Growth Characteristics of Polyporales Mushrooms for the Mycelial Mat Formation

  • Mushroom strains of Polyporales from the genera Coriolus, Trametes, Pycnoporus, Ganoderma, and Formitella were explored in terms of mycelial growth characteristics for the application of mushroom mycelia as alternative sources of materials replacing fossil fuel-based materials. Among the 64 strains of Polyporales, G. lucidum LBS5496GL was selected as the best candidate because it showed fast mycelial growth with high mycelial strength in both the sawdust-based solid medium and the potato dextrose liquid plate medium. Some of the Polyporales in this study have shown good mycelial growth, however, they mostly formed mycelial mat of weak physical strength. The higher physical strength of mycelial mat by G. lucidum LBS5496GL was attributed to its thick hyphae with the diameter of 13 mm as revealed by scanning electron microscopic analysis whereas the hyphae of others exhibited less than 2 mm. Glycerol and skim milk supported the best mycelial growth of LBS5496GL as a carbon and a nitrogen source, respectively.

  • This study investigated the microscopic characteristics and genetic relationships of Ganoderma applanatum fruiting bodies. Basidiospores were brown, ellipsoid, and had one or two large vacuoles and a double wall. The surface of basidiospores was smooth or wrinkled and most had numerous small and shallow holes. The length and width of basidiospores of Ganoderma applanatum isolates GBGA-01, GBGA-02, ASI 50167, ASI 52821, ASI 52822, ASI 52823, and ASI 53399 were on average 7.6×4.8 ㎛, 7.9×4.6 ㎛, 7.7×4.9 ㎛, 8.2×5.3 ㎛, 7.7×5.0 ㎛, 8.0×4.9 ㎛, and 7.9×4.9 mm, respectively. In contrast, the basidiospores of Ganoderma lucidum isolate ASI 7125 were 7.7×5.2 ㎛. Using the universal ITS1/ITS4 primer set, the ITS region of the isolates were amplified and sequenced. The ITS sequences were very closely related to G. applantum isolate GBGA-01, GBGA-02, ASI 50167, ASI 52821, ASI 52822, ASI 52823 and ASI 53399, but were not the same species. Whereas, G. lucidum isolate ASI 7125 belongs to different group.

Effect of Fermented Ginseng Extract by Mushroom Mycelia on Antiproliferation of Cancer Cells (버섯균사체로 발효시킨 인삼 추출물의 암세포 증식억제 효과)

  • This study was conducted to investigate the effects of fermented ginseng extract by mushroom mycelia on antiproliferation of cancer cells. Phellinus linteus, Ganoderma lucidum, and Hericium erinaceum mycelia were inoculated to ginseng. The effects of fermented ginseng extract on antiproliferation of stomach (MKN-45), colon (HCT116), mammary (MCF-7), lung (NCIH460), prostate (PC-3), and liver (HepG2) cancer cells were investigated by MTT assay. Fermented ginseng extract showed significant antiproliferation effects compared with fresh ginseng extract. Fermented ginseng extract by P. linteus, G. lucidum, and H. erinaceum mycelia showed growth-inhibitory effect of 44.50, 17.75 and 43.98% viability at 1.5 mg/mL on the MKN-45 cell line, 62.86, 3.73, and 54.55% at 1.5 mg/mL on the HCT116 cell line, 41.81, 7.01, and 37.84% at 1.5 mg/mL on the MCF-7 cell line, 53.52, 5.31, and 35.27% at 1.5 mg/mL on the NCIH460 cell line, 35.05, 3.07, and 44.29% at 1.5 mg/mL on the PC-3 cell line, and 59.57, 6.34, and 4.97% at 1.5 mg/mL on the HepG2 cell line, respectively. These results indicated that fermented ginseng by G. lucidum mycelium showed the highest antiproliferation effect against various cancer cells.

Single- and Repeated-dose Toxicities of Acanthopanax senticosus Fermentation Products in Rats (흰쥐에서 가시오가피 발효물의 단회 및 반복투여 독성평가)

  • In this study, the products of Acanthopanax senticosus fermentation were derived from the mycelia of 2 mushrooms, Ganoderma lucidum and Phellinus linteus, to determine their safety in Sprague-Dawley rats. Rats were orally administered the water extracts of A. senticosus fermentation products with G. lucidum (FM-5111) or P. linteus (FM-5131) at dose levels of 0.5, 1.0, or 2.0 g/kg for the single-dose toxicity test and 0.25, 0.5, or 1.0 g/kg for the repeated-dose toxicity test. There were no significant differences in body weight gain, feed intake, or water consumption between control and FM-5111- or FM-5131-treated rats. Hematological and blood biochemistry analysis revealed that none of the investigated parameters were affected by the A. senticosus fermentation products, and no remarkable lesions were observed upon histopathological analysis. We conclude that the A. senticosus fermentation products obtained from mushroom mycelia are safe for long-term administration and could be considered as multi-functional nutrients for the improvement of liver function and immunity.

  • The immune activities of Ganoderma lucidum Mycelium added garlic extracts (GAM), Ganoderma lucidum Mycelium (GM), garlic extracts (GS) and standard $({\beta}-glucan)$ were compared. GAM enhanced the growth of human immune T cell up to $1.25{\sim}1.46$ times, compared to control group. GAM showed relatively lower cytotoxicity in using normal human lung cell, while GAM showed the most potent inhibitory effect on the human lung carcinoma, compared to GM and GS. The selectivity of GAM was also higher than that of GM and GS. GAM increased the secretion of cytokines, IL-6 and TNF- from human B cell as well as the growth of human immune cells. It can imply that GAM has higher immune activity than GM or GS.

