  • Fungicides and antagonists were tested for their efficacy in the management of fungal pathogens of watermelon. The fungal species in different genera were isolated from the seeds of watermelon and their vulnerability was assessed against an array of chemicals and bioagents. Among the fungal pathogens, Fusarium species were effectively controlled by Bavistin. Topsin also showed the promising effects against all the fungal pathogens, and Dithane M-45 effectively controlled Didymella bryoniae. Seed treatment with antagonists like Trichoderma harzianum and T. viride improved the seed germination, seedling vigour and reduced the incidence of seed-borne fungal pathogens. Bavistin and Topsin among chemicals increased significantly the seed germination and vigour index. Trichoderma harzianum showed its efficacy against all Fusarium species and even stood effective than Captan and Blitox. However, Pseudomonas fIuorescens also showed promising effect against Didymella bryoniae over fungicides. Under field condition, Topsin and Dithane M-45 showed better yield than Bioagents.

  • Fungal occurrence during production of dried red pepper was investigated using red pepper samples collected at harvest, before and after washing, and before, during or after drying. Fungal incidence was evaluated by counting the number of fungal colonies grown after incubating random pepper cuts on potato dextrose agar plates. Washing with ground water had no significant effect on reduction of fungal contamination. Fungal increase was observed in some samples, and the insides of washer and containers were contaminated with fungi. Drying caused significant fungal increase regardless of drying method although the fungal incidence after machine drying was lower than that after greenhouse drying. Fungal increase was observed in the samples being dried in a greenhouse and some mycotoxigenic species were also detected. Therefore, the most important control point for fungal contamination during dried pepper production appears to be the drying process, especially in a greenhouse.

  • The mycobiota of the sandy soil of Egyptian beaches was investigated in thirty six sand samples collected from nine different localities in Egypt. The filamentous fungi were identified and assigned to thirty one genera and fifty one species. Greater populations as well as a wider spectrum range of fungal genera and species were obtained in sandy soil of Alexandria beach while Balteem beach was the poorest one. The total count of the genus or species was not depended upon cases of isolation. Most of the genera detected belonged to the Deuteromycotina with fewer proportions belonging to the Ascomycotina and Zygomycotina. The genera of highest incidence and their respective numbers of species were: Penicillium(35.72%, 6 species) and Aspergillus(30.28%, 16 species). The species which showed the highest incidence in all cases was P chrysogenum, followed by P citrinum, A. flavus, Chaetomium murorum and Trichoderma viride. A few number of other genera and species were also detected.

  • In recent years the immunocompromised population has increased rapidly to include people with acquired immune deficiency syndrome (AIDS), drug abusers, and transplant patients. Accordingly, the incidence of intracranial fungal infection has increased. Our institution experienced 2 cases of internal carotid artery (ICA) occlusion due to invasion of the cavernous sinus by an intracranial fungal infection. The first case was a 60-year-old man who presented with headache, eye pain, conjunctival injection, right-sided diplopia, and blurred vision. Infected tissues within the frontal and ethmoid sinuses were removed via bifrontal craniotomy and endoscopic sinus surgery through the Caldwell Luc approach. The second case was a 63-year-old woman who developed right-sided facial pain after a tooth extraction. The infection was not controlled despite continuous use of antifungal agents, resulting in death from sepsis. We believe that when intracranial fungal infection is suspected in a patient with orbital symptoms and a focal neurologic deficit, immediate angiographic investigation of possible ICA occlusion is warranted. Aggressive treatment with antifungal agents is the only way to improve prognosis.

  • This investigation was undertaken to survey toxigenic fungal contamination of various rice samples in 93 rice processing complexes (RPC) in Korea. Rice was grown in 2009 and the samples were collected in 2010. Seven types of rice samples such as unhusked, brown, blue-tinged, discolored, polished, half-crushed, and rice husks were obtained from each RPC. One-hundred and five grains of each sample were placed on PDA plates after surface disinfection. The incidence of fungal contaminants was 26.8%. Aspergillus spp. was the most dominant fungal contaminants and Fusarium spp. was the most frequently occurred in samples. The heaviest Fusarium contamination was found in unhusked grain, rice husks, and bare blue-tinged rice and followed by colored rice whereas broken rice was the least contaminated. Regional difference of fungal contamination was distinctive. Fusarium incidence in the rice samples from southern region of Korea including Jeolla and Gyeongsang Provinces was higher than those from central region including Chungcheong, Gyeonggi, and Gangwon Provinces. In contrast to Fusarium spp., Aspergillus spp. and Penicillium spp. were dominated in brown and polished rice samples and their incidences were more severe in central region than southern region. The major contaminants shown more than 1% of kernels infected were Aspergillus (5.0%), Fusarium (2.0%), Alternaria (1.4%), Dreschlera (1.3%), Penicillium spp. (1.3%), and Nigrospora spp. (1.0%). Collectotrichum, Pyricularia, Myrothecium, Epicoccum, Cladosporium, Moniliella, Gloeocercospora, Chaeto- mium, Curvularia, Phialopora, Acremonium, Gliomastix, Trichoderma, Rhizopus, Phomopsis, Paecilomyces, Genicularia, Geotrichum, Acremoniella, Rhizoctonia, Phoma, Oidiodendran, and Candida spp. were among the rest observed at low incidence. The major contaminants of rice samples were well-known as toxigenic fungal genera so toxin producibility of these fungal isolates is necessary to be examined in future. It is also needed to study Myrothecium spp. on species level as it was detected for the first time in rice.

