Chrysanthemum plants are one of the most economically important plants in South Korea. Both virus and viroid can cause diseases and economic damage to the plants. In this study, we investigated the detection of seven viruses and two viroids in 350 chrysanthemum plants cultivated in Yesan-gun, Chungcheongnam-do. Two viruses, chrysanthemum virus B (CVB) and tomato aspermy virus (TAV), and two viroids, chrysanthemum chlorotic mottle viroid (CChMVd) and chrysanthemum stunt viroid (CSVd), were detected in this study. The two viruses were detected in six samples and one sample, respectively. The two viroids were detected in 97 samples and 21 samples, respectively. The nucleotide sequences of the CVB-CN-Y, TAV-CN-Y, CChMVd-CN-Y, and CSVd-CN-Y obtained in this study showed 83.7-86.9%, 99.2-100.0%, 94.4-99.5%, and 95.7-99.7% identity, respectively, compared to their other strains/isolates. The CVB-CN-Y and TAV-CN-Y showed the greatest nucleotide sequence homology to CVB-GS1 and three TAV isolates (TAV-V, TAV-P, and TAV-ChJ), respectively. The CChMVd-CN-Y and CSVd-CN-Y showed the greatest nucleotide sequence homology to CChMVd-Horst and four CSVd isolates (Au1.1, K4pop, Sagae, and Tochigi), respectively. This study is the report on the infection rate of viruses and viroids in chrysanthemum plants cultivated in Yesan-gun in 2021.
A study was carried out to determine the effect of ginseng saponin an its related materials on aflatoxin production by Aspergillus parasiticus NRRL2999 in glucose-salts(GS) medium. Maximal growth of the mold and AF froduction in the medium occurred after 5 and 9 days at $28^{\circ}C$, respectively. When various concentrations of saponin added to the medium aflatoxin synthesis were significantly reduced (p<0.05) compared to the control after 9 days at $28^{\circ}C$. 0.05% of saponin inhibited aflatoxin production most effectively in the low concerntrations of saponin (0.01-0.2%) and the toxin synthesis reduced with an increasing concentrations of saponin in the high concentrations (0.03-5.0%). Various concentrations (0.01-1.0%) of saponin diol and triol in the media also caused to reduce aflatoxin synthesis by the mold (p<0.05). All saponin fractions were found to decrease aflatoxin production significantly. Saponin fraction numbers of 1,2,4 and 6 were shown to reduce aflatoxin production effectively, and the number 1 was the most effective. Addition of 0.05% of nucleic acid related materials to the medium reduced aflatoxin production (p<0.05). Aflatoxins could not be found in broth at all, but in mycelia when 0.05% of caffeine was added to the medium. Aflatoxin synthesis was well correlated with total lipid synthesis, growth and glucose uptake. When aflatoxin synthesis inhibited (5.0% of saponin) both total lipid synthesis and growth were stimulated and the efficiency of glucose utilization was reduced.

No analytical solution exists for evaluating in-situ hydraulic conductivity of vertical cutoff walls by analyzing slug test results with consideration of transient flow. There is an analytical solution proposed to interpret a slug test performed in a partially penetrated well within an aquifer. However, this analytical solution cannot be directly applied to the cutoff wall because the solution has been developed exclusively for an infinite aquifer instead of a narrow cutoff wall. To consider the cutoff wall boundary conditions (i.e, constant head boundary and no flux boundary condition), the analytical solution has been modified in this study to take into account the narrow boundaries by introducing the imaginary well theory. Type curves are constructed from the currently derived analytical solution and compared with those of a partially penetrated well within an aquifer. The constant head boundary condition provides faster hydraulic head recovery curve than the aquifer case. On the other hand, no flux boundary condition leads to slower hydraulic head recovery. The bigger the shape factor and deviation of the well and the smaller the width of the vertical cutoff wall are, the more effect of boundary condition was observed. The type curves obtained from the analytical solution for a cutoff wall are similar to those made by the numerical method in the literature.
