활성산소로 손상된 골모세포에 대한 해백의 영향 (Effect of Alii Macrostemi Bulbus on Cultured Mouse Osteoblasts Damaged by Reactive Oxygen Species)

  • It has been demonstrated that oxidative stress of reactive oxygen species(ROS) may be a causative factor in the pathogenesis of bony disorder. The purpose of this study was to evaluate the oxidative stress of glucose oxidase(GO) in the cultured mouse osteoblasts and the protective effect of Alii Macrostemi Bulbus(AMB) on ROS-induced osteotoxicity. Toxic effect of GO and protective effect of AMB were carried out by colorimetric assay. 20mU/mi GO decreased cell viability dose-dependently, and AMB increased cell viability against GO-induced cytotoxicity in these cultures. From above the results. GO has toxic effect, and AMB is very effective on GO-induced osteotoxicity in cultured osteoblasts of neonatal mouse.

진간식풍탕 전탕액이 GO에 의해 손상된 배양 척수감각신경세포의 LDH 활성도에 미치는 영향 (Effects of Jingansikpung-tang Water Extract on LDH Activity of Cultured Spinal Sensory Neurons Damaged by GO)

  • To evaluate the effect of Jingansikpung-tang(JST) water extract on cultured mouse spinal sensory neuron which was inhibited by glucose oxidase(GO)-induced cytotoxicity, MTT and LDH activity assay were carried out after the cultured mouse spinal sensory neuron were pre-incubated with various concentrations of JST extract for 3 hours prior to exposure of GO. The results obtained were as follows: GO, a oxygen radical, decreased the survival rate of the cultured mouse spinal sensory neuron cells on MTT assay. JST water extract have efficacy of decreasing LDH activity increasing by GO in cultured mouse spinal sensory neuron. From above the results, it is concluded that JST water extract has marked efficacy as a treatment for the damages caused in the GO-mediated oxidative process.

흐름주입분석 기술에 의한 글루코우스와 전분의 온라인 모니터링 (On-line Monitoring of Glucose and Starch by a Flow Injection Analysis Technique)

  • 본 연구에서는 생물공정에서 주요 기질로 이용되는 글루코오스와 전분을 온라인 모니터링 하기 이하여 GOD, AMG를 이용한 흐름 주입분석(Flow Injection Analysis : FIA) 기술을 개발하였고, 효소활성 변화를 비교, 고찰하였다. 특히, epoxy 고분자 담체에 고정화된 COD-FIA와 AMG/GOD-FIA 장치의 성능을 조사하였고, FIA의 조작온도, pH, 운반용액의 첨가제, 염 그리고 각종 신진대사물질의 고정화된 GOD, AMG의 활성에 대한 영향을 소형 반응기내 글루코오스와 전분의 농도 변화를 온라인 모니터링하였다. GOD-FIA에 의한 온라인 모니터링 결과는 오프라인 분석과 비교적 잘 일치하였으며, AMG/GOD-FIA에의한 전분 농도의 온라인 모니터링은 단일 효소 반응기를 사용한 경우 두 개의 효소 반응기를 사용한 경우보다 더 효과적임을 알 수 있었다.

Highly Sensitive and Selective Glucose Sensor Realized by Conducting Polymer Modified Nanoporous PtZn Alloy Electrode

  • Platinum is a well known element which shows a significant electrocatalytic activity in many important applications. In glucose sensor, because of the poisoning effect of reaction intermediates and the low surface area, the electrocatalytic activity towards the glucose oxidation is low which cause the low sensitivity. So, we fabricate a nanoporous PtZn alloy electrode by deposition-dissolution method. It provides a high active surface and a large enzyme encapsulating space per unit area when it used for an enzymatic glucose sensor. Glucose oxidase was immobilized on the electrode surface by capping with PEDOT composite and PPDA. The composite and PPDA also can exclude the interference ion such as ascorbic acid and uric acid to improve the selectivity. The surface area was determined by cyclic voltametry method and the surface structure and the element were analyzed by Scanning Electron Microscope (SEM) and Energy Dispersive X-ray spectroscopy (EDX), respectively. The sensitivity is $13.5{\mu}A/mM\;cm^2$. It is a remarkable value with such simply prepared senor has high selectivity.

  • 다양한 아미노산과 당을 $90^{\circ}C$에서 반응시켜 생성한 마이얄반응 생성물(Maillard reaction product)은 쑥갓에서 추출한 폴리페놀산화효소(polyphenol Oxidase, PPO)를 강하게 저해하였다. 글리신(glycine)과 포도당을 함유한 반응물으로부터 MRP의 생성은 $90^{\circ}C$에서 반응시간에 따라 직선적으로 증가하였으나 글리신과 포도당의 양은 상대적으로 감소하였다. 반응액에서 MRP의 증가함에 따라 반응액은 쑥갓 PPO활성을 비례적으로 강하게 저해하였다. 실험에 사용한 아미노산과 당중에서 글루타민과 자일로스를 아미노산과 당으로 이용했을 경우에 생성된 MRP가 쑥갓 PPO의 활성에 강한 저해제로 작용하였다. 카테콜을 기질로 사용한 쑥갓 PPO의 마이클레스 상수(Michaelis Menten constant)는 22.0 mM이었으며, MRP는 쑥갓 PPO의 비경쟁적 저해제로 작용하였다.