  • The emergence of multidrug-resistant microorganisms, as well as fungal infectious diseases that further threaten health, especially in immunodeficient populations, is a major global problem. The development of new antifungal agents in clinical trials is inferior to the incidence of drug resistance, and the available antifungal agents are restricted. Their mechanisms aim at certain characteristics of the fungus in order to avoid biological similarities with the host. Synthesis of the cell wall and ergosterol are mainly targeted in clinical use. The need for new approaches to antifungal therapeutic agents or development alternatives has increased. This review explores new perspectives on mechanisms to effectively combat fungal infections and effective antifungal activity. The clinical drug have a common feature that ultimately causes caspase-dependent cell death. The drugs-induced cell death pathway is associated with mitochondrial dysfunction, including mitochondrial membrane depolarization and cytochrome c release. This mechanism of action also reveals antimicrobial peptides, the primary effector molecules of innate systems, to highlight new alternatives. Furthermore, drug combination therapy is suggested as another strategy to combat fungal infection. The proposal for a new approach to antifungal agents is not only important from a basic scientific point of view, but will also assist in the selection of molecules for combination therapy.

  • dsRNA was found in malformed cultures of Lentinula edodes strain FMRI0339, one of the three most popular sawdust cultivated commercial strains of shiitake, and was also found in healthy-looking fruiting bodies and actively growing mycelia. Cloning of the partial genome of the dsRNA revealed the presence of the RdRp sequence of a novel L. edodes mycovirus (LeV), and sequence comparison of the cloned amplicon showed identical sequences sequence to known RNA-dependent RNA polymerase genes of LeV found in strain HKA. The meiotic stability of dsRNA was examined by measuring the ratio of the presence of dsRNA among sexual monokaryotic progeny. More than 40% of the monokaryotic progeny still contained the dsRNA, indicating the persistence of dsRNA during sexual reproduction. Comparing the mycelia growth of monokaryotic progeny suggested that there appeared to be a tendency toward a lower frequency of virus incidence in actively growing progeny.

  • The Korean sawmills have recently recognized the importance of prevention of fungal discoloration due to increased losses in revenue. Before establishing integrated control strategies of fungal discoloration, more complete knowledge about causal organisms is needed. As a first step, we initiated a through survey of fungi colonizing commercially important softwood(Pinus dens flora, Pinus koraiensis, and Pinus radiata) logs and lumber in Korea. In this paper we report results obtained from Japanese red pine(Pinus densiflora) log study. In summer 2000, fungi were isolated from Japanese red pine logs in storage, and identified based on their cultural and morphological characteristics. A total of 595 fungi were isolated, representing 21 genera and 30 species. Mold fungi, mostly Trichoderma species, were the most frequently isolating fungi, representing more than half of all isolates. Dematiaceous fungi represented approximately one fifth of the isolates, and Rhinocladiella atorvirens was the most abundant in all samples. Opiostoma species represented 7% of all isolates from cores planted on malt extract agar(MEA) and the incidence of these species doubled with the addition of streptomycin and cycloheximide to MEA. The results indicate that Japanese red pine sapwood is susceptible to colonization by a variety of fungal species. As a result, control strategies that concentrate on one fungus may have limited success because of interference from competing flora.

  • Plantation forests are established by planting Eucalyptus tree species to provide timber and pulp for the construction industries and to meet the energy needs in Ethiopia. Besides the extensive Eucalyptus plantations in the country, fungal pathogen-related diseases are the main challenges to successful production and management. The disease survey was conducted in the Eucalyptus growing areas of Ethiopia during 2019/2020 and 2020/2021. The objective of this research was to assess the diseases associated with Eucalyptus plant species and identify the causal fungal species. Plants of E. camaldulensis were the dominant species in the survey fields and were severely associated with stem and branch canker diseases. Diseased samples were collected and fungal species were identified as Didymellaceae according to culture morphology and affirmed by internal transcribed spacer sequence analysis. In phylogeny, isolates in the study and a reference strain formed supportive monophyletic clades with strong 90% and 95% support with Didymella coffeae-arabicae and Didymella pinodella respectively. Pathogenicity tests revealed that Didymellaceae could infect E. camaldulensis. The findings are the first reports of Eucalyptus stem canker disease caused by Didymellaceae in Ethiopia.