Degradation behavior of the three commercial biodegradable polymers, namely poly(3-hydroxybutyrate) (PHB) Sky-Green/sup R/ (SG) and Mater-Bi/sup R/ (MB) was investigated using bacteria isolated from activated sludge and farm soil. Three PHB degrading bacteria, three SG degrading bacteria and one MB degrading bacteria were isolated. The PHB degrading bacteria were identified to be Flavimonas oryzihabitans, Corynebacterium pseudodiphtheriticum and Micrococcus diversus, while Pseudomonas vesicuraris, Pasteurlla multocida and Flavobacterium odoratum were identified as SG degrading bacteria. As for MB, Pseudomonas vesicuraris was isolated. The shake flask test for 28 days indicated that the rate of biodegradation of PHB, SG and MB in terms of weight loss were about 44∼69% 25∼32% and 29% respectively. The surface morphology of PHB, SG andMB films before and after degradation by microorganisms in an activated sludge soil was observed under SEM, demonstrating that the film surface had a very porous structure, and that microorganisms colonized heavily on the film surface. TOC and pH variation as a result of abiotic hydrolysis, or microbial growth in the absence of the polymers were compared to those due to degradation by F. oryzihabitans. Abiotic hydrolysis of PHB was three times as fast as that of SG and MB. Addition of yeast extract to the basal liquid medium accelerated the biodegradation of the polymers. Biodegradation of PHB was always faster than that of SG and MB irrespectively of the presence of yeast extract in the basal liquid medium.
Oxypetalum coeruleum, commonly known as Tweedia, is a perennial herbaceous plant of the Apocynaceae family native to southern Brazil and Uruguay. Tweedia plants are grown as one of the most popular ornamental flowers for floral arrangement in Korea. In May 2021, several tweedia plants in a single greenhouse in Gimje, Jeollabuk-do were found to show virus-like symptoms including necrotic rings, vein-clearing, chlorotic mottle, and mosaic on the leaves, and necrosis on the stems. Here, we have identified tomato spotted wilt virus (TSWV) in symptomatic tweedia leaves by applying high-throughput RNA sequencing. In the result, a single infection by TSWV was verified without mixed infections of different virus species. To confirm the presence of TSWV, a reverse transcription polymerase chain reaction was performed with a specific primer set to the N gene of TSWV. The complete genomic sequence of L, M, and S segments of TSWV 'Oxy' isolate were determined and deposited in GenBank under accession numbers LC671525, LC671638, and LC671639, respectively. In the phylogenetic tree analysis by maximum likelihood method, 'Oxy' isolate showed a high relationship with TSWV 'Gumi' isolate from Gerbera jamesonii in Gyeongsangbuk-do, Korea; for all three RNA segments. To our knowledge, this is the first report of TSWV infection of O. coeruleum in Korea.

The problem of interaction between the structures interconnected at discrete points as like composite structures, has a attracted considerable attention for a prolonged period of time. Recently, mixed structures are applied for overcoming structural limits by developed countries. In this paper, advanced connection type of mixed structures are presented by numerical approach. Also it is performed on extensive literature review from theoretical method to numerical analysis. For analysing behaviors of mixed structures according to connection type, 2 different connections and 1 reinforced connection are compared by 3D nonlinear numerical analysis. Nonlinear analysis of mixed structures is carried out by utilizing contact elements of a general purpose structural analysis computer program(ABAQUS). By using 6 criteria, each connections are investigated. From this result, proper reinforcing and well designed connection type are proposed. And results also show that the deflections which are induced by discontinuity on mixed structures, has a linear distribution that should decrease as applying proposed connection type.
