1, 2-Hexanediol과 1, 2-Hexanediol Galactoside의 HaCaT Cell에 대한 세포독성 (Cytotoxic Effects of 1, 2-Hexanediol and 1, 2-Hexanediol Galactoside on HaCaT Cell)

  • 화장품에 방부제(살균/보존제)로 사용되는 1, 2-hexanediol (HD)로 인한 부작용을 극복하기 위하여, Escherichia coli (E. coli)의 ${\beta}-galactosidase$ (${\beta}-gal$)를 이용하여 transgalactosylation 반응으로 1, 2-hexanediol galactoside (HD-Gal)를 합성하였다. 본 연구에서는 합성된 HD-Gal의 인간 피부세포에 대한 독성이 어느 정도인지를 HD와 비교하여 관찰하였다. HD-Gal과 HD의 세포독성은 인간 피부각질형성세포(HaCaT cell line)에 HD와 HD-Gal을 처리한 후, cell proliferation assay를 이용하여 비교 분석하였다. 또한 이때, 위상차 현미경으로 HD-Gal과 HD로 처리한 세포의 상태를 비교 관찰하였다. 그 결과, HD-Gal은 42.2 mM에서 211 mM의 농도 범위에서 세포독성이 관찰되지 않았으며, 현미경 관찰에서도 큰 변화를 관찰할 수 없었다. 그러나, HD의 경우에는 저농도에서(42.2 mM and 84.4 mM)는 세포독성이 관찰되지 않았으나, 고농도(168.8 and 211 mM)에서 매우 높은 세포독성을 나타내었고, 현미경 관찰에서는 고농도에서는 물론이고, 세포독성이 관찰되지 않은 HD의 저농도에서도 세포모양과 세포 수에서의 변화가 관찰되었다. 앞으로 세포독성이 감소된 HD-Gal이 HD의 대체제로서 안전, 건강 및 웰빙 개념의 새로운 용도로 개발될 수 있을 것으로 생각된다.

대장균 베타-갈락토시데이즈를 이용하여 합성된 1, 2-Hexanediol Galactoside의 NMR Spectroscopy 및 Mass spectrometry (NMR Spectroscopy and Mass Spectrometry of 1, 2-Hexanediol Galactoside synthesized using Escherichia coli β-Galactosidase)

  • 화장품용 방부제로 사용하고 있는 1, 2-hexanediol (HD)에 높은 농도의 lactose (300 g/l)를 넣고, 재조합 대장균 ${\beta}$-galactosidase (${\beta}$-gal)를 이용하여 galactose 한 분자를 결합시키는 transgalactosylation 반응을 시켜서, 1, 2-hexanediol galactoside (HD-gal)을 합성하였다. 그리고, 합성된 HD-gal 분자를 확인하기 위하여, HD-gal에 대한 NMR ($^1H$- and $^{13}C$-) 스펙트럼 분석과 mass 스펙트럼 분석을 실시하였다. HD-gal의 $^1H$ NMR 스펙트럼에서 HD에 갈락토실화가 되었음을 보여주는 다양한 피크를 확인하였다. $^1H$ NMR 스펙트럼의 다운필드인 ${\delta}_H$ 4.44 ppm과 ${\delta}_H$ 3.96~3.58 ppm에서 나타나는 다양한 피크들은 HD에 갈락토실화가 되었다는 것을 잘 암시하고 있으며, 또한 $^1H$ NMR 스펙트럼의 업필드에서 나타나는 ${\delta}_H$ 1.60~1.35 ppm과 0.92 ppm의 피크는 HD의 $CH_2$$CH_3$ 작용기로부터 나타나는 피크로써 HD가 본 물질에 존재한다는 것을 나타내고 있다. $^{13}C$ NMR 스펙트럼에서는 HD-gal의 알파-아노머와 베타-아노머의 구조에서 기인하는 총 21의 카본피크가 나타났고, 각 아노머마다 12개의 카본이 존재하는데 이중 ${\delta}_C$ 68.6, 60.9 and 13.2 ppm에 보이는 3개의 카본은 겹쳐서 나타나 총 24개의 피크 중 21개가 나타났다. 또한, 질량스펙트럼 분석에서는 protonated HD-gal인 281.1601 (m/z)의 peak를 확인할 수 있었다. 이를 종합하면, NMR ($^1H$- and $^{13}C$-) 스펙트럼 분석 결과와 질량분석 결과들은 우리가 기대했던 HD-gal의 구조와 매우 잘 일치하고 있다는 것을 알 수 있었다. 앞으로 추가적으로, 세균에 대한 minimum inhibitory concentrations (MICs) 조사와 human skin cell에 대한 독성연구를 추가적으로 진행할 예정이며, 이러한 결과를 근거로 항균력을 유지하면서 피부세포에 대한 독성이 감소된 화장품용 방부제의 연구/개발이 계속되기를 기대하고 있다.

