  • The activity of fucosidase shows different value depending on disease, eg) fucosidosis and I cell disease are characterized by the absence or deficiences of .alpha.-fucosidase, and sera of ovarian cancer patients exhibited a statistically significant deficiency of .alpha.-L-fucosidase activity (Zielke et al., 1972; Kress et al., 1980; Barlow et al., 1981). For the purpose of diagnosis of these disease easily, the manual of test can be developed by preparing kits of hydrophobic-binding substrate of fucosidase that bind C18-column.

Effects of Sardine Oil Fedding and Vitamin E Supplementation on Histopathological Changes and $\alpha$-L-Fucosidase Activity in Experimental Hepatocarcinogenesis (정어리유 섭취와 비타민 E 보충이 실험적 간 발암과정의 병리조직학적 변화와 $\alpha$-L-fucosidase의 활성도에 미치는 영향)

  • Effects of Sardine Oil Feeding and Vitamin E Supplementation on Histopathological Changes and $\alpha$-L-fucosidase activity in experimental hepatocarcinogenesis. Sprague-Dawley rats weighing 80~90 g were fed the diet containing either 15% corn oil (CO) or sardine oil (SO) with or without vitamin E supplements (dl-$\alpha$-tocopherol acetate 800 IU/kg diet) for 8 weeks. After 2 weeks of feeding, the rats were given a single intraperitoneal injectin of diethylnitrosamine (DEN, 200 mg/kg BW). From the fifth week, rats were given 0.02% acetylaminofluorene (AAF) in diet for 4 weeks. At the seventh week, 0.05% phenobarbital in liver and hepatic glutathione S-transferase palcental form positive (GST-P+) foci were examined by Hematoxylin& Eosin (H&E) staining and immunohistochemical method, respectively. Serum $\alpha$-L-fucosidase activity was determined. The livers fromt he carcinogen treated rats showed significantly increased formation of GST-P+ foci at sacrifice points while the livers fromthe non-carcinogen treated groups showed almost no foci. Although GST-P+ foci formation was not affected by dietary oil, it was increased unexpectedly by vitamin E supplementation. Histopathological changes were similar to patterns of GST-P+ foci formation in almost all groups. Serum $\alpha$-L-fucosidase activities were increased by carcinogen treatment in all dietary groups. $\alpha$-L-fucosidase activities were positively correlated with GST-P+ foci formation. There results suggest that excessive vitamin E supplementation can enhance hepatocarcinogenesis although the mechanisms involved are not clearly understood.

  • Bacteroides is the most predominant bacteria in the human large intestinal tract. Bacteroides fragilis RoidB isolated from a Korean faecal sample showed high $\alpha-fucosidase$ activity compared to other bacterial species. The optimized meidum for the production of $\alpha-fucosidase$ from 8. fragilis contained BHI 37g, hemin 10mg, cysteine 0.5g, resazurin 1mg, vitamin K1mg, and starch 5g per 1 L. The crude $\alpha-fucosidase$ obtained through DEAE-sepharose and CM- cellulose chromatography showed optimum temperature and pH at $40^{\circ}C$ and 7.0, respectively. Among several metals, $Co^{++}\;and\;Zn^{++}$ showed strong inhibition on enzyme activity.

Effect of Monosaccharide L-fucose and Polysaccharide Fucoidan on Sperm ${\alpha}$-L-fucosidase Activity and Relation to Sperm-oocyte Interaction in Pig

  • Carbohydrate-protein interactions are known to be important in gamete interactions. Several evidence indicated that a fucose-containing sulfated polysaccharide fucoidan was potential inhibitor of fertilization in vitro and thus fucose seemed to be part of the recognition signal of gamete interaction in mammals. In recent investigation we found that ${\alpha}$-L-fucosidase activity was present in boar spermatozoa and it was related to sperm binding to and penetration into zona pellucida (ZP) in vitro. The objective of this study was to determine the effects of monosaccharide L-fucose and polysaccharide fucoidan on sperm ${\alpha}$-L-fucosidase activity and relation to sperm-oocyte interaction in pig. Results indicated that the activity of sperm ${\alpha}$-L-fucosidase was largely inhibited (62%) when sperm suspension was treated with monosaccharide L-fucose. It also significantly inhibited the number of sperm binding to ZP (32%) and penetration into zona-intact oocytes (72%), but did not inhibit penetration into zona-free oocytes when fertilization medium contained L-fucose. The chlorotetracycline (CTC) assessment showed that L-fucose did not affect induction of sperm capacitation and acrosome reaction. In contrast, the activity of sperm ${\alpha}$-L-fucosidase was not inhibited when sperm suspension was treated with polysaccharide fucoidan but sperm-ZP binding was greatly inhibited (85%) and completely blocked sperm penetration into zona-intact or zona-free oocytes. The CTC assessment showed that fucoidan increased the F pattern and decreased the AR pattern sperm. These results suggested that the different inhibitory mechanisms were present between monosaccharide L-fucose and polysaccharide fucoidan on sperm-oocyte interaction, the inhibition effect of ${\alpha}$-L-fucose on sperm binding and penetrating into ZP caused sperm ${\alpha}$-L-fucosidase inhibited by ${\alpha}$-L-fucose.

