Antidiabetic Effect of Glechoma hederacea LINNAEUS in Streptozotocin-Induced Diabetic Rats (Streptozotocin 유발 당뇨 흰쥐에 대한 금전초 추출물의 혈당 강하 효과)

  • This study was done to investigate the antidiabetic effect of Glechoma hederacea LINNAEUS in Streptozotocin (STZ)-induced diabetic rats. Diabetes was induced by intravenous injection of STZ at a dose of 45 mg/kg dissolved in citrate buffer. The methanol extract of Glechoma hederacea was orally administrated once a day for 6 days. The contents of serum glucose, triglyceride (TG), total cholesterol and Atherogenic Index (Al) were significantly decreased, but high density lipoprotein (HDL)-cholesterol and HDL-cholesterol/total cholesterol ratio (HTR) were significantly increased in Glechoma hederacea treated STZ-sample group compared to the those of STZ-control group. The content of hepatic lipid peroxide and activity of catalase were decreased, but content of glutathione as well as activities of glutathione-S-transferase and superoxide dismutase were increased in Glechoma hederacea treated STZ-sample group compared to the those of STZ-control group. The content of hepatic glycogen and activities of glucose-6-phosphate dehydrogenase, glucokinase were significantly increased, but activity of glucose-6-phosphatase was decreased in Glechoma hederacea treated STZ-sample group compared to the those of STZ-control group. These results indicated that methanol extract of Glechoma hederacea would have antidiabetic effect in STZ-induced diabetic rats.

Antidiabetic Metabolism Effect on the water Extract of $Cordyceps$ $Militalis$ in Streptozotocin-Induced Diabetic Rats (밀리타리스 동충하초($Cordyceps$ $Militalis$)열수 추출물이 스트렙토 조토신으로 유발된 당뇨 흰쥐의 항 당뇨 대사에 미치는 영향)

  • This study was carried to investigate the antidiabetic metabolism effect of water extract $Cordyceps$ $Militalis$(C.M.) in Streptozotocin(STZ) induced diabetic rats. Diabetes were induced by intravenous injection of STZ at a dose of 42mg/kg,b.w. dissolved in citrate buffer. The water extract of C.M. was orally administrated once a day for 7 days at a dose of 500mg/kg,b.w(body weight). or 1,000mg/kg.b.w. The content serum glucose was significantly decreased in C.M. treated group compared to the those of STZ-control group. The content of hepatic glycogen and activities of glucose-6-phosphate dehydrogenase(G-6-PDH), glucokinase(GK) were significantly increased, but activity of glucose-6-phoshatase(G-6-Pase) was significantly decreased in C.M. treated group compared to the those of STZ-control group. These results indicated that water extract of C.M. would have antidiabetic metabolism effect in STZ-induced diabetic rats.

Antidiabetic Effect of Yacon(Smallanthus sonchifolius) of Root and Leaf in Streptozotocin-Induced Diabetic Rats (Yacon의 뿌리와 잎의 추출물이 streptozotocin으로 유발된 흰쥐의 항당뇨 효과)

  • This study was carried to investigate the antidiabetic effect of ethanol extract of Smallanthus sonchifolius(S.S) in Streptozotocin(STZ)- induced diabetic rats. Diabetes was induced by intravenous injection of STZ at a dose of 43mg/kg dissolved in citrate buffer. The ethanol extract of S,S in leaves and root were orally administrated once a day for 7 days at a dose of 1,000mg/kg. The contents of serum glucose, triglyceride(TG), total cholesterol were significantly decreased in S.S treated group compared to the those of STZ-control group. The content of hepatic glycogen and activity of glucokinase(GK) were significantly increased,and activity of glucose-6-phoshatase(G-6-Pase) was significantly decreased in S.S treated group compared to the those of STZ-control group, but activity of glucose-6-phosphate dehydrogenase(G-6-PDH) was not significantly increased, These results indicated that ethanol extract of S.S would have antidiabetic effect in STZ-induced diabetic rats.

