Effects of Hyolbuchukeo-tang Extracts on Blood and liver of Hyperlipidemia Rats Induced by High Fat Diet (혈부축어탕(血府逐瘀湯) 추출물(抽出物)이 고지방식이(高脂肪食餌)로 유발(誘潑)시킨 고지혈증(高脂血症) 흰쥐의 혈액(血液) 및 간(肝) 기능(機能)에 미치는 영향(影響))

  • Objectives: The purpose of this study was to investigate the effects of Hyolbuchukeo-tang extracts on the hyperlipidemia rats induced by high fat diet. Methods and Materials: In vitro; The extracts prepared for Hyolbuchukeo-tang by hot water extraction (HH), fermentation(HF) and UMPM extraction(HU) method. The extracts were examined for levels of polyphenol compounds, antioxidant activities, and inhibitory potencies for HMG-CoA reductase. In vivo; Sprague-Dawley male rats of weighing $150{\pm}5g$ were randomly divided into six groups ; normal control diet(NC), and high fat diet(HC), high fat diet with treated lovastatin of 10mg/kg(PC), high fat diet with Hyolbuchukeo-tang extracts; HH, HF and HU treated extracts of 300mg/kg, respectively. Also, we compared the effects of the extracts of HH, HF and HU on rats fed high fat diet for four weeks. Results: 1. The content of polyphenol compounds and electron donating abilities(EDAs) was the HF higher than HH and HU. The superoxide dismutase(SOD)-like activities were proportionate in consistency and they appeared highly from all extracts. The HMG-CoA reductase inhibition activities was highest activities in the HU. 2. The activities of serum GOT and GPT were significantly lower in the HH and HF groups. The level of serum triglyceride was significantly decreased in the HF group. HH and HU groups were significantly decreased in the atherogenic index(AI). The total cholesterol concentration in liver was significantly decreased in the HF group, and HU showed more significantly decreased in the triglyceride than of the lovastatin. Also, photomicrographs of liver tissue showed higher fat accumulation in the HC group than in the HH, HF and HU groups. Conclusions: These result suggest that the hyper-lipidemia caused by a high fat diet was effectively inhibited the administration of HF and HU.

Comparative Morphology of Minute Intestinal Fluke Eggs That Can Occur in Human Stools in the Republic of Korea

  • The egg morphology of minute intestinal flukes (MIF) that can occur as human infections in the Republic of Korea, i.e., Metagonimus yokogawai, M. miyatai, M. takahashii, Heterophyes nocens, Heterophyopsis continua, Stellantchasmus falcatus, Stictodora fuscata, Pygidiopsis summa, and Gymnophalloides seoi, was studied in comparison with Clonorchis sinensis. The adult worms were obtained from residents of endemic areas, and their intrauterine eggs were studied and measured using light microscopy; the length, width, length-width ratio (LWR), and Faust-Meleney index (FMI). Several specimens were processed for scanning electron microscopy (SEM), and before gold-coating, the uterine portion of each fluke was etched with a sharp pin in order to expose the eggs. The MIF eggs were ovoid, pyriform, or elliptical with a size range of $21-35{\times}12-21{\mu}m$. S. fuscata eggs revealed the highest FMI (largest in the area) and lowest LWR, whereas P. summa eggs showed the lowest FMI and medium LWR. SEM revealed that G. seoi and S. fuscata had remarkably clean shell surface lacking the muskmelon-like structure which is prominent in C. sinensis eggs. In Metagonimus spp., H. continua, H. nocens, and S. falcatus eggs, minute surface ridges were recognizable though less prominent compared with C. sinensis. On the surface of P. summa eggs, thread-like curly structures were characteristically seen. The results revealed that important differential keys for MIF eggs include the length, width, area (FMI), shape of the eggs, and the extent of the muskmelon-like structure or ridges on their shell surface and operculum.

Effect of Paclobutrazol Application on the Growth and Characters Related with Lodging of Paddy Rice Plant (Paclobutrazol처리(處理)가 수도생육(水稻生育) 및 도복(倒伏) 관련형질(關連形質)에 미치는 영향(影響))

  • This experiment was conducted to find the effect of paclobutrazol application of 12, 18, 24g/10a at 35, 25 and 15 days before heading on the characters related with lodging, growth and yield of paddy rice plants in 1985. Culm lengths were reduced from 7% to 12% by paclobtrazol application of 35, 25 and 15 days before heading. The paclobutrazol caused more short internode, thick well of internode and wide section area of internode than the untreated control. The lodging index were remarkably decreased by paclobutrazol application and also the field lodging of rice plant was not observed. The number of spikelet per panicle was decreased by paclobutrazol application at 35 days before heading and 25, but ripened grain ratio and 1000 grain weight were higher than the untreated control. Paclobutrazol applications increased more from 5% to 8% than untreated control on the grain yield.

