Effect of ionizing radiation on cultured submandibular gland (SG) cells of mouse (전리 방사선 조사에 의한 마우스 배양 악하선 세포의 변화)

  • The present study was undertaken to investigate effects of ionizing radiation on DNA synthesis and chromosomal abnormality in cultured submandibular gland(SG) cells. SG cells from C57BL/6N Crj mice were cultured in Dulbecco's modified Eagle's medium (DME) supplemented with $10\%$ fetal bovine serum, antibiotics and fungizone. The cultured SG cells were irradiated with graded doses of gamma ray ($^{60}Co$) at a dose rate of 58.4rad/min. The effect of irradiation of $^{60}Co$ on DNA synthesis in cultured cells was evaluated by measuring the incorporation of 3H-TdR. Using conventional chromosome techniques and Giemsa staining methods, chromosomal abnormalities in cultured SG cells, induced by irradiation of $^{60}Co$ werw examined. Cytological observations were carried out by a light microscope with high resolving power. The results obtained were as follows : 1. DNA synthesis of SG cells was quantitatively dependent on a radiation dose compare to control. 2. A polyploids and few chromosome-type break, such as single and double breaks, deltions and triradial figures were more predominantly in irradiated SG cells than in control. This increase of chromosomal abnormality was in the proposition to the irradiation doses.

  • Cryptosporidium, a coccidian protozoa, commonly causes a self-limiting diarrheal illness in humans and animals. Fecal samples from various animals in Chonbuk district were observed using Sheather's aoatation technique, Kinyoun's modified acid-fast staining, and osmic acid pre-fixed Giemsa staining. The oocysts were detected in 74 cages(29.6%) out of 250 cages of mature mice, 26 (13.3%) out of 195 mature house rats, 75 (15.0%) out of 4-week-old 500 fowls, 98(19.9%) out of 6 to 8-month-old 500 pigs, and 111(22,2%) out of 2 to 5-year-old 500 dairy cattle, respectively. The degree of prevalence was slight in general, but actual prevalence was higher than infection rate because the detection rates were higher in repeated-preparation examinations in comparison to the first examination. Meanwhile, large and small types of oocysts were detected from mice, house rats, pigs, and cattle, and midium type from fowls.

  • The induction of polyhedroses in the silkworm, Bambyx mari L., was investigated treating the 5th instar larvae just after eodysis with high temperature (hot water bath at 40$^{\circ}C$ for 5 minutes or dry heat shock at 40$^{\circ}C$ for 30 minutes) and low temperature (5$^{\circ}C$ for 24 hours). The results obtained were as follows; 1. Comparing between the frequency of nuclear and cytoplasmic polyhedroses induced by cold and heat treatments (hot water bath at 40$^{\circ}C$ for 5 minutes), the induction ratio of the former is clearly less than that of the latter. But if the larvae tested with cold were left at room temperature (25$^{\circ}C$) for 30-120 minutes till the next hot water bath (40$^{\circ}C$) for 5 minutes and water bath (20$^{\circ}C$) for 5 minutes, treatments, the frequency of induced cytoplasmic polyhedrosis was more than that in the case of cold or hot water bath treatment alone. 2. The frequency of nuclear and cytoplasmic polyhedrosis induced by cold and successive heat (dry heat shock at 40$^{\circ}C$ for 30 minutes), left at room temperature (25$^{\circ}C$) ti11 the second treatment, the frequency of nuclear polyhedrosis was less than that of cytoplasmic polyhedrosis. 3. The reaction of nuclear polyhedra to stains also differs sharply from that of the cytoplasmic type. In a smear of nuclear polyhedra on a slide staining with Giemsa solution remains unstained against a stained back ground, in contrast to this, the cytoplasmic polyhedra take up stain readly.

Methodology of Chromosome Preparation and Banding Analysis in Gallus domesticus (닭 염색체의 분리 분석 방법에 관한 연구)

  • The purpose of this paper to present morphological normal chick chromosomes and develope avian cytogenetic techniques including chromosome preparation and banding technique. The early chick embryos provide a consistent source of material with hish mitotic cells. Although chick embryo tissue gives excellent preparations, the 4-5 days embryo is somewhat incovenient materials, Most imp of ant for avian Chromosome analysis are the technical protocols to achieve adequate preservation, spreading, and staining of the full chromosome complement. To precise chromosome analysis, pro-metaphase states are required. Best results of banding will be obtained from air dried slides prepared from early chick embryos that have been aged at least 1 week. Good G-banding differentiation is achieved by adequate trypsin digestion fellowed by staining in Goe,sa dye. The results of C-banding is influenced by many factors including the conditions of Ba(OH)$_2$, HCl treatment, and state of rinsing. In addition to precisely interprets banding patterns, the densitometric analysis is recommended.

3-Hydrogenkwadaphnin Induces Monocytic Differentiation and Enhances Retinoic Acid-mediated Granulocytic Differentiation in NB4 Cell Line

  • Recently, we have reported that 3-hydrogenkwadaphnin (3-HK), a diterpene ester isolated from Dendrostellera lessertii (Thymealeaceae), is very effective against leukemia cell lines without any detectable effects on normal cells (Moosavi et al., 2005b). In this study, we report that 3-HK induces $G_1$ cell-cycle arrest, differentiation and apoptosis in APL NB4 cell line. Indeed, the drug between 24 to 96 h induced 7-65% growth inhibition of NB4 cells. Cell viability was also decreased by 2-55% between 24 to 96 h treatments with the drug, respectively. These effects of the drug were also dose-dependent. According to flow cytomtry results, 3-HK (15 nM) induced a significant G1-arrest up to 24 h which was consequently followed with appearance of sub-$G_1$ peak at 72 to 96 h. Hoechst 33258 staining and DNA fragmentation assays confirmed the occurrence of apoptosis among the treated cells. On the other hand, NBT reducing assay, Wright-Giemsa staining, phagocytic activity and expression of cell surface markers (CD11b and CD14) confirmed that the inhibition of proliferation is associated with differentiation especially toward macrophage-like morphology. Interestingly, 3-HK at 5 and 10 nM enhanced the effects of all-trans retinoic acid (ATRA) in NB4 cells. Based on these results, 3-HK might become an ideal candidate for treatment of APL patients pending full exploration of its biological functions.

