Antibacterial Activity of Water Soluble Components of Elfvingia applanata Alone and in Combinations with Quinolones

  • A preparation of water soluble components(EA) was made from carpophores of Elfvingia applanata(Pers.) Karst and its in vitro antibacterial activity on a number of bacterial species was examined by macrobroth dilution assay. Among 16 species of bacteria tested, the most potent antibacterial activity was observed against Staphylococcus epiderrnidis and Proteus vulgaris, of which MICs were 1.25 mg/ml. To investigate the antibacterial effects in combinations of EA with quinolone antibiotics, such as ciprofloxacin, enoxacin, lomefloxacin, norfloxacin, and ofloxacin, the fractional inhibitory concentrations(FICs) and the fractional inhibitory concentration indices(FICIs) for four bacterial strains were determined by macrobroth dilution checkerboard assay. Combinations of EA and quinolones exhibited either additive or indifferent effects of antibacterial activity in most instances. However, both synergistic and antagonistic effects were not observed in any cases.

Miconazole과 Amphotericin B의 항진균 활성에 대한 Anethole의 선택적 병용 효과 (Selective Combination Effect of Anethole to the Antifungal Activities of Miconazole and Amphotericin B)

  • The combination effect of anethole with amphotericin B, fluconazole, miconazole, or 5-fluorocytosine was investigated against Saccharomyces cerevisiae. When combined with $\frac{1}{2}$ minimum inhibitory concentration (MIC) or $\frac{1}{2}$ minimum fungicidal concentration (MFC) of anethole, the antifungal activities of fluconazole and 5-fluorocytosine were not changed, but the fungistatic and the fungicidal activities of miconazole were increased 64-fold, respectively. In the case of amphotericin B, the fungistatic activity was increased 2-fold, while the fungicidal activity was decreased 2-fold. The combination effect of anethole with miconazole or amphotericin B was also investigated at the various concentrations using the macrobroth dilution checkerboard method. The fractional inhibitory concentration (FIC) and the fractional fungicidal concentration (FFC) index between B exhibited the FIC index of 8.25 and the FFC of 32.06, respectively. Thus, it is analyzed that the combination of anethole with miconazole or amphotericin B on the antifungal action shows synergism and antagonism, respectively.

Effect of Natural Extracts on Oral Care Probiotics Weissella cibaria CMU and Periodontal Pathogens

  • The purpose of this study is to determine if natural extracts could be used as an additive in oral health food made with Weissella cibaria CMU (oraCMU). Natural extracts of green tea, mulberry leaf, licorice, and propolis, which are reported to have antimicrobial activities, were selected and used in this study. The minimum inhibitory concentrations (MIC) of extracts on periodontal pathogens such as Fusobacterium nucleatum and Porphyromonas gingivalis and their synergy effects with oraCMU by the fractional inhibitory concentrations methods were measured. From the results obtained, all the extracts showed no effect on the growth of oraCMU. Green tea extract showed the best antibacterial activity with MIC of 1.8 mg/ml against both F. nucleatum and P. gingivalis. In addition, green tea extract had a synergistic effect with oraCMU against F. nucleatum. Therefore, these results suggested that green tea extract is available as an additive in oral health food made with oraCMU.

  • 천연의 붉은 색소인 prodigiosin은 Serratia marcescens에 의해서 생산되며 이는 pyrrolylpyrromethane 골격으로 구성되어 있다. 이 색소는 그 분자가 가진 특성 때문에 넓은 범위에서 활용되고 있다. 또한 항암제, 면역억제제, 항진균제, 살조제 등 다양한 분야의 효과가 보고되소 있다. Methicillin-resistant Staphylococcus aureus (MRSA)는 세계적으로 가장 주요한 원내 감염균으로써, 미국에서 해마다 HIV로 사망하는 수보다 더 많은 사망률을 보이고 있다. 그러므로 MRSA에 대한 새로운 치료제의 개발이 매우 중요하며 급박한 문제이다. 본 연구에서는 중증오염도를 가진 하천수로부터 Serratia sp. PDGS 120915를 분리하였으며 항 MRSA 활성을 가진 prodigiosin에 대하여 연구하였다. 본 연구를 위하여 HPLC를 이용하여 물질을 정제하였으며, 분리된 물질의 항 MRSA 활성을 확인하였다. Prodigiosin의 MRSA에 대한 최소억제농도(Minimal inhibitory concentration; MIC)는 $32{\mu}g/ml$이었으며, 분할저해농도(Fractional inhibitory concentration; FIC)는 ampicillin과 penicillin에서 상승작용이 있는 것으로 나타났다.

