Standardizing GC-FID Measurement of Nonmethane Hydrocarbons in Air for International Intercomparison Using Retention Index and Effective Carbon Number Concept

  • Accurate measurements of ozone precursors are required to understand the process and extent of ozone formation in rural and urban areas. Nonmethane hydrocarbons (NMHCs) have been identified as important ozone precursors. Identification and quantification of NMHCs are difficult because of the large number present and the wide molecular weight range encountered in typical air samples. A major plan of the research team of the Climate and Air Quality Taiwan Station (CATs) was the measurement of atmospheric nonmethane hydrocarbons. An analytical method has been development for the analysis of the individual nonmethane hydrocarbons in ambient air at ppb (v) and subppb(v) levels. The whole ambient air samples were collected in canisters and analyzed by GC-FID with $Al_2O_3$/KCl PLOT column. Our targeted for quantitative analysis 43 compounds that may be substantial contributors to ozone formation. The retention indices and molar response factors of some commercially available $C_2{\sim}C_{10}$ hydrocarbons were determined and used to identify and quantify air samples. A quality assurance program was instituted to ensure that good measurements were made by participating in the International Nonmethane Hydrocarbon Intercomparison Experiments (NOMHICE).

수원지역 PM-10 중 다환방향족 탄화수소의 농도 변화에 관한 연구 (Trends in Concentrations of Polycyclic Aromatic Hydrocarbons of PM-10 in Suwon Area)

  • We determined 6 polycyclic aromatic hydrocarbons (PAHs) fluranthene, pyrene, benz(a)anthracene, chrysene, benzo(a)pyrene, dibenzo(a,h)anthracene). A total of 129 samples has been collected from September 1990 to September 1994 on 2 different types of filters (quartz fiber filter, glass fiber filter) by a PM-10 high volume air sampler at the Kyung Hee University-Suwon campus. The organic components in the PM-10 were extracted by an ultrasonication process with benzene:ethanol(4:1, v/v) prior to the analysis by using a GC/FID. We had also investigated the decaying quantity of 6PAHs at the room temperature. For example, chrysene was decayed by 56.7% after 4 days and benzo(a)anthracene by 84.2% after 30 days. All of PAHs were almost completely decayed after a year. We extensively estimated the decay rates by regression analyses for existing 18 raw data sets. Based on the decay rate constants $(\beta)$, pyrene was rapidly decayed by 19.0 $\times 10^{-2}$/day; on the other hand, dibenzo(a,h)anthracene slowly by 0.7 $\times 10^{-2}$/day. Applying the decay rates of PAHs on stored and dated samples, we could reasonably determine annual and seasonal concentration average of PAHs in particulate matters smaller than 10 $\mu$m.

A High-speed Miniature Screening Gaschromatograph with Flame Ionization Detector

  • The combination of Gas chromatography (GC) for separation and Flame Ionization Detection (FID) for detection and identification of the components of a mixture of compounds is a fast and strongly proved method of analytic chemistry. The objective of this research was to design a combined High-speed miniature screening Gas chromatograph along with a Flame Ionization Detector for quick, quantitative and qualitative analysis of gas components. This combined GC-FID system is suitable to detect the volatile and semi-volatile hydrocarbons present in a gas mixture. The construction made it less expensive, easy to use and movable. The complete gas path was developed. On/off valves, temperature and flow sensors and their interface electronics were used for controlling purpose. A Microcontroller was programmed to measure the temperature and gas flow using the sensors and to control and regulate them using the electronics and valves. A pocket PC with its touch screen served as a user interface for the system. Software was developed for the pocket PC, which makes the communication possible with the Microcontroller. The system parameters can be indicated in the Pocket PC as simple text and also the analysis result can be displayed.

