Objective: Fatty acid composition is one of the most important meat quality traits because it can contribute to functional, sensorial, and nutritional factors. In this study, quantitative trait locus (QTL) analyses for fatty acid composition traits were investigated in thigh and breast meat of Korean native chicken (KNC). Methods: In total, 18 fatty acid composition traits were investigated from each meat sample using 83 parents, and 595 $F_1$ chicks of 20 week old. Genotype assessment was performed using 171 informative DNA markers on 26 autosomes. The KNC linkage map was constructed by CRI-MAP software, which calculated genetic distances, with map orders between markers. The half-sib and full-sib QTL analyses were performed using GridQTL and SOLAR programs, respectively. Results: In total, 30 QTLs (12 in the thigh and 18 in the breast meat) were detected by the half-sib analysis and 7 QTLs (3 in the thigh and 4 in the breast meat) were identified by the full-sib analysis. Conclusion: With further verification of the QTL regions using additional markers and positional candidate gene studies, these results can provide valuable information for determining causative mutations affecting the fatty acid composition of KNC meat. Moreover, these findings may aid in the selection of birds with favorable fatty acid composition traits.
To identify the seedlings from controlled pollination between one paternal tree and three maternal trees of Japanese red pine, cpSSR markers of the paternally inherited haploid genome were analyzed in two year old 114 seedlings of full sib families. Individual specific DNA fingerprint like haplotypes of the parental trees were determined by PCR with three cpSSR primers. Haplotypes of the 114 seedlings were also identified by PCR with the same primers. On the basis of the comparison of cpDNA haplotypes of the 114 seedlings with those of the parental trees, 14 seedlings revealed to have distinguished haplotypes from those of the paternal tree. It was tentatively concluded that they were generated via pollination with the non-paternal trees. A seedling of Gangwon30 revealing non-paternal haplotype might have been generated via self pollination with the pollens of maternal tree through improper emasculation or contamination during artificial pollination. DNA fingerprint like cpSSR profiles observed in this study could be successfully applied to the various plant forensic analyses, such as identification of siblings of individual trees, asexually reproduced ramets of a specific clone, vegetatively propagated individuals via tissue culture, and pure full sib progenies.
To analyze the possibility for the genetic improvement at the early period at 60 days of age in tilapia, Oreochromis niloticus. Genetic parameters on eight traits, total length (TL), standrad length (SL), head length (HL), body depth (BD), body height at the origin of dorsal fin (BH1), length from the origin of dorsal fin to the origin of dorsal fin (BH2), snout length (SNL), and body weight (BW), were estimated by sib analysis. Heritabilities estimated from sire, dam and full-sib components were moderately high in all metric traits, ranged 0.08$\~$0.70, 0.22$\~$0.41 and 0.18$\~$0.55, respectively. Those of SL from sire, dam, and full-sib component were estimated as 0.13, 0.22, and 0.18, respectively, Besides BH1, BH2 also showed high heritabilities, $h^2_s$ (0.08), $h^2_d$ (0.38) and $h^2_{s+d}$ (0.23), indicating that it would be a new production-related metric trait for selection. Among the metric traits, phenotypic and genetic correlation coefficients were ranged from 0,86 to 0.97 and from 0.90 to 0.99, respectively. Thus, genetic improvement would be possible at the early growth rate by the individual selection in tilapia.
Pedigree data were used to predict the performance of 1,166 imported Americas Brahman breeder cattle, composed of 104 bulls and 1,062 heifers with an average age of 24.4 months (range of 17 to 40 months). A total of 13 full sib, 10 maternal half sib, and 228 paternal half sib groups were identified, each with average group size of 2.08, 2.00, and 4.49, respectively. Only 758 (64.9% of total) imported cattle were found to have at least one ancestor with expected progeny differences (EPDs) reported in the Spring 1995 Brahman Sire Summary. Moderate average accuracy values of .71, .69, .52, and .52 for birth weight, weaning weight, yearling weight, and maternal milk, respectively, were noted for EPDs of the ancestors. Prediction equations were derived by multiple regression analysis of available EPDs of sire, paternal grand sire, and maternal grand sire. Based on pedigree indexes that involve various combinations of available ancestral information, the average predicted EPDs (lbs) for imported cattle were $1.76{\pm}0.54$, $14.93{\pm}4.86$, $25.10{\pm}9.50$, and $5.86{\pm}2.08$ for birth weight, weaning weight, yearling weight, and maternal milk, respectively. Significant correlations (p < .05) were also found between sire and son EPDs (+.27) for yearling weight; and between sire and paternal grand sire EPDs for birth weight (+.34), weaning weight (+.51), yearling weight (+.49), and maternal milk (+.55).