Anti-Aspergillus Activities of Plant Essential Oils and Their Combination Effects with Ketoconazole or Amphotericin B

  • The essential oils from Cedrus atlantica, Styrax tonkinensis, Juniperus communis, Lavandula angustifolia, Melaleuca alternifolia, Pelargonium graveolens, Pogestemon patchouli and Rosmarinus officinalis were analyzed by GC-MS. Antifungal activities of the oils were investigated by disk diffusion assay and the broth dilution method against Aspergillus niger and A. flavus. The effects of geraniol and the essential oil fraction from P. graveolens on the antifungal activity of amphotericin Band ketoconazole were examined using a checkerboard microtiter assay against both Aspergillus fungi. Most of the tested essential oils, with the exception of C. atlantica, J. communis, and P. patchouli, significantly inhibited growth of A. niger and to a lesser extent that of A. fIavus, with MICs (minimal inhibitory concentrations) in the range 0.78-12.5 mg/mL. The essential oil fraction of P. graveolens and its main components, geraniol and citronellol, exhibited additive effects with amphotericin B and with ketoconazole against both Aspergillus species, resulting in fractional inhibitory concentration (FIC) indices ranging from 0.52 to 1.00.

갯방풍 지상부 정유 및 그 주성분의 항상제 감수성 및 내성 Streptococcus pneumoniae 균주에 대한 억제효과 (In Vitro Inhibitory Activities of Essential Oils from the Aerial Parts of Glehnia littoralis and its Main Components Against Antibiotic-Susceptible and -Resistant Strains of Streptococcus pneumoniae)

  • To develop a new natural antibiotics from Koran plant resources for dealing with the current situation regarding the rapid increase in antibiotic-resistant pathogen, the in vitro inhibitory activities of essential oils from the young leaves of Glehnia littoralis (Umbelliferae) as well as its main constituents were evaluated against susceptible and resistant species of Streptococcus pneumoniae. The essential oil fraction of G. littoralis and its main components, $\alpha-and\;\beta-pinene$, exhibited significant inhibitory activities against the antibiotic-susceptible and resistant strains of S. pneumoniae, with MICs (minimum inhibiting concentrations) ranging from 4.0mg/ml to 16mg/ml. No remarkable differences were shown between the susceptible and resistant strains. Moreover, the disk diffusion test disclosed that these inhibitory activities were dose­dependent. Furthermore, data from the checkerboard titer test with FICIs (fractional inhibiting concentration indices) from 0.15 to 0.28 indicated synergisms between norfloxacin and $\alpha-or\;{\beta}-pinene$ in activity against S. pneumoniae KCCM49629 and S. pneumoniae CCARM4059.

Activity of Allyl Isothiocyanate and Its Synergy with Fluconazole against Candida albicans Biofilms

  • Candidiasis involving the biofilms of Candida albicans is a threat to immunocompromised patients. Candida biofilms are intrinsically resistant to the antifungal drugs and hence novel treatment strategies are desired. The study intended to evaluate the anti-Candida activity of allyl isothiocyanate (AITC) alone and with fluconazole (FLC), particularly against the biofilms. Results revealed the concentration-dependent activity of AITC against the planktonic growth and virulence factors of C. albicans. Significant (p <0.05) inhibition of the biofilms was evident at ${\leq}1mg/ml$ concentrations of AITC. Notably, a combination of 0.004 mg/ml of FLC and 0.125 mg/ml of AITC prevented the biofilm formation. Similarly, the preformed biofilms were significantly (p <0.05) inhibited by the AITC-FLC combination. The fractional inhibitory concentration indices ranging from 0.132 to 0.312 indicated the synergistic activity of AITC and FLC against the biofilm formation and the preformed biofilms. No hemolytic activity at the biofilm inhibitory concentrations of AITC and the AITC-FLC combination suggested the absence of cytotoxic effects. The recognizable synergy between AITC and FLC offers a potential therapeutic strategy against biofilm-associated Candida infections.

