The Effects of Dietary Fermented Fruit Pomace and Angelica keiskei Koidz Pomace on Shelf Life, Cholesterol and Fatty Acid Composition in Broiler (발효 과일박 및 신선초박의 급여가 계육 내 지방산 조성, 콜레스테롤 및 저장 기간 중 지방산패도에 미치는 영향)

  • This study investigated the effects of dietary supplementation of fermented apple pomace (FAP), fermented pear pomace (FPP), fermented orange pomace (FOP), and fermented Angelica keiskei Koidz pomace (FAKP) on performance, shelf life, fatty acid composition and cholesterol in broiler chickens. A total of 600, 1-day-old male broiler chicks (Cobb strain) were randomly divided into six groups with four replicates of 30 birds each. There were five treatment groups: control (C), FAP (1.0%, T1), FPP (1.0%, T2), FOP (1.0%, T3), and FAKP (1.0%, T4). The body weight of the broiler chickens fed FAP diet was higher (1,758 g) than the other treatments. There was no difference in the thiobarbituric acid reactive substances (TBARS) in chicken meat between all groups at days 1, 3, and 5 of storage, while the FAP-supplemented group displayed lower TBARS values at day 7. There was no significant difference in fatty acid composition between the groups but the cholesterol content of chicken meat was lower than the control groups. These results suggest the possibility that FAP could be used as a functional feed to improvement the quality performance of broiler chickens.

Dietary maximum exposure assessment of vitamins and minerals from various sources in Korean adolescents (한국 청소년의 다양한 급원을 통한 비타민과 무기질 최대섭취량 평가)

  • Dietary supplement use is prevalent and represents an important source of nutrition. This study was conducted in order to assess the dietary maximum exposure of vitamins and minerals from various sources including regular diet, vitamin mineral supplements for non-prescription drug (VMS-NPD), vitamin mineral supplements for health functional foods (VMS-HFF), and fortified foods (FF). A total of 1,407 adolescent boys and girls attending middle or high schools were chosen from various cities and rural communities in Korea. Users of vitamin and mineral supplements (n = 60, 15-18 years of age) were chosen from the above 1,407 students. Intake of vitamins and minerals from a regular diet and FF was assessed by both food record method and direct interview for three days of two weekdays and one weekend, and those from VMS-NPD and VMS-HFF were assessed by both questionnaire and direct interview, and compared with the recommended nutrient intake (RNI) and the tolerable upper intake level (UL) for Korean adolescents. Daily average exposure range of vitamins and minerals from a regular diet was 0.3 to 4.4 times of the RNI. Some subjects had an excessive exposure to the UL in the following areas: from regular diets, vitamin A (1.7%) and niacin (5.0%); from only VMS-NPD, vitamin C (9.1%) and iron (5.6%); and from only VMS-HFF, niacin (8.6%) > vitamin $B_6$ (7.5%) > folic acid (2.9%) > vitamin C (2.3%). Nutrients of daily total intake from regular diet, VMS-NPD, VMS-HFF, and FF higher than the UL included nicotinic acid for 33.3% of subjects, and, then, in order, vitamin C (26.6%) > vitamin A (13.3%), iron (13.3%) > zinc (11.7%) > calcium (5.0%) > vitamin E (1.7%), vitamin $B_6$ (1.7%). Thus, findings of this study showed that subjects may potentially be at risk due to overuse of supplements, even though most of them took enough vitamins and minerals from their regular diet. Therefore, we should encourage adolescents to have sound health care habits through systematic and educational aspects.

Effects of Green Tea Powder on the Disorders of Lipid Metabolism and Hepatic Functions in Rats treated by 2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD) (녹차가 다이옥신계 TCDD(2,3,7,8-tetrachlorodibenzo-p-dioxin)에 노출된 흰쥐의 지질대사 및 간 독성물질대사에 미치는 영향)

