발효법을 달리하여 제조한 치자꽃 발효주의 특성 (Manufacturing of Korean Traditional Rice Wine by using Gardenia jasminoides)

  • To develop a new traditional rice wine by using Gardenia jasminoides, various fermentation methods such as without cooking, with cooking and with starter seed methods were studied. The condition of alcohol fermentation was investigated by addition of 1% Gardenia jasminoides into mash. Among the fermented methods, the fermentation with starter seed was the best as the alcohol was 19%. The acceptability of the Gardenia jasminoides rice wine with different methods were compared. The starter seed method which was prepared by adding 1% Gardenia jasminoides into mash showed the best acceptability in the sensory evaluation test and color test.

치자색소의 염료화 및 염색성 (The Acquisition and dyeability of Gardenia jasminoides Colorant)

  • The acquistion and dyeability of the Gardenia jasminoides were examined to establish the optimum condition for extraction and storage in the process of obtaining the natural dye, Gardenia jasminoides colorant. Also the dyeability and colorfastness of Gardenia jasminoides were investigated. The results of this study are as follows. The optimum part-removed seed. In all experiments, the part of fat-removed seed and pericarp was used. The optimum condition for extraction of Gardenia jasminoides colorant was at 40℃ and for 90 min. in methanol. As storage temperature was higher, the absorbance of colorant extract decreased rapidly. The Gardenia jasminoides colorant exhibited dyeability to cotton, silk, wool, and nylon. The dyeability was the greatest in wool, and then nylon, silk, and cotton. Both wool and nylon had the greatest K/S value at pH3, however, nylon and cotton at pH 6 and pH 8 respectively. In addition, the increase in K/S value corresponded to temperature in wool and nylon, but the silk and cotton had the greatest K/S value at 60℃. Also, the K/S value increased in concomitant with the increased number of dyeing repetition. In the most cases, colorfastness of light was weak but colorfastness of laundry, sweat was relatively excellent.

남해산 치자(Gardenia jasminoides Ellis fructus) 씨 추출물의 항산화 능 및 생리활성에 미치는 영향 (Effect on Antioxidant Ability and Physiological Activities from Seed Extract of Gardenia jasminoides Ellis in Namhae Korea)

  • 치자(Gardenia jasminoides Ellis fructus, GJE) 씨에서의 70% 에탄올, CM (chloroform: methanol, 부피 비 2:1(CM)) 및 노르말 부탄올 추출 용매별 flavonoid 함량 및 항산화 능, 금속 킬레이트 능력 측정을 통하여 치자 씨의 기능성 식품 소재로서의 가치를 검토한 결과, 치자 씨의 proanthocyanidin 함량은 건조 무게(g) 중 카테킨(catechin) 당량으로 계산하여 $70.035{\pm}0.772mg$으로 나타났으며, 추출 수율은 CM (36.39%), 70% 에탄올(27.32%), 노르말 부탄올(26.23%) 순으로 나타났다. 추출 용매별 항산화 능은 농도(0.2, 0.4, 0.6 mg/mL)가 증가할수록 유의적으로 증가하였으며(p<0.05), 양성대조군으로 사용된 ascorbic acid, butylated hydroxyanisole (BHA), ethylenediamnetetraacetic acid (EDTA) 보다는 낮은 활성이 확인되었다. Flavonoid 함량(mg)은 시료 g당 케르세틴(quercetin) 당량으로 계산하여 70% 에탄올(0.830), CM (0.752), 노르말 부탄올(0.105) 순으로 확인되었으며, 항산화 능 분석 실험과 금속 킬레이트 능력 실험에서도 이와 유사한 양상으로 동정되어 모든 분석에서 70% 에탄올과 CM 추출물이 강한 생리활성을 나타내었다. 따라서 치자 씨의 용매별 flavonoid 함량에 따라 항산화 능 및 금속 킬레이트 능력이 증가하는 것으로 관찰되었다. 이에, 치자씨는 proanthocyanidin과 flavonoid 화합물을 다량 함유하고 있으며, 높은 항산화 능과 생리활성을 가지고 있어 기능성 식품 및 천연 항산화제로서의 가치가 있을 것으로 추정된다.