메티실린-내성 포도상구균에 대하여 Brazilein 혼합에 따른 항생제 Hygromycin-b의 상승효과 (Synergistic Effect of Brazilein in Combination with Hygromycin-b against Staphylococcus aureus)

  • Methicillin-resistant Staphylococcus aureus (MRSA) is a substantial contributor to morbidity and mortality. In search of a natural products capable of inhibiting this multidrug resistant bacteria, we have investigated the antimicrobial activity of brazilein (BRZ) isolated from Caesalpinia sappan L. (Leguminosae) against 8 different strains of Staphylococcus aureus (S. aureus). New antimicrobial activity was found using the minimum inhibitory concentrations (MICs), broth dilution as well as checkerboard method. Against the 8 strains, the minimum inhibitory concentrations of BRZ were in the range of $62.5-500{\mu}g/mL$. From those results we performed the checkerboard test to determine the synergism of BRZ in combination with Hygromycin-b (HgB) against 4 strains. The combined activity of BRZ and HgB against 4 strains resulted in a fractional inhibitory concentrations index (FICI) ranging from 0.18-0.5. The effect of BRZ with HgB was found to be synergistic. We found that BRZ reduced the MICs of HgB. BRZ and HgB could lead to the development of new combination antibiotics against MRSA infection.

치아우식증유발세균에 대한 단삼 메탄올추출물과 항생제와의 병용효과 (Synergistic Effect of Methanol Extract of Salvia Miltiorrhiza and Antibiotics against Dental Caries Pathogens)

  • 단삼(Salvia miltiorrhiza Bunge)은 우리나라에서 전통적으로 부종, 관절염, 간염 등과 같은 염증질환의 치료에 사용해왔다. 본 연구에서는 단삼 메탄올 추출물을 이용하여 구강미생물에 대한 항균활성을 확인하였다. 그 결과 메탄올추출물은 실험 되어진 모든 구강미생물에서 강한 항균활성을 나타내었다(MICs, 8 to $64\;{\mu}g/mL$; MBCs, 16 to $64\;{\mu}g/mL$). 추가적으로 ampicillin이나 gentamicin과의 병용투여서 최소억제농도(MIC)와 최소살균농도(MBC)가 감소하는 병용효과를 나타내었다. 더불어 메탄올추출물 단독 사용시 보다 항생제와 병용투여시 빠른 사멸효과를 보였다. 결론적으로 단삼 메탄올추출물이 치아우식을 유발하는 많은 세균들에 대한 항균효과가 뛰어남을 확인 할 수 있었으며, 안정성이 높은 천연 치아우식예방제로서의 개발가능성을 확인하였다.

녹농균에 대한 Aminoglycoside계와 Cephalosporin계의 병합작용 (Combined action of Aminoglycoside and Cephalosporin Against Pseudomonas aeruginosa)

  • Thirty-one strains of Pseudomonas aeruginosa were submitted to the synergistic activity test of amikacin(AK) and gentamicin(GM) combined with moxalactam(MX), ceftizoxime(CTZ) or cefoperazone(CFZ). The minimal inhibitory concentrations(MICs) of each drug and drugs combined in various ratios were measured by checkerboard dilution method. The synergism was determined through analysing the MIC distribution curve on isobologram and calculating the fractional inhibitory concentration index(FICI). MICs of GM, AK, MX, CFZ and CTZ against the 31 tested strains were distributed from $12.5{\mu}g/ml$ to $800{\mu}g/ml$, from $0.8{\mu}g/ml$ to $25{\mu}g/ml$, from $3.1{\mu}g/ml$ to $50{\mu}g/ml$, from $3.1{\mu}g/ml$ to $400{\mu}g/ml$, and from $12.5{\mu}g/ml$ to $100{\mu}g/ml$, respectively. The rate synergism of each drug combination by means of FICl was 45.5% in GM-MX, 36.4% in GM-CFZ, 63.6% in GM-CTZ, 48.6% in AK-MX, 35.3% in AK-CFZ, and 35.7% in AK-CTZ combination. Thus, it is suggested that Pseudomonas aeruginosa may effectively be inhibited by various aminoglycoside and cephalosporin combinations.
