Potential Reasons for Prevalence of Fusarium Wilt in Oriental Melon in Korea

  • This study aims to examine the potential reasons for the current prevalence of the fusarium wilt in the oriental melon. Twenty-seven Fusarium isolates obtained from oriental melon greenhouses in 2010-2011 were identified morphologically and by analysis of elongation factor-1 alpha gene (EF-$1{\alpha}$) and internal transcribed spacer (ITS) rDNA sequences as 6 Fusarium species (8 isolates of F. oxysporum, 8 F. commune, 5 F. proliferatum, 3 F. equiseti, 2 F. delphinoides, and 1 F. andiyazi), which were classified as same into 6 EF-$1{\alpha}$ sequence-based phylogenetic clades. Pathogenicity of the Fusarium isolates on the oriental melon was highest in F. proliferatum, next in F. oxysporum and F. andiyazi, and lowest in the other Fusarium species tested, suggesting F. proliferatum and F. oxysporum were major pathogens of the oriental melon, inducing stem rots and vascular wilts, respectively. Oriental melon and watermelon were more susceptible to F. oxysporum than shintosa and cucumber; and cucumber was most, oriental melon and watermelon, medially, and shintosa was least susceptible to F. proliferatum, whose virulence varied among and within their phylogenetic subclades. Severe root-knot galls were formed on all the crops infected with Meloidogyne incognita; however, little indication of vascular wilts or stem and/or root rots was shown by the nematode infection. These results suggest the current fungal disease in the oriental melon may be rarely due to virulence changes of the fusarium wilt pathogen and the direct cause of the severe root-knot nematode infection, but may be potentially from other Fusarium pathogen infection that produces seemingly wilting caused by severe stem rotting.

국내 수수 알곡에서의 Fusarium속 균의 발생현황 (Occurrence of Fusarium Species in Korean Sorghum Grains)

  • 2017년, 2018년에 단양과 영월에서 재배된 수수에서 1,159점의 Fusarium 균주를 분리하여 유전적, 독성학적, 병리학적 특성을 규명하였다. TEF-1α와 RPB2 유전자를 이용한 계통수 분석결과 수수는 17종의 Fusarium 종들로 오염되었으며, 이 중 F. graminearum, F. proliferatum, F. thapsinum, F. incarnatum, F. asiaticum 등이 주요 우점종이었다. F. graminearum과 F. asiaticum 중에는 F. graminearum-15-acetyl deoxynivalenol 화학형과 F. asiaticum-nivalenol 화학형 균들이 우점하였다. F. thapsinum과 FTSC 11을 제외한 6개의 Fusarium 종이 하나 이상의 독소를 생성하였다. F. proliferatum과 F. fujikuroi 균주 들 중 각각 76.0%와 81.6%가 FUM1 유전자를 가지고 있으며, 일부 균주는 고농도(1,000 ㎍ 이상)의 푸모니신을 생성하였다. F. proliferatum과 F. thapsinum이 다른 종에 비해 높은 수수 병원성을 나타내었다. 이러한 결과는 수수의 Fusarium종들이 여러 종의 독소를 생성할 수 있다는 것을 시사한다.

Fusarium oxysporum Protects Douglas-fir (Pseudotsuga menziesii) Seedlings from Root Disease Caused by Fusarium commune

  • Fusarium root disease can be a serious problem in forest and conservation nurseries in the western United States. Fusarium inoculum is commonly found in most container and bareroot nurseries on healthy and diseased seedlings, in nursery soils, and on conifer seeds. Fusarium spp. within the F. oxysporum species complex have been recognized as pathogens for more than a century, but attempts to distinguish virulence by correlating morphological characteristics with results of pathogenicity tests were unsuccessful. Recent molecular characterization and pathogenicity tests, however, revealed that selected isolates of F. oxysporum are benign on Douglas-fir (Pseudotsuga menziesii) seedlings. Other morphologically indistinguishable isolates, which can be virulent, were identified as F. commune, a recently described species. In a replicated greenhouse study, inoculating Douglas-fir seedlings with one isolate of F. oxysporum prevented expression of disease caused by a virulent isolate of F. commune. Moreover, seedling survival and growth was unaffected by the presence of the F. oxysporum isolate, and this isolate yielded better biological control than a commercial formulation of Bacillus subtilis. These results demonstrate that an isolate of nonpathogenic F. oxysporum can effectively reduce Fusarium root disease of Douglas-fir caused by F. commune under nursery settings, and this biological control approach has potential for further development.

