밤나무품종에 따른 밤나무혹벌의 산란 선호성 (Oviposition Preferences of the Oriental Chestnut Gall Wasp, Dryocosmus kuriphilus, on Various Chestnut Varieties)

  • 밤나무혹벌 산란동아수의 품종별 차이를 비교한 결과, 석추 및 다압에서 가장 적었고 전체적으로 신초의 하부로 갈수록 산란동아수가 많았으며 특히, 옥광, 축파, 유마 에서 이러한 현상이 뚜렷하였다. 대부분의 품종에서 동아당 평균 1.0개 이상 다중 산란되었으며 신초하부의 경우 산란수가 재래종, 축파, 옥광, 평기에서는 3.0개 이상이었으며 석추와 다압은 1.0개 미만이었다. 산란동아별 난수는 모든 품종에서 상부의 동아에 산란된 난수가 가장 적었고, 하부의 동아에 많은 경향을 보였다. 신초하부의 경우 축파는 산란된 알이 평균 15개 이상으로 가장 많았던 반면, 다압은 한개 이하로 가장 적었다. 품종별 산란비율은 모든 품종에서 밤나무혹벌이 산란한 동아를 발견 할 수 있었으며, 재래종이 $83.8\%$로 가장 높았고, 축파와 유마에서는 각각 78.5와 $75.9\%$ 이상의 산란율을 나타내었다.

  • 남색긴꼬리좀벌은 밤나무혹벌의 유충을 공격하는 외부기생성 천적으로 판명되었다. 남색긴꼬리좀벌의 수명은 꿀 원액을 공급한 경우 $20^{\circ}C$에서의 암컷성충 $42.8{\pm}9.8$일이었으며 아무것도 공급하지 않은 경우 $3.3{\pm}0.8$일이었다. 산란수는 $20^{\circ}C$에서 $22.3{\pm}12.5$개체, $25^{\circ}C$에서 $42.2{\pm}18.4$개체이었으며 산란초기에 다수 산란하는 경향을 보였다. 알은 장타원형으로 백색이며, 길이는 $0.56{\pm}0.33mm$ 이었고, 충방(gall chamber)당 1개의 알을 산란하나 중복하여 산란하는 경우도 관찰되었다. 유충은 백색이며 길이는 $2.94{\pm}0.18mm$이었고, 암컷 용의 길이는 $2.73{\pm}0.09mm$, 숫컷 용의 길이는 $2.01{\pm}0.18mm$로 암컷이 수컷에 비하여 컸다. 강원도 춘천지역에서는 년 2세대 발생했으며, 월동세대 성충발생기간은 5월 하순-6월 상순이었고 제1세대 성충발생기간은 6월 하순-7월 상순이었다. 경상남도 하동지역에서는 년 3세대 발생했으며, 월동세대 성충발생기간은 5월 중순-6월 상순, 제 1세대는 6월 하순-7월 상순, 제 2세대는 7월 하순-8월 상순이었다.

  • The newly sequenced complete mitochondrial genome of the brown marmorated stink bug, Halyomorpha halys($St{\aa}l$) (Hemiptera: Pentatomidae), is a circular molecule of 16,518 bp with a total A+T content of 76.4% and two extensive repeat regions in A+T rich region. Nucleotide composition and codon usage of H. halys are about average when compared with values observed in 19 other published hemipteran mitochondrial genomes. Phylogenetic analyses using these 20 hemipteran mitochondrial genomes support the currently accepted hypothesis that suborders Heteroptera and Auchenorrhyncha form a monophyletic group. The mitochondrial gene arrangements of the 20 genomes are also consistent with our results.

국내 산림해충 페로몬 연구현황과 발전 방향 (Current Status of Pheromone Research of Forest Insect Pests in Korea and Development Direction)

  • 페로몬을 비롯한 화학통신물질들은 곤충의 화학 통신에 이용되는 물질들로 개체군 모니터링, 대량 포획 및 교미교란 등에 폭넓게 이용되고 있다. 본 종설에서는 북방수염하늘소, 솔수염하늘소, 솔껍질깍지벌레, 광릉긴나무좀, 회양목명나방, 솔알락명나방, 매미나방, 복숭아유리나방, 별박이자나방 등 국내 주요 산림해충의 페로몬 연구현황을 소개하고 국외의 연구 결과와 비교하였다. 국내 연구현황 분석을 통해 추후 산림해충 페로몬 연구 방향을 제시하고 발전 방향에 대해 제언하였다.

