Atomisation and vacuum drying studies on Malaysian honey encapsulation

  • Malaysian honey is rich in nutrients and bioactive compounds, which can be a healthy alternative to refined sugar in food production. However, liquid honey's viscous and sticky nature makes it unpreferable in industrial handling. This study, an atomization system coupled with vacuum drying to produce honey powders to overcome the problem. Three types of Malaysian honey, namely Acacia, Gelam, and Tualang, were encapsulated in Ca-alginate gel beads using the atomization system. The density viscosity, and surface tension of the honey-alginate solutions were measured, and the concentration of honey and alginate influenced the physical properties of the solutions. Honey-encapsulated gel beads in the size range of 2.16-2.92 mm were produced using the atomization system with the air-liquid mass flow rate ratios of 0.22-0.31, Weber number (We) of 112-545, and Ohnersorges number (Oh) of 0.35-10.46. Gel bead diameter can be predicted using a simple mathematical model. After vacuum drying, the honey gel powder produced was in the size range of 1.50-1.79 mm. Results showed that honey gel powders with good encapsulation efficiency and high honey loading could be produced using the atomization system and vacuum drying.

  • The experiment was designed to investigate the sustained release dosage form of silymarin (SL) from chitosan (CS) carrier. Solid dispersed system was prepared by mixing the drug with chitosan. This solid dispersed system was cross-linked by glutaraldehyde, formaldehyde, acetaldehyde and butylaldehyde, respectively. The dissolution rates of these preparations were compared with each other in vitro. The silymarin was mired with anionic alginate gel and bead was prepared by dropping this mixture to cationic chitosan solution including calcium chloride. Chitosan encapsulated alginate bead after drying in the oven was investigated for the dissolution rate. The dissolution rate of SL-CS mixture was delayed with increase in the amounts of CS and the concentration of aldehyde. The effect on the delay of dissolution rate was in the increasing order of formaldehyde, glutaraldehyde, acetaldehyde, butylaldehyde. The dissolution rate of chitosan encapsulated alginate bead was parallel with the concentration of chitosan in diluted hydrochloric acid solution and delayed with increase in the concentration of chitosan in phosphate buffer solution.

  • Immobilized cells of Arthrohacter nitroguajacolicus ZJUTB06-99 capable of producing nitrilase were used for biotransformation of acrylonitrile to acrylic acid. Six different entrapment matrixes were chosen to search for a suitable support in terms of nitrilase activity. Ca-alginate proved to be more advantageous over other counterparts in improvement of the biocatalyst activity and bead mechanical strength. The effects of sodium alginate concentration, $CaCl_2$ concentration, bead diameter, and ratio by weight of cells to alginate, on biosynthesis of acrylic acid by immobilized cells were investigated. Maximum activity was obtained under the conditions of 1.5% sodium alginate concentration, 3.0% $CaCl_2$ concentration, and 2-mm bead size. The beads coated with 0.10% polyethylenimine (PEI) and 0.75% glutaraldehyde (GA) could tolerate more phosphate and decrease leakage amounts of cells from the gel. The beads treated with PEI/GA could be reused up to 20 batches without obvious decrease in activities, which increased about 100% compared with the untreated beads with a longevity of 11 batches.

  • The purpose of the present investigation was to develop a novel method for cell immobilization. Aureobasidium pullulans cells were mixed with an alginate solution, and the mixture was extruded to form small gel beads as hydrated- immobilized cells. The beads were then placed at $-15^{\circ}C$ for 6-24 h to induce freeze-dehydration. The freeze-dehydration resulted in shrinkage of beads due to water removal reducing bead volume by 82% and bead weight by 85%. The dehydrated beads were successfully used for the production of fructo-oligosaccharides in a model reactor system. This study showed that bioreactor performance can be improved up to 2 times by the use of the dehydrated beads.

  • Alginate-chitosan (anion-cationic polymeric complex) was prepared to control the release rate of silver sulfadiazine (AgSD). Na-alginate (2%) solution containing AgSD was gelled in $CaCl_2$ solution. The gel beads formed were immediately encapsulated with chitosan (CS). The gel matrix and membrane were then reinforced with chondroitin-6-sulfate (Ch6S). Release rate of AgSD from the gel matrix was investigated by placing alginate beads in the sac of cellulose membrane simmered in HEPES-buffer solution. The concentration of AgSD released was analyzed by UV at 264 nm. Incorporation capacity of AgSD in Ca-alginate gel was more than 90%. Alginate-Ch6S-CS could control the release rate of AgSD. The amount of AgSD release was dependent on the AgSD loading dose. Incorporation of tripolyphosphate (polyanionic crosslinker) onto the alginate-Ch6S-CS bead increased the release rate of AgSD. Collagen-coating had no influence on the AgSD release rate. Alginate-Ch6S-CS beads with a sufficiently high AgSD encapsulation were capable of controlling the release of the drug over 10 days. In summary, alginate-Ch6S-CS beads could be used as a sustained delivery for AgSD and provide local targeting with low silver toxicity and patient discomfort.

