Effect of Storage Conditions and Scarification on in vitro Seed Germination in Lorathus tanakae Hosok

  • Loranthus tanakae (Franch. & Sav.) is an endangered species of mistletoe, distributed in Korean peninsula. The objective of our research is to determine the effect of storage duration and conditions [air flow (AF) and air tight (AT)] at different temperatures for survivability and germination of mistletoe seeds, and also to monitor the effect of seed scarification on germination in vitro. The result revealed that the seeds stored in natural conditions (no stratification) showed highest survival rate of 100% and retained up to 93.3% even after two months of storage in natural conditions and showed higher germination percentage (90%) compare to after ripened seeds. However, the seed stored at $0^{\circ}C$ decreased the germination percentage (ranged from 63 to 73%). Therefore, it can be confirmed that mistletoe does not need after ripened treatment to promote germination. Our research also showed that the storage of L. tanaka seeds in freezing temperatures of $-20^{\circ}C$ and in room temperature for long time either in AT or AF conditions caused the loss of survival and germination rate. On the other hand, the chemical scarification (0.01N HCl incubation for 12 hrs. at $38^{\circ}C$) method was proven more effective to enhance germination percentage of L. tanakae. Regarding the temperature regime, $22^{\circ}C$ showed early germination of mistletoe seeds in vitro.

Real-Time Measurement of the Liquid Amount in Cryo-Electron Microscopy Grids Using Laser Diffraction of Regular 2-D Holes of the Grids

  • Cryo-electron microscopy (cryo-EM) is now the first choice to determine the high-resolution structures of huge protein complexes. Grids with two-dimensional arrays of holes covered with a carbon film are typically used in cryo-EM. Although semi-automatic plungers are available, notable trial-and-error is still required to obtain a suitable grid specimen. Herein, we introduce a new method to obtain thin ice specimens using real-time measurement of the liquid amounts in cryo-EM grids. The grids for cryo-EM strongly diffracted laser light, and the diffraction intensity of each spot was measurable in real-time. The measured diffraction patterns represented the states of the liquid in the holes due to the curvature of the liquid around them. Using the diffraction patterns, the optimal time point for freezing the grids for cryo-EM was obtained in real-time. This development will help researchers rapidly determine high-resolution protein structures using the limited resource of cryo-EM instrument access.

Durability Performance Evaluation of PolyUrea for Seismic Retrofitting of RC Structures (구조물 내진 보강용 폴리우레아의 내구 성능 평가)

  • An experimental study is needed a reinforcing method for seismic load to apply for RC structures because a lot of earthquakes have frequently happened in the world and those also collapsed infrastructures or damaged human lives. The reinforcing effect of PolyUrea (PU) appeared to be excellent under blast and impact about RC structures. In this study, Stiff Type PolyUrea (STPU) had developed by manipulating the ratio of the components of prepolymer and hardener of PU. And the durability performance evaluation of STPU for deterioration and chemical resistance has been performed. Acid environmental exposure test and ultraviolet (UV) exposure test have been performed as the durability performance evaluation for STPU. Concrete carbonation exposure test and freezing and thawing test for concrete coated with STPU have been performed. The experimental result showed that STPU has high resisting capacity and durability in all tests. Therefore, STPU would be used as seismic reinforcement materials.

Production and Transfer of In Vitro fertilized Hanwoo Embryos with Serum-free Media

  • As a simple and economical method for in vitro produced embryos, we have used BSA instead of serum for the production and embryo transfer of Hanwoo in vitro fertilized (IVF) embryos and obtained the following results: 1) When using serum (FBS; fetal bovine serum) or BSA-containing culture media as the initial culture media for immature oocytes, it is regarded as inappropriate to add only BSA to the culture solutions from maturation of the immature oocytes to development stage culture, but serum still needs be added though there is no significant difference in the concentration, with a change from 5% to 10%. 2) The results of culturing IVF embryos after development (4 cell stage) in the Medium199 solutions containing BSA instead of serum (FBS) showed that 0.3% BSA concentration is not optimal and 0.5% or higher BSA concentration has no significant difference among 0.5%, 0.7%, 1% and 2% (p > 0.05). 3) The post-freezing survival ratio after development in 5% FBS-Medium199 showed that 1% BSA concentration of the culture solution is the most suitable in the BSA concentrations of 0.3% (51%), 0.5% (67%), 0.7% (69%), 1% (77%) and 2% (75%). 4) The pregnancy rates of the transplanted fresh(not frozen) blastocyst had no significant concentration dependency (p > 0.5), and the average pregnancy rate was 63.8%. 14% of overweight calves were found among the calves given birth to by the transfer of IVF blastocysts cultured in the serum-added culture solution, but none was found in the experimental groups in which BSA was added instead of serum.

The effect of various heat covering materials on the sprouting and occurrence of disease, insects during wintering of grape young tree (Shigyoku) (포도(자옥) 유목의 월동시 피복한 보온피복재 종류가 발아 및 병해 발생에 미치는 영향)

  • The effect of heat covering materials during cultivation of Shigyoku grape was examined on the effect of protection from freezing damage, labor reduction, and disease resistance during the wintering. This experimentwas accomplished using different combination of heat covering materials from the end of Nov as follows: straw and heat conservation cover B, kilding, color fabric A, and white needle punching fabric. Capacity of heat conservation was the least form white needle punching fabric, and the differences of other materials was slight. Early sprouting occurred by the treatment with heat conservation cover B, and the rate of sprouting was about 70.7% after 9 days of sprouting, showing 2.3-12.5% increase. Growth effect was not differ between heat conservation materials. Disease and insects occurred by the in dice 3.0-3.2 of crown gall from the heat conservation cover B treatment. Other damages were not observed or very little from other treatments. Demand of labor during steps of these treatments was more observed by 6% for kilding than heat conservation cover B, and those of color fabric A and white needle punching fabric were reduced by 6 and 15%, respectively.