Development of primary reference gas mixtures of 18 volatile organic compounds in hazardous air pollutants (5 nmol/mol level) and their analytical methods

  • Volatile organic compounds (VOCs) in hazardous air pollutants (HAPs) have been regulated by the Air Pollution Control Act (1978) and their atmospheric concentrations have been monitored in 39 monitor sites in Korea. However, measurement standards of volatile organic compounds (VOCs) in HAPs at ambient levels have not been established in Korea. Primary reference gas mixtures (measurement standards) at ambient levels are required for accurately monitoring atmospheric VOCs in HAPs and managing their emissions. In this study, primary reference gas mixtures (PRMs) at 5 nmol/mol were developed in order to establish primary national standards of VOCs in HAPs at ambient levels. Primary reference gas mixtures (PRMs) were prepared in pressurized aluminum cylinders with special internal surface treatment using gravimetric method. Analytical methods using gas chromatography-flame ionization detector (GC-FID) coupled with a cryogenic preconcentrator were also developed to verify the consistency of gravimetrically prepared HAP VOCs PRMs. Three different columns installed in the GC-FID were evaluated and compared for the retention times and separation of eighteen target components in a chromatogram. Results show that the HAP VOCs PRMs at 5 nmol/mol were consistent within a relative expanded uncertainty (k=2) of less than 3 % except acrylonitrile (less than 6 %) and the 18 VOCs were stable for 1 year within their associated uncertainties.

저온열탈착기술을 이용한 에틸렌 및 저분자 탄화수소 분석방법 연구 (A Study of Analytical Method for Ethylene and Low Weight Hydrocarbons (LWHC) using Thermal Desorption and Gas Chromatography-Flame Ionization Detector with (TD-GC-FID))

  • In this study, an experimental approach to measure a suite of low weight hydrocarbons was investigated with an emphasis on ethylene (EL) along with many others (ethane (EA), propane (PA), propylene (PL), n-butane (BA), acetylene (AL), methyl acetylene (ML)). Their concentrations were quantified using GC-FID system equipped with thermal desorption (TD) system. The TD-based analysis was conducted using both Link Tube/Thermal Desorber (LT/TD) method and Modified Injection through a Thermal Desorption (MITD) method. The results of these analyses were evaluated in a number of respects. The system allowed the detection of all compounds except methane with the mean response factor (RF) of 10.28 (EA) to 11.94 (PL). The method detection limits of target compounds were seen in the range of 0.027 (ML) to 0.146 ng (BA). The emission flux of some environmental samples (fruits), when measured using a small flux chamber system, fell in the range of 0.14 (AL: Kiwi) to $181ng{\cdot}g^{-1}{\cdot}hr^{-1}$ (EL: Apple Peel). The results of this study confirm that the experimental approach developed in this study allows to accurately measure emissions of low weight hydrocarbons (LWHC) like ethylene from various natural and man-made source processes.

초식동물 쓸개즙 추출물의 천연항산화 성분; 생물학적인 기능 및 특성규명 (Natural Antioxidant Activity of Ethanol Extracted from Bovine Bile ; Biological Effects and Characterization)

  • 본 연구에서는 Bovine bile에서 항산화 물질분획을 분리하고 이에 따른 항산화 효소들의 여러 활성효과를 측정하였으며 GC/FID에 의한 항산화성 물질의 패턴을 확인하였다. 항산화력은 Butylated hydroxy toluene(BHT)에 비해 Bovine bile의 EtOAc추출물(BBE)이 51.2%로 나타났고 xanthine oxidase를 측정 48.3%의 저해효과를 보였으며 glutatione-S-transferase의 활성은 85.7%의 활성이 나타났다. MTT방법에 의한 세포생존율의 조사에서는 6mM glutathione 투여에서 95%라는 생존율과 비교하여 20mg, 60mg의 BBE를 처리했을 때 56%와 65%, 140mg처리에서는 78%의 세포생존율이 나타났다. Bovine bile EtOAc추출물을 TLC로 분석한 결과 Rf값이 0.72에서 항산화 활성이 나타났고 이 분획을 methylation과 trimethysilyl(TMS) 유도체화를 시켜서 Gas Chromatography(FID)와 mass spectrum을 통한 성분 분석을 하고 bile acid의 구성분과 비교하여 계통분석을 실시한 결과 Coprostan-3ol로 추정되었다.

