Experimental Studies on the Antidiarrheal Effects of Anjang-san

  • Objectives: The purpose of the present study was to investigate the antidiarrheal effects of Anjang-san in mice and rats. Methods: We measured the content of condensed tannin in Anjang-san extract, and observed the effects of Anjang-san on the small intestinal and colonic transport of mice, as well as on mice models of diarrhea induced by castor oil and $MgSO_4$, and on rat models of castor oil-induced enteropooling. Results: Anjang-san showed significant inhibitory effects on abnormally increased small intestinal transit induced by pyridostigmine and neostigmine, and inhibitory effects on large intestinal transit. Anjang-san also exhibited antidiarrheal effects on diarrhea induced by $MgSO_4$, and inhibitory effects on castor oil-induced enteropooling. Anjang-san also improved castor oil-induced diarrhea based on simple numbers without statistical significance. Conclusion: These results demonstrate that Anjang-san has significant antidiarrheal properties and attests to its possible utility in functional diarrheas, irritable bowel syndrome and other gastrointestinal disorders based upon further studies.

  • Objectives: The purpose of this study was to describe the effectiveness of Jeoreongchajeonja-tang as a treatment for patients with diarrhea-predominant irritable bowel syndrome (IBS-D). Methods: Two irritable bowel syndrome patients who had visited Semyung University Korean Medical Hospital were enrolled. The patients were diagnosed according to the Rome III criteria, and the treatment effect was assessed using the Gastrointestinal Symptom Rating Scale (GSRS) and the colon transit time. Results: After treatment with Jeoreongchajeonja-tang for 4 weeks, the GSRS score was improved and colon transit time was delayed. Conclusions: This study suggests that Jeoreongchajeonja-tang is effective for improving diarrhea-predominant irritable bowel syndrome. More extensive study is needed in the future.

Small Intestinal Transit Does Not Adequately Represent Postoperative Paralytic Ileus in Rats

  • Small intestinal transit (SIT) has often been regarded as an index of pathophysiological state of postoperative ileus (PI) in rats. The reliability of SIT as an index of PI was examined in the present study. PI was induced via abdominal surgery (i.e., laparotomy with evisceration and manipulation) in rats. For one group of PI-induced rats, SIT of a charcoal test meal was measured. When necessary, the physical state (i.e., severity and site of distension) of the gastrointestinal (GI) tract in each rat was visually examined. For another group of PI-induced rats, abdominal X-ray radiographs were obtained after introducing the barium sulfate suspension. The abdomen was then opened and the physical state of the GI tract was visually examined. The SIT was decreased in most of the PI-induced rats, and the GI distension was observed, with substantial intersubject variations, in all of the PI-induced rats. However, no linear relationship was evident between the SIT and the severity of GI distension (e.g., at 20 h after PI induction). Instead, the severity and site of GI distension could be monitored by the X-ray radiology. Therefore, the use of SIT as an index of PI should be substantially limited.

동물모델을 이용한 MMSC(DL-Methionine Methylsulfonium Chloride)의 기능성소화불량증 개선효과 (The Improvement Effect of MMSC (DL-Methionine Methylsulfonium Chloride) in Functional Dyspepsia Animal Models)

  • 본 연구는 cisplatin 및 atropine으로 유발된 기능성소화불량증 동물모델에서 MMSC의 위 배출능 및 위장관 전이 지연의 개선 효과를 연구한 후, 기능성소화불량증을 치료하기 위한 약물의 기초자료를 제공하고자 하였다. 기능성소화불량은 뚜렷한 기질적 병변 없이 여러 가지 다양한 소화불량 증상에 의한 진단이기 때문에 치료 또한 단순하지가 않다. 대부분의 증상이 호전과 악화를 반복하며 음식, 스트레스 등에 의해 변화가 심하므로 임상적으로 효과 판정이 어렵고, 더욱이 위약만을 투여하더라도 증상의 호전이 있을 수 있으므로 어떠한 치료가 의미 있게 효과가 있는지를 판단하는데 애로점이 많다. 이번 연구에 대조물질로 비교한 약물도 현재 사용되는 위장질환 치료물질로 위 배출능에서 시험물질보다 좋은 결과 값을 얻었지만, 위장관 전이 지연 개선 실험에서 시험물질 MMSC와 비교하였을 때 약물의 효과가 다르게 나타났다. 결론적으로 MMSC를 투여한 그룹이 유발그룹에 비해 위 배출능 및 위장관의 전이 비율이 유의적으로 증가하였음을 확인하였다. 위장관 운동 촉진제로 시판되는 약물을 투여한 그룹보다는 그 비율이 다소 낮게 관찰되었지만 시판 약물의 고용량 투여와 비교하여 저용량의 MMSC에도 불구하고 위배출능의 효과를 확인하였으므로 고용량의 MMSC투여 시 위 배출능의 개선 효과가 높게 관찰될 가능성 역시 배제할 수 없어 향후 보다 구체적인 연구가 수행되어야 할 것으로 판단된다.

