Development of High-Functional Hyaluronic Acid/Salmon Extract Formulation Using Gamma-Ray (감마선을 이용한 고기능성 히알루론산/연어추출물 제형개발)

  • Hangover after drinking is different from person to person symptoms and degree, but usually thirst, fatigue, headache, general boredom, gastrointestinal disorder, vomiting, diarrhea, deficiency of vitamin appears. This hanging phenomenon is caused by the action of precursors such as ethyl acetate and acetaldehyde, which are the by products of fermentation contained in alcohol and alcohol accumulated in hepatocytes and body. In order to solve the hangover phenomenon, the same Origin as polysaccharide Polydeoxyribonucleotide, which is a nucleic acid-sugar-phosphate complex, which is a semen or testicular extract in salmon extract, and a water soluble salmon extract powder having the same structure and lower price than Polydeoxyribonucleotide And D-Glucuronic acid and N-Acetyl glucosamine. It has excellent biocompatibility, viscoelasticity and moisturizing power. It has effect on reduction of body water loss and skin moisture content in hangover phenomenon. It is antioxidant and skin moisturizing effect Hyaluronic acid was irradiated with gamma rays, and the composition was prepared by using the salmon extract powder and the main raw material. The ethanol degradation, the acetaldehyde reduction amount, the blood acetaldehyde concentration and the acetic acid concentration were measured to evaluate the alcoholysis effect, Skin moisture evaporation rate To examine the evaluation unit water content of the skin was improved determine whether the antioxidant and provide skin moisturizing effect. The addition of ethanol extracts of salmon extracts showed a decrease of 5 to 7 times compared with no addition, and a decrease of 3 to 5 times of acetaldehyde. Therefore, hyaluronic acid, which is a polysaccharide polymer, has excellent viscoelasticity and moisturizing ability, It is considered to provide antioxidant and skin moisturizing effect. Therefore, it can be said that the composition containing salmon extract powder and hyaluronic acid as a main ingredient is effective for the hangover phenomenon which occurs after drinking.

  • Purpose : Whole blood interferon-$\gamma$ assay was developed and many studies showed its usefulness in diagnosing tuberculosis (TB) including latent tuberculosis infection (LTBI). However, assessment in children has been limited. This study was undertaken to evaluate the usefulness of QuantiFERON-TB Gold for the diagnosis of LTBI in children exposed to pulmonary TB. Methods : Children who visited Seoul National University Bundang Hospital with a history of TB exposure were enrolled from January 2006 to December 2007. They were evaluated with chest x-rays, tuberculin skin test (TST) and QuantiFERON-TB Gold test. TST was retested 3 months later for those with initial negative reactivity. Definition of LTBI was made on the basis of the TST reactivity. Results : Among the 103 children with a history of TB exposure, 49 children were tested with chest x-ray, TST, and QuantiFERON-TB Gold. Twenty-two were males. Median age was 7.5 years (range; 3 months to 14.7 years). According to TST reactivity, LTBI was in 8 (19%), no infection was in 21 (50%), possible LTBI was in 13 (31%). QuantiFERON-TB Gold test was positive in 5 of the 49 subjects (10%); 3 of the 13 subjects (23.1%) in unknown status, 1 of the 8 subjects (13%) in LTBI, and 1 of the 21 subjects (5%) without infection. The agreement between the QuantiFERON-TB Gold and the TST was poor (${\kappa}=0.101$). Conclusion : QuantiFERON-TB Gold showed poor sensitivity for the diagnosis of LTBI in children with exposure to TB. QuantiFERON-TB Gold alone does not seem to be useful in the diagnosis of LTBI in children.

Radioprotective Effect of Red Ginseng in Irradiated Mice with ${\gamma}$-ray (생쥐에서 홍삼의 감마선조사에 의한 방어효과)

  • Recently, the incidents of direct or indirect radiation exposure due to increase of use of radiation or radioisotope are on the increase in medical and industrial circles. If cells are irradiated, free radicals are created through biological process, and cells are directly or indirectly damaged. This research intends to explore into the effect of saponin at the level of cell (in vitro) and entity (in vivo), using red ginseng extract "saponin", as radioprotective agent. In the experiment implemented at the level of cell (in vitro), degree of cell activity was measures by adding mouse mesenchymal stem cells "C3H/10T1/2 cells" into red ginseng extract "saponin(0, 0.05, 0.2, and 0.4 g/L)", and then the optimal concentration of saponin influencing cells was calculated, in 24, 48, 72, and 96 hours after gamma irradiation at the optimal concentration of saponin, each cell survival rate was observed through XTT assay. The best time period of cultivation for the optimal activity of C3H/10T1/2 cells was as 48 hours, and the degree of optimal activity was shown at 0.05 g/L. In 48 hours after irradiation of 5 Gy to C3H/10T1/2 cells at 0.05 g/L, the degree of activity of cells increased by 10%. In the experiment implemented at the level of entity (in vivo), red ginseng extract "saponin" at a dose of 100 mg/kg/day was injected into the abdominal cavity of six-week immature mouse for two weeks. Right after the last abdominal injection, total body irradiation of gamma rays was carried out at a dose of 5 Gy and 10 Gy. And after irradiation, the blood sample was taken, and then the number of red corpuscles was counted. In result, the decrement of experimental group treated with red ginseng extract "saponin" was 2.3 times larger than that of control group. In view of the results so far achieved, it was revealed that red ginseng extract "saponin" has a radiation exposure protection effect in the experiment implemented at the level of cell (in vitro). In case of animal experiment, the decrement of number of red corpuscles decreased. Finally, it is necessary to carry out more various researches continuously.