Galectin-1 from redlip mullet Liza haematocheilia: identification, immune responses, and functional characterization as pattern recognition receptors (PRRs) in host immune defense system

  • Galectins, a family of ß-galactoside-binding lectins, have emerged as soluble mediators in infected cells and pattern recognition receptors (PRRs) responsible for evoking and regulating innate immunity. The present study aimed to evaluate the role of galectin-1 in the host immune response of redlip mullet (Liza haematocheilia). We established a cDNA database for redlip mullet, and the cDNA sequence of galectin-1 (LhGal-1) was characterized. In silico analysis was performed, and the spatial and temporal expression patterns in gills and blood in response to lipopolysaccharide polyinosinic:polycytidylic acid, and Lactococcus garvieae were estimated via quantitative real-time PCR. Functional assays were conducted using recombinant protein to investigate carbohydrate binding, bacterial binding, and bacterial agglutination activity. LhGal-1 was composed of 135 amino acids. Conserved motifs (H-NPR, -N- and -W-E-R) within the carbohydrate recognition domain were found in LhGal-1. The tissue distribution revealed that the healthy stomach expressed high levels of LhGal-1. The temporal monitoring of LhGal-1 mRNA expression in the gill and blood showed its significant upregulation in response to immune challenges with different stimulants. rLhGal-1 exhibited binding activity in response to carbohydrates and bacteria. Moreover, the agglutination of rLhGal-1 against Escherichia coli was observed. Collectively, our findings suggest that LhGal-1 may function as a PRR in redlip mullet. Furthermore, LhGal-1 can be considered a significant gene to play a protective role in redlip mullet immune system.

Developmental Patterns of Gal$\beta$1,3(4)GlcNAc $\alpha$2,3-Sialyltransferase (ST3Gal III) Expression in the Mouse: In Situ Hybridization Using DIG-labeled RNA Probes

  • Sialic acids are key determinants for biological processes, such as cell-cell interaction and differentiation. Sialyltransferases contribute to the diversity in carbohydrate structure through their attachment of sialic acid in various terminal positions on glycolipid and glycoprotein (N-linked and O-linked) carbohydrate groups. Gal$\beta$ 1,3(4)GlcNAc $\alpha$2,3-sialyltransferase (ST3Gal III) is involved in the biosynthesis of $sLe^{X}$ and sLe^{a}$ known as selection ligands and tumor-associated carbohydrate structures. The appearance and differential distribution of ST3Gal III mRNA during mice embryogenesis [embryonic (E) days; E9, E11, E13, E15] were investigated by in situ hybridization with digoxigenin-labeled RNA probes coupled with alkaline phosphatase detection. On E9, all tissues were positive for ST3Gal III mRNA expression whereas ST3Gal III mRNA on E11 was not detected throughout all tissues. On E13, ST3GAl III mRNA was expressed in different manner in various tissues. In this stage, ST3Gal III mRNA was positive only in the liver, pancreas and bladder. On E15, specific signal for ST3GAl III was detected in the liver, lung and forebrain. These results indicate that ST3Gal III is differently expressed at developmental stages of mice embryo, and this may be importantly related with regulation of organogenesis in mice.

중합도별 gum류 가수분해 올리고당과 urea관련화합물과의 반응혼합물이 항산화능에 미치는 영향 (Antioxidant Action of Reaction Mixtures of Gums Hydrolysates and Urea Derivatives)