  • The purpose of this study was to investigate the bioactivity of extracts from the seeds of Gardenia jasminoides Ellis fructus (GJE) found in Namhae, Korea. Extraction was performed using three solvents, 70% methanol, Distilled Water (DW), and Ethyl Acetate (EA). We determined the total phenol and phytic acid contents of the extracts to evaluate their nitrogen oxide scavenging activity, antioxidant activity, reducing power, and lipid peroxidation inhibition ability. The phytic acid content of GJE was found to be 1.157 mg PAE (Phytic Acid Equivalent) /g DW. The yields of the three extraction processes were as follows: DW, 36.61%; 70% methanol, 30.10%; and EA, 20.40%. The physiological activities of the extract solvents increased significantly with increasing concentrations (0.2, 0.4, and 0.6 mg/mL) (p<0.05), but were lower than those of ascorbic acid, BHA, and trolox. Total phenol content was the highest in the 70% methanol extract, followed by DW and EA extracts. Further, nitrogen oxide scavenging activity and antioxidant activity were the highest for the 70% methanol extract followed by DW and EA extracts. Based on these results, the bioactivities of the 70% methanol and DW extracts of GJ seeds were excellent. These extracts can be used as natural antioxidants.

치자(Gardenia jasminoides Ellis) 씨 추출물의 질소산화물 소거능 및 지질과산화 저해능에 관한 연구 (Effect of Extracts produced from Gardenia jasminoides Seed Using Different Types of Solvents on Nitrogen Oxide Scavenging Activities and Lipid Peroxidation Inhibition)

  • 치자 씨 추출물이 항산화력 및 지질과산화 저해능에 미치는 영향을 알아 보고 치자의 기능성 식품 소재로서의 가치를 검토하기 위하여 실험을 수행한 결과, 치자 씨의 anthocyanin 함량을 측정한 결과 $2.201{\pm}0.516mg/100g\;DW$로 나타났으며, 치자 씨의 용매 별 추출 수율은 chloroform:methanol(CM, 2:1, v/v) 36.39%, 70% ethanol (27.32%), n-butanol (26.23%) 로 확인되었다. 추출 용매 별 항산화 활성은 농도(0.2, 0.4, 0.6 mg/mL)가 증가할수록 유의적으로 증가하였으며 positive control로 사용된 ascorbic acid, BHA, trolox 보다는 낮은 활성이 관찰되었다. 치자 씨의 total phenol 함량(mg CAE/g)은 CM (32.50), 70% ethanol (30.09), n-butanol (11.07) 추출물 순으로 n-butanol 추출물에서 가장 적은 함량을 보였으며, Nitric oxide (NO) radical 소거능에서는 CM (76.97~84.24%), 70% ethanol (74.10~79.99%), n-butanol (30.66~37.15%) 추출물 순으로 관찰되었다. Nitrite ($NO_2$) 소거능은 CM (33.53~43.23%), 70% ethanol (32.40~35.98%), n-butanol (24.72~28.14%) 순으로 관찰되었다. ${\beta}-carotene$ 탈색 저해능은 CM (23.73~44.70%), 70% ethanol (22.03~41.32%), n-butanol (16.00~27.87%) 순으로 확인되었다. Reducing power (optical density)는 70% ethanol (0.073~0.182), CM (0.057~0.154), n-butanol (0.028~0.079) 순으로 관찰되었다. 지질과산화 저해능은 씨 추출물 중 CM (53.26~76.56%), 70% ethanol (52.97~76.56%), n-butanol (38.54~53.33%) 순으로 나타났다. 이에, 치자씨 추출물은 천연 항산화제로서 기능성 식품의 가치가 높을 것으로 판단된다.