Performance Test of Portable Hand-Held HPGe Detector Prototype for Safeguard Inspection (안전조치 사찰을 위한 휴대형 HPGe 검출기 시제품 성능평가 실험)

  • IAEA has employed various types of radiation detectors - HPGe, NaI, CZT - for accountancy of nuclear material. Among them, HPGe has been mainly used in verification activities required for high accuracy. Due to its essential cooling component(a liquid-nitrogen cooling or a mechanical cooling system), it is large and heavy and needs long cooling time before use. New hand-held portable HPGe has been developed to address such problems. This paper deals with results of performance evaluation test of the new hand-held portable HPGe prototype which was used during IAEA's inspection activities. Radioactive spectra obtained with the new portable HPGe showed different characteristics depending on types and enrichments of nuclear materials inspected. Also, Gamma-rays from daughter radioisotopes in the decay series of $^{235}U$ and $^{238}U$ and characteristic x-rays from uranium were able to be remarkably separated from other peaks in the spectra. A relative error of enrichment measured by the new portable HPGe was in the range of 9 to 27%. The enrichment measurement results didn't meet partially requirement of IAEA because of a small size of a radiation sensing material. This problem might be solved through a further study. This paper discusses how to determine enrichment of nuclear material as well as how to apply the new hand-held portable HPGe to safeguard inspection. There have been few papers to deal with IAEA inspection activity in Korea to verify accountancy of nuclear material in national nuclear facilities. This paper would contribute to analyzing results of safeguards inspection. Also, it is expected that things discussed about further improvement of a radiation detector would make contribution to development of a radiation detector in the related field.

  • The low-flow grouting injection technique, the target construction method for this study, is a method of pouring mortar into the ground by non-emission replacement principle, which can be expected to increase the density of the ground, and, in some cases, be used as a base file using the strength of the high injection solids, along with low noise, low pollution, and high durability. To verify that the dynamic characteristics of the ground are improved by the low-flow injection technique, the test work was conducted on the site and physical tests were performed, and the quality of the improvement formed in the ground was verified through the indoor test on the core and core recovery rate was analyzed. The density logs test layer calculated the volume density of the ground layer by using the Compton scattering of gamma-rays, and the sonic logs was tested on the ground around the drill hole using a detector consisting of sonar and receiver devices inside the drill hole. As a result of the measurement of the change in physical properties (density and sonic logs) before and after grouting, both properties were basically increased after infusion of grout agent. However, the variation in density increase was greater than the increase in speed after grouting, and the ground density measurement method was thought to be effective in measuring the fill effect of the filler. Strength and core recovery rates were measured from specimens taken after the age of 28 days, and the results of the test results of the diffusion and strength test of the improved products were verified to satisfy the design criteria, thereby satisfying the seismic performance reinforcement.

Domestic Intercomparison Study for the Performance of Personnel Dosimeters (개인선량계 성능의 국내 상호비교)

  • The Korea Atomic Energy Research Institute(KAERI) conducted a intercomparison study for personnel dosimetry services in Korea to enhance the accuracy and precision of the dosimetry system. Nine types of dosimeters(6 TLD, 3 film badge) from 7 institutions took part in this intercomparison study. Each participant submitted 30 dosimeters including transit control for irradiations. Both TLDs and film badges were irradiated with Cs-137 gamma, Sr/Y-90 beta and 4 X-ray beams in ISO wide series. Four dosimeters were irradiated on phantom with same dose equivalent for each field category. The delivered dose equivalent was in the range of $0.1{\sim}10mSv$. The participants assessed the results of their dosimeter readings in terms of the ICRU operational quantities for personal monitoring, Hp(10) and Hp(0.07). Most participants except 1 dosimeter estimated the delivered dose equivalent with biases less than ${\pm}25%$ for Cs-137 and Sr/Y-90. But for X-rays, the biases exceeded ${\pm}35%$ in some cases bacause the dose evaluation algorithm was based on the ANSI N13.11 X-ray fields which are different from those given by ISO.

