Antidiabetic Effect of Ethanol Extract of Liriope platyphylla in Streptozotocin Induced Diabetic Rats (Streptozotocin으로 유발된 당뇨쥐에서 맥문동 에탄올 추출물의 당뇨개선효과)

  • The ethanol extraction yield of Liriope platyphylla(Lp) was about 30.7% by extract apparatus. This study was done to investigate the carbohydrate metabolism related enzyme activities effects of Lp in streptozotocin (STZ)-induced diabetic rats. The contents of serum glucose, triglyceride (TG) were significantly decreaed in Lp treated group compared to the those of STZ-control group, also content of Total cholesterol was decreased. High density lipoprotein (HDL)-cholesterol was increased in Lp treated group. The activity of glucose-6-pase(G-6-Pase) was significantly decreased in Lp treated group. Also the activity of glucokinase(Gk) was increaed in Lp treated group. The content of hepatic glycogen was significantly increaed in Lp treated group. These results indicated that ethanol extract of Lp would have antidiabetic effect in STZ-induced diabetic rats.

Antidiabetic effect of ethanol extract on Codonopsis lanceolata root in streptozotocin induced diabetic rats (더덕 뿌리 에탄올 추출물이 streptozotocin으로 유발된 흰쥐의 항 당뇨효과)

  • This study was done to investigate the antidiabetic effect of ethanol extract from Codonopsis lanceolata root in Streptozotocin (STZ)-induced diabetic rats. Diabetes was induced by intravenous injection of STZ at a dose 45mg/kg.b.w. dissolved in citrate buffer(pH4.5). The ethanol extract of Codonopsis lanceolata root was orally administrated once a day for 7 days. The contents of serum glucose, triglyceride(TG) and total cholesterol were significantly decreased(p<0.05) in Codonopsis lanceolata root treated group compared to the those of STZ-control group. Also the content of hepatic glycogen and activities of glucose-phosphate dehydrogenase(G-6-PDH) and glucokinase(GK) were significamtly increased(p<0.05). These results indicated that ethanol extract of Codonopsis lanceolata root would have antidiabetic effect in STZ-induced diabetic rats.

Increased glucose metabolism and alpha-glucosidase inhibition in Cordyceps militaris water extract-treated HepG2 cells

  • BACKGROUND/OBJECTIVES: Recent living condition improvements, changes in dietary habits, and reductions in physical activity are contributing to an increase in metabolic syndrome symptoms including diabetes and obesity. Through such societal developments, humankind is continuously exposed to metabolic diseases such as diabetes, and the number of the victims is increasing. This study investigated Cordyceps militaris water extract (CMW)-induced glucose uptake in HepG2 cells and the effect of CMW treatment on glucose metabolism. MATERIALS/METHODS: Colorimetric assay kits were used to determine the glucokinase (GK) and pyruvate dehydrogenase (PDH) activities, glucose uptake, and glycogen content. Either RT-PCR or western blot analysis was performed for quantitation of glucose transporter 2 (GLUT2), hepatocyte nuclear factor 1 alpha ($HNF-1{\alpha}$), phosphatidylinositol 3-kinase (PI3k), protein kinase B (Akt), phosphorylated AMP-activated protein kinase (pAMPK), phosphoenolpyruvate carboxykinase, GK, PDH, and glycogen synthase kinase 3 beta ($GSK-3{\beta}$) expression levels. The ${\alpha}-glucosidase$ inhibitory activities of acarbose and CMW were evaluated by absorbance measurement. RESULTS: CMW induced glucose uptake in HepG2 cells by increasing GLUT2 through $HNF-1{\alpha}$ expression stimulation. Glucose in the cells increased the CMW-induced phosphorylation of AMPK. In turn, glycolysis was stimulated, and glyconeogenesis was inhibited. Furthermore, by studying the mechanism of action of PI3k, Akt, and $GSK-3{\beta}$, and measuring glycogen content, the study confirmed that the glucose was stored in the liver as glycogen. Finally, CMW resulted in a higher level of ${\alpha}-glucosidase$ inhibitory activity than that from acarbose. CONCLUSION: CMW induced the uptake of glucose into HepG2 cells, as well, it induced metabolism of the absorbed glucose. It is concluded that CMW is a candidate or potential use in diabetes prevention and treatment.

Effects of Cordyceps militaris on Key Enzymes of Carbohydrate Metabolism (Cordyceps militaris 성분이 당대사 관련 효소 활성에 미치는 영향)

  • The present study was designed to investigate the antidiabetic effect of Cordyceps militaris on activities of hepatic glucose-regulating enzymes, such as glucokinase (GK), pyruvate dehydrogenase (PDH) and acetyI-CoA carboxylase (ACC). We have isolated the active compounds, CM-A and CM-B from C. militaris and the extracts are under investigation to determine their mechanism of action. Hepatic GK, PDH and ACC activities were significantly (p < 0.05) increased compared with the control. Treatment with CM-A led to a rise in percentage of enzymes by 380$\%$,396$\%$ and 286$\%$, respectively, relative to control levels. CM-B were 329$\%$, 312$\%$ and 239$\%$, respectively. The increase in GK and ACC activities was linearly proportional with increased ratio of CM-A. Our findings suggest that C. militaris exerted antihyperglycemic potency, which is thought to be mediated through activation of GK, PDH and ACC enzymes related to glucose metabolism directly or indirectly and therefore C. militaris is promising as antidiabetic functional foods.

