Polyphosphate의 진균 성장 억제 작용에 관한 연구 (Studies on Antifungal Effect of Polyphosphate)

  • Polyphosphate의 Candida albicans와 Trichophyton mentagrophytes에 대한 항진균 작용에 대하여 연구하였다. 평균 사슬길이가 15, 45, 75인 polyphosphate는 진균에 대하여 성장억제 효과를 나타낸 반면, pyrophosphate는 성장억제 효과가 없었다. 사슬길이가 증가함에 따라서 진균 성장억제 효과가 증가하는 것으로 관찰되었으며 $800\;{\mu}g/ml$ 농도에서는 균주의 성장을 완전히 억제시켰다. $Mg^{2+}$$Ca^{2+}$의 첨가는 polyphosphate의 성장억제 효과를 감소시켰다. Polyphosphate를 처리한 C. albicans에서는 핵산물질의 유리가 관찰되었으며, 세포 swelling이나 표면돌출과 같은 세포형태 변화도 관찰되었다. 세포막 불투과성 물질인 propidium iodide는 Polyphosphate를 처리한 C. albicans의 핵을 염색하는 것이 관찰되었다. Polyphosphate의 항진균 작용은 polyphosphate가 진균 세포벽이나 세포막의 필수 양이온에 대한 chelation 효과에 의한 것으로 사려된다.

Identification and Characterization of Pathogenic and Endophytic Fungal Species Associated with Pokkah Boeng Disease of Sugarcane

  • Pokkah Boeng is a serious disease of sugarcane, which can lead to devastating yield losses in crop-producing regions, including southern China. However, there is still uncertainty about the causal agent of the disease. Our aim was to isolate and characterize the pathogen through morphological, physiological, and molecular analyses. We isolated sugarcane-colonizing fungi in Fujian, China. Isolated fungi were first assessed for their cell wall degrading enzyme capabilities, and five isolates were identified for further analysis. Internal transcribed spacer sequencing revealed that these five strains are Fusarium, Alternaria, Phoma, Phomopsis, and Epicoccum. The Fusarium isolate was further identified as F. verticillioides after Calmodulin and EF-$1{\alpha}$ gene sequencing and microscopic morphology study. Pathogenicity assay confirmed that F. verticillioides was directly responsible for disease on sugarcane. Co-inoculation of F. verticillioides with other isolated fungi did not lead to a significant difference in disease severity, refuting the idea that other cellulolytic fungi can increase disease severity as an endophyte. This is the first report characterizing pathogenic F. verticillioides on sugarcane in southern China.

진균 세포벽 형성 저해물질 생성 Streptomyces속 세균의 분리 및 수리동정 (Isolation and Numerical Identification of Streptomyces Strains Producing Inhibitors of Fungal Cell Wall Synthesis)

  • The aim of the present research program was to identify and develop strains of actinomycetes producing antifungal antibiotics which inhibit cell wall biosynthesis. 860 strains of Actinomycetes were isolated from various soil samples. Three isolates, EMS4, EMP22, and L234 were selected as the strains producing antifungal antibiotics inducing abnormal morphology against Penicillium cyclopium, Cryptococcus laurentii, and Aspergillus flavus, respectively. Taxonomic unit characters of the strains were tested and the data were analyzed numerically using TAXON program. EMS4, EMP22, and L234 were indentified to be a member of Streptomyces lavendulae, Streptomyces willmorei, and Streptomyces aburaviensis, respectively.

$18{\beta}$-Glycyrrhetinic Acid의 면역보조제효능에 의한 항 전신성캔디다증 효과 ($18{\beta}$-Glycyrrhetinic Acid Induces Protective Anti-Candida albicans Antibody by Its Immunoadjuvant Activity)

  • The role of antibody in the fungal infections is controversial. However, our previous reports showed a certain epitope in Candida albicans cell wall (CACW) induces protective antibody. A major problem is that the epitope isolation requires tremendous time with high cost. This aspect led us to investigate a simple way inducing protective antibodies against C. albicans. In the present study, we determined if $18{\beta}$-glycyrrhetinic acid ($18{\beta}$-GA) from Glabrae Radix (a family of Leguminosae) has immunoadjuvant activity. Data displayed that the $18{\beta}$-GA suppressed proliferations of both T- and Blymphocytes at high concentrations, whereas below 20 ${\mu}M$ concentration the compound supported the proliferations. These observations indicate that $18{\beta}$-GA has immunoregulatory activity. Based on this observation, an immunoadjuvant effect was examined at the low concentration. Results from animal experiments showed that CACW combined with or without $18{\beta}$-GA produced the anti-C. albicans antiserum in mice. Nevertheless, the CACW combined with $18{\beta}$-GA formula only protected mice against disseminated candidiasis (P<0.05). These data implicate that $18{\beta}$-GA has immunoadjuvant activity, which may provoke the CACW antigen to induce protective antibody. Currently, we are investigating possible mechanism of how the $18{\beta}$-GA provokes such protective immunity against the disseminated disease.