저항전분 투여가 loperamide 유도 변비 쥐의 체중변화와 배변상태에 미치는 영향 (Effect of Resistant Starch on Body Weight and Defecation in Loperamide-induced Constipated Rats)

  • 본 연구는 2종류의 저항전분(RS2, RS3)이 loperamide 유도 변비 쥐의 체중변화와 배변상태에 미치는 영향을 다른 식이섬유 급원과 비교하여 실험 관찰하였다. 실험동물은 Sprague-Dawley계 수컷 쥐 60마리를 사용하였으며 정상 대조군, 변비 셀룰로오스군, 변비 펙틴군, 변비 RS2군, 변비 고RS2군, 변비 RS3군의 6개군으로 나누었다. AIN-76 기본식이에 각각 다른 식이섬유를 첨가하고 에너지 밀도가 동일하게 조제한 실험식을 5주 동안 투여하여 변비 쥐의 식이효율과 체중 변화 및 배변상태와 장통과시간을 분석 비교하여 다음과 같은 결과를 얻었다. 저항전분 투여군들은 셀룰로오스군에 비해서 식이효율과 체중 증가가 유의적으로 낮았고 펙틴군과는 차이가 없었다. 소장의 길이는 모든 식이섬유군간에 유의적 차이가 없었으나, 대장의 길이는 저항전분군들에서 펙틴군에 비해 유의적으로 길었고, 셀룰로오스군과는 유의적인 차이가 없었다. 사료섭취량 대비 분변 배설량은 저항전분 RS2의 경우는 펙틴군 보다는 높았고 셀룰로오스군 보다는 유의적으로 낮았으나 RS3군은 셀룰로오스군과 유의적인 차이가 없이 많았다. 분변의 외형은 RS3군과 고RS2군은 셀룰로오스군과 유사하였으나 저RS2군은 펙틴군과 유사하였다. 변 중 수분함량은 식이섬유 간에 유의적 차이가 없었으나 장 통과시간은 셀룰로오스군과 RS3 투여군이 펙틴군에 비해 유의적으로 단축되는 것으로 나타났다. 본 연구 결과는 변비 동물에서 저항전분의 투여는 식이효율과 체중 증가를 억제하는 작용이 있고, 저항전분중 RS3는 셀룰로오스와 마찬가지로 변비 동물의 배변량을 증가시키고 장 통과시간을 단축시켜 배변 상태를 개선시킬 수 있는 가능성을 나타내었다.

  • The intestinal microbiota has been shown to have a vital role in various aspects of human health, and accumulating evidence has shown the beneficial effects of supplementation with bifidobacteria for the improvement of human health, ranging from protection against infection to various positive effects. However, maintaining bacterial cell viability during storage and gastrointestinal transit remains a challenge. Microencapsulation of probiotic bacterial cells provides protection against adverse conditions during processing, storage, and gastrointestinal passage. In this paper, we review the current knowledge, future prospects, and challenges of microencapsulation of probiotic Bifidobacterium spp.

  • This research aims to study the changes In gastrointestinal function attributed to aging In human. The thresholds for recognition and detection of flavors became elevated and salivary gland acinar cells decreased in the old age. But most esophageal function remained relatively Intact. Although gastric emptying time has been slowed with aging, the total intestinal transit time did not differ. Atropic gastritis due to H. pylori in old man decreased secretion of acid and Intrinsic factor and absorbability of calcium and iron. Pancreatic secretion is droned in older persons. Prevalence of gallstones rised with age. Liver size and portal blood flow decreased significantly with age. Mucosal surface area has been reported to be slightly diminished in the aging man. Glucose transporters decreased and Insulin tolerance Increased. Absorption of aromatic amino acid is diminished with age. Dietary protein In that aging human increased fecal nitrogen excretion. Vitamin A tolerance increased. Vitamin D receptor concentration decreased and resistance to 1,25-(OH)2D3 action increased. Permeability of aging small Intestine Increased. Zinc balance dirt not differ Copper absorption appeared not to be significantly affected by age. Neurotensin secretion decreased thus slowed colonic peristaltic movements and Intestinal mucosal growth.

