시설재배 오이의 품종별, 생육단계별 항산화능 비교 (Comparison of the Antioxidative Abilities of Greenhouse-Grown Cucumber According to Cultivars and Growth Stages)

  • 시설오이로 많이 재배되고 있는 4 품종을 선발하여 생육단계별로 DPPH radical 소거활성, 아질산염 소거활성, ABTS 소거활성 그리고 항산화효소 활성에 대하여 비교 분석하였다. 장죽청장 품종이 모든 생육단계에서 DPPH radical 소거활성이 상대적으로 높게 나타났으며, 그 외 품종들 간에는 큰 차이를 보이지는 않았다. 또한 생육단계별로는 수확기에 못 미친 미성숙단계에서 DPPH radical 소거 활성이 높게 나타났다. 아질산염 소거활성은 pH가 1.2일 경우 모든 품종과 생육단계에서 40% 이상 분해시켰으며, 특히 장형낙합과 신조은백다다기 품종이 상대적으로 높은 활성을 보였다. 또한 pH 4.0에서는 추출물 모두에서 30% 이상의 아질산염 소거활성을 나타냈으며, pH 6.0에서는 대부분 활성이 없었다. ABTS 소거활성은 장죽청장과 늘푸른 청장 품종이 다른 품종보다 상대적으로 다소 높은 소거능을 나타냈고, 생육단계별로는 대부분의 품종에서 ABTS 소거능에 큰 차이를 보이지는 않았다. SOD 활성은 모든 품종과 전 생육단계에서 95% 이상의 높은 활성을 보였으며, 품종 간에 거의 유의한 차이를 나타내지 않았다. CAT 활성은 전체적으로 장죽청장과 늘푸른청장 품종이 그리고 APX 활성은 늘푸른청장, 장형낙합 품종에서 상대적으로 높은 활성을 나타냈고, POX 활성은 생육단계별로 뚜렷한 경향을 보여 수확기에 이를수록 활성이 현저히 감소하는 결과를 나타냈다. 본 실험의 결과, 오이의 높은 항산화능을 확인함으로서 향후 향장소재 및 식품소재로서의 이용 가치성을 증대시킬 수 있음을 시사한다.

Neuroprotective and Free Radical Scavenging Activities of Phenolic Compounds from Hovenia dulcis

  • The EtOAc-soluble fraction from a methanolic extract of Hovenia dulcis Thunb. exhibited neuroprotective activity against glutamate-induced neurotoxicity in mouse hippocampal HT22 cells. The neuroprotective activity-guided isolation resulted in 8 phenolic compounds (1-8), such as vanillic acid (1), ferulic acid (2), 3,5-dihydroxystilbene (3), (+)-aromadendrin (4), methyl vanillate (5), (-)-catechin (6), 2,3,4-trihydrobenzoic acid (7), and (+)-afzelechin (8). Among these, compounds 6 and 8 had a neuroprotective effect on the glutamate-induced neurotoxicity in HT22 cells. Furthermore, compound 6 had a DPPH free radical scavenging effect with an $IC_{50}$ value of $57.7{\mu}M$, and a superoxide anion radical scavenging effect with an $IC_{50}$ value of $8.0{\mu}M$. Both compounds 6 and 8 had ABTS cation radical scavenging effects with $IC_{50}$ values of $7.8{\mu}M\;and\;23.7${\mu}M$, respectively. These results suggest that compounds 6 and 8 could be neuroprotectants owing to their free radical scavenging activities.