Effects of Fermented Milk Containing Herb Extract from Acanthopanax divaricatus var. albeofructus and Codonopsis Ianceolata on the Immune Status of Mouse (흰털오가피와 더덕 추출물을 첨가한 발효유 급여가 마우스의 면역기능에 미치는 영향)

  • We have investigated the immunomodulatory activity of water extracts of Acanthopanax divaricatus var. alveofructus in male ICR mice. Mice were administrated with fermented milk containing freeze-dried extract 3 mg/Kg (A), 9 mg/kg (B), 27 mg/Kg (C) per body weight with A. divaricatus var. alveofructus (loots : leaves : stem) : Codonopsis lanceolata = (5 : 2 : 1.5) : 1.5 for 7 and 10 weeks, respectively. Body weight, relative organ weight, cellularity of lymphoid organs, plaque- forming cell (PFC) assay, agglutination (AGG) test and lymphoproliferation were examined in various groups of animals. Any significant differences of body weight gain were not recorded in the tested ICR mice. There was significant different (p<0.05) in the spleen index in B group of 10 weeks and C group of 7 weeks fed mouse. The thymus gain weight was increased during administration of the extract, but there was no significant increase on other organs gain. Humoral immunity as measured by PFC showed more decreased PFC level in 10 weeks than in 7 weeks. In the HT, A. divaricatus var. albeofructus extract also showed a significant increase (p<0.05) in C group of 10 weeks. Administration of extracts from A. divaricatus var. albeofructus increased significantly in the production of IgG antibodies on the mice immunized with SRBC in B group of 7 and 10 weeks (p<0.05).

Effects of Ethanol and Phenobarbital on Hemoglobin Adducts Formation in Rats Exposed to Direct Black 38 (Direct Black 38 염료를 흰쥐에 투여 시 형성되는 헤모글로빈 부가체에 에탄올과 Phenobarbital이 미치는 영향)

  • Objectives : To evaluate the effects on the formation of benzidine-hemoglobin, and benzidine metabolite-hemoglobin adducts, caused by pretreatment with the known xenobiotic metabolism effectors, ethanol and phenobarbital, in rats administered Direct Black 38 dye. Methods : The experimental rats were divided into three groups: a control group, an ethanol group and a phenobarbital group. Rats were pretreated with ethanol (1g/kg) or phenobarbital (80mg/kg) 24 hours prior to the oral administration of Direct Black 38 (0.5mmol/kg), with the control group being administered the same amount of distilled water. Blood samples were obtained from the vena cava of 5 rats from each group prior to, and at 30 min, 3h, 5h, 9h, 12h, 24h, 48h, 72h, 96h, and 144h following the oral administration of Direct Black 38. Directly after sampling the blood was separated into hemoglobin and plasma, with the adducts being converted into aromatic amines by basic hydrolysis. Hydrolyzed benzidiene, monoacetylbenzidine and 4-aminobiphenyl were analyzed by reverse-phase liquid chromatography with an electrochemical detector, The quantitative amount of the metabolites was expressed by the hemoglobin binding index (HBI). Results : In the ethanol group, benzidine-, monoacetylbenzidine-, and 4-aminobiphenyl-HBI were increased to a greater extent than those in the control group. These results were attributed to the ethanol inducing N-hydrgxylation, which is related to the formation of the hemoglobin adduct, In the phenobarbital group, all the HBIs, with the exception of the benzidine-HBI, were increased to a greater extent than those of the control group. These results were attributed to the phenobarbital inducing N-hydroxylation related to the formation of the hemoglobin adduct. The N-acetylation ratio was only increased with the phenobarbital pretreatment due to the lower benzidine-HBI of the phenobarbital group compared to these of the control and ethanol groups. The N-acetylation ratios for all groups were higher than f for the duration of the experimental period. Although the azo reduction was unaffected by the ethanol, it was inhibited by the phenobarbital, The ratio of the benzidine-HBI in the phenobarbital group was lower than those of the ethanol the control groups for the entire experiment. Conclusion : Our results indicate that both ethanol and phenobarbital increase the formation of adducts by the induction of N-hydroxylation, but also induced N-acetylation. Phenobarbital decreased the formation of benzidine-HBI due to the decrease of the azo reduction. These results suggest that the effects or ethanol and phenobarbital need to be considered in the biochemical monitoring of Direct Black 38.