Glycosidase Pattern of Bacteroides fragilis Roid 8 Isolated from a Korean Adult Feces (한국인 분변으로부터 분리된 Bacteroides fragilis Roid 8의 Glycosidase 패턴)

  • The intestinal microflora of humans is an extraordinarily complex mixture of microorganisms, the majority of which are anaerobic bacteria. Amongst them, most prevalent bacteria are Bacteroides, Eubacterium, Peptococcus, Bifidobacteria. We isolated a Bacteroides fragilis strain from a Korean adult and examined various glycosidase activities of this strain. The activities of $N-acetyl-{\beta}-glucosaminidase,\;{\alpha}-fucosidase$, ${\beta}-glucuronidase$, chitobiase and PNPCase were stronger in Bacteroides fragilis Roid 8 than in other intestinal anaerobic bacteria. $N-acetyl-{\beta}-glucosaminidase$ was strongest, followed by ${\alpha}-fucosidase$, ${\beta}-glucuronidase$ and PNPCase. The activities of ${\beta}-galactosidase$, ${\beta}-xylosidase,\;{\alpha}-arabinofuranosidase$ were not present or very low. The activities of ${\alpha}-glucosidase$, ${\beta}-glucosidase$ and ${\alpha}-galactosidase$ were present but at a lower level than in Bifidobacterium. The effect of the carbon sources on the production of $N-acetyl-{\beta}-glucosaminidase$, ${\alpha}-fucosidase$, ${\beta}-glucuronidase$ and PNPCase of Bacteroides fragilis Roid 8 was investigated. :.actose and glucose lowered the production of the varous glycosidase enzymes studied in this work. In addition, we investigated the optimum temperature and pH of each glycosidase from Bacteroides fragilis Roid-8 using crude enzyme preparations.

A Simple Enzymatic Method for Quantitation of 2'-Fucosyllactose

  • 2'-Fucosyllactose (2'-FL) is one of the most important human milk oligosaccharides and has several health benefits for infants. The levels of 2'-FL in breast milk or samples from other sources can be quantified by high-performance liquid chromatography. However, this method cannot be used for simultaneous detection of the target compound in numerous samples. Here, we developed a simple method for quantifying 2'-FL in a microplate format. The method involves two steps: (i) release of $\text\tiny{L}$-fucose from 2'-FL by ${\alpha}$-(1-2,3,4,6)-$\text\tiny{L}$-fucosidase and (ii) measurement of NADPH formed during the oxidation of $\text\tiny{L}$-fucose by $\text\tiny{L}$-fucose dehydrogenase. This method enables measurement of up to 5 g/l 2'-FL in 50 min using a 96-well microplate. The efficiency and simplicity of the proposed method make it suitable for the analyses of a large number of samples simultaneously.

Anti-carcinogenic Effects of Korean Mistletoe Extract and Lectin in Experimental Hepatocarcinogenesis (실험적 간암모델에서 한국산 겨우살이(Mistletoe)추출물 및 렉틴의 발암 억제효과 탐색)

  • This study was done to investigate effects of Korean mistletoe extract and lectin on serum GOT, GPT and $\alpha$-L-fucosidase activities and the preneoplastic lesion in chemically induced rat hepatocarcinogenesis. To attain the above objectives weanling Sprangue-Dawley male rats were fed modified AIN-76 diets containing 10% corn oil for 9 weeks. One week after feeding rats were intraperitonealy injected twice with a dose of diethylnitrosamine (DEN, 50 mg/kg body weight(BW)) and were provided 0.05% phenobarbita (PB) with drinking water from one week after DEN treatment until the end of experiment. For the same period as PB treatment, rats were injected mistletoe extract (10 $\mu\textrm{g}$/kg BW European mistletoe, 10 $\mu\textrm{g}$/kg BW and 100 $\mu\textrm{g}$/kg BW Korean mistletoe) and lectin(1 ng/kg BW, 10 ng/kg BW) twice a week. At the end of 9th week rats were sacrificed and the formation of hepatic glutthione S-transferase placental form positive (GST-P+) foci serum GOT, GPT and $\alpha$-L-fucosidase activities were determined. By treatment of mistletoe extract or lectin there were no significant effects on serum GOP, GPT and $\alpha$-L-fucosidase activities whereas those activities showed a tendency to increase by DEN treatment. The formation of GST-P+ foci was significantly decreased by mistletoe extract or lectin treatment especially in group of 100$\mu\textrm{g}$/kg BW Korean mistletoe. These results suggest that Korean mistletoe extract and lectin have a possibility to inhibit hepatocarcinogenesis of animals.