  • $100^{\circ}C$에서 반응시켜 2, 4, 6, 8시간의 갈변도 변화는 ${\beta}-1,4-mannobiose$와 urea의 반응혼합물이 시간의 경과함에 따라 강한 착색도의 증가를 나타냈으나 phenylthiourea, thiourea의 반응액은 착색속도가 늦었다. 같은 조건하에서 $Gal^3Man_4$와 urea 관련화합물의 각 반응혼합물의 갈변도를 나타낸 것으로 urea가 강한 착색도의 중가를 보였고 phenylthiourea, thiourea의 반응액은 비교적 높은 착색도를 보였다. DP 7의 galactosyl mannooligosaccharide와의 각 반응혼합물의 갈변도는 urea의 반응액이 강한 착색을 보였으며 phenylthiourea, thiourea의 반응액의 순으로 착색를 나타내었다. 반응혼합물의 TLC 결과에서 ${\beta}-1,4-mannobiose$와 urea의 반응혼합물 이외에 phenylthiourea, thiourea의 반응액은 새로운 화합물이 생성되었고, $Gal^3Man_4$와 D.P 7 galactosyl manooligosaccharide와의 반응혼합물에서는 모든 반응액에서 새로운 화합물이 출현되었다. 반응혼합물의 환원력은 urea관련화합물 중에서 특히 phenylthiourea, thiourea가 ${\beta}-1,4-mannobiose,\;Gal^3Man_4$, DP 7의 galactosyl manooligosaccharide와의 반응혼합물에서 강한 환원력이 나타났다. Linoleic acid에 대한 반응혼합물의 항산화력 측정에서 ${\beta}-1,4-mannobiose$와 thiourea의 반응혼합물의 항산화력이 Ascorbic acid(AsA)수준의 항산화력을 나타내었고, $Gal^3Man_4$와 thiourea, phenylthiourea의 반응혼합물은 AsA와 유사한 항산화능을 나타내었다. DP 7의 galactosyl manooligosaccharide와 thiourea, phenylthiourea의 반응혼합물의 항산화력은 AsA에 미치지는 못하였다. ${\beta}-1,4-Mannobiose$와 thiourea, phenylthiourea의 반응혼합물이 강한 전자공여능을 나타내고 있고, $Gal^3Man_4$와 phenylthiourea의 반응혼합물 및 D.P 7의 galactosyl manooligosaccharide와 phenylthiourea와의 반응혼합물이 전자공여능을 나타내고 있다.

한국 고유음료류 개발에 관한 연구중 갈수에 관한 연구 (A study on the development of Korean Indigenous beverages in a research on GAL-Soo)

  • 1) The characteristic of the recipe for GAL-SOO is to make odorous, pharmaceutical cereal into plaster by honey-mixed boiling so as to be drunken whenever it needed. But the cereal which included sugar was boiled without adding sugar. 2) The part of cereal which was used mostly in making GAL-SOO was seeds of cereals. 3) The history of GAL-SOO was derived from GU GA PIL YONG, a document of WON dynasty of GHINA, but it was developped according to our taste in this county. 4) The pharmaceutical action of GAL-SOO was mainly to strengthen stomach, digestion and appeasing thirst. 5) GAL-SOO tastes so sour and sweet that it was splendid to drink especially in summer as a beverage. 6) The Value of developping GAL-SOO as a beverage is highly approved because of its expediency of recipe and tough endurance storing under ordinary temperature.

Inhibitory Effects of Oriental Medicinal Herbs on Enzymatic Activity of Sialyltransferases (ST3Gal I and ST6Gal I) Associated with Cancer

  • In many cases of human cancer, the appearance of hypersialylated glycan structures is related to a precise stage of the disease ; this may depend on the elebated sialyltransferase activity during carcinogenesis. The aim of this study was to investigate the inhibitory effects of Oriental medicinal herbs on enzymatic activities of two kinds ofsialyltransferase, Gal $\beta$ 1,3GalNAc$\alpha$2,3-sialyltransferase(ST3Gal I) and Gal $\beta$ 1,4GlcNAc $\alpha$2,6-sialyltransterases(ST6Gal I), which are well known as glycosyltransterases associated with cancer. The aqueous extracts of Scutellaria Baicalensis Georgi, Coptidis Rhizoma, Glycyrrhiza urlensis Fisch, Bupleuri Radix and Platycodi Radix were prepared and tested, respectively. At concentration of 100$\mu$g, Glycyrrhiza uralensis Fisch showed the highest inhibitory effects(about 42% and 57%, respectively) on ST3Gal Iand ST6Gal Iactivities. ST3GAl I was inhibited about 23% by Scutellaria baicalensi G댁햐, but not by the other samples, whereas ST6Gal I was inhibited about 20% and 40%,respectively, by Scutellaria baicalensis Georgi and Bupleuri Radix. All inhibitory effects were obtained in a concentration-dependent manner.