  • This study investigated the effects of green tea on the disorders of lipid metabolism, oxidative system and hepatic functions induced by 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD), using adult male rats (SD) for 3 weeks. These 36 animals were divided into four groups. TCDD ($50{\mu}g/kg$ BW) was intraperitoneally injected at the beginning of experiment. Green tea powder was added 1% or 3% levels in basal diets respectively. Relative weights of thymus were decreased about one-third of control group, but those of liver, brain and testis were significantly increased in rats treated TCDD. Neutrophill% and lymphocyte% by TCDD treatment was improved by green tea diets. In liver functional enzyme, elevation of glutamic oxaloacetic transaminase (GOT) and alkaline phosphatase (ALP) activities due to TCDD treatment was lowered by green tea diets. The concentrations of serum and liver lipids were significantly increased by TCDD treatment, however, those of serum and liver triglyceride tended to decrease by green tea diets. Fecal lipid excretion was increased in rats fed green tea diets. Especially, fecal total cholesterol level was significantly elevated by 3% green tea diets. The activities of superoxide dismutase (SOD) were increased in rats fed 3% green tea diets. Increment of benzphetamine N-demethylase (BPND) activity by TCDD treatment was declined by 1% green tea diets. These results indicate that green tea can exert improving effects on liver lipid accumulation and unfavorable hepatic functions, and elevate antioxidation.

Preparation and Characterization of Enzymatic Oyster Hydrolysates-added Yogurt (굴 효소 가수분해물 첨가 요구르트의 제조 및 특성)

  • The base for preparing oyster hydrolysate-added yogurt was consisted of whole milk (1,000 mL), skim milk (44.05 to 42.05 g), enzymatic oyster hydrolysates powder (OHP, 0 to 2.0 g) and pectin. The yogurt base was fermented with 7 kinds of starter cultures (3% based on yogurt volume), such as Lactobacillus acidophilus, lactobacillus bulgaricus, lactobacillus casei, Lactobacillus fermentum, Lactobacillus pentosus, Streptcoccus thermophilus and the mixed starters (L. bulgaricus and S. thermophilus) at optimal temperature. Processing condition and quality characteristics of the yogurt were evaluated by analyzing pH, titratable acidity, viscosity, viable cell count, functional properties and sensory evaluation. The results suggested that the optimal conditions for preparing the good quality yogurt revealed the mixed starters (L. bulgaricus and S. thermophilus) for starter culture, 1.0 g of 3 kDa hydrolysate for amount, and 5.5 hrs for fermentation time. The good quality yogurt showed 4.31 for pH, 1.07% for titratable acidity, 469 cps for viscosity and $4.9{\times}10^8\;CFU/mL$ for viable cell count. The hydrolysate-added yogurt was 2 times higher in ACE inhibitory and antioxidant activities than commercial yogurt, and kept good quality during storage of 15 days at $5^{\circ}C$.

Screening for Fittest Miscellaneous Cereals for Reclaimed Land and Functionality Improvement of Sorghum bicolor Cultivated in Reclaimed Land (간척지 적응성 잡곡 선발 및 간척지 재배 수수의 기능성 향상 효과)

  • Genetic resources of 84 species of Setaria italica BEAUVOIS, Sorghum bicolor, and Panicum miliaceum were collected to select the adaptable miscellaneous cereals in Saemangeum reclaimed land. The adaptability of Sorghum bicolor in reclaimed land was the highest among the three cereals cultivated on reclaimed land. The ratio of the average height of Sorghum bicolor plants cultivated in reclaimed land/normal field was 0.82, that of Panicum miliaceum was 0.61, and that of Setaria italica BEAUVOIS was 0.51. Three species of Sorghum bicolor, Satangdajuk, Kkamansusu, and Nampungcharl, were selected as potential genetic resources as they had excellent adaptability to reclaimed land. The yield of Satandaejuk on reclaimed land was 229.4 kg/10a, and the yield ratio of reclaimed land/normal field was 89.3%. The yield of Kkamansusu was 227.4 kg/10a, with reclaimed land/normal field ratio of 87.8%, and yield of Nampungcharl was 239.6 kg/10a, and reclaimed land/normal field ratio of 86%. In order to study the salt tolerance of selected genetic resources, we conducted salinity test. Salinity tolerance of Sorghum bicolor species-Satangdajuk, Kkamansusu, Nampungcharl was excellent compared to that of the other cereals. Among these, Satandaejuk had to highest salt tolerance level. Polyphenols, flavonoids, and detoxification of free radical were also studied. The anti-diabetic property of the cereals was also analyzed by ${\alpha}$-glucosidase inhibitory activity. We confirmed that the functionality of 3 lines in reclaimed land had improved in all the functional analysis categories when compared to that with yield in the normal field. Polyphenol, an antioxidant, increased in the range of 2~26% when cultivated in reclaimed land and the flavonoid content also increased from 8.5 to 55.6%. DPPH elimination capability, the ability to scavenge harmful reactive oxygen, also increased from 16.7 to 47% when cultivated in reclaimed land. The anti-diabetic activity and ${\alpha}$-glucosidase inhibition activity of selected Sorghum bicolor species-Satangdajuk, Kkamansusu, Nampungcharl also increased from 18.4 to 19.9% when cultivated on reclaimed land.