Identification of Fusarium Basal Rot Pathogens of Onion and Evaluation of Fungicides against the Pathogens

  • Onion (Allium cepa L.) is an economically important vegetable crop worldwide. However, various fungal diseases, including Fusarium basal rot (FBR), neck rot, and white rot, reduce onion production or bulb storage life. FBR caused by Fusarium species is among the most destructive onion diseases. In this study, we identified Fusarium species associated with FBR in Jeolla and Gyeongsang Provinces in South Korea and evaluated fungicides against the pathogens. Our morphological and molecular analyses showed that FBR in onions is associated with Fusarium commune, Fusarium oxysporum, and Fusarium proliferatum. We selected seven fungicides (fludioxonil, hexaconazole, mandestrobin, penthiopyrad, prochloraz-manganese, pydiflumetofen, and tebuconazole) and evaluated their inhibitory effects on mycelial growth of the pathogens at three different concentrations (0.01, 0.1, and 1 mg/mL). We found that prochloraz-manganese was highly effective, inhibiting 100% of the mycelial growth of the pathogens at all concentrations, followed by tebuconazole. Fludioxonil showed < 50% inhibition at 1 mg/mL for the tested isolates.

율무 이삭마름 증상에서 분리한 Fusarium속 균의 다양성 및 병원성 (Diversity and Pathogenicity of Fusarium Species Associated with Head Blight of Job's Tears)

  • 율무(Coix lacryma-jobi)는 동아시아에 자생하는 작물로, 종실은 주로 건강식품이나 의약용으로 사용되고 있다. 2006년부터 2008년까지 국내 율무 재배 지역에서 이삭마름 증상이 관찰되었다. 병징은 겉껍질이 변색되고, 내부의 속씨는 마르고 쭉정이가 되는 것이다. 159개의 Fusarium균을 분리하였고, 분리 빈도별로 F. graminearum(34%), F. proliferatum(14.5%), F. verticillioides(10.1%), F. equiseti(6.9%), F. fujikuroi(6.3%) 순으로 분리되었고, 그 밖에 F. subglutinans, F. semitectum, F. poae, F. sporodochioides가 분리되었다. 분리균주의 elongation factor 1 alpha 유전자의 염기서열 분석을 통한 계통분석을 수행한 결과, NCBI GenBank에 등록된 균주와 동일한 것을 확인하였다. 분리균주의 병원성 검정을 수행한 결과, F. graminearum, F. proliferatum, F. verticillioides가 병원성이 강하였다. 이 연구는 Fusarium균에 의해 발생한 율무 이삭마름 증상에 대한 첫 번째 보고이다.

종 특이 primer를 이용한 옥수수 오염 Fusarium verticillioides의 PCR 검출 (Detection of Fusarium verticillioides Contaminated in Corn Using a New Species-specific Primer)

  • Fusarium verticillioides(완전세대: Gibberella moniliformis)는 Gibberellea fujikuroi 종 복합체에 속하는 식물병원균으로서 옥수수의 줄기와 이삭에 썩음병을 일으킬 뿐 아니라 인축에 중독증을 일으키는 fumonisin 곰팡이 독소를 생산한다. 본 연구의 목적은 옥수수에 주로 발생하는 fumonisin 생성가능 G. fujikuori 종 복합체 소속 Fusarium 곰팡이 중 F. verticillioides와 그 외 F. proliferatum, F. fujikuori 등을 서로 구별할 수 있는 종 특이적 PCR primer 조합을 개발하는 것이다. RNA polymerase II beta subunit 유전자(RPB2)의 염기서열로부터 제작된 특이 primer 조합(RVERT1와 RVERT2)은 우리나라 옥수수에서 분리한 잠재적인 fumonisin 생성 G. fujikuori 종 복합체 균주 중 오직 F. verticillioides로부터 208 bp 크기의 단일 DNA 절편을 증폭하였다. 한편 F. verticillioides를 포함한 모든 조사균주는 fumonisin 생합성에 필수적인 FUM1 유전자를 포함하고 있었다. 개발된 특이 primer 조합의 검출한계는 분석 곰팡이 DNA 0.125 pg/${\mu}l$ 수준이었다. 한편, 같은 primer 조합으로 Fusarium spp.에 오염된 옥수수 시료의 게놈 DNA로부터 F. verticillioides 특이 DNA 절편이 증폭되었다. 이와 같은 결과를 종합할 때, 본 연구에서 개발된 primer 조합은 여러 곡물 시료에 오염되어 있는 F. verticillioides 균주의 검출과 종 동정에 유용하게 사용될 것이다.