Hypoglycemic effect of Rehmannie Radix Preparata (Sookjihwang) extract in streptozotocin-induced diabetic rats

  • Rhemannie Radix Preparata (RRP) has been previously employed in traditional oriental medicine as a treatment for diabetic thirst and improving blood flow. The aim of this study was to evaluate its hypoglycemic control by assaying the activities of key enzymes of carbohydrate metabolism in streptozotocin-(STZ)-induced diabetic rats. Further, RRP extracts were prepared in water (RRPW), in 50% ethanol (RRP50), and in 100% ethanol (RRP100), respectively, and compared for their actions in diabetic rats. The oral treatment of RRP (5 mg/kg b.w./d) to diabetic rats for 21 days resulted in a significant decline in blood glucose by 67% compared to diabetic control rats (P < 0.05). The altered activities of glucokinase, glucose-6-phosphate dehydrogenase (G6PD), 6-phosphogluconate dehydrogenase (6PGD), and acetyl CoA carboxylase (ACC) in the livers of diabetic rats were reversed significantly to near-normal levels by the administration of RRP (P < 0.05). Among the three RRP extracts, RRP100 was the most effective in terms of hypoglycemic action. However, the administration of RRP to diabetic rats did not improve insulin production. The modulatory effects of RRP100 on the attenuation of carbohydrate enzyme activities appear to hold promise for widespread use for the treatment of diabetes in the future.

Effects of Yanggyuksanhwa-tang on Diabetic Rats Induced by Streptozoticin (양격산화탕(凉膈散火湯)이 실험적 당뇨에 미치는 영향)

  • This study evaluated effects of Yanggyuksanhwa-tang(YST) on diabetic rats. Diabetic condition in rats was induced by streptozotocin injection. Then control effect of YST was observed with changes of serum glucose, insulin, and triglyceride levels and hepatic glucokinse activity. YST treatment was resulted significant decrease of high serum glucose level of diabetic rats induced by streptozotocin at 5th day of YST treatment. YST treatment was resulted increase of low serum insulin level of diabetic rats induced by streptozotocin, but it was not significant statistically. YST treatment was resulted significant decrease of high serum triglyceride level of diabetic rats induced by streptozotocin at 3rd and 5th of YST treatment. YST treatment was resulted significant increase of low hepatic glucokinase activity of diabetic rats induced by streptozotocin at 5th day of YST treatment. These results suggest that YST has effects on serum glucose control against cerebral inchemic damage under diabetic condition.

Antidiabetic and Antioxidative Effects of Lycii fructus in Streptozotocin-Induced Diabetic Rats (Streptozotocin으로 유발된 흰쥐에 대한 구기자 추출물의 혈당강하 및 항산화 작용에 미치는 효과)

  • This study was done to investigate the antidiabetic and antioxidant effects of Lycii fructus in Streptozotocin (STZ)-induced diabetic rats. Diabetes was induced by intravenous injection of STZ at a dose 45mg/kg.b.w. dissolved in citrate buffer(pH4.5). The ethanol extract of Lycii fructus was orally administrated once a day for 7 days. The contents of serum glucose, total cholesterol were significantly decreaed (p<0.05) in Lycii fructus treated group compared to the those of STZ-control group, also content of Triglyceride (TG), Altherogenic index (AI) were decreased, but not statistically significant. But high density lipoprotein (HDL)-cholesterol and HDL-cholesterol/total cholesterol ratio (HTR) were increased in Lycii fructus treated group compared to the those of STZ-control group. The content of hepatic glycogen and activities of glucose-6-phosphate dehydrogenase(G-6PDH), glutathione-s-transferase(GST), superoxide dismutase(SOD) were significantly increased(p<0.05), glucose-6-phos-phatase(G-6pase)was significantly decreased(p<0.05) and The glutathione(GSH), glucokinase(GK) were increased. But not statistically significant In Lycii fructus treated group compared to the those of STZ-control group These results indicated that ethanol extract of Lycii fructus would have antidiabetic and antioxidant effects in STZ-induced diabetic rats.

Study on the Hypoglycemic Action of the Fat Soluble Fraction of Panax ginseng C.A. Meyer in Streptozotocin Induced Diabetic Rats (인삼 지용성분획의 고혈당 강하작용에 관한 연구)

  • We attempted in this study to understand the hypoglycemic action of the fat soluble fraction of red ginseng roots in streptozotocin injected diabetic rats, through its actions on several enzymes relating to carbohydrate metabolism of the 1eve1 to compare with those of ginsenosides in streptozotocin injected diabetic rats. It was realized that the increased level of glucose, ketone bodies, lactate, nonesterified fatty acids and triacylglycerol in blood was significantly decreased and the decreased liver glycogen content of streptozotocin injected rats were appreciably moderated by intraperitoneal injection of the fat soluble fraction of red ginseng roots as shown in the saponin injected diabetic rats. The deceased activities of liver enzymes relating to carbohydrate metabolism such as phosphofructokinase, glucokinase, glucose-6-phosphate dehydrogenase, 6-phosphogluconate dehydrogenase and acetyl CoA carboxylase of streptozotocin induced diabetic rats were also sufficiently modified by the intraperitoneal injection of the above fat soluble fraction as shown in the ginsenoside injected streptozotocin induced rats.