  • Biological control of turfgrass insect pest Blitopertha orientalis, forest insect pests, Agelastica coerulea, Meganola melancholia, and Glyphodes perspectalis,vegetable insect pests, Plutella xylostella and Agrotis segetum were conducted with entonopathogenic fungus, Metarhizium anisopliae J-22 isolated from black pine forest soil in Cheju province. Mortality of B. orientalis larvae was 53.3% at the rate of 3.4 $\times$ 1O 7 conidia /ml. A. coerulea and M melancholia larvae showed 100% mortalities at 9.6 $\times$ 106 conidia /ml and 2.7 $\times$ 10 7 conidia /ml as well. However, G. perspectalis larvae were not dead even at 4$\times$ 1O 7 conidia /ml. On the other hand, M anisopliae J-22 was effective against P. xylostella larvae showing 100% mortality at 4 $\times$ 10 7 conidia /ml. Filamentous Fungi Isolated from Platypus koryoensis, the Insect Vector of Oak Wilt Disease in Korea

  • The ambrosia beetle, Platypus koryoensis, is a serious pest of oak trees in Korea. In this study we investigated filamentous fungi present in the body of the beetle. Fourteen genera of filamentous fungi belonging to Ascomycota and Basidiomycota were isolated. Among the isolated fungi, some were able to produce wood degrading enzymes. This is first report of fungi associated with P. koryoensis.

Identification of Entomopathogenic Fungus, Beauveria spp. F-101 Isolated from Thecodiplosis japonensis Using Internal Transcribed Spacer Sequence

  • For the development of the alternative control system against the major forest pests, Beauveria spp. F-101, isolated from a dead larva of Thecodiplosis japonensis, was selected because this isolate showed high pathogenicities against T. japonensis and Acantholyda parki. Beauveria spp. F-101 had irregular clustered conidio-phores and conidia borne on a distinctive apical zigzag extension, and it showed typical characteristic of the genus, Beauveria in morphology. For molecular based-identification, the ribosomal ITS region of Beauveria spp. F-101 was amplified with ITS1 and ITS4 primers, and cloned into pGEM- T Easy vector. The amplified PCR product was 569 bp in size and completely sequenced. The similarities of the cloned ITS sequence were 99 % and 97% to those of B. bassiana and B. brongniartii, respectively. In comparison to other species among the genus Beauveria, the ITS region of Beauveria spp. F-101 showed a similarity of 95% to B. amorpha, 95% to B. tenella, 89% to B. vermiconia and 69% to B. alba, respectively. In addition, in comparison to different genus, it had 95% similarities to Cordyceps militaris and 91% to Paecilomyces tenuipes. Accordingly, the current result suggests that Beauveria spp. F-101 was a variant of B. bassiana and it seems to be a new isolate considering sequence variation in ITS region.

Identification of a pr 1-like Gene of Entomopathogenic Fungus, Beauveria bassiana F-101 Isolated from Thecodiplosis japonensis

  • Beauveria bassiana F-101, which has high toxicity toward Acantholyda parki as well as Thecodiplosis japonensis, was an isolate to develop an alternative control system against the major forest pests. Up to now, in B. bassiana, only one pr1 gene has been isolated and characterized. Therefore, we here reported the identification of a pr1-like gene, which would be a factor of toxicity from B. bassiana F-101. The oligonucleotides for the amplification of the pr1-like gene, were chosen based on the conserved regions of the subtilisin family enzymes, pr1 genes of B. bassiana and Metarhizium anisopliae, and proteinase K of Tritirachium album. The cloned PCR fragment had 1111 bp including 52 bp intron. The deduced Pr1-like peptide showed a low identity with Pr1s of entomopathogenic fungi such as B. bassiana Pr1 (BbPr1) and M. anisopliae Pr1 (MaPr1) as well as the proteinase K of T. album (TaPrK). Instead, the deduced peptide had a substantially high amino acid sequence identity $(>65\%)$ with the serine proteases of Magnaporthe grisea (MgSPM1) and Podospora anserina (PaPspA). These results, therefore, appear to suggest that the putative Pr1-like peptide of B. bassiana F-101 belongs to the subtilisin-like serine protease family and may be a novel gene.

  • Three chalcones and a stilbenoid have been isolated from the stem bark of Lindera erythrocarpa. They were elucidated as 2'-hydroxy-3',4',5',6'- tetramethoxychalcone, 2',4'-dihydroxy-3',6'-dimethoxy chalcone, 2',4',5',6'- tetrahydroxy-3'-methoxychalcone, and 5,6-dihydroxy-2,3,4-trimethoxystilbene. Chemical structures were elucidated on the basis of MS, $^1H,\;^{13}C$ and 2D-NMR spectroscopic data. This is the first report on the isolation of these compounds from the L. erythrocarpa.
