White mutant of Ganoderma lucidum(G4142) induced the non-basediocarpous basidiospores(NBB) from the aerial mycelia on agar media by the light illumination. Light was found to be necessary for NBB formation, but it also inhibited the growth of mycelium. The best sporulation was obtained at the periodic exposure of 16 hour light and 8 hour dark. Blue and yellow light were the most effective on sporulation, however, near UV and red light did not induce any spores. Effective light intensity for NBB bearing was about 1,000 lux as white light. Even after 16 days of culture, this strain did not form the pinhead nor chlamydospore. Optimum temperature for the mycelial growth and NBB formation were 30$\circ $C. Ganoderma lucidum G4142 exhibited the formation of stroma after five days of incubation at 30$\circ $C.
Ganoderan, antitumor ${\beta}-glucan$ from Ganoderma lucidum was extracted from the mycelium of G.lucidum IY009 which was cultured in various carbon sources. The mycelium was shown to be capable of utilizing various carbon sources, e.g., soluble starch, fructose and glucose, and differs in morphology on carbon sources. In radioisotope assay, about $5.2{\sim}16%$ of glucose was to be incorporated in ganoderan of the mycelium. The monosugars of these ganoderan were mainly consisted of glucose, mannose, galactose. The galactose was not good carbon source for growing the mycelium but the best carbon source for producing the potentialized-ganoderan on the antitumor and anticomplementary activity. The tumor inhibition ratio of ganoderan-GAL, obtained from galactose medium, was 83.6% at the dose of 20 mg/kg/day. This crude polysaccaride was composed of five monosaccharide and the protein contained 16 amino acids. Also, ganoderan-GAL increased the anticomplementary activity than that obtained from any other media. This fact suggests that the structural differences of ganoderan influence the antitumor and anticomplementary activity.
It was discovered that ammonium phosphate in the medium played an important role in both growing mycelium and producing exopolysaccharides (EPS) from G. lucidum. In lower concentration levels of ammonium phosphate (0-3 g/l), an improved mycelial growth was observed by maintaining more filamentous morphology than in high concentrations (5-11 g/l). In addition, it was confirmed by comparing the factual dimension and frequency of the area regarding the mycelial pellets. This must be attributed to limitations of nutrient transfer by maintaining filamentous mycelium during the cultivation in a low ammonium phosphate containing medium. On the other hand, the best EPS production was observed in medium with the absence or low concentration of ammonium phosphate. The shear stress of the culture broth was greatly affected by the shear rate, as compared with that of the culture broth with high ammonium phosphate concentration. The rheological characteristics of the fermentation broth and filtrate worked well according to the Herschel-Bulkley model. It was also found that the morphological changes of the mycelium resulting from the ammonium phosphate concentration directly affected the rheological characteristics of the system and resulted in reversely affecting the EPS production levels. Based on these results, it can be concluded that delicate regulation of the ammonium phosphate concentration in the culture media should be provided in order to obtain optimal mycelial growth and/or EPS production.
The ginseng residue, a by-product of ginseng tea manufacture, was used as growth promoting substance in the submerged cultivation of Basidiomycetes for its effective utilization. Ginseng residue contained about 46% of total sugar, 14% of crude protein, 12% of ash, and 0.16% of crude saponin. Among inorganic substances in ginseng residue, amount of Mg, Na, K and Ca were much more than other inorganic substances. All ginsenosides existed in red ginseng residue. Ganoderma lucidum and Pleurotus ostreatus could be excellently cultured in potato dextrose broth. Most effective additional concentration of residue extract for growth of these fungi was shown to be 0.2%, and the contents of crude protein and amino acid in mycelium were increased when 0.5% of residue extract were added to the medium.

Isozyme patterns of leucine aminopeptidase, esterase and protein from 16 isolates of Ganoderma lucidum were observed by electrophoresis for characterization of the isolates. Even in the same isolate, the patterns of the isozymes from mycelium and cap were different. The esterase patterns from the mycelium could differentiate the isolates. Of 16 isolates, four isolates showed identical patterns. It was assumed that these had the same genetic background.
Auxotrophic mutants were obtained by UV-irradiation to mycelium of Ganoderma lucidum. Induction rate of auxotrophs was 5.78%. Interspecific fusion products of protoplasts were obtained by polyethylene glycol induced fusion of protoplasts from auxotrophic mutants of Ganoderma lucidum and Ganoderma applanatum. Fusion products were selected by means of the comparison with the mycelial growth rate and colony morphology. Fusion products were confirmed by mycelial morphology and esterase isozyme pattern. Some segregants were observed and fusion product produced fruit bodies.