  • As a series of systematic classification for Korean common liver fluke, Fasciola sp., karyotype was investigated by means of the modified air-drying technique and of the regular Giemsa staining. Also, C-staining method was applied for detailed karyological analysis from the germ cells of the fluke. The following is a brief summary of the leading facts gained through the experiment. 1. Korean Pasciola sp. was classified into three types based on their chromosomal complements; individuals with 20 or 30 chromosomes and with a 20/30 mosaic constitution. Worms having 30 chromosomes represent a triploid form with 3 sets of 10 basic chromosomes, while those with 20 chromosomes were diploid and mosaic individuals were 2n/3n mixoploid. 2. The frequency of the individual type calculated is as follows; 67.45% of 212 flukes examined was of diploid, 10.85%, triploid, and the rest, 21.7%, mixoploid, respectively. In many cases, two or three types were found in the peculiar bovine host while single type inhabitant was about 20% out of 52 cases. 3. The twenty chromosomes consisted of 1 pair of large metacentrics, 4 pairs of medium-sized subtelocentrics, and 5 pairs of small submetacentrics, while constitution of the thirty chromosomes was nearly interpreted as a triploid form with 3 sets of 10 basic chromosomes. The high centromeric indexes of both types are the first Pairs among all the examined, and 37.93% was of diploid and 47.93%, triploid, respectively. 4. In mixoploid individuals, constitution of the chromosomes of diploid or triploid cells was the same as that of diploid or triploid individuals. 5. All the chromosomes of the germ cells in both types showed C-band around the centromeric region and especially the chromosomes no's 3,7, and 8 showed a remarkable C-band distinguished from other chromosomes. 6. The variance for the sizes of the worms and the eggs were not parallel with three different genotypes in Korean common liver fluke.

Effects of Peppermint Oil on Apoptosis of Astrocytes (신경교(神經膠) 성상세포(星狀細胞)의 세포자감사(細胞自減死)에 있어서 박하오일의 효과(效果))

  • Recently, essential oils are used for aromatherapy. Most essential oils are said to be anti-bacterial; some may be anti-viral or anti-fungal. I investigated the effects of peppermint pure essential oil on the heat shock-induced apoptosis in human astrocyte cell line CCF-STTGI. In previous studies, heat shock has been reported to induce the apoptosis or programmed cell death through the activation of caspase-3. We studied the heat shock-induced apoptosis through flow cytometry, DNA electrophoresis, and giemsa staining. Interestingly, these events were inhibited by pretreatment of peppermint pure essential oils in CCF-STTGl cells. Peppermint oil also inhibited the heat shock-induced apoptosis in primary cultured rat astrocytes. In addition, this Peppermint essential oil inhibited the heat shock-induced activation of caspase-3. These results suggest that peppermint pure essential oils may modulate the apoptosis through the activation of the interleukin-I -converting enzyme-like protease.

  • The possibility of strain improvement of cellulolytic fungus, Penicillium verruculosum via protoplast fusion was investigated. The cellulolytic activities of the six fusants, finally selected for their hyper-cellulolytics were 2 times of those of wild type and 1.2 to 4.4 times of those parental auxotrophs. It was confirmed that the nuclear fusion occurred in fusants by their DNA contents and nuclear staining with Giemsa. It was also found that the fusants were aneuploids, and their genetic stability was demonstrated from the subculture for four months.

Inhibitory Effect of Apoptosis of Human Astrocytes by Juniper Oil (신경교(神經膠) 성상세포(星狀細胞)에서 쥬니퍼오일에 의한 세포자멸사(細胞自滅死)의 억제(抑制) 효과(效果))

  • In previous studies, heat shock has been reported to induce the apoptosis or programmed cell death through the activation of caspase-3. 1 investigated the effect of juniper pure essential oil on the heat shock-induced apoptosis in human astrocyte cell line CCF-STTGI. Treatment of the astrocytes with heat shock markedly induced apoptotic cell death. However, pretreatment of the astrocytes with juniper oil ingibited the heat shock-induced apoptosis. To determine whether juniper inhibits the heat shock-induced activation of these apoptotic proteases, activation of CPP32 was assessed by Western blotting. Consistent with flow cytometry. DNA fragmentation and giemsa staining, heat shock-induced activation of CPP32 was blocked by juniper oil. Poly(ADP-ribose) polymerase (PARP), cysteine protease substrates were fragmented as a consequence of apoptosis by heat shock. Juniper oil inhibited the PARP fragmentation. This juniper oil also inhibited the heat shock-induced activation of caspase-3. These results suggest that juniper oil may modulate the apoptosis through the activation of the interleukin-1-converting enzyme-like protease.

Lemon also inhibited the heat shock-induced apoptosis in primary cultured rat astrocytes. To determine whether lemon inhibits the heat shock-induced activation of these apoptotic proteases, activation of CPP32 was assessed by Western blotting.