Mycelial Growth of Ganoderma lucidum and Grifola frondosa in Milk Whey (유청을 이용한 영지버섯과 잎새버섯의 균사체 배양)

  • For the production of mycelia of mushroom by submerged culture, the experiment was carried out. mushroom. The optimum culture broth for Ganoderma lucidum NG-L were compo­sed of CMC 0.8%(w/v) and $(NH_4)_2SO_4$ 0.2%, with 10%(v/v) of inoculum size and pH 5.5 when the milk whey was used as basal medium. In case of Grifola frondosa ATCC48688, the optimum broth were composed of soluble starch 2%(w/v) and $KNO_3$ 0.l%(w/v), with 8%(v/v) of inoculum size and pH 5.2. Among several plant growth hormones,indole-3-acetic acid and gibberellin $A_3-3-acetate$ stimulated the mycelial growth of Ganoderma lucidum NG-L and Grifola frondosa ATCC 48688 respectively. The culture broth of these mushrooms inhibited the growth of B. subtilis and P. aeruginosa.

  • To determine contents of general constituents of Ganoderma lucidum in Korea, the dried carpophores were analyzed. The contents of water, ash, crude lipid, crude protein and crude cellulose were 14.6, 2.0, 3.3, 23.6, and 59.0%, respectively. Among reducing sugars, maltose was the most abundant. Seventeen free amino acids were detected, showing alanine the highest value. The pH of hot water extract was 4.1-4.2. The spores of Ganoderma lucidum was flat and ovoidal long. Their size was $6.3-7.1{\times}3.5-4.3{\times}2.0-2.5\;{\mu}m$ long. To examine effects of life span against sarcoma-180 cells, Fractions A, B and C, were obtained from the extract of Ganoderma lucidum. The survival rates of Fractions A, B and C were 131.7, 162.5, and 141.7 %, respectively. In addition, to examine effects of Fraction B on cell-mediated immunity, delayed type hypersensitivity (=DTH) test was conducted. It restored the suppressed DTH in the sarcoma-180 bearing group up to 66.7%.

Effect of Far-infrared Irradiation on the Antioxidant Activity of Extracts from Phellinus igniarius and Ganoderma lucidum (원적외선 처리가 상황버섯과 영지버섯 추출물의 항산화능에 미치는 영향)

  • The antioxidant activities of Phellinus igniarius (PI) and Ganoderma lucidum (GL) extracts were evaluated after far-infrared (FIR) irradiation. PI and GL were irradiated at $120^{\circ}C$ for 30, 60, 90 and 120 min with a FIR heater (2-14 ${\mu}m$), and then extracted by distilled water for 1 hr. Total sugar contents (TSC), total phenolic contents (TPC), radical scavenging ability (RSA), and reducing power (RP) were significantly increased by FIR irradiation. When PI was irradiated for 120 min, the TSC, TPC, RSA, and RP values of the PI extracts increased from 10.1 to 25.9 mg/g, 6.1 to 13.4 mg/g, 23.8 to 51.5%, and 0.222 to 0.363, respectively, compared to the untreated controls. The GL extracts, under the same conditions ($120^{\circ}C$ for 120 min), also increased in TSC, TPC, RSA, and RP from 11.1 to 22.6 mg/g, 2.5 to 5.8 mg/g, 14.5 to 18.6%, and 0.271 to 0.296, respectively. These results indicate that appropriate FIR irradiation of PI and GL could enhance the antioxidant activities of their extracts by increasing amounts of phenolic and sugar compounds.

  • Ganoderma lucidum의 균사체로 부터 얻어진 G009의 이화학적 특성을 규명하였다. G009는 넓은 범위의 분자량 분포를 가지고 있는 다당체로 분자량 9.4kD의 물질이 주성분이었다 G009의 당의 함량은 약 70%이었고 단백질은 약 2.4%인 것으로 나타났다. G009의 주요 구성당은 glucose, xylose, mannose, galactose 등이었고, glycine, leucine, alanine, valine, isoleucine, proline 등 17종의 아미노산이 확인되었다. 유기원소 분석 결과 G009는 탄소 약 40%, 수소 5.7%, 질소 1.8%의 구성비를 보였으며, 무기원소 분석결과 Ca, Mg, Zn 등이 확인되었다. 이상의 결과로 부터 G009는 9.4 kD의 분자량을 갖는 단백다당체를 주성분으로 하는 당화합물로 추정된다.

Hepatoprotective effect of G009, a protein - polysaccharide fraction from Ganoderma lucidum. (영지 단백다당체 G009의 간장 보호 효과)

  • 정상 동물군의 GOT값 및 GPT 값이 각각 137.8$\pm$23.4 및 67.4$\pm$10.0 이었고 acetaminophen 투여군은 418.6$\pm$69.1 및 447.4$\pm$7.7로 증가하였으나 G009 투여군은 그 증가가 현저히 억제되었다. 즉 G009 10mg/kg 투여군은 GOT 및 GPT 값이 192.6$\pm$19.2 및 144.8$\pm$28.7에 그쳐 각각 GOT 및 GPT 값의 상승이 80.5% 및 79.6% 억제되었고 20mg/kg 투여군은 90.8% 및 86.6% 억제되었다. 그러나 Licovek 은 50mg/kg의 투여군에서 이와 유사한 효과가 관찰되었다. 또한 간 절편의 조직학적 관찰 결과 acetaminophen 투여 대조군이 간세포 괴사등 심한 조직 괴사를 보인 반면 G009 10mg/kg 및 20mg/kg 농도에서는 간장 손상이 매우 경미함을 확인할 수 있었다.