Characterization of Immobilized Denitrifying Bacteria Isolated from Municipal Sewage

  • As a component for a recirculating aquaculture system, a new strain of denitrifying bacterium was isolated from municipal sewage. The isolate was motile by means of one polar flagellum, catalase-positive, and a Gram-negative rod-shaped cell measuring $0.5-0.6{\mu}m$ in width and $1.3-1.9{\mu}m$ in length. The isolate was identified as Pseudomonas fluorescens and produced dinitrogen gas via the reduction of nitrate. The optimal growth conditions (pH, temperature, carbon source, and C/N ratio) of the isolate were found to be 6.8, $30^{\circ}C$, malate, and 3, respectively. Under optimal growth conditions of P. fluorescens, dinitrogen gas was first detected in the exponential growth phase, then a small amount of nitrite was developed and converted to dinitrogen gas in the stationary phase. Pseudomonas fluorescens cells were immobilized in modified polyvinyl alcohol (PVA) gel beads, and the maximum denitrification rate was measured as $36.6 {\mu}lN_2h^-1$ per bead with an optimum cell loading of $20mg {\mu}l^-1$ and $2\%$ sodium alginate added to the PVA gel. The operating stability of the modified PVA gel beads remained unchanged for up to 43 repeated batches.

Simulated Nitrogen Removal for Double-Layered PVA/Alginate Structure for Autotrophic Single-Stage Nitrogen Removal (2중 구조의 PVA/alginate 겔 비드에서의 독립영양 단일공정 질소제거효율 시뮬레이션)

  • Recently, an autotrophic single-stage nitrogen removal (ASSNR) process based on the anaerobic ammonium oxidation (ANAMMOX) reaction has been proven as an economical ammonia treatment. It is highly evident that double-layered gel beads are a promising alternative to the natural biofilm for ASSNR because of the high mechanical strength of poly(vinyl alcohol) (PVA)/alginate structure and efficient protection of ANAMMOX bacteria from dissolved oxygen (DO) due to the thick outer layer. However, the thick outer layer results in severe mass transport limitation and consequent lowered bacterial activity. Therefore, the effects of the thickness of the outer layer on the overall reaction rate were tested in the biofilm model using AQUASIM for ammonia-oxidizing bacteria (AOB), nitrite-oxidizing bacteria (NOB) and ANAMMOX bacteria. A thickness of 0.5~1.0 mm is preferred for the maximum total nitrogen (TN) removal. In addition, a DO of 0.5 mg/L resulted in the best total nitrogen removal. A higher DO induces NOB activity and consequent lower TN removal efficiency. The optimal density of AO B and NO B density was 1~10% for a 10% ANAMMOX bacterial in the double-layered PVA/alginate gel beads. The real effects of operating parameters of the thickness of the outer layer, DO and concentrations of biomass balance should be intensively investigated in the controlled experiments in batch and continuous modes.

Vinegar Production by Acetobacter aceti Cell Immobilized in Calcium Alginate (Calcium Alginate로 고정화된 Acetobacter aceti에 의한 식초생산)

  • This study is to investigate for obtaining the operating conditions of continuous vinegar production using fluidized bed reactor by Acetobacter aceti cell immobilized in Ca-alginate gel. The optimum conditions obtaining by batch fermentation using fluidized bed reactor were as follows; The fermentation temperature and aeration rate were 3$0^{\circ}C$ and 1.0VVM and the initial concentration of ethanol and acetic acid in medium were 33g/l and 27g/l respectively. The amount of bead used was 25%(w/v). The overall acetic acid productivities of batch fermentations by free cell and immobilized cell were 0.31g/l-hr and 0.48g/l-hr, respectively, at the final acetic acid concentration of 50g/l. In the continuous vinegar production using fluidized bed reactor by immobilized cell under optimum conditions, it was possible to produce 23g/l acetic acid continuously up to 90 days with maximum acetic acid productivity of 2.76g/l-hr at dilution rate 0.12hr-1.

Cadmium Adsorption Characteristic of Cellulose-gel Manufacture using Alkali Solvent (알칼리 용제를 이용하여 제조한 셀룰로오스 겔의 카드뮴 흡착특성)

  • This study was carried out to investigate the characterization of cadmium adsorption by cellulose hydrogel and aerogel. Hydrogel and aerogel were made from ashless pulp dissolved in alkali hydroxide-urea aqueous solution and manufactured in film and bead types. After regeneration of cellulose, hydrogel went through the process of substitution of organic solvent and freeze-dry in order to make aerogel. SEM was used to analyze the microstructure of hydrogel and aerogel. Experiment was conducted in various concentrations and pH conditions to find out the characteristic of cadmium adsorption. After that, EDS was used to identify existence and distribution of cadmium in hydrogel and aerogel. The result from comparisons of cadmium adsorption shows that bead type aerogel has the maximum cadmium adsorption and film type hydrogel has the minimum cadmium adsorption.

  • The denitrification characteristics of free- and gel- D photosynthetic bacteria were studied. At various salt concentrations (0, 1, 2, 3.5%), the maximum specific growth rates were found to be 0.22, 0.22, 0.20 and 0.11 $h^{-1}$, respectively. At lower pH, pH affected the growth significantly. In experiments of PVA beads, the maximum $N_2$ production rate $(dN_2/dt)$ were calculated to be 0.09-0.10 (D= 0.7cm) and 0.09-0.11(D= 1.0cm) ml/h, respectively, and those per bead were 5 and $5.5\;{\mu}l$/h/bead.