Objective: When evaluating individuals with the same parent and no phenotype by pedigree best linear unbiased prediction (BLUP), it is difficult to explain carcass grade difference and select individuals because they have the same value in pedigree BLUP (PBLUP). However, single step GBLUP (ssGBLUP), which can estimate the breeding value suitable for the individual by adding genotype, is more accurate than the existing method. Methods: The breeding value and accuracy were estimated with pedigree BLUP and ssGBLUP using pedigree and genotype of 408 Hanwoo cattle from 16 families with the same parent among siblings produced by fertilized egg transplantation. A total of 14,225 Hanwoo cattle with pedigree, genotype and phenotype were used as the reference population. PBLUP obtained estimated breeding value (EBV) using the pedigree of the test and reference populations, and ssGBLUP obtained genomic EBV (GEBV) after constructing and H-matrix by integrating the pedigree and genotype of the test and reference populations. Results: For all traits, the accuracy of GEBV using ssGBLUP is 0.18 to 0.20 higher than the accuracy of EBV obtained with PBLUP. Comparison of EBV and GEBV of individuals without phenotype, since the value of EBV is estimated based on expected values of alleles passed down from common ancestors. It does not take Mendelian sampling into consideration, so the EBV of all individuals within the same family is estimated to be the same value. However, GEBV makes estimating true kinship coefficient based on different genotypes of individuals possible, so GEBV that corresponds to each individual is estimated rather than a uniform GEBV for each individual. Conclusion: Since Hanwoo cows bred through embryo transfer have a high possibility of having the same parent, if ssGBLUP after adding genotype is used, estimating true kinship coefficient corresponding to each individual becomes possible, allowing for more accurate estimation of breeding value.

The purpose of the study was to develop an optimum SNP marker set to be utilized for domestic pork traceability. The study tested 51 SNP markers analyzed for origin of farm to be determined from genotypes of offspring and parents in pigs. With the simulation data through random mating population (PI), half sib mating population ($PI_{half-sib}$) and full sib mating population ($PI_{sibs}$), probability of identical genotypes were analyzed as $5.63{\times}10^{-33}$, $4.35{\times}10^{-15}$ and $1.32{\times}10^{-15}$, respectively. The 51 SNP markers also had 100% accuracy for parental determination. These results suggest that if the pig breeding stock is genotyped with the 51 SNP markers, the genotype information of individual offspring can be checked for farm origins by tracing parental sow and sire. Therefore, these SNP markers will be useful to trace the pork from production to consumption in pigs.

Background and Purpose: The efficacy and safety of GV1001 have been demonstrated in patients with moderate-to-severe Alzheimer's disease (AD). In this study, we aimed to further demonstrate the effectiveness of GV1001 using subscales of the Severe Impairment Battery (SIB), which is a validated measure to assess cognitive function in patients with moderate-to-severe AD. Methods: We performed a post hoc analysis of data from a 6 month, multicenter, phase 2, randomized, double-blind, placebo-controlled trial with GV1001 (ClinicalTrials.gov, NCT03184467). Patients were randomized to receive either GV1001 or a placebo for 24 weeks. In the current study, nine subscales of SIB-social interaction, memory, orientation, language, attention, praxis, visuospatial ability, construction, and orientation to name-were compared between the treatment (GV1001 1.12 mg) and placebo groups at weeks 12 and 24. The safety endpoints for these patients were also determined based on adverse events. Results: In addition to the considerable beneficial effect of GV1001 on the SIB total score, GV1001 1.12 mg showed the most significant effect on language function at 24 weeks compared to placebo in both the full analysis set (FAS) and per-protocol set (PPS) (p=0.017 and p=0.011, respectively). The rate of adverse events did not differ significantly between the 2 groups. Conclusions: Patients with moderate-to-severe AD receiving GV1001 had greater language benefits than those receiving placebo, as measured using the SIB language subscale.