The immune activities of Ganoderma lucidum Mycelium added garlic extracts (GAM), Ganoderma lucidum Mycelium (GM), garlic extracts (GS) and standard $({\beta}-glucan)$ were compared. GAM enhanced the growth of human immune T cell up to $1.25{\sim}1.46$ times, compared to control group. GAM showed relatively lower cytotoxicity in using normal human lung cell, while GAM showed the most potent inhibitory effect on the human lung carcinoma, compared to GM and GS. The selectivity of GAM was also higher than that of GM and GS. GAM increased the secretion of cytokines, IL-6 and TNF- from human B cell as well as the growth of human immune cells. It can imply that GAM has higher immune activity than GM or GS.

Ganoderma lucidum BCCM 31549 has a long established role for its therapeutic activities. In this context, much interest has focused on the possible functions of the (1,3)-β-D-glucan (G) produced by these cultures in a stirred-tank bioreactor and extracted from their underutilized mycelium. In the existing study, we report on the systematic production of G, and its sulfated derivative (GS). The aim of this study was to investigate G and its GS from G. lucidum in terms of their antibacterial properties and cytotoxicity spectrum against human prostate cells (PN2TA) and human caucasian histiocytic lymphoma cells (U937). 1H NMR for both G and GS compounds showed β-glycosidic linkages and structural similarities when compared with two standards (laminarin and fucoidan). The existence of characteristic absorptions at 1,170 and 867 cm-1 in the FTIR (Fourier Transform Infrared Spectroscopy) for GS demonstrated the successful sulfation of G. Only GS exhibited antimicrobial activity against a varied range of test bacteria of relevance to foodstuffs and human health. Moreover, both G and GS did not show any cytotoxic effects on PN2TA cells, thus helping demonstrate the safety of these polymers. Moreover, GS showed 40% antiproliferation against cancerous U937 cells at the low concentration (60 μg/ ml) applied in this study compared with G (10%). Together, this demonstrates that sulfation clearly improved the solubility and therapeutic activities of G. The water-soluble GS demonstrates the potential multifunctional effects of these materials in foodstuffs.
For the screening and the development of the new bio-material, cultural conditions for the exo-biopolymer (EBP) production throught the submerged cultivation of Ganoderma lucidum mycelium were investigated. Also, the fractionations and the purifications of the exo-biopolymer were carried out and the chemical compositions of the exo-biopolymer were examined. The optimal culture conditions for the exo-biopolymer production were pH 5.0, 30$^{\circ}C$ and 100 rpm of agitation speed in the medium containing of 5% (w/v) glucose, 0.5%(w/v) yeast extract, 0.1% (w/v) ($(NH_4)_2HPO_4$, and 0.05% (w/v) $KH_2PO_4$. In the flask cultivation for 7 days under these conditions, the concentration of the maximum exo-biopolymer and the cell mass were 15.4g/l and 18.8g/l, respectively. The specific growth rate was 0.039 $hr^{-1}$. In addition, the substrate consumption rate, and the exo-biopolymer production rate were 0.043$gg^{-1}$$hr^{-1}$ and 0.025$gg^{-1}$$hr^{-1}$, respectively. The exo-biopolymer was fractionated into BWS (water soluble exo-biopolymer) and BWI (water insoluble exo-biopolymer) by the water extraction, and the sugar contents of two fractions were higher than 97% (based on dry basis). The components sugar of BWS and BWI fractions were glucose, galactose, mannose, xylose, and fucose. Their molar ratios were 3.6:1.5:2.1:0.5: trace and 2.9:3.1:2.0:1.6:0.3, respectively.
This study was conducted to develop a renewable and sustainable bio-material to replace polystyrene (EPS) in fungal-mycelium-based composite using agricultural by-products. Four mushrooms (Ganoderma lucidum, Fomitella fraxinea, Phellinus linteus, and Schizophyllum commune) were cultured in an oak sawdust plus rice bran substrate to select the mushroom with the best growth. The mycelia of G. lucidum showed the best growth. To investigate the optimal mixing ratio with spent mushroom substrate (SM) and oak sawdust (OS), samples were prepared by mixing SM and OS at ratios of 50%:50%, 60%:40%, and 80%:20% (w/w). Each substrate was then inoculated with G. lucidum. G. lucidum showed the best mycelial growth of 140.0 mm in the substrate with SM and OS mixed at a 60%:40% ratio. It was also found that the substrate with SM and OS mixed at a 60%:40% ratio had the best handling properties. The compressive strength of mycelial materials inoculated with G. lucidum was in the range of 300-302 kgf mm-1, and the materials were four times stronger than polystyrene materials. These results indicate that substrates comprising spent mushroom substrate mixed with oak sawdust can be successfully upcycled to mycelium-based composite materials using G. lucidum. This represents a sustainable approach.