High Food Efficiency Ratio of Prepubertal Growth Period Leads to a Long-Term Susceptibility for Obesity and Insulin Resistance in Obesity-Prone and Obesity-Resistant Sprague Dawley Rats

  • Excessive body weight gain during the growth period of early life may predispose individuals towards obesity and metabolic disorder in later life. We investigated the possibility of using the food efficiency ratio as an early indicator for predicting susceptibility to diet-induced obesity and insulin resistance. Four-week-old, prepubertal, male Sprague Dawley rats were divided into obesity-prone and obesity-resistant groups based on food efficiency ratio values after five days on a high-fat diet. Metabolic parameters measured after 2, 6, and 10 weeks, and specific phenotypes were compared with each group. Obesity-prone rats had higher increases in body weight and fat mass compared to obesity-resistant rats over the study period. Obesity-prone rats became glucose intolerant early in this study and remained so throughout the experimental period, with increases in fat weight and leptin levels occurring first, followed by increases in insulin level. Gluconeogenesis and insulin resistance significantly increased in obesity-prone groups in which activities of glucose-6-phosphatase and phosphoenolpyruvate carboxykinase were increased and glucokinase activity decreased. Higher food efficiency ratio at an early age was closely correlated with body fat accumulation, hyperleptinemia, and hyperinsulinemia of middle and elderly age. We suggest a high food efficiency ratio in prepubertal subjects may be a useful predictor of future obesity and insulin resistance.

Sargassum coreanum extract alleviates hyperglycemia and improves insulin resistance in db/db diabetic mice

  • BACKGROUND/OBJECTIVES: The goal of this study was to examine the effect of Sargassum coreanum extract (SCE) on blood glucose concentration and insulin resistance in C57BL-KsJ-db/db mice. MATERIALS/METHODS: For 6 weeks, male C57BL/KsJ-db/db mice were administrated SCE (0.5%, w/w), and rosiglitazone (0.005%, w/w). RESULTS: A supplement of the SCE for 6 weeks induced a significant reduction in blood glucose and glycosylated hemoglobin concentrations, and it improved hyperinsulinemia compared to the diabetic control db/db mice. The glucokinase activity in the hepatic glucose metabolism increased in the SCE-supplemented db/db mice, while phosphoenolpyruvate carboxykinase and glucose-6-phosphatase activities in the SCE-supplemented db/db mice were significantly lower than those in the diabetic control db/db mice. The homeostatic index of insulin resistance was lower in the SCE-supplemented db/db mice than in the diabetic control db/db mice. CONCLUSIONS: These results suggest that a supplement of the SCE lowers the blood glucose concentration by altering the hepatic glucose metabolic enzyme activities and improves insulin resistance.

Ethanol Effect of Nelumbo nucifera Root on Carbohydrate Methabolism Related Enzyme Activities and Antioxidative Effect in Streptozotocin-Induced Diabetic Rats (Streptozotocin으로 유발된 당뇨 흰쥐에 대한 연근 에탄올 추출물의 당대사 효소활성과 항산화 작용에 미치는 영향)

  • This study was done to investigate the carbohydrate metabolism related enzyme activities and antioxidative effects of Nelumbo nucifera(N.N) Root in streptozotocin (STZ)-induced diabetic rats. The contents of serum glucose, triglyceride (TG) and Total cholesterol were significantly decreaed in N.n treated group compared to the those of STZ-control group. The activity of glucose-6-pase(G-6-Pase) was significantly decreased in N.N treated group. Also the activity of glucokinase(Gk) was significantly increaed in N.N treated group. The content of hepatic glycogen was significantly increaed in N.N treated group, in addition, content of malondialdehyde(MDA) was significanly decreased in N.N treated group. Also, content of glutathione(GSH) was significanly increased in N.N treated froup. whereas, activity of catalase(CAT) was significantly decreaed in N.N treated group compared to the those of STZ-control group. activity of glutathione peroxidase(GSH-Px) was inecreaed. In conclusion, these results indicated that ethanol extract of N.N would have carbohydrate metabolism antioxidative effects in STZ-induced diabetic rats.
