Changes in Sporophore Appearance of Cultivated Mushroom, Agaricus bisporus, during the Harvesting Period (양송이 자실체(子實體) 발생(發生)의 경시적(經時的) 변동(變動)에 관한 연구(硏究))

  • This stuy was conducted to analyse the variation in occurrance of fruit body of Agaricus bisporus (Lange) Sing, depending on difference of cultivation and harvesting method and to find out a method for early predicting the mushroom yield potential at the base of results of the analysis. The results obtained were as follows ; 1. Total mushroom yield was highly correlated with picking amount of fruit bodies during first 30 days of picking period, and yield potentiality in the certain high yielding mushroom house was markedly affected with the yield of middle and late period of picking. 2. Variation of mushroom yield with varying the degree of maturity of fruit bodies appeared to be different between spring and autumn cropping : in case autumn cropping was usally undertaken with normal cropping scheme, mushroom yield showed a reduction in early period of picking but an increase in late period when the mushrooms where picked at pre-matured stage. 3. When the picking rate of button form was laid in the range of 20~45%, and increase of total mushroom yield was accompanied by an increasing tendency of picking rate of button form. 4. Cropping results from mushroom houses could be early predicted from the regression equations, which was based on the analysis of variation in fruit body occurance throughout the picking period.

A Study on the Method of Culture for Paecilomyces japonica Using an Egg (계란을 이용한 눈꽃동충하초 재배방법에 관한 연구)

  • This study was conducted to established method of culture for Paecilomyces japonica using an egg. Mycelia grew favorably at the temperature of 22~26$^{\circ}C$ on eggs. 5.1g of dry matter basis(average 7.2cm of longer and 199.6 of numbers) of artificial fruiting bodies were harvested at 60 days after inoculation from one of egg. Commercial fruiting bodies of Paecilomyces japonica from silkworms was used for comparative nutriental contents. Cordycepin contents of fruit bodies of Paecilomyces japonica cultivated on eggs and silkworms were not significantly different. Crude fat contents of fruiting bodies of Paeilomyces japonica cultivated from eggs was significantly higher than from silkworms(P<0.05). Mn and Cu contents of fruiting bodies of Paecilomyces japonica cultivated from silkworms were significantly higher than from eggs(P<0.05), but Na, Mg, Fe and Zn contents were significantly higher from eggs(P<0.05). Glycine, Arginine and Proline contents in the fruiting bodies of Paecilomyces japonica cultivated from silkworms were tend to higher than from eggs, but Serine, cystein. methionine, isoleucine and phenylalanin were tend to higher from eggs. These results were made possible that possible mass production of artificial fruiting bodies of Paecilomyces Japonica cultivated on eggs.

  • Auxotrophic mutants were obtained by UV-irradiation to mycelium of Ganoderma lucidum. Induction rate of auxotrophs was 5.78%. Interspecific fusion products of protoplasts were obtained by polyethylene glycol induced fusion of protoplasts from auxotrophic mutants of Ganoderma lucidum and Ganoderma applanatum. Fusion products were selected by means of the comparison with the mycelial growth rate and colony morphology. Fusion products were confirmed by mycelial morphology and esterase isozyme pattern. Some segregants were observed and fusion product produced fruit bodies.

Cultivation characteristics and genetic diversity of wild-type collections of Hericium erinaceus in Korea (국내 노루궁뎅이버섯 야생수집균주의 자실체 특성 및 유전적 다양성)

  • Wild-type isolates of H. erinaceus were collected from different geographical areas of Korea. Nineteen isolates were cultured on mushroom substrate for producing fruiting bodies. Of these, 14 isolates formed pinheads and fruiting bodies on the substrate. The morphological and cultivation characteristics of fruiting bodies were categorized by pinheading, fruit body formation, spine types, fresh weights, and colors. Microstructures, including spines, spores, and basidia on the fruiting bodies were observed using scanning electron microscopy. The H. erinaceus isolates demonstrated different PCR polymorphisms produced by universal fungal primers (UFPs) and were classified into four groups based on their high genetic diversity.