GC/MS를 이용한 잎담배 중 알칼로이드 함량 분석 (Determination of Nicotine and Other Minor Alkaloids in Tobacco Leaves by GC/MS)

  • To obtain the optimum condition for analysis of 10 alkaloids in tobacco leaves, such as nicotine, nornicotine, anatabine, anabasine, myosmine, cotinine, 2,3'-dipyridyl, $\beta-nicotyrine,\;\beta-nornicotyrine\;and\;\beta-formylnornicotin$, 5 types of extraction method were investigated by GC-FID and GC/MS. The optimum condition of alkaloid extraction was achieved by using methanol:dichloromethane(1:3, v/v) after NaOH treatment. The use of mass selective detector (MSD) provided unambiguous nicotine related alkaloid analysis. Alkaloids in various tobacco leaves were extracted with the optimum extraction condition and quantified by GC/MS/SIM mode. Compared with concentrations of alkaloids among the various tobacco leaves, the concentration of alkaloids was generally in the order burley > flue-cured > oriental tobacco. In flue-cured tobacco leaves, the order of concentration of alkaloids was nicotine > anatabine > nornicotine > $\beta-nicotyrine\;>\;\beta-formylnornicotine\; >\;myosmine\;>\;2,3'-dipyridyl\;>\;cotinine\;>\;anabasine\;>\;\beta-nornicotyrine$. However, in the case of burley and oriental tobacco leaves, the concentration of nornicotine was higher than that of anatabine.

Quantitative Analysis of Radiation-induced Hydrocarbons in Irradiated Chicken at Various Dose Levels

  • Gamma-irradiated chicken at dose levels of 0.1 to 10 kGy was subjected to detection of radiation-induced hydrocarbons whether irradiated or not. The hydrocarbons extracted from chicken fat were separated by florishil column chromatography and identified with GC-FID and GC/MS methods. Eight kinds of hydrocarbons were identified from irradiated chicken, among which 1, 7-hexadecadiene and 8-heptadencene were detected as major compounds , Remarkably radiation-induced hydrocarbons in irradiated chiken were detected at 0.5kGy and over. The concentration of radiation-induced hydrocarbons was relatively constant during 16 weeks.

Comparative Studies on Medicinal Constituents of Korean and Chinese Angelicae Dahuricae Radix

  • The medicinal constituents contained in Korean and Chinese Angelicae dahuricae Radix were compared by confirming their qualities. From the extracts of Angelicae dahuricae Radix, oxypeucedanin, imperatorin, and alloisoimperatorin etc. furanocoumarin derivatives were identified by GC/MS analysis. Through GC/FID analysis, the furanocoumarin derivatives content of Baizi cultivated in Korea was more than that cultivated in China, except for one cultivated at Ankuk province, and so confirmed to possess, on the whole, good quality medicinal constituents by content, as compared with the ones cultivated in China.

사망사고 관련된 유해화학 물질 남용자들의 혈액 중 톨루엔과 부탄의 분석 (Analysis of Blood Toluene and Butane in Death Cases of Inhalant Abusers)

  • 혈액 중에서 톨루엔을 HS-SPME 법을 이용하여 GC/MSD로 분석하였으며, 부탄가스류의 분석은 HS 법을 이용하여 CC/FID로 분석하였다. 우리 나라의 경우 지난 3년 동안 청소년들이 본드류나 부탄가스류 등을 흡입한 상태에서 사망한 경우가 총 75건이 발생하였다. 본드류를 흡입하다가 사망한 사람은 27명이었으며, 그 중에서 9명은 톨루엔 중독상태에서 자제력의 상실 등으로 인하여 고층에서 떨어지는 추락사를 하였으며, 이때의 혈중 농도 분포는 $1.3{\sim}21.6{\mu}g/mL$으로 상당히 높은 농도였다. 또한 부탄가스를 흡입하다가 질식 사망 또는 흡입 사망한 사람은 54명이었으며, 이 중에는 본드류와 부탄가스를 동시에 흡입하다가 사망한 6명도 포함되었다.