Citronellol 및 Thymol이 Candida albicans 세포막 조성에 미치는 영향 (Effects of Citronellol and Thymol on Cell Membrane Composition of Candida albicans)

  • Phospholipids are crucially important in a cell membrane function and could thereby influence antibiotic susceptibility. In order to investigate the antifungal mechanism the total lipid was extracted from C. albicans treated with citronellol or thymol in concentration of their minimum inhibiting concentration and the changes in phospholipids composition were analyzed using ketoconazole as control. The cell growth and total lipid synthesis in cell walls of C. albicans were inhibited by treatment with citronellol. The levels of total lipids were decreased by 35.85% compared to the control. They also showed a significant decrease in the contents of phospholipid, phosphatidylcholine(PC), phosphatidyl ethanolamine(PE) and phosphatidylinositol(PI). As the result of GC assay for total fatty acid methyl esters of PC, PE and PI in C. albicans treated with citronellol, it was found that the major fatty acid composed of three phospholipid were palmitic acid, stearic acid and oleic acid. Moreover, the pattern of the fatty acid compositions of PC, PE and PI were changed by the oil. Based on the results, the anti-Candida mechanism of citronellol or thymol might be closely associated with disrupting the permeability barriers of the fungal cell wall composition or construction.

감마선 조사에 의한 목재성질 변화에 관한 연구 (Study of the Changes in Wood Properties by Gamma Irradiation)

  • 본 연구는 감마선 조사가 소나무(Pinus densiflrora), 느티나무(Zelkovas serrenata), 오동나무(Paulownia tomentosa)의 세포벽 열화, 셀룰로오스 결정화도, 휨 강도에 미치는 영향을 구명하기 위해 수행되었다. 각 목재 시편들을 100 kGy 선량의 감마선 조사 후 주사전자현미경(scanning electron microscope; SEM) 관찰 결과, 목재 세포벽 열화는 관찰되지 않았으며 휨 강도 역시 현저한 변화가 나타나지 않았다. 감마선 조사에 의한 셀룰로오스 결정화도 역시 유의한 변화가 관찰되지 않았다. 이러한 감마선에 대한 목재의 높은 안전성은 감마선 조사 기술을 재질손상 없이 목재를 가해 중인 균류와 충류 구제를 위해 적용할 수 있음을 의미한다.

Ectopic Expression of Wild Rice OgGRP Gene Encoding a Glycine Rich Cell Wall Protein Confers Resistance to Botrytis cinerea Pathogen on Arabidopsis

  • A full-length cDNA of OgGRP gene encoding a glycinerich cell wall protein was isolated from wild rice (Oryza grandiglumis). Deduced amino acid sequences of OgGRP are composed of 148 amino acids (16.3 kDa), and show 85.9% homology with Osgrp-2 (Oryza sativa). RT-PCR analysis showed that RNA expression of OgGRP was regulated by defense-related signaling chemicals, such as cantharidin, endothall, jasmonic acid, wounding, or yeast extract treatment. In relation to pathogen stress, the function of OgGRP was analyzed in OgGRP over-expressing Arabidopsis thaliana. Overexpression of OgGRP in Arabidopsis contributed to moderate resistance against fungal pathogen, Botrytis cinerea, by lowering disease rate and necrosis size. In the analysis of the transgenic Arabidopsis lines to check the change of gene expression profile, induction of PR1, PR5 and PDF1.2 was confirmed. The induction seemed to be caused by the interaction of ectopic expression of OgGRP with SA-and JA-dependent signaling pathways.