The Effect of Atorvastatin on the Development of Puromycin Aminonucleoside(PAN)-induced Nephrosis in Rats (Puromycin을 투여한 백서에서 지질 변화가 신증의 진행에 미치는 영향)

  • Purpose : Several studies have suggested that hyperlipidemia might be a causative factor contributing to the progression of initial glomerular injury through the development of glomerulosclerosis. We examined the potential beneficial effect of atorvastatin - which blocks the rate limiting step of cholesterol synthesis by inhibiting HMG-CoA reductase - in PAN-induced nephrosis. Materials and Methods : Glomerulosclerosis was induced in Sprague-Dawley male rats by repeated administration of PAN. Sprague-Dawley male rats were divided into 3 groups : group I(control), group II(PAN 20 mg/kg, subcutaneous injection), group III(PAN 20 mg/kg subcutaneous injection and atorvastatin 50 mg/kg/day per oral). On the 11th week, upon sacrifice of the experimental animals, blood sampling, 24-hr urine collection and nephrectomy were performed. Results : Group III had significantly lower BUN and higher serum albumin($30.9{\pm}17.2\;vs.\;17.3{\pm}2.5\;mg/dL;\;2.3{\pm}0.1\;vs.\;2.5{\pm}0.2\;g/dL$, P<0.05) compared with group II. In the lipid profiles, group III was associated with a reduction in total cholesterol and LDL($291{\pm}173\;vs.\;167{\pm}72\;mg/dL:\;57{\pm}53\;vs.\;27{\pm}12\;mg/dL$, P>0.05) compared with group II. Atorvastatin administration lowered the glomerular sclerosing index significantly(26.2% vs. 13.3%, P<0.05). Conclusion : Puromycin-induced glomerulosclerosis could be ameliorated by the reduction of hyperlipidemia with atorvastatin. This suggests that hyperlipidemia contributes to the pathogenesis of glomerulosclerosis.

Factors Associated with a Prolonged Length of Hospital Stay in Patients with Diabetic Foot: A Single-Center Retrospective Study

  • Background We conducted this study to identify factors that may prolong the length of the hospital stay (LHS) in patients with diabetic foot (DF) in a single-institution setting. Methods In this single-center retrospective study, we evaluated a total of 164 patients with DF, and conducted an intergroup comparison of their baseline demographic and clinical characteristics, including sex, age, duration of diabetes, smoking status, body mass index, underlying comorbidities (e.g., hypertension or diabetic nephropathy), wound characteristics, type of surgery, the total medical cost, white blood cell (WBC) count, C-reactive protein (CRP) levels, erythrocyte sedimentation rate, and albumin, protein, glycated hemoglobin, and 7-day mean blood glucose (BG) levels. Results Pearson correlation analysis showed that an LHS of >5 weeks had a significant positive correlation with the severity of the wound (r=0.647), WBC count (r=0.571), CRP levels (r=0.390), DN (r=0.020), and 7-day mean BG levels (r=0.120) (P<0.05). In multiple regression analysis, an LHS of >5 weeks had a significant positive correlation with the severity of the wound (odds ratio [OR]=3.297; 95% confidence interval [CI], 1.324-10.483; P=0.020), WBC count (OR=1.423; 95% CI, 0.046.0-356; P=0.000), CRP levels (OR=1.079; 95% CI, 1.015-1.147; P=0.014), albumin levels (OR=0.263; 95% CI, 0.113.3-673; P=0.007), and 7-day mean BG levels (OR=1.018; 95% CI, 1.001-1.035; P=0.020). Conclusions Surgeons should consider the factors associated with a prolonged LHS in the early management of patients with DF. Moreover, this should also be accompanied by a multidisciplinary approach to reducing the LHS.