The Effect of Platycodon grandiflorum Root Ethanol Extract on Blood Glucose, Lipid, Activities of Carbohydrate Metabolism Related Enzyme in Streptozotocin-Induced Diabetic Rats (도라지 뿌리 에탄올 추출물이 streptozotocin으로 유발된 흰쥐의 혈당지질, 당대사에 미치는 영향)

Antidiabetic Effect of Ethanol Extract on Astragali Radix (황기 에탄올 추출물의 항 당뇨 효과)

  • This study was carried to investigate the antidiabetic effect of ethanol extract of Astragali Radix(A.R) in Streptozotocin(STZ) induced diabetic rats. Diabetes was induced by intravenous injection of STZ at a dose of 45mg/kg dissolved in citrate buffer. The ethanol extract of A. R was orally administrated once a day for 7 days at a dose of 1,000mg/kg. The contents of serum glucose, triglyceride(TG), total cholesterol were significantly decreased in A.R treated group compared to the those of STZ-control group. The content of hepatic glycogen and activities of glucokinase(GK) and glucose-6-phosphate dehydrogenase(G-6-PDH) were significantly increased, and activity of glucose-6-phoshatase(G-6-Pase) was significantly decreased in A.R treated group compared to the those of STZ-control group, These results indicated that ethanol extract of A.R would have antidiabetic effect in STZ-induced diabetic rats.

  • Matrix Assisted Laser Desorption ionization Time of Flight (MALDI-TOF) was used for enzymes identification related to B -1,3-glucan synthesis. Agrobacterium sp. ATCC31750 was cultivated with two stage Continuous Stirrer Tank Reactor (CSTR) and the cells were harvested and their protein profiles were analysed by two dimensional electrophoresis. The specific enzyme spot was treated with trypsin and ana lysed by MALDI-TOF to get peptide molecular weight. The peptide molecular weights were matched with Agrobacterium tumefacience's Data Base from the matrix science site, then could identify the avaliable key enzymes. In this study, we identified key metabolite of synthesis of beta-1,3-glucan, such as glucose-6-phosphate isomerase, phosphoglucomutase, B-1,3-glucan synthase and glucokinase, and we also identified uracil phosphoribocyl transferase and Ribosome recycling factor also.

호염성 세균의 생리적 특성

  • 국내 염장식품 및 염전으로부터 세균을 분리하여, 호염성 세균의 NaCl 농도에 따른 성장범위, 생리적 및 효소학적 특성을 조사하고자 했다. 염전으로부터 NaCl 20%배지에서 14주와 총 16종류의 젓갈류에서 NaCl 10% 배지로 56균주의 호염성 세균을 분리하여 0, 5, 10, 15, 20, 25% NaCl농도에서 성장률을 조사하고 최적온도 및 배지조성과 함께 동정에 필요한 생리실험을 하였다. 또한 세포의 효소로서 Lactate dehydrogenase, Glucokinase, Glucose-6-phosphate dehydrogenase, Alanine dehydrogenase, Isocitrate dehydrogenase 등의 특성도 조사하였다. 선별한 균주중 Acinetobacter sp, 등이 관찰 조사되었으며 최적 성장 NaCl농도는 10%이고, 최적온도는 3$0^{\circ}C$이며, 25% NaCl, 45$^{\circ}C$에서 자란 Halobacterium sp. 등이 분리되었다. 그중 Acinetobacter strain H6는 단백분해효소와 탄수화물 분해효소의 생성능이 15>10>20% NaCl순이며, 특히 Lactate dehydrogenase 활성은 2>3>1>OM NaCl 순으로 나타났고, NaCl 대신 KCl을 사용했을 때는 3>2>1> OM순으로 활성이 나타났다.

  • This study was performed to investigate the antidiabetic study effect in ethanol extract of Ailanthicortex(A.C) radici in Streptozotocin(STZ)-induced diabetic rats. Diabetes was induced by intravenous injection of STZ at a dose of 45mg/kg dissolved in citrate buffer. The ethanol extract of A.C radici was orally administrated once a day for 7 days at a dose of 500mg/kg. The content of serum glucose, triglyceride(T.G), total cholesterol were significantly decreased in treated group compared to the those of STZ-control group. The content of hepatic glycogen and activity of glucokinase(GK) were significantly increased in treated group compared to the those of STZ-control group. but activity of glucose-6-phosphate dehydrogenase(G-6-PDH) was significantly increased. These results indicated that ethanol extract of A.C would have antidiabetic effect in STZ-induced diabetic rats.

Antidiabetic Effect of Ha-Su-O(polygoni radix) (하수오 추출물의 혈당강하작용)

Diabetes was induced by intravenous injection of STZ at a dose of 45mg/kg dissolved in citrate buffer. The ethanol extract of polygoni radix was orally administrated once a day for 7 days. The content of serum glucose, triglyceride(TG), total cholesterol were significantly decreased in polygoni radix treated STZ-sample group compared to the those of STZ-control group. The content of hepatic glycogen and activities of glucose-6-phosphate dehydrogenase(Glucose-6-PDH), glucokinase were significantly increased, but activity of glucose-6-phoshatase was decreased in polygoni radix treated STZ-sample group compared to the those of STZ-control group. These results indicated that ethanol extract of polygoni radix would have antidiabetic effect in STZ-induced diabetic rats.