"Sukang", a winter wheat (Triticum aestivum L.) cultivar was developed by the National Institute of Crop Science, RDA. It was derived from the cross "Suwon266" / "Asakaze" during 1994. "Sukang" was evaluated as "Iksan312" in Advanced Yield Trial Test in 2005. It was tested in the regional yield trial test between 2006 and 2008. "Sukang" is an awned, semi-dwarf and hard winter wheat, similar to "Keumkang" (check cultivar). The heading and maturing date of "Sukang" were similar to "Keumkang". Culm and spike length of "Sukang" were 90 cm and 8.1 cm, longer culm length and similar spike length compared to "Keumkang" (80 cm and 7.9 cm, respectively). "Sukang" had similar test weight (819 g/L) and lower 1,000-grain weight (40.2 g) than "Keumkang" (813 g/L and 44.9 g, respectively). "Sukang" showed resistance to winter hardiness and pre-harvest sprouting, which lower withering rate on the high ridge (4.5%) and rate of pre-harvest sprouting (0.2%) than "Keumkang" (21.9% and 30.4%, respectively). "Sukang" had lower flour yield (71.1%) and higher ash content (0.45%) than "Keumkang" (74.1% and 0.42%, respectively). "Sukang" showed lower lightness (89.13) and higher yellowness (10.93) in flour color than "Keumkang" (90.02 and 9.28, respectively). It showed higher protein content (12.8%) and gluten content (11.1%) and lower SDS-sedimentation volume (56.8 ml) and mixing time of mixograph (2.6 min) than "Keumkang" (11.9%, 10.2%, 62.3 ml and 4.7 min, respectively). Fermentation properties, amylose content and pasting properties of "Sukang" were similar to "Keumkang". "Sukang" showed different compositions in high molecular weight glutenin subunits (HMW-GS, $2^{\ast}$, 13+16, 2+12) and puroindolines (pina-1b/pinb-1a) compared to "Keumkang" ($2^{\ast}$, 7+8, 5+10 in HMW-GS and Pina-1a/Pinb-1b in puroindolines, respectively). "Sukang" showed lower hardness (4.53 N) and similar springiness and cohesiveness of cooked noodles (0.94 and 0.63) compared to "Keumkang" (4.65 N, 0.93 and 0.64, respectively). Average yield of "Sukang" in the regional adaptation yield trial was 5.34 MT/ha in upland and 4.72 MT/ha in paddy field, which was 4% and 1% lower than those of "Keumkang" (5.55 MT/ha and 4.77 MT/ha, respectively). "Sukang" would be suitable for the area above $-10^{\circ}C$ of daily minimum temperature in January in Korean peninsula.

A gene coding for an endoglucanase (EglA), of the glycosyl hydrolase family 12 and derived from Aspergillus niger VTCC-F021, was cloned and sequenced. The cDNA sequence, 717 bp, and its putative endoglucanase, a 238 aa protein with a predicted molecular mass of 26 kDa and a pI of 4.35, exhibited 98.3-98.7% and 98.3-98.6% identities, respectively, with cDNA sequences and their corresponding endoglucanases from Aspergillus niger strains from the GenBank. The cDNA was overexpressed in Pichia pastoris GS115 under the control of an AOX1 promoter with a level of 1.59 U/ml culture supernatant, after 72 h of growth in a YP medium induced with 1% (v/v) of methanol. The molecular mass of the purified EglA, determined by SDS-PAGE, was 33 kDa, with a specific activity of 100.16 and 19.91 U/mg toward 1% (w/v) of ${\beta}$-glucan and CMC, respectively. Optimal enzymatic activity was noted at a temperature of $55^{\circ}C$ and a pH of 5. The recombinant EglA (rEglA) was stable over a temperature range of $30-37^{\circ}C$ and at pH range of 3.5-4.5. Metal ions, detergents, and solvents tested indicated a slightly inhibitory effect on rEglA activity. Kinetic constants ($K_m$, $V_{max}$, $k_{cat}$, and $k_{cat}/K_m$) determined for rEglA with ${\beta}$-glucan as a substrate were 4.04 mg/ml, 102.04 U/mg, 2,040.82 $min^{-1}$, and 505.05, whereas they were 10.17 mg/ml, 28.99 U/mg, 571.71 $min^{-1}$, and 57.01 with CMC as a substrate, respectively. The results thus indicate that the rEglA obtained in this study is highly specific toward ${\beta}$-glucan. The biochemical properties of rEglA make it highly valuable for downstream biotechnological applications, including potential use as a feed enzyme.
