Antioxidant effect of complex extracts from pine needle, green tea, and sea buckthorn leaves (솔잎, 녹차 및 비타민나무 잎 혼합추출물의 항산화효과)

  • This study investigated the antioxidant effects of complex extracts from pine needles (PN), green tea (GT), and sea buckthorn leaves (SL). Measurement of total polyphenol and flavonoid content, DPPH radical scavenging activity (DPPH), ABTS+ radical scavenging activity (ABTS), and reducing power (RP), showed that SL extract (SE) had the highest values. The superoxide radical scavenging activity (SSA) and nitrite scavenging activity (NSA) were the highest in the GT extract (GE). In most experiments, the PN extract (PE) showed higher activity in complex extracts than in single extracts. The combination of GT and SL led to a higher activity than that exhibited by GT (DPPH and ABTS at 300 ppm, RP at 100 and 500 ppm) and SL (SSA at 100 ppm and NSA at 300 ppm) alone. These results suggest that the combination of PE, GE, and SE may be a useful functional food material in the food industry.

The immune enhancement effect of Cheonggukjang Water Extract (CWE) via activation of NF-κB pathways in murine macrophage RAW 264.7 cells (RAW 264.7 대식세포에서 청국장 열수 추출물(Cheonggukjang Water Extract, CWE)의 면역 증강 효과)

  • Due to the COVID-19 pandemic, the immuneenhancing health functional food market that protects our bodies from pathogens such as viruses continues to grow. In this study, we aimed to prove the Cheonggukjang, a high-nutrient food with high protein, fat, and dietary fiber content, as an immuneenhancing nutraceutical. Cheonggukjang water extract (CWE) increased the production of nitric oxide, reactive oxygen species, and cytokines such interleukin (IL)-6, IL-1β, and tumor necrosis factor-α without affecting viability in RAW 264.7 cells. Furthermore, CWE significantly upregulated the expression of inducible nitric oxide synthase and cyclooxygenase-2 in RAW 264.7 cells. CWE enhanced the phosphorylation of I kappa B kinase α/β and I kappa B (IκB)α, as well as the degradation of IκBα. CWE also induced increased phosphorylation of nuclear factor-kappa B p65 and facilitated the redistribution of p65 from the cytoplasm to the nucleus in RAW 264.7 cells. These findings suggest that CWE has potential as a health functional food material that can enhance the innate immune response.

Effects of Salt Concentration in Soybean Sauce on the Physicochemical Properties of Pre-rigor Ground Hanwoo Muscle

  • This study was conducted to examine the effect of salt concentration in soybean sauce (ganjang) on pre-rigor ground Hanwoo (Korean native cattle) muscle. Although adding soybean sauce reduced pH of pre-rigor Hanwoo muscle, it improved the water holding capacity (WHC), cooking loss, total protein solubility, myofibrillar fragmentation index (MFI), 2-thiobar-bituric acid (TBA) values, and textural properties by increasing salt concentration in the soybean sauce. In particular, adding soybean sauce inhibited lipid oxidation despite increasing salt concentration. An increased salt concentration in the soybean sauce in pre-rigor Hanwoo muscle tended to make the meat have lower lightness and higher redness and yellowness values. Although soybean sauce had a significantly lower pre-rigor salting effect than sodium chloride in terms of cooking loss and total protein solubility (p<0.05), soybean sauce improved myofibrillar fragmentation and lipid oxidation when compared with sodium chloride. Furthermore, no significant differences in textural properties were observed between adding soybean sauce and sodium chloride at the same salt concentrations. Therefore, soybean sauce can be a functional curing material for pre-rigor muscle.