Immunomodulating activity of Sargassum horneri extracts in RAW264.7 macrophages (RAW264.7 대식세포에서 괭생이 모자반 추출물의 면역활성 증진 효과)

  • Purpose: Sargassum horneri (S. horneri) is a species of brown macroalgae that is common along the coast of Japan and Korea. The present study investigated the immuno-modulatory effects of different types of S. horneri extracts in RAW264.7 macrophages. Methods: S. horneri was extracted by three different methods, hot water extraction, 50% ethanol extraction, and supercritical fluid extraction. Cell viability was then measured by MTT assay, while the production levels of tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), interleukin-6 (IL-6), and nitric oxide (NO) were measured by enzyme-linked immunosorbent assay and Griess assay, respectively. The expression and activation levels of inducible NO synthase (iNOS), mitogen-activated protein kinase (MAPK) and nuclear factor ${\kappa}B$ ($NF-{\kappa}B$) were examined by western blot analysis. Results: The three different S. horneri extracts were nontoxic against RAW 264.7 cells up to $50{\mu}g/mL$, among which treatment with hot water extract (HWE) of S. horneri significantly enhanced the production of TNF-${\alpha}$, IL-6, and NO in a dose-dependent manner. Hot water extract of S. horneri also increased the expression level of iNOS, suggesting that up-regulation of iNOS expression by HWE of S. horneri was responsible for the induction of NO production. In addition, treatment of RAW 264.7 macrophages with HWE of S. horneri increased the phosphorylation levels of ERK, p38 and JNK. Furthermore, the activation and subsequent nuclear translocation of $NF-{\kappa}B$ was enhanced upon treatment with HWE of S. horneri, indicating that HWE of S. horneri activates macrophages to secrete TNF-${\alpha}$, IL-6 and NO and induces iNOS expression via activation of the $NF-{\kappa}B$ and MAPKs signaling pathways. Conclusion: Taken together, these findings suggest that HWE of S. horneri possesses potential as a functional food with immunomodulatory activity.

A Study on the Structure and Function of the Underground Storage Facility in Baekje (백제 지하저장시설(地下貯藏施設)의 구조와 기능에 대한 검토)

  • Increasing discovery cases of underground storage facilities made of earth, wood, or stone are being reported from the recent excavation survey of the Baekje relics. Accordingly, the purpose of this study is to examine the structure and function of the underground storage facilities of Baekje following a classification made by the type and building method as follows: plask shape, wooden box shape, and stone box shape. The plask shape storage is the most representative underground storage of Baekje that has been found in numerous relics more than 600 sets around Hangang(Han River) and Geumgang(Geum River) from the Hansung period to Sabi period in Baekje Dynasty. It is a historical artefact as a part of the unique storage culture of Baekje around Hangang and Geumgang from the 3rd to 7th Century. Considering its structure and the example of Chinese one, it might had been used for a long-term storage of grains and various other items including earth wares. The storage facility in wooden box shape and stone box shape are found mostly in the relics Of Sabi period. Thus it might had taken some functions of the storage in traditional pouch shape which had decreased after the 6th Century. In particular, the wooden box shape and stone box shape storage required enormous labor force to build owing to their structure and building method. Thus, they were considered to had been used for official purposes in province fortress and citadel artefact. The wooden box shape storage facility is classified into flat rectangular type and square type based on the structure, and into Gagu type(架構式) and Juheol type(柱穴式) based on the building method. It might had been decided according to the geography and geological feature of the place where the storage was to be built. Considering the examples of Gwanbuk-ri relics and Weolpyong-dong relics, the wooden box shape storage facility might had been used for various items depending on the needs, including foods such as fruits and essential provisions at the military base. Considering the long-term food storage, the examples in Japan, and the functional characteristics of the underground storage facility, there is a possibility that the wooden and stone box shape storage facilities had been built so as to safely store important items in case of fire. This study is only a rudimentary examination for the storage facility in Baekje. Thus further studies are to be made specifically and comprehensively on the comparison with other regions, distribution pattern, discovered relics and artefacts, and functions.