  • We investigated the cultivation characteristics of Neolentinus lepideus (21 strains derived from Korea and Japan) on Pinus densiflora sawdust medium as follows; period of mycelial growth, fruiting period, productivity of fruit body and color and morphology of fruit body. The periods for mycelial growth and fruiting on the sawdust medium of N. lepideus were 40~65 days and 6~22 days, respectively. It was produced approximately 48.3 g of fresh weight and 9 fruit-bodies per 600 g of sawdust medium. The productivity of fruit body was reduced by scratching off the surface of sawdust medium. We suggest that three strains (KFRI 1199, 1302, 1331) might be good candidates for sawdust cultivation.

Effect of schizandra berry dregs and rice bran treatment on γ-aminobutyric acid (GABA) content enhancement in Pleurotus ostreatus (오미자박과 미강 첨가배지가 느타리버섯 자실체의 γ-aminobutyric acid(GABA) 함량에 미치는 효과)

  • This study was carried out to establish a cultivation technique for increasing the ${\gamma}$-aminobutyric acid (GABA) content in the fruit body of mushrooms by adding processed by-products. For the oyster mushroom 'Heucktari', addition of green tea powder, sea tangle powder, and green tea dregs resulted in very poor primordia formation, fruit body growth, and increased GABA. However, addition of 10% schizandra berry dregs and 1% rice bran to the basal substrate induced 100% and 10% increases, in GABA content in the fruit bodies compared to the control treatment without by-product, respectively. In addition, fruit body growth and primordia formation were greatly increased by these treatments. Therefore, GABA content was increased when the substrate was prepared by mixing an appropriate amount of schizandra berry dregs and rice bran.

Monitoring of Feed-Nutritional Components, Toxic Heavy Metals and Pesticide Residues in Mushroom Substrates According to Bottle Type and Vinyl Bag Type Cultivation (버섯의 봉지재배 및 병재배 시 재배단계별 배지의 사료영양적 성분, 독성중금속 및 잔류농약 모니터링)

  • This study was carried out to monitor feed-nutritional components, toxic heavy metals (Cd, Pb and As) and pesticide residues through three cultivation stages (1st initial culture stage, 2nd mycelial growth stage, and 3rd fruit body-harvested stage) of king oyster mushroom (Pleurotus eryngii) produced by bottle type cultivation and oyster mushroom (Pleurotus osteratus) produced by vinyl bag type cultivation. For both cultivation types, compared with the initial culture, the weight reduction rate in spent mushroom substrates (SMS) after fruit body harvest was 29% for total wet mass, 21~25% for dry and organic matters and 19 ~22% for neutral detergent fiber. Two thirds to 3/4 of organic matter degraded and utilized by mycelia and fruit bodies was originated from fiber, of which the primary source (50~70%) was hemicellulose. The effect of mycelial growth stage on chemical compositional change in culture was little (P>0.05) for bottle type cultivation of king oyster mushroom but considerable (P<0.05) for vinyl type cultivation of oyster mushroom. Culture nutrients uptake by fruit bodies was very active for the bottle type cultivation. Compared with SMS, harvested fruit bodies (mushrooms) contained higher (P<0.05) crude protein, non-fibrous carbohydrate, and crude ash and lower (P<0.05) neutral detergent fiber. Regardless of stages, no culture samples were contaminated with toxic heavy metals and pesticide residues. In conclusion, the increase of fiber (neutral and acid detergent fibers) and indigestible protein contents and the decrease of true protein content in SMS indicated that the feed-nutritional value of SMS was significantly reduced compared with that of the initial culture and they were safe from toxic heavy metals and pesticide residues.