Development of a Noble Dosimetry Using Metaphase Analysis and Micronuclei Assay of Bone Marrow Cells in Mice (마우스 골수세포의 중기염색체 분석 및 미소핵 검사를 이용한 피폭선량 평가법의 개발)

  • Purpose: The purpose of this study was to develop in vivo dosimetries using both chromosomal aberrations and micronuclei in mice to assess biological effects of radiations. Materials and Methods: Five each mice were irradiated with 0, 1, 2, 3, 4, 5, 10 Gy of Cs-137 gamma-rays. We scored numbers of chromosomal aberrations in metaphase spreads and numbers of micronuclei in bone marrow smears under light microscope, and obtained the dose-response relationships. We also examined the relationship between the two dose-response curves. Results: The frequency of both chromosomal aberrations and micronuclei increased with dose, in a linear-quadratic manner The delta, beta, and alpha coefficients were 0.0176, 0.0324, and 0.0567 for metaphase analysis (r=1.0, p<0.001) and 0.0019, 0.0073, and 0.0506 for micronuclei assay (r=1.0, p<0.001). The frequency of chromosomal aberrations and micronuclei in different radiation doses was significantly correlated (r=0.99, p<0.01). Conclusion: In vivo dosimetry using either metaphase analysis or micronucleus assay was feasible in mice. These methods could be useful to evaluate biological effects of radiation.

Post-irradiation Changes in Antioxidant Activity of Spindle Tree Leaves from Different Areas (서식지에 따른 사철나무 잎의 이온화 방사선 조사 후 항산화 활성 변화 차이)

  • As the plants grow in a fixed place, they can be a good indicator which reflects the level of environmental pollution. It is necessary for them to develop a strategy to cope with stress under unfavorable environmental conditions. In this study, spindle trees ($Euonymus$ $japonica$) were collected from a clean area (Kijang) as well as a heavily polluted area (Onsan) to check applicability of irradiation combined with plant bioassay to environmental monitoring. The leaves were irradiated with 0, 50 and 100 Gy of gamma rays, and then evaluated for antioxidative capacity with 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay and superoxide dismutase (SOD) analysis. The result shows that there was no significant changes in SOD and EDA (Electron Donationg Ability) in the samples collected from a polluted area. In the meanwhile, SOD increased in the samples from a clean area until 6 to 10 hours after irradiation, then it decreased gradually until 24 hours after irradiation. In conclusion, the plants in the polluted area have developed higher resistance to oxidative stress induced by ionizing radiation than those in the relatively clean area. Irradiation combined with plant bioassay on enzymatic activities and free radical scavenging capacity has proven to be a possible tool for biomonitoring the environmental pollution.

Safety Simulation of Therapeutic I-131 Capsule Using GEANT4 (GEANT4를 이용한 치료용 I-131 캡슐의 안정성 시뮬레이션)

  • Purpose Iodine (I-131) is one of the most widely used radioactive isotopes for therapeutic in the field of nuclear medicine. Therapeutic I-131 capsule is made out of lead to shield high energy radiation. Accurate dosimetry is necessarily required to perform safe and effective work for relative workers. The Monte Carlo method is known as a method to predict the absorbed dose distribution most accurately in radiation therapy and many researchers constantly attempt to apply this method to the dose calculation of radiotherapy recently. This paper aims to calculate distance dependent and activity dependent therapeutic I-131 capsule using GEANT4. Materials and Methods Therapeutic capsules was implemented on the basis of the design drawings. The simulated dose was determined by generating of gamma rays of energy to more than 364 keV. The simulated dose from the capsule at the distance of 10 cm and 100 cm was measured and calculated in the model of water phantom. The simulated dose were separately calculated for each position of each detector. Results According to the domestic regulation on radiation safety, the dose at 10 cm and 100 cm away from the surface of therapeutic I-131 capsule should not exceed 2.0 mSv/h and 0.02 mSv/h, respectively. The simulated doses turned out to be less than the limit, satisfying the domestic regulation. Conclusion These simulation results may serve as useful data in the prediction of hands dose absorbed by I-131 capsule handling. GEANT4 is considered that it will be effectively used in order to check the radiation dose.

Quality Correction for Ir-192 Gamma Rays in Air Kerma Strength Dosimetry Using Cylindrical Ionization Chambers (원통형 전리함을 이용한 Ir-192 선원에 대한 공기커마세기 측정 시 선질보정에 관한 연구)

  • The quality correction in the air kerma dosimetry for Ir-192 using farmer type ionization chambers calibrated by Co-60 quality is required. In this study we determined quality factor ($k_u$) of two ionization chambers of PTW-N30001 and N23333 for Ir-192 source using dosimetric method. The quality factors for energy spectrum of microSelectron were determined as $k_u$=1.016 and 1.017 for PTW-N30001 and N23333 ionization chambers respectively. We applied quality factors in air kerma dosimetry for microSelectron source and compared with reference values. As a results we found that the differences between reference air kerma rate and measured it with and without quality correction were about -0.5% and -2.0% respectively.
