Synthesis and Characterization of Novel pH-Sensitive Hydrogels Containing Ibuprofen Pen dents for Colon-Specific Drug Delivery

  • The aim of this study was to develop novel intestinal specific drug delivery systems with pH sensitive swelling and drug release properties. The carboxyl group of ibuprofen was converted to a vinyl ester group by reacting ibuprofen and vinyl acetate as an acylating agent in the presence of catalyst. The glucose-6-acrylate-1, 2, 3, 4-tetraacetate (GATA) monomer was prepared under mild conditions. Cubane-1, 4-dicarboxylic acid (CDA) linked to two 2-hydroxyethyl methacrylate (HEMA) group was used as the crosslinking agent (CA). Methacrylic-type polymeric prodrugs were synthesized by the free radical copolymerization of methacrylic acid, vinyl ester derivative of ibuprofen (VIP) and GATA in the presence of cubane cross linking agent. The structure of VIP was characterized and confirmed by FTIR, $^1H$ NMR and $^{13}C$ NMR spectroscopy. The composition of the cross-linked three-dimensional polymers was determined by FTIR spectroscopy. The hydrolysis of drug polymer conjugates was carried out in cel-lophane membrane dialysis bags, and the in vitro release profiles were established separately in enzyme-free simulated gastric and intestinal fluids (SGF, pH 1 and SIF, pH 7.4). The detection of a hydrolysis solution by UV spectroscopy at selected intervals showed that the drug can be released by hydrolysis of the ester bond between the drug and polymer backbone at a low rate. Drug release studies showed that increasing the MAA content in the copolymer enhances the rate of hydrolysis in SIP. These results suggest that these polymeric prodrugs can be useful for the release of ibuprofen in controlled release systems.

  • An innovative biopolymer known as the Rhizobian gum has been developed in France, which shows some dramatic refreshing effect on the skin. The origin of this innovative project takes its source in the natural environment, and in particular the natural environment of the roots of sunflowers and wheat, where a symbiotic bacterium has been discovered. It is a Rhizobium bacterium, which is hosted by the roots, and which is able to synthesize a specific polymer showing a dramatic water binding capacity. This polymer is in particular synthesized in period of drought, and its biological role is to concentrate the small amount water present in the soil in order to take it available for the root, which becomes then able to absorb it. This vital mechanism allows the plant to survive despite a severe climatic environment. This basic research has been conducted in collaboration whit the French National centre of scientific Research (CNRS), and has lead to the isolation of the Rhizobium bacteria. Rhizobian gum is a branched biopolymer consisting in the repetition of a polysaccharide unit of 3 molecules of glucose, 3 molecules of galactose and 1 molecule of glucuronic acid, whit one pyruvate group an average 1.6 acetyl groups. The fresh effect of Rhizobian gum is a strong sensorial impact that 100 % of the consumers are able to perceive, and which is judged very pleasant by most of them. In addition to this, a large majority of consumers are perceived, and which is judge very pleasant by most of them. In addition to this, a large majority of consumers also feel a very pleasant relaxing sensation. Smoothness and softness are also felt by most consumers and qualified positively by most of them. These qualities guarantee a strong impact on today's consumers.

  • 레트로바이러스를 동결 건조과정에서 발생하는 물리적, 화학적인 스트레스로 인한 바이러스의 실활을 보완하기 위해 첨가하여 준 여러 가지 첨가물 중 trehalose가 가장 탁월한 protectant임을 알 수 있었고 최적 첨가농도는 300mM이었다. Trehalose의 역할을 보완하기 위하여 분자량이 큰 polymer들을 첨가 하였으나 trehalose만을 넣어 주었을 때 보다 poly(vinylpyrollidone)(PVP)의 경우 큰 영향은 볼수 없었고 dextran과 PEG를 첨가 하였을 경우엔 오히려 더욱 활성이 감소하였다.

Characterization of an Acidic Polysaccharide from Fruiting Bodies of Lyophyllum shimeji

  • An acidic polysaccharide H-III was extracted from fruiting bodies of Lyophyllum shimeji with hot water. Acid hydrolysis and gas chromatography analysis showed that the polysaccharide was almost exclusively composed of glucose with a very small amount of mannose and galactose. Uronic acid of 8.36% was also detected in H-Ill. Its molecular weight was estimated to be $1{\times} 10^6 Da.\;By\;^{13}C-NMR$ analysis, some repeating units of disaccharide were detected in the polymer H-III. The polysaccharide showed a strong mitogenic activity in a dose-dependent manner.

  • Anion-exchange porous hollow-fiber membranes with a thickness of about 1.2 mm and a pore size of about $0.30{\mu}m$ were used as a supporting matrix to immobilize cycloisomaltooligosaccharide glucanotransferase (CITase). CITase was immobilized to the membrane via anion-exchange adsorption and by subsequent enzymatic cross-linking with transglutaminase, the amount of which ranged from 3 to 110 mg per g of the membrane. The degree of enzyme multilayer binding was equivalent to 0.3 to 9.8. Dextran, as the substrate, was converted into seven- to nine-glucose-membered cycloisomaltooligosaccharides (CI-7, -8, and -9) at a maxi mum yield of $28\%$ in weight at a space velocity of 10 per hour during the permeation of $2.0(w/w)\%$ dextran solution across the CITase-immobilized porous hollow-fiber membrane.

Synthetic bio-actuators and their applications in biomedicine

  • The promise of biomimetic smart structures that can function as sensors and actuators in biomedicine is enormous. Technological development in the field of stimuli-responsive shape memory polymers have opened up a new avenue of applications for polymer-based synthetic actuators. Such synthetic actuators mimic various attributes of living organisms including responsiveness to stimuli, shape memory, selectivity, motility, and organization. This article briefly reviews various stimuli-responsive shape memory polymers and their application as bioactuators. Although the technological advancements have prototyped the potential applications of these smart materials, their widespread commercialization depends on many factors such as sensitivity, versatility, moldability, robustness, and cost.

  • Streptococcus mutans에 의한 sucrose로 부터 축적된 polysaccharide를 조사한 결과 1 균주이 따라 36시간 후 4.41mg/ml로 부터 1.86mg/ml사이의 polysaccharide가 축적되고 2 효소에 의해서도 10시간 후 2.36mg/ml에서 1.4mg/ml의 polysaccharide가 생성되고 3 생성된 polysaccharide는 통일형의 polysaccharide였다.