Safety Evaluation of Tobacco Substitute (Herbrette); Inhalation Toxicity, Mutagenicity and Immunotoxicity

  • Inhalation toxicity, mutagenicity, and immunotoxicity tests were performed using a smoke generation system to investigate the safety of Herbrette, a tobacco substitute made with the leaves of Perilla frutescens. ICR mice were exposed to nicotine-free Herbrette smoke with concentrations of 0 (control), 4.08 $\pm$ 1.32 mg/$m^3$ (low dose), 7.72 $\pm$ 2.14 mg/$m^3$ (medium dose) and 12.83 $\pm$ 1.69 mg/$m^3$ (high dose) total particulate matters (TPM) for 4 weeks. When compared to the control group, the body weights, organ weights in the exposed groups did not show any significant differences. However, certain change of several serum chemical data and biochemical parameters were observed, however, the changes were within normal physiological ranges. Moreover, no changes in organ weight, and no gross/microscopic changes were observed between the exposed and control groups. Salmonella typhimurium reverse mutation, in vivo chromosomal aberration and micronucleus assays revealed that Herbrette did not induce mutagenicity. Upon evaluation of peripheral cellular immunity of mice through in vitro lymphocyte proliferation assay, no significant difference was observed in mean stimulation index between the exposed and control groups. Taken together, our results strongly suggest that Herbrette may not cause toxicity on mice under current condition.

The Effect of Continuous Use of Cow Manure on Productivity of Organic Rice and Feed Value, Stock Carrying Capacity of Organic Rice Straw in Central Region of Korea (한국 중부지역에서 우분의 연용이 유기 벼의 생산성 및 유기 볏짚의 사료가치, 가축사육능력에 미치는 영향)

  • This experiments was conducted to investigate the influence on organic rice cultivation by continuous use of cow manure in the central region of Korea and investigated productivity of organic rice and feed value, stock carrying capacity of organic rice straw. In the results, application of chemical fertilizer showed higher one of 1st and 2nd year in fresh matter yield (15.77, 30.30ton/ha), dry matter yield (9.90, 13.89 ton/ha), dry matter yield of rice straw (4.88, 7.15ton/ha), dry matter weight of kernel per plant (29.81, 39.99g) and number of kernel per plant (1212.58, 1701units), but there were not significantly differences with each experimental plots in 3rd year. And harvest index showed higher one in application of chemical fertilizer (0.52, 0.48, 0.43) until 3 years. But their quantitative difference between application of chemical fertilizer and cow manure was gradually decreased each year. And the average value of $K_{CP}$ and $K_{TDN}$ showed higher one in application of chemical fertilizer (2.01, 2.94, 1.95 head/ha/yr) until 3 years. However, quantitative difference between chemical fertilizer and cow manure was gradually decreased each year. Especially, application of cow manure showed higher value of 3rd year in $K_{ME}$, but there was not significantly difference with each experimental plots. According to the results, continuous use of cow manure will be reduced their quantitative differences between chemical fertilizer as the year progress in the central region of Korea in terms of productivity of organic rice and feed value, stock carrying capacity of organic rice straw.

Determination of Optimal Unit Hydrographs and Infultration Rate Functions from Single Rainfall-Runoff Event (단순 강우-유출 사상으로부터 최적단위도와 침투율의 결정)

  • This paper is to present the determination of the optimal Joss rate parameters and urnt bydrographs from the observed single rainfall-runoff event using optimization models coupled with a stochastic technique for the global solution. Two kinds of the linear program models are formulated to derive the optimal unit hydrographs and loss rate parameters for gaged basins; one mimmizes the summation of the absolute residual between predlCted and observed runoff ordinates and the other, the maximum absolute residuaL Multistart algorithm which is one or stochastic techniques for the global optimum is adopted to perturb the parameters of the loss rate equations. Multistart efficiently searches the feasIble region to identify the global optimlUll for loss rate parameters, which yields the optimal loss rate parameters and unit hydrograph for Kostiakov's, Plulip's, and Horton's equation. The unique unit hydrograph ordinates for a gIven rainfall-runoff event iS exclusrvely obtained WIth $\Phi$ index, but unit hydrograph ordinates depend upon the parameters [or each loss rate equations. The parameters of Green-Ampt's are determined through a trial and error method. In this paper the single rainfall-nmoff event observed from a watershed is considered to test the proposed method. The optimal unit hydrograph herein found has smaller deviations than the ones reported previously by other researchers.