Contents of $\beta$-Glucan in Various Cereals and Its Functional Properties

  • A soluble dietary fiber, $\beta$-glucan, contained in oat and barley has nutritional benefits such as hypocholesterolemic effects and influences blood glucose regulation. The contents of $\beta$-glucan in both cereals range from 3 to 7% with the exception of a certain barley genotype which contains up to 16% $\beta$-glucan. $\beta$-Glucan is distributed mainly in the cell walls of endosperm and the distal (bran) portion of kernel. Various procedures have been developed for increasing the extraction yield of $\beta$-glucan. Oat gum prepared with weak alkali extractionand alcohol proecipitation following protein removal usually contains 80% $\beta$-glucan.The most commonly used method for $\beta$-glucan quantitiation is an enzymatic procedure combining lichenase plus $\beta$-glucosidase followed by measuring the amount of glucos released by glucose oxidase-peroxidase treatment. The increase in foam-and emulsion-stabilizing capacity of $\beta$-glucan is due to the increase in viscosity of the aqueous phase. Therefore, $\beta$-glucan shows great potentials as a thickener and a stabilizer.

Diet of Red Ginseng-Cheonggukjang Improves Streptozotocin-induced Diabetes Symptoms and Oxidative Stress

  • Protective effects of cheonggukjang fermented with 20% red ginseng (RC) were observed in streptozotocin (STZ)-induced diabetic rats by measuring levels of blood glucose, serum lipid profiles, and hepatic reactive oxygen species generating and scavenging enzymc activities. RC diet was prepared by mixing with AIN-76 diet at the final concentration 2%, and it was fed to STZ-induced diabetic rats for 3 weeks. The RC dict was significantly improved body weight, feed efficiency ratio, levels of serum glucose, and serum and hepatic lipids in diabetes. The significantly elevated O type activity of xanthine oxidase in diabetes was also greatly decreased by the RC diet. The treatment of RC showed the improved hepatic glutathione s-transferase activities in the diabetic animals. The present study indicates that cheonggukjang fermented with red ginseng could ameliorate STZ-induccd diabetic symptoms such as aggravated blood glucose levels, serum lipid profiles, and even the conditions of oxidative stress.

  • 소결공정에 의하여 tertiary amine을 함유하는 hydroxypropyl methacrylate 고분자막을 제조하였다. 이 막의 함수율은 수용액의 pH가 감소함에 따라 증가하였다. 인슐린의 투과도 역시 수용액의 pH가 감소함에 따라 증가하였다. 이 막의 투과도는 소결 압력이 증가함에 따라 감소하였다. 이 막에 당산화효소를 고정화시킨 acryl amide 막을 부착하여 복합 matrix device를 제조하였다. 이 막의 인슐린 투과도는 glucose의 농도가 증가함에 따라 증가하였다.

  • A novel type of intelligent microcapsule reactor system was prepared. The reactor can recognize pH change in the medea and control reaction rate by itself. For the reactor system, acrylic acid (AA), N-isopropylacrylamide (NIPAM), and glucose oxidase (GOD) were selected as a pH-responsive device, a gating device according and a reaction device, respectively. Poly(NIPAM-co-AA) (P-NIPAM-co-AA) are known to change its hydrophilicity-hydrophobicity due to pH change. They were integrated in a core-shell microcapsule space. GOD was loaded inside the core space and the pores in the outside shell layer were filled with P-NIPAM-co-AA linear grafted chains as pH-responsive gates by plasma graft filling polymerization method. When P-NIPAM-co-AA gates are hydrophilic at high pH value, this microcapsule permits glucose penetration into the core space and GOD reaction proceeds. However, when P-NIPAM-co-AA gates are hydrophobic at low pH value, this microcapsule forbids glucose penetration and GOD reaction will not occur. The accuracy of this concept was examined.

  • Numerous studies have shown that ascorbic acid is an indispensable micronutrient required to maintain the physiological processes of different animals including most of fishes. L-ascorbyl-2-glucose (AA2G) is an $\alpha$-glucose conjugate of AA on the C-2 position and a newly found AA derivative front Kimchi, Korean traditional fermented vegetable food, AA2G s stable to ascorbate oxidase and heating, it can be effectively hydrolyzed in vitro by the rice seed $\alpha$-glucosidase.
