Effects of Incompatibility on Protoplast Fusion between intra-and inter Species in Basidiomycete, Pleurotus spp. (느타리버섯의 불화합성(不和合性)이 종내(種內) 및 종간(種間) 원형질체(原形質體) 융합(融合)에 미치는 영향(影響))

  • Effects of incompatibility existing between intra-and interspecies in Pleurotus spp. on protoplast fusion, clamp formation of their fusants and fruitbody production were investigated. Protoplast fusion between intra-and interspecies of the fungus was achieved by Poly ethylene glycol treatment. The fusion frequency between intraspecies was a little higher than that of interspecies. Fusion frequency between interspecies was not correlated with their similarities based on isozyme patterns. In case of protoplast fusion between intra-and interspecies, the fusants from the compatible isolates produced normal fruit bodies, while those from the incompatible isolates did not produce clamp connections and fruit bodies except those of a few isolates presumed mutants.

Study on the Genetic Diversity and Biological Characteristics of Wild Agaricus bisporus Strains from China

  • 90 wild Agaricus strains from China, including 44 Agaricus bisporus strains identified preliminarily by isozyme electrophoresis, were studied by the techniques of SRAP and ISSR. 18 special SRAP bands and 12 special ISSR bands were analyzed, the strains were clustered and a demdrogram was obtained. The results showed that the strains were divided into 2 groups, wild A. bisporus group and the other Agaricus group. It is similar to the result of isozyme electrophoresis. 41 wild A. bisporus strains from Sichuan and Tibet were divided into 4 groups based on their growing places, suggesting the regionally difference of the strains to be quite obvious. Some white wild A. bisporus strains from Xinjiang and Tibet had special patterns, resulting in lower coefficient values with other wild A. bisporus strains. The biological characteristics of three wild A. bisporus strains were analyzed, and the results showed: 1. The wild strains grew slowly on PDA medium with weak appressed mycelia, and grew normally in kernel or fermented cottonseed shell substrate. 2. They grew faster than control strain As2796 under lower temperature of $16^{\circ}C$, and higher temperature of $32^{\circ}C$, with optimum growing temperature of $20-24^{\circ}C$, which was $4^{\circ}C$ lower than that of control strain. 3. In the cultivation with manure compost via twice fermentation, the mycelia grew normally in compost and quite slowly in casing soil, and the fruitbodies occurred less and late with easily opening and low production. 4. The fruitbody was off-white with flat and scaled cap, long stipe and dark gill. The bisporus basidia occupied 70-80% and trisporus basidia 20-30% of the total basidia. 5. Heterokaryotic monospore isolates could fruit in cultivation, and the homokaryotic isolates could cross with those derived from overseas wild A.bisporus strains. 6. The electrophoresis phenotype of isozymes such as esterase etc. belonged to high production type (H type). 7. The RAPD patterns made much difference from those of high production, good quality or hybrid strains, which indicated that the wild strains produce a new kind of RAPD type.

Characteristics and breeding of a new pink oyster mushroom variety 『Jeakdan』 (느타리버섯의 신품종 육성 연구 - 분홍느타리 신품종 『적단』의 특성 -)

  • Jeakdan', a new variety of pink oyster mushroom was bred in Mushroom Research Institute, Gyeonggido Agricultural Research & Extension Services in 2006. This pink oyster mushroom was selected one which cross with monokaryon collected from strain KME20063 and KME20193. The major characteristics of the mushroom were showed a lot of pinheadings, the pink-colored and infundibuliform pileus. The optimum temperature for the mycelial growth was around $26{\sim}30^{\circ}C$ and that for the primordia and growth of fruitbody was around $18{\sim}23^{\circ}C$. Primordia and young fruiting bodies were bright pink but become less intensely colored while the mushroom were mature. Incubation period was required about 20 days in bottle culture and 21 days in bag culture. Number of valid stipes was 31.5/850cc bottle and 47.0/2kg bag. The fruiting body yield was 134.5g/850cc bottle and 350.0g/2kg bag.

