  • A new bright yellow gerbera (Gerbera hybrida Hort.), 'Sunmyo' was bred from a cross between red medium-gerbera 'Beauty' and yellow gerbera 'Tamara' in 2001. Characteristics trials were conducted three times from 2004 to 2006. Bred cultivar 'Sunmyo' showed bright yellow color (YG12B), black center, semi-double type, and middle size flower with stable flower shape. The average yield of 'Sunmyo' increased up to 37% (70.0 stems per year) as compared to control cultivar 'Biro' (51.0 stems per year). The vase life is 7.5 days. The survival rate is 85.0%, and days to first flowering are 95 in the plastic house passing the winter season.

  • A new bright yellow gerbera (Gerbera hybrida Hort.) cultivar, 'Joyful', was released by the Flower Research Institute of Gyeongsangnam-do Agricultural Research & Extension Service in 2013. 'Joyful' was developed in Changwon from 2005 to 2006 and is a cross between 'Chowoo', a semi-double type gerbera cultivar with yellow-orange colored flowers, and 'Focus', a semi-double type cultivar with yellow colored flowers. The characteristics trials for selection were conducted from 2007 to 2009. 'Joyful' has large, semi-double type flowers with yellow (RHS, 12-A) ray florets and a brown central part. It also has a stable flower shape, strong peduncles, and a vase life of 12.8 days. The average flower yield of 'Joyful' was determined in greenhouse trials carried out from 2008 to 2010 and is 49.2 stems per plant per year. 'Joyful' was demonstrated to be a new cultivar based on ploidy tests and RAPD analysis and was registered (Grant No. 4574) to the Korea Seed and Variety Service (KSVS) for commercialization in 2013.

  • Gerber (Gerbera hybrida) is a valuable ornamental species grown as a potted plant and cut flowers. However, genetic variability within the gerbera genus is very limited. So it is absolutely needed to introduce and widen genetic resources into gerbera lines by genetic transformation. For the purpose, 18 Korean gerbera lines were screened to establish Agrobacterium-mediated genetic transformation procedure. In an experiment to select Korean gerbera lines which are amenable to Agrobacterium-inoculation, 12 lines turned out to be positive in Agrobacterium-inoculation. More callus were produced from BA 2ppm, Zeatin 2ppm, IAA 0.2ppm in pre-culture and regeneration medium (2X media) but there was no difference in the frequency of GUS expression rate. In another experiment to find out optimal condition for highly efficient Agrobacterium-inoculation, petiole and leaf explants have been treated with four different pre-culture periods, two different co-culture periods and two different Agrobacterium strains. As a result, high GUS expression has been showed from petiole and leaf explants treated no pre-culture period with LBA4404 Agrobacterium tumerfaciens, 5 day co-culture period and dipping treatment.

  • This study was conducted to find out the cause of discoloration after pressing yellow-colored flowers. Plant materials used were Lilium Asiatic Hybrids, Oncidium spp., Rosa hybrida, $Tulipa{\times}gesneriana$, Antirrhinum majus, Dianthus caryophyllus, Freesia hybrida, Zantedeschia elliottiana, Gerbera jamesonii, Helianthus annuus, and Ranunculus asiaticus. Thickness, ratio of length to width, and water content of petals did not considerably affect discoloration of pressed flowers. Expression of yellow color in petals was caused by carotenoids in Lilium Asiatic Hybrids, Oncidium spp., Rosa hybrida and $Tulipa{\times}gesneriana$, by flavonoids in Antirrhinum majus, Dianthus caryophyllus, Freesia hybrida and Zantedeschia elliottiana, and by coexistence of carotenoids and flavonoids in Gerbera jamesonii, Helianthus annuus and Ranunculus asiaticus. Carotenoids that expressed yellow color in petals affected significantly on discoloration in pressed flowers containing carotenoids alone or both carotenoids and flavonoids. However, discoloration was hardly ever found in yellow-colored species as affected by flavonoids. Dark treatment was effective for almost perfect preservation in color of pressed flowers, while light treatment resulted in severe discoloration.

  • This research was conducted to improve the cold tolerance of the gerbera cv. Gold Eye by introduction of the Arabidopsis $Ca^{2+}/H^+$ antiporter gene (CAX1) via Agrobacterium-mediated transformation. Prior to genetic transformation, we optimized a combination of plant growth regulators; $1.0mgl^{-1}$ 6-Benzyladenine (BA) and $0.1mgl^{-1}$3-indole-acetic acid (IAA) were found to lead to proper in vitro shoot regeneration from petiole explants. In addition, $50mgl^{-1}$ kanamycin was determined to be the minimal concentration useful for selection of putative transgenic plants. In this study, transgenic gerbera expressing the Arabidopsis $Ca^{2+}/H^+$ antiporter gene (CAX1) were obtained using the optimized concentrations. We expect that introduction of the gene to the cultivar will improve cold tolerance, which will be important in the winter months.
