusion : These results suggest that GO extract recovers the damage of liver due to bromobenzene intoxication by decreasing the lipid peroxidation AST AL T ${\gamma}$-GTP enzyme activity and increasing epoxide hydrolase glutathioneS-transferase glutathione peroxidase enzyme activity, glutathione levels.

  • The effects of different dietary fatty acids on the hepatic glutathione S-transferase(GST-P) positive foci and glutathione related enzyme system were investigated in carcinogen treated rats. Weaning male Sprague Dawley rats were divided into three groups and fed the diets of 15% corn(CO), perilla(PO), and sardine oil(SO), respectively. Hepatocellular carcinogenesis was initiated with diethylnitrosamine(DEN) and then fed the diet containing 0.02% 2-acetylaminofluorene(2-AAF) followed by 0.05% phenobarbital for 10 weeks. The hepatic tissues were homogenized and centrifugated to prepare microsomal and cytosolic fractions. The enzyme activities of hepatic glutathione S-transferase(GST), glutathione reductase(GR), and glutathione peroxidase(GPx) were determined from cytosolic fractions. The number of GST-P hyperplastic nodules was the highest in corn oil group at 6th week, the early stage of hyperplastic nodule formation. GST activities were increased significantly by carcinogens in all dietary groups after 6th wk. GR activities followed the same trend as GST activities. GPx activities were decreased by carcinogens in all dietary groups at 10th week. In this experiment, corn oil diet may have promotive effect on hyperplastic nodule formation during the early promotional stages of chemical carcinogenesis.

Production of Glutathione by Candida sp. Mutant (Candida sp. 변이주에 의한 Glutathione 생산)

  • For the overproduction of glutathione, Candida sp. mutant was isolated by the treatment with U.V. light. The highest glutathione production of Candida sp. mutant was obtained after shaking culture for 48 hours in the cullture medium containing glucose 1.5%(w/v), yeast extract 4.0% (w/v), KH2PO4 0.04%(w/v), biotin 5 ng/ml, and L-cysteine 0.04%(w/v). The optimal pH and temperature for the glutathione production were pH 6.0 and 25C, respectively.

  • This investigation was aimed at the study of the antidiabetic activity and effect on hepatic glutathione metabolism of polysaccharide from Trichosanthes kirilowii in hyperglycemic rats with aboxan (175 mg/Kg, i.p.). As the results, the polysaccharide inhibited the increase of blood glucose, triglyceride level and lactate dehydrogenase activity, but cholesterol not changed. And it increased protein bound-SH, nonprotein bound-SH, glutathione level and inhibited the decrease of glutathione S-transferase.

  • To evaluate the effect of bromobenzene pretreatment on the bromobenzene metabolism, the animal group was induced the stage of slight liver damage with 7 times bromobenzene injection every two days (400 mg/kg body wt. i.p.). In the present experimental animal model, the single dose of bromobenzene(400 mg/kg body wt. i.p.) was injected to the bromobenzene-pretreated rats and the hepatic aniline hydroxylase(AH) activity, glutathione(GSH) content and glutathione S-transferase (GST) activity were determined at the intervals of 2, 4, 8, 24 hours throughout 24 hr. The activities of hepatic AH and GST were generally higher in bromobenzene-pretreated rats than those in normal group throughout the whole course of experiment. Furthermore, the decreasing rate of hepatic GSH content was also higher in bromobenzene pretreated rats than in normal rats. Moreover, the value of V$_{max}$ in hepatic GST was higher in bromobenzene pretreated rats than that in the normal rats. In conclusion, these results indicate that the pretreatment of bromobenzene may rather enhance the bromobenzene metabolism.

  • Sophorae Flos aqua-acupuncture solution(SFAS) was prepared and tested for the induction of quinone reductase and glutathione S-transferase activities and glutathione. SFAS significantly induced QR activity at the concentrations of $0.5{\times},\;1{\times}\;and\;3{\times}$ in cell culture. However, GST activity in murine Hepa 1c1c7 cells was slightly increased with SFAS. SFAS increased GSH levels.