Diallyl Disulfide 가 사염화탄소에 의한 마우스 간손상에 미치는 영향 (Protective Effect of Diallyl Disulfide on the Carbon Tetrachloride-Induced Hepatotoxicity in Mice)

  • This study was intended to clarify the protective mechanism of diallyl disulfide on the carbon tetrachloride-induced hepatotoxicity in mice. It was observed that a powerfully increment of serum alanine aminotransferase activity and hepatic lipid peroxide content after carbon tetrachloride injection were markedly inhibited by the pretreatment of diallyl disulfide (20mg/kg) for 5 days. It was also observed that hepatic aminopyrine demethylase and xanthine ocidase as free radical generating enzymes as well as superoxide dismutase and catalase activities as free frdical scavenging enzymes and hepatic glutathione content were not changed by the pretreatment with diallyl disulfide. But, treatment with diallyl disulfide did signifiantly increase cytosolic glutathione S-transferase activity. However, glutathione S-transferase activity in the presence of diallyl disulfide was not affected in vitro. Therefore, it is concluded that mechanism for the observed preventive effect ofdiallyl disulfide against the carbon tetrachloride-induced hepatotoxicity can be due to the engancement of glutathione S-transferase activity.

Comparison of Hydroxyl Radical, Peroxyl Radical, and Peroxynitrite Scavenging Capacity of Extracts and Active Components from Selected Medicinal Plants

  • The ability of 80% ethanol extracts from five medicinal plants, Aralia continentalis, Paeonia suffruticosa, Magnolia denudata, Anemarrhena asphodeloides, and Schizonepeta tenuifolia, to neutralize hydroxyl radical, peroxyl radical and peroxynitrite was examined using the total oxyradical scavenging capacity (TOSC) assay. Peroxyl radical was generated from thermal homolysis of 2,2'-azobis(2-methylpropionamidine) dihydrochloride (ABAP); hydroxyl radical by an iron-ascorbate Fenton reaction; peroxynitrite by spontaneous decomposition of 3-morpholinosydnonimine N-ethylcarbamide (SIN-1). The oxidants generated react with $\alpha$-keto-$\gamma$-methiolbutyric acid (KMBA) to yield ethylene, and the TOSC of the substances tested is quantified from their ability to inhibit ethylene formation. Extracts from P. suffruticosa, M. denudata, and S. tenuifolia were determined to be potent peroxyl radical scavenging agents with a specific TOSC (sTOSC) being at least six-fold greater than that of glutathione (GSH). These three plants also showed sTOSCs toward peroxynitrite markedly greater than sTOSC of GSH, however, only P. suffruticosa revealed a significant hydroxyl radical scavenging capacity. Seven major active constituents isolated from P. suffruticosa, quercetin, (+)-catechin, methyl gallate, gallic acid, benzoic acid, benzoyl paeoniflorin and paeoniflorin, were determined for their antioxidant potential toward peroxynitrite, peroxyl and hydroxyl radicals. Quercetin, (+)-catechin, methyl gallate, and gallic acid exhibited sTOSCs 40~85 times greater than sTOSC of GSH. These four components also showed a peroxynitrite scavenging capacity higher than at least 10-fold of GSH. For antioxidant activity against hydroxyl radical, methyl gallate was greatest followed by gallic acid and quercetin. Further studies need to be conducted to substantiate the significance of scavenging a specific oxidant in the prevention of cellular injury and disease states caused by the reactive free radical species.

  • In order to find the new hepatoprotective agents the preliminary screening of 12 prescriptions which have been used for the treatment of hepatic disease in Oriental traditional medicine has been carried out.   • One of the important function of skin is the protection from harmful environments. Many studies were carried out to keep the skin from wrinkling and pigmenting. Skin wrinkle and pigmentation could be caused' by the unusual disruption of connective tissue, the formation of free radicals and the radiation of ultraviolet. In this study, the extracts obtained from 23 different kinds of medicinal plants were examined whether they have inhibitory effect on tyrosinase and elastase as well as free radical scavenging effect. All the extracts examined were obtained by using 70% (v/v) ethanol at $60^{\circ}C$. It has been found that there were two medicinal plants which have positive effects matching with the purpose of this study. The extract of Ephedra sinica stapf shows an inhibitory effect on tyrosinase ($IC_{50}$ = 83.7~$\mu\textrm{g}$/ml), on elastase ($IC_{50}$ = $690{\mu}\textrm{g}$/ml), and also shows free radical scavenging effect ($IC_{50}$ = 29.6$\mu\textrm{g}$/ml). The extract of Betula platyphylla Var. shows an inhibitory effect on elastase ($IC_{50}$ = 498.1$\mu\textrm{g}$/ml), and free radical scavenging effect ($IC_{50}$ = 9$\mu\textrm{g}$/ ml). The aim of the study is to find the natural compounds which have function in skin-lightening, anti-wrinkling and anti-oxidation, with safe and active ingredients in cosmetic.

