Studies were conducted to develop new breeding resources using wild soybean (Glycine soja Sieb. & Zucc.) species. In 132 wild soybeans, the average crude protein content was $45.4\%$, ranged from $37.4\%\;to\;50.2\%$ and average crude oil content was $9.3\%$ showing lower than cultivated soybean. The average total amino-acids content was $19,214\;{\mu}g/g$, containing $252\;{\mu}g/g$ of methionine and $103\;{\mu}g/g$ of cysteine, a sulfur containing amino-acid. In ratio of individual amino-acids, the glutamic acid, methionine and cystein content was $16.1\%,\;1.3\%,\;and\;0.5\%$, respectively. The correlation between total amino-acid and sulfur containing amino-acid was not significant. Total isoflavone concentration was, on average, $1,105\;{\mu}g/g$ in wild soybean germplasm. As a result of comparison with total isoflavone and genestein contents, significant positive correlations were observed between total isoflavone and genistein concentration.
The cytotoxic activity of methanol extracts of 25 leguminous seeds in vitro was evaluated by sulforhodamine B assay, using the five human solid A549 lung, SK-OV-2 ovarian, SK-MEL-2 melanoma, XF-498 CNS and HCT-15 colon tumor cell lines. The responses varied with both cell line arid leguminous seed used. Extracts of Canavalia lineata and Glycine soja revealed potent cytotoxic activity against A549 arid SK-MEL-2 cell lines. Moderate activity was observed in the extracts of Cassia obtusifolia and Glyeine max var. chungtae, and C. lineata and Vigna angulasis against SK-MEL-2 and HCT-15 cell lines, respectively. The other seed extracts were ineffective against model tumor cell lines. Because of their potent cytotoxic activities, the activity of each solvent fraction from C. lineata and G. soja was determined and the potent activity was produced from their chloroform fractions. As a naturally occurring therapeutic agent, leguminous seeds described could be useful for developing new types of anti-tumor agents.
In surveys of weed occurrence undertaken from 2006 to 2007, near to the Daegu experimental fields of the National Institute of Crop Science, plants belonging to 31 families, 74 genera and 96 species were found. For the investigation of the natural or alternative hosts of Soybean yellow mottle mosaic virus (SYMMV), 495 plant samples belonging to 26 families 84 species were subjected to RT-PCR. SYMMV was detected only from legume plants such as Glycine soja, Vigna angularis var. nipponensis, Trifolium repens, and Lespedeza cuneata. Among legume plants tested, more than a third of G. soja (wild soybean) contained SYMMV, indicating that the wild soybean played an important role as a reservoir of SYMMV. Wild soybeans may be infected with SYMMV as early as mid-July. Considering the results of early infection and the high infection rate of seed and seed transmission of SYMMV in G. soja, wild soybeans may have played an important role in the completion of disease cycle of the virus.

The variation of the esterase isozyme, germination rate, chemical composition and digestibility of field bean(G1ycine soja S. and Z.) were estimated. The results are as follows; 1. The banding patterns of the esterase isozyme in field bean were varied with the tissue and habitat. 2. The enzyme activity of the Est-I, Est-2, Est-3 and Est-4 in field bean showed a high value compared with the other enzyme. 3. The range of germination temperature in field bean was 10-40C and the optimum germination temperature was 25- 38^{\circ}C.$. 4. The crude protein(CP) contents was 19.9% in the whole plant, 27.8% in the leaf and 45.9% in the seed, the cellulose contents was 29.5% in the whole plant, 23.0% in the leaf and 13.8% in the seed, the neutral detergent fiber(NDF) was 62.6% in the whole plant, 47.9% in the leaf and 47.9% in the seed and the acid detergent fiber(ADF) was 44.5% in the whole plant, 28.4% in the leaf and 28.4% in the seed, respectively. 5. The digestibility of the field bean was 44.1% in the whole plant, 49.6% in the leaf and 75.1% in the seed, NDF was 26.2% in the whole plant 46.2% in the leaf, ADF was 29.0% in the whole plant, 47.7% in the leaf and 58.0% in the seed and Cellulose was 48.7% in the whole plant, 58.0% in the leaf and 70.2% in the seed, respectively. 6. Total digestible nutrients(TDN) of the field bean was 47.4% in the whole plant, 51.5% in the leaf and 70.2% in the seed, respectively. The digestible energy(DE) value was 2.1 kcal/g in the whole plant, 2.27 kcal/g in the leaf and 3.10 kcal/g in the seed and the metabolizable energy(ME) value was 1.72 kcal/g in the whole plant, 1.86 kcal/g in the leaf and 3.23 kcal/g in the seed, respectively.
