The formation of osmiophilic globules related to the granal lysis has been investigated with a shade plant ginseng (Panax ginseng C. A. Meyer) exposed to full sunlight. The changes of chloroplast were examined as a function of time over 9 days under full sunlight exposure. The ultrastructure of ginseng leaf showed swelling of the granal thylakoid during an early stage of the light exposure. The thylakoid membrane faded and small electron-opaque dots were aggregated on the edges of the granal thylakoid membrane when the exposure time was increased over 1 day. Then, the sahpe of the grana changed into round. After the exposure over 3 days, there appeared many osmiophilic globules with multi-lamellated concentric structure. The globules at this stage were partly accumulated with osmiophilic substances. The outermost membrane of these multi-lamellated osmiophilic globules was attached to the stromal thylakoid membrane connecting to the deforming grana. The osmiophilic globules were elongated after 9 days. In this stage, the multi-lamellated structure was difficult to identify due to severe accumulation of osmiophilic substances. The number of the osmiophilic globules also increased along with the full sunlight exposure time. This observation leads us to believe that the multi-lamellated osmiophilic globules came from the deformation of grana.
Effect of growth light intensity (5% and 20%) and planting density (40, 56, 90/33㎡) on yield and red ginseng quality was investigated in a sandy loam field of ginseng plantation. High growth light intensity had no effect on yield and quality but decreased stem length, leaf area and chlorophyll content and increased specific leaf weight. The optimum planting density was 64/3.3㎡ for both yield and Heaven + Earth grade (HE grade) of red ginseng. HE-shape ginseng rate showed significant positive correlation (P=0.0l) with HE grade rate.
This study was investigated and analyzed the physiological reaction mechanisms and the factors of the chlorophyll bleaching phenomenon on leaf burning-disease of the Ginseng (Panax ginseng C.A. Meyer). Chlorophyll bleaching phenomenon was mainly caused by the photooxidation of singlet oxygen and the autooxidation of hydrogen peroxide($H_2O_2$) accumulation resulted from inactivation of catalase and peroxidase. Chlorophyll bleaching phenomenon was remarkably accelerated by addition of saponin.
The experiment was conducted to obtain basic information on breeding of flue-cured tobacco varieties. Nine cultivars and partial diallel set of 36 F1 hybrides were grown at Taegu Experiment Station, Korea Ginseng & Tobacco Research Institute in 1983. Estimated heritability in the narrow-sense ranged from 66.21% to 94.12% for yield, leaves per plant, days to flower, leaf weight, leaf width, leaf shape, nicotine content and reducing sugar content, while that for stalk height, leaf length, midrib weight and midrib width ranged from 28.12% to 56.25%. The genotypic correlations were positive among yield, leaf weight, leaf length, leaf width, midrib weight and midrib width in the middle leaves, days to flower, nicotine content and reducing correlated with leaf length than leaf width. At the path coefficient analysis of the effect of leaf and midrib characters upon yield according to stalk position, leaf weight, leaf width and midrib width in the middle leaves, and leaf weight, and leaf width in the top leaves showed high direct effects.
Panax ginseng leaf tea was developed for the functional benefit of health, preference and convenience. The leaves of 4-year-old ginseng were selected in July and August. The ginseng leaf was treated by three methods : heat processed tea(HPT), aged tea(AGT) and hot-air dried tea(DRT). The contents and compositions of their crude saponin of ginseng leaves were measured. 1. The content of crude saponin of HPT was the higher than other treatments. The content of HPT was 18.72∼18.82%, ACT 18.24∼18.29% and DRT 17.02∼17.17%. 2. The harvest time and treatment methods were not affect the composition of ginsenoside in ginseng leaf tea. The ginsenoside-Re was shown the highest value as 1.97∼2.15. And ginsenoside-Rd was 1.48∼1.79, -Rg$_1$ 1.33∼1.58 and -Rb, -Rb$_2$, -Rc in the order. 3. The content of protopanaxadiol(PD) and protopanaxatriol(PT) was shown that DRT was 1.11∼1.13, HPT 1.09~l.12 and AGT 0.92∼1.02. The content of PD and PT were shown similar result at any harvest time. 4. The contents of crude saponin extracted by hot-water at 5 min was the higher ratios in HPT and harvested in July than other treatments. The content of crude saponin of ginseng leaf harvested in July was 15.88% and HPT was 16.88%. The order of contents of ginsenoside were -Re, -Rd, -Rg$_1$, -Rb$_1$, -Rb$_2$, and - Rc. The extraction ratio of crude saponin extracted by the circulated extraction method in 8 hours and 5 min extraction were 81.74∼84.38%. And HPT of ginseng leaf harvested in July was the highest value 84.3% but the extraction ratio of ginsenoside was 78.00~88.13%. But the extraction ratio of ginsenoside was similar trend in all treatments.
The production of ginseng cultivation is decreasing due to shortage of cultivated land and climate change, so additional methods are needed. Therefore, the physiological and morphological characteristics of finebubble were analyzed by applying them to ginseng cultivation which is the representative crop of Korea. The application of fine bubble water to 2 year old ginseng showed that stem length and leaf were increased, and weight increased as main root increased in root. In particular, it is shown that the weight of roots increases with the increase of leaf size. This is because the total chlorophyll content is high and it is judged to be related to the increase of photosynthesis efficiency. When the results of this study were confirmed, it was confirmed that ginseng cultivation using fine bubbles showed no inhibition of growth of ginseng. According to the results of physiological characteristics and subdividing results, It was confirmed that the growth of about 10% to 15% of the growth was increased and that the growth of ginseng was increased by applying the fine bubble when growing ginseng.