Protective effect of matcha green tea (Camellia sinensis) extract on high glucose- and oleic acid-induced hepatic inflammatory effect (고당 및 올레산으로 유도된 간세포에서의 염증반응에 대한 말차(Camellia sinensis) 추출물의 보호효과)

  • To evaluate hepatoprotective effects, the antioxidant capacities of matcha green tea extract (Camellia sinenesis) were compared to those of green leaf tea and the anti-inflammatory activities in HepG2 cells were investigated. Evaluation of the total phenolic and total flavonoid content, 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, and inhibitory effect on lipid peroxidation indicated that the aqueous extract of matcha green tea presented significant catechin content and antioxidant capacity compared to those of green leaf tea. In addition, the extract had considerable inhibitory effects on α-glucosidase, α-amylase, and advanced glycation end-products. The matcha green tea extract significantly increased cell viability and reduced reactive oxygen species in H2O2- and high-glucose-treated HepG2 cells. Furthermore, in response to oleic acid-induced HepG2 cell injury, treatment with matcha green tea aqueous extract inhibited lipid accumulation and regulated the expression of inflammatory proteins such as p-JNK, p-Akt, p-GSK-3β, caspase-3, COX-2, iNOS, and TNF-α. Matcha green tea could be used as a functional material to ameliorate hepatic lipid accumulation and inflammation.

Comparison of Antioxidant Activity According to Silkworm Cultivars (품종에 따른 국내산 누에의 항산화 활성 비교)

  • Although varieties of silkworms, which have recently attracted attention as a health functional food, are being produced, studies on the differences in the functionality of different silkworm varieties are insufficient. Therefore, in this study, the antioxidant activities of different silkworm breeds bred in domestic farms were analyzed, and the potential for their cultivation as specialized varieties with excellent antioxidant function was investigated. To compare antioxidant activity, four varieties of silkworms, white Bakokjam, Golden silk, Yeonokjam, and Hanseongjam, were bred; water and ethanol extracts of these silkworms were prepared on the 3rd and 5th days of the 5th instar larval stage. The highest extraction yield was seen for the water extract from the Golden silk variety on the 3rd day of the 5th instar; the highest total phenolic compound and flavonoid contents were observed for the water extract (86.11±4.04 ㎍/mg GE) and 70% (v/v) ethanol extract (46.70±2.81 ㎍/mg QE). Bakokjam and Yeonokjam exhibited DPPH radical scavenging activity of up to 78% and showed the highest nitrite scavenging activity (85%) at pH 1.2. The maximum SOD-like activity of Yeonokjam was about 47%. Furthermore, 48 ㎍/ml of the Yeonokjam extract showed a reducing power of 0.7 abs, which was the best among the four varieties. Considering these results, the Yeonokjam (on the 3rd day of the 5th instar) had antioxidant activity and represents a silkworm cultivar that would be suitable for cultivation as a health food.

Study on the Chemical Composition of Lotus Root and Functional Evaluation of Fermented Lotus Root Drink (연근의 성분분석 및 연근 발효음료의 기능성 평가)

  • This study examined the chemical composition of lotus root and functionally evaluated a fermented lotus root drink. Electron-donating ability using DPPH along with nitrite-scavenging ability were used to compare the antioxidative activities of unfermented and fermented lotus root drinks. The electron-donating abilities of the unfermented lotus root drink (1%) and fermented lotusroot drink (1%) were 22.55% and 23.88%, respectively. At pH 6.0, the nitrite-scavenging abilities of the unfermented lotus root drink and the fermented lotus root drink (100%) were 27.64% and 40.3%, respectively, and their scavenging ability increased in a dose-dependent manner at all pH values. In order to study the anti-obesity effects of the two drinks, male Sprague-Dawley rats were divided into four groups (A: basal diet, B: high fat diet, C: high fat diet+unfermented lotus root drink, D: high fat diet+fermented lotus root drink). Net weight gains were not significantly different among the four groups. Plasma total cholesterol concentrations significantly decreased in the groups receiving the unfermented and fermented lotus root drinks. Also, plasma total lipid and triglyceride contents were lower in the groups receiving the unfermented and fermented lotus root drinks as compared to the high fat diet group; however, the differences among the three groups were not significant.