Soybean sprout trails were evaluated for soybeans for sprout (Glycine max) and wild soybeans (Glycine soja). Soybeans for sprout are characterized with small seed size of 10-14 grams per 100 seeds and are traditionally used only for producing sprouts. No difference was observed in hypocotyl length and sprout yield between two types of soybean. (However total length, hypocotyl hypocotyl, body weight, and the rate of good quality were greater in soybeans for sprout.) Number of lateral roots and root length were greater in wild soybeans. Contents of crude oil, oleic acid, and sucrose were higher in soybean for sprout, while that of protein and linoleic acid were higher in wild soybeans. There was no difference in sprout isoflavone content. Out of 17 amino acids analyzed, 10 amino acids such as aspartic acid, lysine, and arginine. etc. showed significantly Higher content in wild soybeans. Hypocotyl color of sprouts estimated by color meter based on the degree of L(brightness), a(red), and b(yellow) values were better in wild soybeans than soybean for sprout.
This study was conducted to obtain the information of varietal development using wild soybean through investigation of variation and heredity of major agronomic characters in $F_2$ generation of interspecific cross between Glycine max and G. soja. In segregating populations of two crosses, all characters observed except 100-seed weight showed transgressive segregation. Days to flowering showed normal distribution; mean days to flowering in $F_1$, and $F_2$ was approximately mean of parent. Mean of $F_1$ for 100-seed weight was 6.2g and 5.7g for Eunhakong/KLG10084 and Sohaegnamulkong/KLG10084, respectively, which were somewhat skewed distribution to small seeded parents. Mean seed yield of $F_2$ was about mean of two parents. Degree of vine was 6.4 and 5.7 in $F_1$ for two crosses but it was 6.0 in $F_2$ for Eonbakong/KLG10084, which revealed the degree of vine as partial dominance while 4.6 for Sobaegnamulkong/KLG10084, somewhat different results from the previous cross. Broad-sense heritability($h^2B$) for plant height, days to flowering, pods per plant, seed yield, and degree of vine was comparatively high and narrow-sense heritability($h^2B$) for 100-seed weight which is the moot important character in the development of small seed-size sprout soybean was 52.3% and 65.6% for Eunhakong/KLG10084 and Sohaegnamulkong/KLG10084, respectively, which indicated that selection for the character in early generation was possible.
This study was conducted to identify the differences in proteomic characteristics of 'Agakong', recombinant inbred line, and its parental genotypes 'Eunhakong' (Glycine max) and 'KLG10084' (G. soja). The isoflavone content of 'Agakong' was 3 times higher than that of its parental lines. A combined high-throughput proteomic approach was employed to determine the expression profile and identity of proteins using 2-dimensional gel electrophoresis and matrix-assisted laser desorption ionization time-of-flight (MALDI-TOF) mass spectrometry. The overall distribution patterns of proteins are quite similar, but lots of protein spot intensities varied among the genotypes. A total of 41 proteins, representing significant difference in the quantities of protein among the lines, were successfully identified. Among them, more than 50% of the proteins identified were subunits of glycinin and $\beta$-conglycinin, 2 major storage proteins. This study showed that the proteomic analysis could help to define specific changes in protein level and composition, which can occur in the generation of new soybean varieties.

The methanol extracts of 25 leguminous seeds in vitro was evaluated for inhibitory activities against the small intestinal $\alpha$-glucosidase of Sprague Dawley male rats. The responses varied both with leguminous seed types and concentrations used. At the concentration of 0.5 mg/ml, the methanol extracts of Cassia obtusifolia, Glycine max var. yagkong, Glycine max var. hooktae, Glycine max var. geumdu, Glycine max var. mejukong, Glycine soja, Phaseolus multiflorus, Pisum sativum, and Vigna sinensis inhibited over 50% of the enzyme activity. The extracts of G. max var. yagkong and V. sinensis showed relatively strong inhibitory activities against $\alpha$-glucosidase at the concentration of 0.1 mg/ml. The activity of each solvent fraction from G. max var. yagkong and V. sinensis was determined, and potent activities were detected from chloroform and butanol fractions, respectively. $IC_{50}$ values of G. max var. yagkong and V. sinensis were 0.06 and 0.19 mg/ml, respectively. As a naturally occurring therapeutic agents, leguminous seeds examined could be useful for developing new types of antidiabetic agents.