  • The ultrastructural changes of embryo and endosperm cells were observed during the green fruit with embryo about $250{\mu}$ long to germination. 1. In the embryo cells of green fruit with embryo about $250{\mu}$ long, mitochondrial cristae and plastid are undifferentiated and dictyosome are occasionally observed. There are electron-opaque globoids in the vacuole and a lot of spherosomes in the outer layer of smooth endoplasmic reticulum. Endosperm is filled with spherosomes and electron-opaque protein bodies surrounded by spherosomes, and due to these, other organelle are not observed. 2. In the embryo cells of seeds with red seed coat, mitochondrial cristae are well developed, electron-opaque globoids increased, and vacuoles are enlarged. In the endosperm, however, spherosomes increased, protein bodies are enlarged, and electron-opaque globoidal crystals are dispersed within them. 3. In the procambium and epicotyl cells of dehiscent seed, Golgi vacuoles and vesicles are well developed, and mitochondrial cristae are also well differentiated. Spherosomes are numerously present and radicle cells, peripheral cells of hypocotyl, and vacuoles of cotyledon are well differentiated. Endosperm is filled with spherosomes containing electron-opaque granules and protein bodies are surrounded by a single membrane. There are acid phosphatase around globoids and spherosomes. 4. At the time of seeding, spherosomes markedly increased in the outer layer of cotyledon and protein bodies are also observed. Cell organelles are differentiated and plastids containing starch are also present. 5. In the outer $2{\sim}3$ layers of cotyledons, radicle cells, and peripheral cells of hypocotyl during post-seeding to germination, spherosomes and plastids with starch increased, and mitochondria and microbodies are also found around the nucleus of embryo cells. With approaching, the germination stage, in the endosperm contacting with embryo, vacuoles are well differentiated but spherosomes decreased. There increased electron-opaque materials within vacuoles. In other endosperm, with the decrease of spherosome, mitochondria increased and electro n-opaque globular bodies are formed and gradually increased. The outer layer of protein bodies are reduced while electron-transparent portions are enlarged and fused together to occupy the outer layer where small particles are formed. 6. In the endosperm of germination stage, spherosomes decreased while protein bodies, are fused together to form 2 or 3 within a cell.

Yield, Nutritional Content, and Antioxidant Activity of Pleurotus ostreatus on Corncobs Supplemented with Herb Residues

  • Improper disposal of herb residues in China has caused severe problems to the surrounding environment and human safety. Three herb residues, i.e., compound Kushen injection residues (CKI) and part one and part two of Qizhitongluo Capsule residues (QC1 and QC2, respectively), were used for the cultivation of Pleurotus ostreatus. The effect of the supplementation of corncobs (CC) with different herb residues on yield, nutritional composition, and antioxidant activity of P. ostreatus was investigated. Compared to the control, the higher mycelial growth rate was observed on substrates CC +30% CKI and CC +30% QC1, while the higher yield was obtained from substrates CC +30% QC2 and CC +30% CKI. Moreover, chemical analysis of fruit bodies revealed that the addition of herb residues to CC significantly increased proteins, amino acids, ashes, minerals (Na and Ca), and total phenolic contents but significantly reduced carbohydrates and $IC_{50}$ values of DPPH radicals. In addition, no heavy metals (Pb, Cd, and As) were detected in the fruiting bodies harvested from different substrate combinations. These results demonstrated that mixtures of CC with herb residues might be utilized as a novel, practical, and easily available substrate for the cultivation of P. ostreatus, which is beneficial for the effective management of herb residues.

Diversity and Occurrence Frequency of Ectomycorrhizal Fruiting Bodies by Planting Sites (식재지별 외생균근성 버섯의 다양성 및 발생 빈도 비교)

  • This study was conducted to investigate the diversity and occurrence frequency of ectomycorrhizal fruit bodies by planting sites from June 2000 to October 2001. A total of 3 classes 3 subclasses 8 orders 22 families 41 genera and 72 species (including two varieties) including saprophytic and ectomycorrhizal fungi was investigated. The mushrooms are classified into 9 families 21 genera and 48 species in Agaricales, 5 families 11 genera and 13 species in Aphllophorales, 3 families 3 genera and 4 species in Heterobasidiomycetes and 5 families 6 genus and 7 species in Gasteromycetdae. A total of 7 families 11 genera 30 species (2,451 ea.) of ectomycorrhizal mushroom was investigated. The occurrence frequency of mushrooms was 1,225, 179 and 130 times for Laccaria vinaceoavellanea, Amanita longistriata and Laccaria amethystea, respectively. The mushroom occurrence of ectomycorrhizal fungi was closely related to climatic conditions such as high air temperature, relative humidity and lots of rainfall from July to August. Diversity and distribution of ectomycorrhizal fungi by plots were very different because of variable local environments and different host plants in experimental plots. Laccaria vinaceoavellanea has showed very low host range of plant specificity because of mushroom occurrence in only Quercus sp. and Amanita longistriata, Russula bella and Inocybe sp. have showed wide host range of plant specificity because of mushroom occurrence in coniferous and broadleaved trees. The environment which has a favorable influence of mushroom occurrence was soil pH, organic matter and T/N ratio of soil enviromental and humidity of climatic environment.