Inhibitory Activity of Intracellular Lipid Accumulation by Various Marine Extracts in HepG2 Cells (HepG2 cell에서 식용수산자원 추출물의 지질축적억제효과)

  • This study was performed to promote the effective utilization of edible marine resources and to develop functional food material from edible marine resource extracts for inhibiting lipid accumulation in liver. Edible marine resource extracts (ME) were prepared by hot water (MWE) and 80% ethanolic (MEE), and both the MWE and MEE extracts were assessed as to their cell cytotoxicity, and Oil Red O staining. Results demonstrated that ME showed no cytotoxic effects. However, treatment with MEE in the concentration of 1000, 500, and 250 ${\mu}g/mL$ significantly inhibited the lipid accumulation in HepG2 cells compared to MWE. Especially, among the 80% ethanolic extracts, Pagrus major, Larimichthys polyactis, Clupea pallasii, Octopus minor, Enteroctopus dofleini, Styela clava, dried sea mustard, and Enteromorpha intestinalis showed greater lipid accumulation inhibitory activity than the others.

Whitening effects of fermented Trigonotis radicans var. sericea with Lactobacillus brevis in α-MSH-stimulated B16F10 melanoma cells

  • This study was designed to compare the whitening effects of 60% ethanol extracts of Trigonotis radicans var. sericea (TR) and Lactobacillus brevis-fermented T. radicans var. sericea (FTR). Measurement of cytotoxicity in B16-F10 melanoma cells to confirm the whitening effect, FTR showed higher cell viability than TR. FTR showed inhibitory activity on melanin contents similar to the normal group at concentrations of 50 and 100 ㎍/mL. MITF expression was used to confirm the effect on melanogenesis-related protein expression. TR and FTR showed significant concentration-dependent decrease, and FTR showed lower expressions than the normal group at concentrations of 25, 50, and 100 ㎍/mL. Additionally, the mRNA expression of melanogenesis-related genes (MC1R, Rab27a, TGF-β1 and Myo5a) were measured by RT-qPCR to confirm the whitening effect. In MC1R expression at a concentration of 100 ㎍/mL in FTR showed effective inhibitory activities, and in TGF-β1 expression, TR and FTR both showed effective activities compared to normal groups even at low concentrations. Results of myo5a and Rab27a, a similar pattern was shown, and FTR showed effective inhibitory activities at a concentration of 100 ㎍/mL. As a result, FTR had higher whitening effects through bioconversion and is expected to be a good material for whitening functional cosmetics.

Investigation of in vitro Antigenotoxic Effect of Korean Mistletoe(Viscum album coloratum) Using Comet Assay (Comet assay에 의한 한국산 겨우살이(Viscum album coloratum)의 항유전독성 규명)

  • To investigate the antigenotoxicity of Korean mistletoe using Comet assay, the crude extract was divided into 4 fractions, i.e. fraction I (MW range over 14,000), fraction II $(8,000{\sim}14,000)$, fraction III $(3,500{\sim}8,000)$, and fraction IV (below 3,500) by molecular weight fractionation. In the non-tumoral 3T3 cells, fraction IV could reduce DNA damage induced by MNNG in a dose dependent pattern while fraction I and III which were known to contain lectins and viscotoxins, respectively, did not show the activity. By heat treatment, the antigenotoxic activity of faction IV, though was gradually diminished according to heating time, was found to be maintained significantly. From the Sephadex G-25 gel filtration chromatography, a more purified fraction responsible for the activity of faction IV was obtained from the latter part of total elute. Therefore, it was concluded that the antigenotoxic components of Korean mistletoe were water soluble substances of MW below 1,000 and there is a possibility of utilization as a material of functional foods for chemoprevention.

Quality of Extracts from Domestic Stevia(stevia rebaudiana BERTONI) Leaves with Harvest Time (수확시기별 국내산 Stevia 추출물의 품질특성)

  • Consumers were tend to purchase more natural sweeting materials then atificial sweeting materials because of safety and the consumed amount of natural sweeting material was increased sharply with consumers concern to health. The proper harvest time with changes of sweeting material contents was investigated in the purpose of cultivation and utilization on functional foodstuff, Soluble solid content and extraction yield were increased with growth priod, but pH was decreased. Color value, transmittance and browning color showed little. L, a, b values was changed a little. Stevioside ratio($68.97\%$) in sweetness components was decreased to $61.09\%$. Rebaudioside A ratio was increased from $20.83\%\;to\;27.52\%$ stevioside contents was maximized to $12.80\%$ of August. The highest sweetening potencies of stevia leaves was 2530times to sugar. The stevia leaves harvested at May showed the best deliciousness.