Characteristics of a new oyster mushroom variety 『Dajoa』 for the bottle and poly prophylene plastic bag culture (느타리버섯의 신품종 육성 연구 -병·봉지재배용 신품종 『다조아』 느타리버섯의 특성-)

  • 'Dajoa', a new variety of oyster mushroom was developed by Mushroom Research Institute, Gyonggi Province Agricultural Research and Extension Services in 2004. It was bred with mating between monokaryotic strains isolated from Boonli 89-1 and ASI 2018-249. The major characteristics of the mushroom are showing a lot of pinheadings, the gray-colored and infundibuliform pileus. The optimum temperature for the mycelial growth was around $26{\sim}28^{\circ}C$ and that for the pinheading and growth of fruitbody was around $15{\sim}18^{\circ}C$. Incubation period was required around 24 days with bottle culture and about 21days in Poly Prophylene(P.P) plastic bag culture. The yields were shown high by 140.7g/850cc bottle and 260.3g/1kg P.P. plastic bag.

Development of mushroom spawn production technology of low price by the superior bag spawn (저비용 우량봉지종균 생산기술 개발)

  • The word spawn is derived from an old French verb, espandre, meaning to spread out or expand. Spawn is also defined as "the mycelium of fungi, especially of mushrooms grown to be eaten, used for propagation". The effects of bag spawn to sawdust substrate on the growth of Pleurotus ostreatus were conducted. The duration of mycelial growth and days of pinhead formation of bag spawn(2.5kg) were 18~19days and 7~8days, whereas bottle spawn ($1,000m{\ell}$) was 18days and 6days, respectively. The yield of mushroom fruitbody was that bag spawn is 100~118g, bottle spawn is 95~115g. In economical analysis, bag spawn is increased to 50%, compared to bottle spawn in relative income.

  • To develop a new cultivar of King oyster mushroom white variety (Pleurotus ferulae), GW10-95 as parental strain was selected by the method of Di-mon crossing between monokaryotic strain ASI 2850-24 derived from ASI 2850 and dikaryotic strain ASI 2803. The GW10-95(ASI 2803 ${\times}$ ASI 2850-24) was shown the best cultural characteristics, selected to be a new cultivar and designed as 'Beesan No.1'. The 'Beesan No.1' was formed incompatibility line distinctly in the confrontation growth of parental strains ASI 2803 and ASI 2850. Analysis of the genetic characteristics of the new cultivar 'Beesan No.1' showed a different DNA profile as that of the control strains, ASI 2803 and ASI 2850, when RAPD(Random Amplified Polymorphic DNA) primer URP6 was used. The optimum temperature and pH arrange for mycelial growth were $25^{\circ}C$ and pH5~8, respectively. Fruiting body production per bottle was about 245 g using demonstration farms. And also the stipe was thick and long. This new cultivar 'Beesan No.1' of Pleurotus ferulae was characterized by fast fruitbody formation, the stipe was thick and long and high quality yield compared to that of other cultuvars. We therefore expect that this new strain will increase of farmer's income by construction of stabilized production system.

Heating and cooling properties in cultivation room and cultural characteristics of oyster mushroom according to type of air exchange (외부공기 유입방식에 따른 느타리버섯 생육실의 온도변화 및 자실체의 생육특성)

  • This study was carried out to find out suitable input method for air exchange about bottle cultivation of oyster mushroom. There was no difference of average temperature in cultivation room, but T1(direct introduction of outside air) was higher than T2(heat exchanger) and T3(air buffer) in the standard deviation. The ratio of cooling operating was the highest in August and in the descending order, T1 54%, T3 43%, T2 33%. At ratio of energy reduction, T2 and T3 were higher than T1. The operating ratio of heater was highest in January and in the descending order, T1 53%, T3 37%, T2 30%. At ratio of energy reduction(%), T2 and T3 were higher than T1 similar to result of cooling operating. Therefore there were largest in August about Cooling Degree-Hour and in January about Heating Degree-Hour. And fruitbody yields quality was excellent in T2 and T3 than T1. The suitable effective type of air exchange in oyster mushroom cultivation are heat exchange and air buffer system.