The aim of this study is to evaluate the effects of organic materials on soil organisms. Changes in the community structure, and population density of soil organisms (microbes, nematodes, and microarthropods) were studied in a Korean ginseng field. Phospholipid fatty acids analysis showed that the relative abundances of bacteria, fungi, and actinomycetes did not differ significantly. The aerobes/anaerobes ratio was the lowest in soils amended with leaf mold, indicating that the decomposition speed was slow. Further, the addition of leaf mold to the soil enhanced the saturated/monounsaturated fatty acid ratio and cyclopropyl fatty acid/precursor ratio, which indicated an increase in environmental stresses. Application of pig manure compost (PMC) had positive effects on the population density of nematodes, and negative effects on that of oribatid mites. The population densities of nematodes, and microarthropods remained relatively low in the plots that had been treated with leaf mold or pig manure compost. It is suggested that pre-planting soil management directed at enhancing the biological decomposition efficiency should be continued over a long period to increase the soil bioactivity in virgin soils.

Ginseng has been widely used in Korea as a natural medicine due to its saponin contents. Although the total amount of ginseng stem and leaf saponins (GSLS) is 4~5 times higher than that of saponin in the root, the root is mainly used. This is due to two reasons: nervous system-stimulant activity of GSLS and pesticide residues in GSLS. In this study, residual agricultural pesticides were removed from GSLS using two types of bacterial treatments. Two GSLS treatment groups of chickens (GSLS-1 and GSLS-2) were established. The chickens were fed 0.4% GSLS-1 or GSLS-2 mixed with crop. We then evaluated the effects of GSLS on bodyweight and several immune parameters. At the end of the experiments, chickens fed GSLS-1 and red ginseng saponin had significantly higher growth rates (16.6% and 8.0%, respectively) compared to the vaccine control group treated with Noblis Salenvac-T. The group fed GSLS-1 also had the highest IgG titer that was significantly different at the end of experiments compared to the other groups. These findings imply that GSLS-1 is a good candidate feed additive for the chicken industry.
This study evaluated the protective effects of ginseng leaf extract (GLE) against high fat-diet-induced hyperglycemia and hyperlipidemia, and explored the potential mechanism underlying these effects in C57BL/6J mice. The mice were randomly divided into four groups: normal control, high fat diet control (HFD), GLE-treated at 250 mg/kg, and GLE-treated at 500 mg/kg. To induce hyperglycemic and hyperlipidemic states, mice were fed a high fat diet for 6 weeks and then administered GLE once daily for 8 weeks. At the end of the treatment, we examined the effects of GLE on plasma glucose, lipid levels, and the expression of genes related to lipogenesis, lipolysis, and gluconeogenesis. Both GLE groups lowered levels of plasma glucose, insulin, triglycerides, total cholesterol, and non-esterified fatty acids when compared to those in HFD group. Histological analysis revealed significantly fewer lipid droplets in the livers of GLE-treated mice compared with HFD mice. To elucidate the mechanism, Western blots and RT-PCR were performed using liver tissue. Compared with HFD mice, GLE-treated mice showed higher levels of phosphorylation of AMP-activated protein kinase (AMPK) and its substrate, acetyl-CoA carboxylase, but no differences in the expression of lipogenic genes such as sterol regulatory element-binding protein 1a, fatty acid synthase, sterol-CoA desaturase 1 and glycerol-3-phosphate acyltransferase. However, the expression levels of lipolysis and fatty acid uptake genes such as peroxisome proliferator-activated receptor-$\alpha$ and CD36 were increased. In addition, phosphoenolpyruvate carboxykinase gene expression was decreased. These results suggest that GLE ameliorates hyperglycemia and hyperlipidemia by inhibiting gluconeogenesis and stimulating lipolysis, respectively, via AMPK activation.

Background: Abnormal activation of matrix metalloproteinases (MMPs) plays an important role in UV-induced wrinkle formation, which is a major dermatological problem. This formation occurs due to the degeneration of the extracellular matrix (ECM). In this study, we investigated the cutaneous photoprotective effects of Ultraflo L treated ginseng leaf (UTGL) in hairless mice. Methods: SKH-1 hairless mice (6 weeks of age) were randomly divided into four groups (8 mice/group). UTGL formulation was applied topically to the skin of the mice for 10 weeks. The normal control group received nonvehicle and was not irradiated with UVB. The UV control (UVB) group received nonvehicle and was exposed to gradient-UVB irradiation. The groups (GA) receiving topical application of UTGL formulation were subjected to gradient-UVB irradiation on $0.5mg/cm^2$ [GA-low (GA-L)] and $1.0mg/cm^2$ [(GA-high (GA-H)] of dorsal skin area, respectively. Results: We found that topical treatment with UTGL attenuated UVB-induced epidermal thickness and impairment of skin barrier function. Additionally, UTGL suppressed the expression of MMP-2, -3, and -13 induced by UVB irradiation. Our results show that topical application of UTGL protects the skin against UVB-induced damage in hairless mice and suggest that UTGL can act as a potential agent for preventing and/or treating UVB-induced photoaging. Conclusion: UTGL possesses sunscreen properties and may exhibit photochemoprotective activities inside the skin of mice. Therefore, UTGL could be used as a potential therapeutic agent to protect the skin against UVB-induced photoaging.