  • Temperature difference power generation using sea water is a method repeatedly closed liquefaction and gasification by using the ammonia (refrigerant) of the deep sea water and surface water with a temperature difference between turning the turbine. The larger the temperature difference between the nature of the temperature characteristic energy generation development, the better. This is the story that the surface waters of the deep-water temperature difference is large. But the winter is not large temperature difference between surface water and deep water has lowered energy efficiency. And desalination technologies accounted for 97% of the earth, but we can not eat the technology to convert sea water into fresh water, fresh water produced by the desalination technology that is available for various industries such as irrigation, drinking water in the vessel.In this paper, LNG transport vessels, based on the LNG transport ship to the temperature difference power generation using cold energy of thermal energy and LNG marine diesel engines, which use the existing order to improve the temperature of the surface waters of the season that is the current problem we propose that a complex development of desalination and desalination of seawater freezing research into hybrid research and utilizing the cold energy of the engine.

Development of a new mini straw for cryopreservation of boar semen

  • Sperm cryopreservation is a fundamental process for the long-term conservation of livestock genetic resources. Yet, the packaging method has been shown, among other factors, to affect the frozen-thawed (FT) sperm quality. This study aimed to develop a new mini-straw for sperm cryopreservation. In addition, the kinematic patterns, viability, acrosome integrity, and mitochondrial membrane potential (MMP) of boar spermatozoa frozen in the developed 0.25 mL straw, 0.25 mL (minitube, Germany), or 0.5 mL (IMV technologies, France) straws were assessed. Post-thaw kinematic parameters were not different (experiment 1: total motility (33.89%, 32.42%), progressive motility (19.13%, 19.09%), curvilinear velocity (42.32, 42.86), and average path velocity (33.40, 33.62) for minitube and the developed straws, respectively. Further, the viability (38.56%, 34.03%), acrosome integrity (53.38%, 48.88%), MMP (42.32%, 36.71%) of spermatozoa frozen using both straw were not differ statistically (p > 0.05). In experiment two, the quality parameters for semen frozen in the developed straw were compared with the 0.5 mL IMV straw. The total motility (41.26%, 39.1%), progressive motility (24.62%, 23.25%), curvilinear velocity (46.44, 48.25), and average path velocity (37.98, 39.12), respectively, for IMV and the developed straw, did not differ statistically. Additionally, there was no significant difference in the viability (39.60%, 33.17%), acrosome integrity (46.23%, 43.23%), and MMP (39.66, 32.51) for IMV and the developed straw, respectively. These results validate the safety and efficiency of the developed straw and highlight its great potential for clinical application. Moreover, both 0.25 mL and 0.5 mL straws fit the present protocol for cryopreservation of boar spermatozoa.

Evaluation of Liquefaction Resistance of Sandy Soil Layer in Songlim-park, Pohang, Using Undisturbed Sample (불교란시료를 활용한 포항시 송림공원 사질토층의 액상화저항강도 평가)

  • In this study, a set of laboratory liquefaction test has been conducted using undisturbed samples collected from Songlim-park, Pohang where liquefaction phenomenon had been observed in 2017. Soil samples were frozen right after tube sampling to minimize the disturbances during transport, storage, and test preparation. Cyclic triaxial test has been carried out to evaluate the liquefaction resistance ratio of undisturbed soil samples. As a result, the liquefaction resistance ratio of samples collected from 8.0~8.8 m and 11.0~11.8 m were almost similar, and these values were approximately 0.04~0.07 larger than values estimated by simplified assessment method using field test results. It is expected that the application of undisturbed sample for the evaluation of liquefaction resistance could contribute to the economical design of geotechnical structures.

Studies on Cryopreservation of Sperm in Sevenband grouper, Epinephelus septemfasciatus and Kelp grouper, E. bruneus (능성어, Epinephelus septemfasciatus와 자바리, E. bruneus의 정자동결보존에 관한 연구)

  • The purpose of this research is to develop the simple freezing method for sperm cryopreservation in the Sevenband grouper and Kelp grouper. The motility and survival rate were checked to investigate the effects of diluent and cryoprotectant on sperm cryopreservation.In cryopreservation experiments, 300 mM glucose and 15% dimethyl sulfoxide (DMSO) were used as a diluent and a cryoprotectant, respectively. As a results of cryopreservation, the motility rate of Sevenband grouper was found to be more than 90%, and that of Kelp grouper was found to be more that 60% in sperm 5 months after cryopreservation.

Review of Frost-susceptibility Testing Methods and Criteria (동상민감성 판정 실내실험법 및 기준에 대한 고찰)

  • Over 100 countries have conducted research on experimental testing methods to assess the frost susceptibility of soils. This study aims to prevent structural damage caused by frost heave. Notably, the United States and Japan, which encompass cold regions such as Alaska and Hokkaido, have actively pursued frost heave research. Through laboratory investigations and field applications, standard testing methods and criteria for evaluating frost susceptibility have been established. However, these methods are complex and their engineering explanations are vague. This study closely compares and analyzes the frost-susceptibility testing methods proposed by ASTM and JGS, considering temperature conditions, specimen size, freezing direction, and drainage conditions. By conducting this comparative analysis, this study aims to shed light on the similarities and differences between the two methods. Furthermore, based on the findings, this study proposes future research guidance for refining frost-susceptibility testing methods and criteria.