A Study on the Antibacterial and Antifungal Properties of Zeolite/Zinc-polypeptide Coated Polypropylene Film (Zeolite/Zinc-polypeptide를 코팅한 폴리프로필렌필름의 항균 및 항진균 특성에 관한 연구)

  • This study is for the application of functional antibacterial packaging to fresh food. Zeolite/Zinc-polypeptide was coated on PP film at concentrations of 5%, 10%, and 15%, degree of dispersion was verified through FESEM and FT-IR analysis. In addition, the antibacterial and antifungal properties of the films were analyzed according to the control group and the concentration of coating materials. As a result, the degree of dispersion of coating material was irregular but wide, depending on the concentration of Zeolite/Zinc-polypeptide on the surface of PP film. The antibacterial effect against E. coli was over 99.9%, and the growth of R. oryzae was inhibited about 70%. Therefore, it was confirmed that Zeolite/Zinc-polypeptide had antibacterial and antifungal properties against E. coli and R. oryzae even after coated on PP film. In conclusion, Zeolite/Zinc-polypeptide coating film is expected to be effective in preventing corruption and improving the shelf life of fresh food as a functional packaging material. In order to be applied to various fresh foods in the future, storage experiments are additionally required with temperature and humidity conditions according to fresh foods.

Corn silk extracts did not alter the adiposity and underlying substrate utilization in high-fat diet-induced obese C57BL/6 mice

  • Corn silk (Okmi-su) was anciently adopted as a material for tea or beverage. Corn silk extracts (CSE) contain bioactive phytochemicals such as phenolic acid, flavonoids, ascorbic acid, tannins, and glycosides. Under the impact of these functional components, CSE has benefits for antioxidation, diuresis, anti-diabetes, and dyslipidemia recovery. Nonetheless, its role in whole-body adiposity was not investigated; therefore, the effects of CSE on obesity were evaluated in high-fat diet-induced obese mice. Mice were assigned to either group (n=12); 1) normal diet (18% kcal from fat), 2) high-fat diet (45% kcal from fat, the control), 3) high-fat diet with CSE (800 mg/kg diet), and 4) high-fat diet with orlistat (500 mg/kg diet, a comparable control for weight loss). Our results showed that body weight, adiposity, and energy expenditure in obese mice were not altered by CSE. Lean body mass tended to decrease by CSE, which can be explained by stimulation of diuresis (p=0.06). In conclusion, our results suggest that dietary consumption of CSE does not influence the adiposity and underlying substrate utilization in high-fat diet-induced obese mice.

Quality Characteristics and Antioxidant Activities of Chocolate Added with Mulberry Pomace (오디박 분말 첨가 초콜릿의 품질 특성 및 항산화성)

  • Mulberry pomace, a by-product obtained from mulberry juice, has not been utilized as a food material for food processing. In this study, the quality characteristics and antioxidant activities of chocolate to which mulberry pomace has been added were evaluated. Freeze-dried mulberry pomace was added to white chocolate at the level of 0, 0.5, 1.5 or 2.5%, and then the white chocolate ganache was put between two layers of dark chocolate. The moisture content and sweetness were not significantly different among treatments, but reducing sugar content was increased, according to the amount of mulberry pomace powder. The Hunter color L(lightness) and b(yellowness) values decreased, according to the mulberry pomace powder amount, whereas the a(redness) increased. The hardness increased according to the mulberry pomace powder amount. Total phenol and flavonoid contents increased, according to the mulberry pomace powder amount. Antioxidant activities(DPPH and hydroxyl radical scavenging activities) also increased, according to the mulberry pomace powder amount. The result of the sensory intensity test revealed that the scores of color, mulberry taste, hardness and chewiness were the highest in the 2.5% mulberry pomace powder added chocolate. Sensory preference test results showed that chocolate added with 1.5% mulberry pomace powder had the highest scores in color, flavor, taste, texture and overall preference. It is suggested that chocolate to which mulberry pomace powder has been added may be a functional chocolate, with high antioxidant activity.