Ultrastructure of the Rust Fungus Puccinia miscanthi in the Teliospore Stage Interacting with the Biofuel Plant Miscanthus sinensis

  • Interaction of the the rust fungus Puccinia miscanthi with the biofuel plant Miscanthus sinensis during the teliospore phase was investigated by light and electron microscopy. P. miscanthi telia were oval-shaped and present on both the adaxial and abaxial leaf surfaces. Teliospores were brown, one-septate (two-celled), and had pedicels attached to one end. Transmission electron microscopy revealed numerous electron-translucent lipid globules in the cytoplasm of teliospores. Extensive cell wall dissolution around hyphae was not observed in the host tissues beneath the telia. Hyphae were found between mesophyll cells in the leaf tissues as well as in host cells. Intracellular hyphae, possibly haustoria, possessed electron-dense fungal cell walls encased by an electron-transparent fibrillar extrahaustorial sheath that had an electron-dense extrahaustorial membrane. The infected host cells appeared to maintain their membrane-bound structures such as nuclei and chloroplasts. These results suggest that the rust fungus maintains its biotrophic phase with most mesophyll cells of M. sinensis. Such a nutritional mode would permit the rust fungus to obtain food reserves for transient growth in the course of host alteration.

Ultrastructures of the Loaves of Cucumber Plane Treated with DL-3-Aminobutyric Acid at the Vascular Bundle and the Penetration Sites after Inoculation with Colletotrichum orbiculare

  • Pre-treatment with DL-3-aminobutyric acid (BABA) in the cucumber plants caused the decrease of disease severity after inoculation with anthracnose pathogen Colletotrichum orbiculare. In this study, ultrastructures of the vascular bundle and the infection structures in the leaves of BABA-treated as well as untreated cucumber plants were observed after inoculation with the anthracnose pathogen by electron microscopy. The ultrastructures of vascular bundle in the leaves of BABA-treated plants were similar to those of the untreated plants except plasmodesmata. In the BABA-treated plants, the plasmodesmata were more numerous than in the untreated plants, suggesting that the BABA treatment may cause the active transfer of metabolites through the vascular bundle. In the leaves of untreated plants, the fungal hyphae were spread widely in the plant tissues at 5 days after pathogen inoculation. Most cellular organelles in the hyphae were intact, indicating a compatible interaction between the plant and the parasite. In contrast, in the leaves of BABA pre-treated plants the growth of most hyphae was restricted to the epidermal cell layer at 5 days after inoculation. Most hyphae cytoplasm and nucleoplasm was electron dense or the intracellular organelles were degenerated. The cell walls of some plant cells became thick at the site adjacent to the intercellular hyphae, indicating a mechanical defense reaction of the plant cells against the fungal attack. Furthermore, hypersensitive reaction (HR) of the epidermal cells was often observed, in which the intracellular hyphae were degenerated. Based on these results it is suggested that BABA causes the enhancement of defense mechanisms in the cucumber plants such as cell wall apposition or HR against the invasion of C. orbiculare.

Effect of method of synthesis on antifungal ability of ZnO nanoparticles: Chemical route vs green route

  • To compare the antifungal effect of two nanomaterials (NMs), nanoparticles of zinc oxide were synthesized by a chemical route and zinc oxide-based nanobiohybrids were obtained using green synthesis in an extract of garlic (Allium sativum). The techniques of X-Ray Diffraction (XRD), Infrared (IR) and Ultraviolet Visible (UV-Vis) absorption spectroscopies and Scanning (SEM) and Transmission Electron Microscopies (TEM) were used to determine the characteristics of the nanomaterials synthesized. The results showed that the samples obtained were of nanometric size (< 100 nm). To compare their antifungal capacity, their effect on Cercospora sp. was evaluated. Test results showed that both nanomaterials had an antifungal capacity. The nanobiohybrids (green route) gave an inhibition of fungal growth of ~72.4% while with the ZnO-NPs (chemical route), inhibition was ~87.1%. Microstructural studies using High Resolution Optical Microscopy (HROM) and ultra-structural analysis using TEM carried out on the treated strains demonstrated the effect of the nanofungicides on the vegetative and reproductive structures, as well as on their cell wall. To account for the antifungal effect presented by ZnO-NPs and ZnO nanobiohybrids on the fungi tested, effects reported in the literature related to the action of nanomaterials on biological entities were considered. Specifically, we discuss the electrical interaction of the ZnO-NPs with the cell membrane and the biomolecules (proteins) present in the fungi, taking into account the n-type nature of the ZnO semiconductor and the electrical behavior of the fungal cell membrane and that of the proteins that make up the protein crown.