When 14 microsatellite (MS) markers were applied in the identifying test for 480 Hanwoo, the discriminating power was estimated as $3.43{\times}10^{-27}$ based on the assumption of a random mating group (PI). This rate is 1,000 times higher than that of 60 single nucleotide polymorphism (SNP) markers. On the other hand, the power of the 60 SNP markers was estimated as $4.69{\times}10^{-20}$ and $8.02{\times}10^{-12}$ on the assumption of a half-sib mating group ($PI_{half-sibs}$) and a full-sib mating group ($PI_{sibs}$), respectively. These powers were 10 times and 10,000 times higher than those of the 14 MS markers. The results indicated that the total number of alleles (MS vs SNP = 146 vs 120) acted as a key factor for the discriminating power in a random mating population, and the total number of markers (MS vs SNP = 14 vs 60) was a dominant influence on the power in half-sib and full-sib populations. In the Hanwoo population, in which it was assumed that the entire population is the enormous half-sib group formed by the absolute genetic contribution of a few nuclear bulls, there will be only a 10 times difference in the discriminating power between the 14 MS markers and the 60 SNP makers. However, the probability of not excluding a candidate parent pair from the parentage of an arbitrary offspring, given that only the genotype of the offspring ($PNE_{pp}$) was 1,000 times higher as shown by the 14 MS markers than that by the 60 SNP markers. The strong points of SNP makers are the stability of the variation (low mutation rate) and automation of high-throughput genotyping. In order to apply these merits for the practical and constant Hanwoo identity test, research and development are required to set a cost-effective platform and produce a homemade apparatus for SNP genotyping.
We conducted QTL mapping for 6-year growths of open-pollinated half-sib progenies from a selected clone 7-1037 in Pinus taeda. With an AFLP marker analysis on haploid DNA samples from the megagametophytes of the open-pollinated seeds, we constructed 20 framework maps spanning a total of 1,869 cM in total length and 18.5 cM in an average interval length between markers. Composite interval mapping reveals that one QTL explains 5.9% of the total phenotypic variation of height, and three QTLs account for 3.9~5.6% of the variation of diameter at breast height (DBH). There are no correlations between the QTLs. The genetic effects of the QTLs are 39.6 cm in height and 7.20~9.41 mm in DBH, respectively, The average effects of gene substitution of the markers closely linked with the QTLs are 44.3 cm in height and 8.38~11.81 m in DBH. Under an assumption that the within-family heritability for the growth traits of loblolly pine is less than 0.2, the QTLs account for 26.8% of the additive genetic variance of the progenies. In terms of relative selection efficiency, the individual selection based on QTL markers could be 5 times as high as phenotypic selection. The results in this study indicate that the QTL mapping method with open-pollinated half-sib family could be more practical and applicable to the conventional seed orchard-based selection work than other mapping methods with a single full-sib family, in particular from the viewpoint that it can provide crucial information for within-family individual selection such as breeding value.
Parentage tests using polymorphic DNA marker are commonly performed to avoid incorrect recording of the parental information of livestock animals, and single-nucleotide polymorphisms (SNPs) are becoming the method of choice. In Japanese Black cattle, parentage tests based on the exclusion method using microsatellite markers are currently conducted; however, an alternative SNP system aimed at parentage tests has recently been developed. In the present study, two types of simulations were conducted using the pedigree data of two subpopulations in the breed (subpopulations of Hyogo and Shimane prefectures) in order to examine the effect of actual genetic and breeding structures. The first simulation (simulation 1) investigated the usefulness of SNPs for excluding a close relative of the true sire; the second one (simulation 2) investigated the accuracy of sire identification tests for multiple full-sib putative sires by a combined method of exclusion and paternity assignment based on the LOD score. The success rates of excluding a single fullsib and sire of the true sires were, respectively, 0.9915 and 0.9852 in Hyogo and 0.9848 and 0.9852 in Shimane, when 50 SNPs with minor allele frequency (MAF: q) of 0.25${\leq}$q${\leq}$0.35 were used in simulation 1. The success rates of sire identification tests based solely on the exclusion method were relatively low in simulation 2. However, assuming that 50 SNPs with MAF of 0.25${\leq}$q${\leq}$0.35 or 0.45${\leq}$q${\leq}$0.5 were available, the total success rates including achievements due to paternity assignment were, respectively, 0.9430 and 0.9681 in Hyogo and 0.8999 and 0.9399 for Shimane, even when each true sire was assumed to compete with 50 full-sibs.