The purpose of this study was to determine the effect of regular exercise during dexamethasone injection on the body weight, weight of hindlimb muslces, myofibrillar protein content and glutamine synthetase activity. 180-200g female Wistar rats were divided into four groups : control, exercise, dexamethasone injection (dexa), and exercise during dexamethasone injection(D+E) group. The dexa group received daily subcutaneous injection of dexamethasone at a dose of 4mg/kg body weight for 7 days. The exercise group ran on a treadmill for 60min/day(20minutes every 4 hours) at 10m/min and a 10$^{\circ}$grade. The control group received daily subcutaneous injection of normal saline at a dose of 4mg/kg body weight for 7 days. The D+E group ran on a treadmill for 60min/day(20minutes every 4 hours) at 10m/min and a 10$^{\circ}$ grade during dexamethasone injection. Body weight of the control group increased significantly from days of experiment, that of the dexa group decreased significantly from day 4 of the experiment resulting in a 82.4% decrease compared to the first day of the experiment. Body weight of the D+E group decreased significantly from day 5 of experiment resulting in a 81.77% decrease comprared to the first day of the experiment. Body weights, muscle weight and myofibrillar protein content of the plantaris and gastrocnemius decreased significantly and muscle weight of the soleus tended to decrease with dexamethasone injection. Glutamine synthetase activity of the hindlimb muscles increased significantly with the dexamethasone injection. The relative weight of the soleus was comparable to the control group and that of plantaris decreased significantly and that of gastrocnemius tended to decrease compared to that of the control in the dexa group. Body weight and muscle weight of the plantaris and gastrocnemius of the excrcise group were comparable to the control group, and the muscle weight of soleus showed a tendencey to increase. The relative weight of the soleus increased significantly and that of the plantaris and gastrocnemius were comparable to the control in the exercise group. Myofibrillar protein content of the soleus and plantaris increased significantly and there was no change of GS activity of the hindlimb muscles compared to the control in the exercise group. Body weight of the D+E group was comparable to the dexa group, muscle weight of the plantaris increased significantly and that of the soleus and gastrocnemius showed a tendency to increase. The relative weight of the hindlimb muscles increased significantly. Myofibrillar protein content of the soleus and plantaris increased significantly and that of the gastrocnemius tended to increase compared to the dexa group. Body weight and muscle weight of the plantaris and gastrocnemius of the D+E group did not recover to that of the control group. Muscle weight of the soleus recovered to that of the control group. The relative weight and of myofibrillar protein content of the hindlimb muscles recovered to that of the control group. From these results, it is suggested that regular exercise during dexamethasone injection might attenuate the muscle atrophy of the hindlimb muscles.
Vertical cutoff walls such as steel pipe sheet piles (SPSPs) have been commonly applied for the construction of the offshore waste landfill site. Because the SPSPs are sequentially installed by connecting their joints to those of adjacent piles, their mechanical stability should be ensured against the inherent external forces on the sea. The objective of this study is to evaluate the structural performances of the newly developed types of SPSP joint compared with those of other joint types. The problems of the traditional SPSP joints are investigated, and an advanced joint shape of SPSP, which is named double H with L-T (DHLT) joint, are designed for improving the constructability and maintenance. Full-scale models of the DHLT joint are manufactured, and then its joint areas are filled with grout material. After 28 days of curing time, compressive and tensile strength tests were performed on the joint models and the test results were compared with those of the traditional joints. Experimental results show that the structural capacities of the DHLT joint models are lower than those of traditional joints due to the influence of grout and steel members. In the cases of the compressive strength test, especially, bending occurs on steel H-beam with no distinct cracks in grout due to the asymmetrical structure of joint which has no reaction force. This study shows that the performance of the SPSP joint can be improved by considering the influence factors on the structural capacities estimated by the experimental tests.

제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