Exo-O-Glycosylhydrolases in Korea Ginseng Roots

  • WB were screening the stele and the cortex of the ginseng roots (Panax ginseng C.A.Meyer) on the exo-0-glycosylhydrolase activities during vegetation period of 1999 year. The following p-nitrophenylglycosides were used to test exe-0-glycosylhydrolase activities: $\alpha$- and $\beta$-D-galactopyranosides,$\alpha$- and $\beta$-D-glucopyranosides, $\alpha$- and $\beta$-D-mannopyranosides, N-acetyl-$\beta$-D-glucosaminide, $\alpha$- and $\beta$-D-xylopyranosides $\alpha$- L-rhamnopyranoside, $\beta$-D-glucuronide, $\beta$-D-galacturonide, $\beta$-L-,$\alpha$-L- and $\beta$-D-fucopyranosides, $\alpha$-L-arabinopyranoside. Only $\beta$-D-galactosidase, $\alpha$-L-mannosi-dase , N- acetyl- ${\beta}$-D-slucosarninidase, $\alpha$-D-galacto sidase, $\alpha$-L-arabinosidase, and $\beta$-D-fuco sidase were found in both partsof ginseng roots. Their contents during the vegetation period were shown to differ considerably, being dependent not only on plant development stage but on plant tissue and environmental conditions too.

Pectolytic Enzymes of the Industrial Fungus Aspergillus kawachii

  • Aspergillus kawachii extracellular pectinases were screened in liquid cultures with different carbon sources. The fungus grown on citrus pectin or lemon pomace produced at least one of these inducible pectinases: acidic polygalacturonase, pectin lyase, pectin methylesterase, $\alpha$-L-arabinofuranosidase, $\alpha$-1,5-endoarabinase, $\beta$-D-galactosidase/exogalactanase, and $\beta$-1,4-endogalactanase. The lemon-pomace filtrates also contained significant $\alpha$-L-rhamnosidase and $\beta$-D-fucosidase activities. Most of the screened pectinases were active at pH 2.0-2.5, indicating that the A. kawachii enzymes were acidophilic. Under the culture conditions employed we could not detect enzymatic degradation of soybean rhamnogalacturonan. The A. kawachii pectinase-production-related regulatory phenomena of induction-repression resemble those described for other Aspergillus sp.

Phenotypic characteristics and antimicrobial susceptibilities of motile aeromonads isolated from freshwater fish in Korea and Japan (한국과 일본의 담수어에서 분리되는 운동성 aeromonads의 표현형적 특성과 약제감수성)

  • We investigated the phenotypic characteristics by using API20E, APIZYM and determined minimum inhibitory concentrations (MICs) of 7 antibiotics in motile aeromonads isolated from freshwater fishes in Korea and Japan, and 4 American Type Culture Collection (ATCC) strains. All isolates (n=7) were identified as motile Aeromonas species according to API20E test. Lysine decarboxylase activity and acid production from 4 different carbohydrates including mannitol, rhamnose, amygdalin and arabinose were observed in various strains. In enzymatic activities by APIZYM, all isolates showed negative reactions in valine and cystine arylamidases, α-chymotrypsin, α-galactosidase, β-glucuronidase, α-glucosidase, α-mannosidase and α-fucosidase. Although the intensities of each enzymatic activity were diverse in alkaline phosphatase, esterase-lipase, leucine arylamidase, β-galactosidase and N-acetyl-β-glucosaminidase, all isolates showed positive reactions. All isolates were resistant to ampicillin sodium (MIC>100㎍/ml), but sensitive to chloramphenicol (MIC≤1.6㎍/ml). However, recently isolated strains (AC9804, AC0202 and GMA0361) were commonly resistant to tetracycline (MIC=50㎍/ml). Furthermore, AC9804 was resistant to oxolinic acid (MIC=12.5㎍/ml). GMA0361 was resistant to kanamycin sulfate (MIC>100㎍/ml) and streptomycin sulfate (MIC>100 ㎍/ml).