  • 치자 씨의 에틸 아세테이트, 증류수 및 70% 메탄올(v/v)의 3가지 용매를 사용한 추출물의 용매 별 플라보노이드 함량 및 항산화 활성, 금속 킬레이트 능력 측정을 통하여 치자의 기능성 식품 재료로서의 가치를 검토하기 위하여 본 실험을 수행하였다. 치자 씨의 탄닌 함량은 $1.517{\pm}0.003mg\;CE/g$(mg of catechin equivalents) 건조 중량 으로 나타났으며, 치자 씨 용매 별 추출 수율은 증류수(36.61%), 70% 메탄올(30.10%), 에틸아세테이트(20.40%) 순으로 나타났다. 추출 용매 별 항산화 활성은 농도(0.2, 0.4, 0.6 mg/mL)가 증가할수록 유의적으로 증가하였으며(p<0.05), 양성대조구로 사용된 ascorbic acid, BHA, EDTA 보다 낮은 활성이 관찰되었다. 플라보노이드 함량(mg QE/g, mg of quercetin equivalents)은 70% 메탄올(0.799), 증류수(0.565), 에틸 아세테이트(0.117) 순으로 관찰되었으며, 항산화 활성은 대부분의 분석에서 용매 별 플라보노이드 함량 순과 비슷한 양상으로 나타났지만 ABTS 라디칼 소거능과 SOD 유사활성은 증류수 추출물이 70% 메탄올 추출물의 항산화 활성 보다 높은 것으로 나타났다. 치자 씨의 각 용매별 수율과 항산화 활성을 비교하였을 때, 증류수와 70% 메탄올 용매를 사용하여 추출 이용하는 것이 바람직하다고 생각된다. 본 실험 결과 치자 씨는 플라보노이드 화합물을 다량 함유하고 있으며 그로 인한 높은 수준의 항산화 활성과 생리활성을 가지고 있어 기능성 식품 및 천연 항산화제로서의 가치가 매우 기대된다.

치자(梔子) 약침(藥鍼)이 백서(白鼠) 모델 족과 염좌(捻挫) 통증(痛症)에 미치는 영향(影響) (Effect of Frutus gardeniae herbal acupuncture on the rat model of ankle sprain pain)

  • Objective : Frutus gardeniae, seed of Gardenia jasminoides Ellis is one of the crude drugs used for the treatment of inflammatory condition in oriental medicine. Methodes : The present study aimed to examine the analgesic effect and anti-inflammatory effect of Frutus gardeniae extract (FGE) on a rat model of ankle sprain pain, and the relations between FGE-induced effect and endogenous nitric oxide (NO) and inducible NO synthase (iNOS), cyclooxygenase-2 (COX-2), and c-Fos protein expression in the spinal cord. As a chronic pain model, ankle sprain pain model was used to test the effect of FCE injection applied to acupuncture point. After the induction of ankle sprain, rats subsequently showed a reduced stepping force of the affected limb for at least the next 4 days. The reduced stepping force of the limb was presumably due to a painful knee. FGE dissolved in normal saline was injected several acupoints. Results : After the treatment, behavioral tests measuring stepping force were periodically conducted during the next 8 hours. FGE produced significant improvement of stepping force of the hindlimb affected by the ankle sprain lasting at least 4 hours. FGE produced the improvement of stepping force of the affected hindlimb in a dose-dependent manner. In addition, FGE injection showed inhibitory effect on the paw edema induced by ankle sprain. Both NO production and iNOS, COX-2 protein expression increased by ankle sprain were suppressed by FGE. FGE on combination with electroacupuncture (EA) produced more powerful and longer lasting improvement of stepping force of the hindlimb affected by the ankle sprain than either FGE or EA did. The present study suggest that FGE produces a potent analgesic effect on the ankle sprain pain model of the rat and that FGE-induced analgesia modulate endogenous NO through the suppression of iNOS/COX-2 protein expression.