식이섬유 첨가식이 Streptozotocin-유도 당뇨쥐의 장기능과 지질 및 당질대사에 미치는 영향 (The Effects of Dietary Fiber Feeding on Gastrointestinal Functions and Lipid and Glucose Metabolism in Streptozotocin-induced Diabetic Rats)

  • This study was conducted to compare the effects of four types of dietary fiber supplementations (cellulose, pectin, guar gum, and polydextrose) on gastrointestinal function, diabetic symptom amelioration and lipid & glucose metabolism in streptozotocin-induced diabetic rats. Six groups of male rats were fed ad libitum dietary fiber-free control diet or one of experimental diets containing 5% dietary fiber for four weeks. All types of dietary fiber supplementation seemed to protect the diabetic animals from the loss of body weight. The primary diabetic symptoms such as polydipsia, polyphasia, polyuria and urinary glucose excretion were ameliorated by cellulose, pectin, and guar gum, but not by polydextrose. Gastrointestinal transit time was significantly shortened and fecal dry weight was significantly increased in all the dietary fiber-supplemented groups except the polydextrose group. Large intestine was significantly lengthened by dietary fiber feeding. The serum triglyceride and total cholesterol levels were effectively lowered by pectin, guar gum and polydextrose. Regardless of their types, the fiber supplementation had no effect on serum HDL-cholesterol. Whereas fasting blood glucose level was significantly lowered by all types of fiber supplementations, glucose tolerance was more effectively improved by pectin and guar gum.

당뇨병 환자를 위한 고식이섬유 보충물의 개발을 위한 연구(I) -해조류 투여가 당뇨쥐의 장기능과 증세호전도에 미치는 영향- (A Study on the development of high-fiber supplements for the diabetic patients (I) -Effect of Seaweed Supplementation on the Gastrointestinal Function and Diabetic Symptom Control in Streptozotocin-induced Diabetic Rats-)

  • The present study was conducted to evaluate the usefulness of four kinds of seaweeds (mixture of purple laver & sea lettuce, sea tangle, sea mustard, agar agar) as a high- fiber supplment in the therapeutic deit for the diabetic patients. Seven groups of normal and streptozotocin-induced diabetic rats were fed dietary fiber-free control diet or one of experimental diets containing 7% of one of the four seaweeds for 6 weeks. The effect of seaweeds supplementation on the body weight change, gastrointestinal function, and the control of diabetic symptoms were examined and compared with the effect of fiber-free diet or pectin diet used as references. The body weight gains of all the diabetic groups were significantly suppressed compared to the normal group. Feed efficiency ratios and body weight gains of seaweed groups were relatively higher than those of the pectin group. Sea tangle appeared to have an effect of alleviating the typical diabetic symptoms such as polyphasia, polydipsia, polyuria, urinary glucose excretion and hyperglycemia indicating its beneficial acition of improving glucose metabolism even though the degree of effectiveness was less than that with pectin. All the supplemntations of seaweeds and pectin ressulted in the significant changes in gastronitestinal functins ; shortening of GI transit time, increase of fecal volume and the length of intestine. Based on their effects of the significant changes in GI function in may be suggested that seaweeds may influence the process of digestion and absorption of nutrients in diabetic animals.

소청룡탕 추출물의 당뇨병 질환에서 위장관 운동 기능 개선에 관한 연구 (Effects of Socheongryong-Tang, a Traditional Chinese Medicine, on Gastrointestinal Motility Disorders (Diabetic Models) in Mice)

  • Objectives: The purpose of this study was to investigate the effects of Socheongryong-tang, a traditional herbal medicine, on gastrointestinal (GI) motility disorders (inflammatory and diabetic models) in mice. Methods: The in vivo effects of Socheongryong-tang on GI motility were investigated by calculating percent intestinal transit rates (ITR) and gastric emptying (GE) values using Evans Blue and phenol red, respectively, in normal mice and in mice with experimentally induced GI motility dysfunction (GMD). GMD models were induced by acetic acid and streptozotocin. Results: In normal ICR mice, ITR and GE values were significantly and dose-dependently increased by the intragastric administration of Socheongryong-tang (0.0025~0.25 g/kg). The ITR values (%) of GMD mice were significantly lower than those of normal mice, and these reductions were significantly and dose-dependently inhibited by Socheongryong-tang. Conclusions: These results suggest that Socheongryong-tang is a good candidate for the development of a prokinetic agent that may prevent or alleviate GMD.