소갈(消渴)에 관한 문헌적(文獻的) 고찰(考察) (Study of oriental medical science documentory records of "So-gal")

  • 1. So-gal IS also called as so-jung, So-dan, Pe-so, Jung-so. Gyuk-so. 2. Cause of So-gal is impairment of Jin-Aek. induced by mal intake. stress, overstrain, intoxication, aging. The process is done by Cho-yeol. 3. Symptoms of So-gal is classified m to three categories Sang-So stands for polydipsia, Jung-So stands for polyphagia and weight loss, and Ha-So stands for polyuria. 4. In treating of So-gal, Chung-Hwa Bo-eum Bo-yang is the main principle. The fundamental basis of treating So-gal is Chi-shin. 5. It is not easy to perfectly classify symptoms of So-gal into three categories Therefore further inquiry is required on classification methods of So-gal.

Massachusetts General Hospital 미니돼지 유래 이종이식용 형질전환 돼지의 번식 특성 분석 (Reproductive Characteristic of Transgenic Massachusetts General Hospital Miniature Pigs for Xenotransplantation)

  • Pigs have been extensively used as mediators of xenotransplantation research. Specifically, the Massachusetts General Hospital (MGH) miniature pig was developed to fix major histocompatibility antigens for use in xenotransplantation studies. We generated transgenic pigs for xenotransplantation using MGH pigs. However, it has not been studied yet whether these pigs show similarity of reproductive physiological characteristics to wild types of MGH miniature pig. In this study we analyzed the estrous cycles and pregnancy characteristics of wild type (WT) and transgenic MGH miniature pigs, which were ${\alpha}1,3$-galactosyltransferase (GalT) heterozygous and homozygous knock-out, and membrane cofactor protein (MCP) inserted in its locus, $GalT^{-MCP/+}$ and $GalT^{-MCP/-MCP}$ pigs. Estrous cycles of WT, $GalT^{-MCP/+}$ and $GalT^{-MCP/-MCP}$ pigs were $20.9{\pm}0.74$, $20.1{\pm}1.26$, and $17.3{\pm}0.87days$, respectively, and periods of estrous were $3.2{\pm}0.10$, $3.1{\pm}0.12$, and $3.1{\pm}0.11days$. The periods of gestation of WT, $GalT^{-MCP/+}$ and $GalT^{-MCP/-MCP}$ pigs were $114.2{\pm}0.37$, $113.3{\pm}0.67$, and $115.4{\pm}0.51days$, respectively. Litter sizes of WT, $GalT^{-MCP/+}$ and $GalT^{-MCP/-MCP}$ pigs were $4.8{\pm}0.35$, $4.8{\pm}1.11$ and $3.0{\pm}0.32$ respectively. There were no significant differences on estrous cycle, periods of estrous and gestation, and litter size among WT, $GalT^{-MCP/+}$ and $GalT^{-MCP/-MCP}$ pigs, meaning that GalT knock-out and additional expression MCP of the MGH miniature pig did not effect on reproduction traits. These results provide relevant information to establish breeding system for MGH transgenic pig, and for propagation of $GalT^{-MCP/-MCP}$ pig to supply for xenotransplantation research.

베타-갈락토시데이즈를 이용하여 합성된 Benzyl Alcohol Galactoside의 NMR Spectroscopy 및 Mass spectrometry (NMR Spectroscopy and Mass Spectrometry of Benzyl Alcohol Galactoside synthesized using β-Galactosidase)

  • 대장균 효소 ${\beta}$-gal를 이용하여 합성된 BzO-gal의 분자구조를 NMR ($^1H$-와 $^{13}C$-)과 고성능 mass spectrometry를 이용하여 분석하였다. BzO-gal은 $^1H$ NMR에서 14개의 proton으로부터 12개의 피크를 나타내었다. 방향족 고리에서 오는 5개의 proton 피크와 벤질기의 $CH_2$에서 오는 2개의 proton 피크는 벤질알코올이 존재함을 나타낸다. 지방족 사슬 영역인 ${\delta}_H$ 4.32 ~ 3.46 ppm에서 나타나는 7개의 proton 피크로부터 단당류가 도입되었음을 확인할 수 있었다. $^{13}C$ NMR 스팩트럼에서 나타난 11개의 carbon 피크도 또한 벤질알코올에 단당이 도입되었음을 나타낸다. BzO-gal의 분자량을 확인하기 위하여 mass spectrometry 로 분석한 결과 m/z가 293.0994인 BzO-gal의 sodium adduct ion($[M+Na]^+$)을 확인할 수 있었다. 이러한 결과를 바탕으로 세포독성이 감소된 첨가물 개발을 기대하고 있으며, 추가적인 후속연구를 진행할 예정이다.