Purification and Some Characteristics of the Proteolytic Enzyme in Fruitbody of Neungee [Sarcodon aspratus (Berk.) S. Ito] (능이 [Sarcodon aspratus (Berk.) S. Ito]중 단백질(蛋白質) 가수분해(加水分解) 효소(酵素)의 정제(精製) 및 성질(性質)에 관하여)

  • This study was undertaken to investigate the characteristics of the proteolytic enzyme extracted from Neungee mushroom [Sarcodon aspratus (Berk.) S. Ito]. The enzyme was purified by using Tris-acryl CM-cellulose ion exchange, gel filtration on Ultrogel AcA 54, Hydroxy apatite column chromatography and preparative isoelectic focusing. The specific activity of the purified enzyme increased 8 times as compared with that of the crude enzyme. The enzyme was homogeneous on polyacrylamide gel electrophoresis (PAGE). The optimum pH was 10.1, indicating the enzyme to be alkaline protease and the optimum temperature was $57^{\circ}C$. The enzyme was stable at temperatures lower than $50^{\circ}C$and at pH values ranging from 4.0 to 10.8. However, the enzyme activity decreased by 26 and 65% at 60 and $65^{\circ}C$, respectively, when incubated for 30 minutes. The enzyme activity was activated by $Mn^{++}$ and inhibited by $Cu^{++}$ and $Hg^{++}$. The enzyme was consisted of monomer and its molecular weight estimated to be about 30,100 when determined by sodium dodecyl sulfate PAGE. Isoelectric point of the enzyme was determined to be 9.80.

Changes of saccharide content in fruitbody composition of Flammulina velutipes during storage (팽이버섯의 저장기간 및 품종별 자실체내의 당 함량 변화)

  • Winter mushroom was monitored to investigate the influence of storage temperature on its quality during the storage and distribution phase. In measuring its quality, the contents of saccharides were quantified with its fruiting bodies using HPLC. Although it has been known to be difficult to separate saccharide isomers, our results indicated that Grace Prevail carbohydrate ES $5{\mu}column$ was the best in the separation to analyze the saccharide out of six columns used in this study. In our results, xylose was the main component of saccharide in the fruiting body of winter mushroom(White line mushroom:47.68mg/g, brown line mushroom: 63.28mg/g). In long-term storage, the total amount of saccharide tended to increase, but trehalose content of the disaccharide decreased. In comparison with the paramount amount of lactose and myo-inositol contents in long-term storage at $4^{\circ}C$, lactose wasn't detected when stored at $-1^{\circ}C$.

Comparisons of Protein-bound Polysaccharide Contents Obtained from Mycelial Cultured Broth and Fruit body of Coriolus versieolor (구름버섯의 균사배양액 및 자실체로부터 얻어진 단백다당체의 수율비교)

  • This study was performed to obtain higher yield rate of protein-bound polysac-charide extracted from cultured broth and fruit body of Coriolus versieolor, basidiomycetes. 16002 strain produced 0.58%, the highest yield rate among four strains, and there was no significantly different yield in other three strains. The optimal liquid media were CVT-III to obtain higher yield of protein-bound polysaccharide comparing with the various culture media. Compared with several materials for substitution of medium source, the residue extract media of Coriolus versieolor fruit-body were yield rate similar to control, and addition of the extracts of ginseng residues increased 11.4-48%. And also compared with fruit bodies that were artificially cultured on the oak longs, 16002 strain produced the highest yield similar to the result of cultured broth. Fruit body of the prematures stage was more effective to obtain high yield rate.