Assessment of free-radical-scavenging and antibacterial activities, and brine shrimp toxicity of Scutellaria pinnatifida (Lamiaceae)

  • Scutellaria pinnatifida A. Hamilt. (Lamiaceae) is an endemic Turkish herb. This plant is also endemic to Iran, and grows abundantly in other central and western Asian countries. Several species of the Scutellaria are known for their traditional uses in the treatment of hypertension, arteriosclerosis, inflammatory diseases, hepatitis, allergy, cancer and diarrhoea. Free-radical-scavenging property, antibacterial activity and brine shrimp toxicity of the n-hexane, dichloromethane (DCM) and methanol (MeOH) extracts of S. pinnatifida were assessed using the 2,2-diphenyl-1-picryl-hydrazyl (DPPH) assay, the resazurin microtitre plate based assay, and the brine shrimp lethality assay, respectively. The DCM and MeOH extracts exhibited free-radical-scavenging property, with the $RC_{50}$ values of 0.362 and 0.127 mg/ml, respectively. Among the solid-phase extraction fractions of the MeOH extract, the 50% aqueous-MeOH fraction showed the highest level of free-radicalscavenging activity ($RC_{50}$ = 0.039 mg/ml). While the DCM extract showed low level of antibacterial activity against Bacillus subtilis and ampicillin-resistant Escherichia coli, the MeOH extract was active against B. cereus, B. subtilis, E. coli and ampicillin-resistant E. coli. However, the minimum inhibitory concentrations (MIC) of the MeOH extract against these bacterial strains were >10 mg/ml. None of the extracts showed any significant toxicity towards brine shrimps ($LD_{50}$ = > 1.00 mg/ml).

  • The steamed root of Rehmannia glutinosa has been used for treatment of inner ear diseases, such as tinnitus and hearing loss in traditional Oriental Medicine. In the present study, we investigated the effect of ethanol extract of steamed root R. glutinosa (SRG) on cisplatin cytotoxicity of HEI-OC1 auditory cells. In addition, to investigate the mechanism of SRG on cisplatin cytotoxicity, the effects of SRG on lipid peroxidation as well as scavenging activities against various free radicals were measured in cisplatin-treated cells. Treatment of SRG protected cells from cisplatin and reduced lipid peroxidation in a dose-dependent manner. Furthermore, SRG demonstrated significant scavenging activity against various free radicals, including superoxide radical, hydroxyl radical, hydrogen peroxide, and DPPH radical. These results indicate that SRG protects cisplatin-induced damages of HEI-OC1 cells through inhibition of lipid peroxidation and augmenting scavenging activities against free radials.

  • The antioxidant activity of Desmodium oldhami (Leguminosae) was determined by measuring the radical scavenging effect on DPPH (1,1-diphenyl-2-picrylhydrazyl). The methanolic extract of D. oldhami showed the strong antioxidant activity. Five compounds, kaempferol-3-O-rhamnopyranoside (afzeline) (1), quercetin-3-O-[$\alpha$-L-rhamnopyranosyl($1{\rightarrow}6$)-$\beta$-Dglucopyranoside] (rutin) (2), kaempferol-3-O-glucopyranoside (astragalin) (3), genistein-7-O-$\beta$-D-glucopyranoside (genistin) (4), kaempferol-3-O-rutinoside (5) were isolated from the active ethyl acetate soluble fraction of D. oldhami through repeated silica gel and Sephadex LH-20 column chromatography. Among them, compound 2 showed the most significant antioxidative effect on DPPH free radical scavenging test. Compounds 1-5 are reported for the first time from this plant.