수수 이삭곰팡이 증상에서 분리한 Fusarium속 균의 다양성 및 병원성 (Diversity and Pathogenicity of Fusarium Species Associated with Grain Mold of Sorghum)

  • 국내의 수수재배는 주로 소면적으로 이루어지며, 대부분 식용이나 사료용으로 사용되고 있다. 2007년부터 2009년까지 전국의 수수 재배 포장을 조사한 결과, 이삭에 곰팡이가 발생하는 증상을 다수 관찰하였다. 병징은 매우 다양한데, 심하게 감염된 이삭의 경우 곰팡이가 발생하였고, 부분적으로 감염된 경우에는 수수 종실이 변색되거나 불임립이 형성되었다. 총 90개의 Fusarium균을 분리하였으며, 형태적 배양적 특성 조사에 의한 동정 결과, F. thapsinum이 41개, F. proliferatum이 23개, F. graminearum이 12개 분리되었으며, F. incarnatum과 F. equiseti가 각각 5개, 3개 분리되었다. Elongation factor 1 alpha 유전자의 염기분석을 이용한 계통분석 결과, 대부분이 NCBI GenBank에 등록된 각 유전자와 일치하는 결과를 나타냈다. 분리된 병원균의 병원성 검정 결과, 우점종인 F. thapsinum, F. proliferatum, F. graminearum종이 병원성이 강한 것으로 조사되었다. 이 연구는 Fusarium균에 의해 발생한 수수 이삭곰팡이증상에 대한 첫 번째 보고이다.

Detection of Fusarium Species by Enzyme-Linked Immunosorbent Assay Using Monoclonal Antibody

  • Enzyme-linked immunosorbent assay (ELISA) was developed for the rapid detection of Fusarium species, known as harmful fungi in food. One of the hybridoma cell lines (lB8) which produced a monoclonal antibody (Mab) specific to Fusarium extracellular polysaccharide (EPS) was screened and the Mab was produced and purified. A detection limit of the sandwich ELISA against F. moniliforme EPS was $0.001\;\mu\textrm{g}/ml$ in the standard curve. Among the 59 strains tested, most Fusarium species showed hight reactivity with Mab lB8, even when the culture broths were diluted 100,000 times. On the other hand, the other genera, except A. versicolor and Trichoderma viride, had no reactivity. When 1 to $100\;\mu\textrm{g}$ of F. moniliforme EPS was spiked into rice, potato, and mandarine orange, the average recoveries were 151%, 84%, and 94%, respectively, determined by sandwich ELISA. The correlation coefficients between the EPS levels determined by sandwich ELISA and the dry mycelial weight of the liquid culture of F. moniliforme, as well as between the EPS and colony forming unit in solid culture of potato, were 0.97 and 0.91, respectively.

Genotypic Identification of Fusarium subglutinans, F. proliferatum and F. verticillioides Strains Isolated from Maize in Austria

  • Gibberella fujikuroi is species complex. This species complex includes Fusarium tabacinum, F. moniliforme(=F. verticillioides), F. nygamai, F. proliferatum as well as F. subglutinans. Our objective was to develop a technique to differentiate between isolates of F. subglutinans, F. proliferatum and F. verticillioides. Thirty-two strains of F. subglutinans, six strains from F. verticillioides and five strains of F. Proliferatum isolated from maize in Austria were studied using random amplified polymorphic DNA(RAPD). F. subglutinans strains clustered very closely, with similarity ranging from $87{\sim}100%$. On the other hand, all the amplification patterns of F. verticillioides were identical, as well as in the case of F. proliferatum. Our results indicated that these Fusaria species are distinct species and hence RAPD markers can be quick and reliable for differentiating them.

  • F. napiforme, F. beomiforme and F. nygamai could not be classified in any of the existing sections of the genus Fusarium. To discuss of the exact taxonomic relationships among these species, the cellular polypeptide patterns were compared by using 2-D PAGE. Polypeptide pattern of F. beomiforme was different from those of other two species and was more similar to F. oxysporum in section Elegans. F. nygamai and F. napiforme might be another same section which would lie between section Liseola and section Elegans. The results were consistent with the comparison of isoenzyme patterns in these species.
