Evaluation of Chemical Composition and In vitro Digestibility of Appennine Pasture Plants Using Yak (Bos grunniens) Rumen Fluid or Faecal Extract as Inoculum Source

  • Pastures of the Apennines of Central Italy contribute to feed resources of high altitude (above 1,300 m sea level) grazing systems. The objective of this study was to evaluate the effectiveness of faecal extract from the yak (Bos grunnienes) as an alternative microbial inoculum to rumen fluid for estimation of digestibility of several forage species. Forage samples produced at high altitude were tested in this study: four legumes (Lathyrus sativus L., Lotus corniculatus L., Onobrychis viciaefolia L. and Trifolium pratense L.), three forbs (Achillea millefolium L., Potentilla reptans L. and Teucrium flavum L.) and one grass (Brachipodyum pinnatum L.) were incubated with yak rumen fluid or faecal extract. A large variability in chemical composition was observed among the species collected. Rumen liquor and faecal samples were collected from adult healthy yak. The $Daisy^{II}$ incubator was used to evaluate the nutrient digestibility of forages using rumen liquor as control and faecal extract as alternative microbial inoculum sources. Filter bags containing samples of browse species were added to the four digestion vessels along with their respective inoculum and then incubated for 48 h and dry matter (DM), organic matter (OM), crude protein (CP), neutral and detergent fiber (NDF) digestibility was determined. There was a significant relationship between estimates, indicating that faecal liquor has the potential to be used instead of rumen fluid for estimation of in vitro digestibility of plants. It is concluded that the $Daisy^{II}$ incubator results are appropriate for the determination of in vitro digestibility of nutrients using faecal liquor to define the potential for adaptation of yak to new pastures.

  • In order to provide recommendations on the most useful forage species to smallholder farmers, eleven grass and eleven legume forages grown in Abomey-Calavi in Republic of Benin were investigated for nutritive value (i.e. chemical composition and energy content) and fermentation characteristics (i.e. gas and volatile fatty acid production, organic matter degradability). The in vitro gas production technique was used, incubating the forages for 120 h under anaerobic condition with buffalo rumen fluid. Compared to legume, tropical grass forages showed lower energy (8.07 vs 10.57 MJ/kg dry matter [DM]) and crude protein level (16.10% vs 19.91% DM) and higher cell wall content (neutral detergent fiber: 63.8% vs 40.45% DM), respectively. In grass forages, the chemical composition showed a quite high crude protein content; the in vitro degradability was slightly lower than the range of tropical pasture. The woody legumes were richer in protein and energy and lower in structural carbohydrates than herbaceous plants, however, their in vitro results are influenced by the presence of complex compounds (i.e. tannins). Significant correlations were found between chemical composition and in vitro fermentation characteristics. The in vitro gas production method appears to be a suitable technique for the evaluation of the nutritive value of forages in developing countries.

  • 본 실험은 이른봄에 많이 생산이용되고 있는 호맥을 수영기, 출영기, 유숙기, 호숙기 및 황숙기의 5단계로 예취하여 이들을 임과로 silage를 만들었을 때 그 품질을 분석비교한 것으로써 그 결과를 요약하면 다음과 같다. 1. Silage의 건물회수율은 난숙기부터 높아졌고 (P<0.01), Total-N에 대한 ammonia 태실표의 비율은 난숙기(9.37%)와 호숙기(12.7%)가 낮았다. (P<0.01). 2.Silage의 건물과 건물회수율 및 건물과 pH의 상관관계는 각각 0.782및 0.785로서 정(+)의 상관이 있었고(P<0.01), 건물회수율과 수분 및 $NH_3-N$간에는 차(일)의 상관이 있었다 (P<0.05). 3. 생율단계별 silage의 유기산함량에 의한 품질평가로서는 난숙기와 호숙기가 우수한 것으로 나타났다. 4. 호맥은 수량과 silage의 품질면에서 볼 때 호숙기에 예취하여 silage로 이용하는 것이 가장 적합하다고 사료된다.

Prediction of the Chemical Composition and Fermentation Parameters of Winter Rye Silages by Near Infrared Spectroscopy

  • This study was carried out to explore the accuracy of near infrared spectroscopy (NIRS) for the prediction of chemical and fermentation parameters of whole crop winter rye silages. A representative population of 216 fresh winter rye silages was used as database for studying the possibilities of NIRS to predict chemical composition and fermentation parameters. Samples of silage were scanned at 1 nm intervals over the wavelength range 680~2,500 nm and the optical data recorded as log 1/Reflectance (log 1/R) and scanned in fresh condition. NIRS calibrations were developed by means of partial least-squares (PLS) regression. NIRS analysis of fresh winter rye silages provided accurate predictions of moisture, acid detergent fiber (ADF), neutral detergent fiber (NDF), crude protein (CP) and pH as well as lactic acid content with correlation coefficients of cross-validation ($R^2cv$) of 0.96, 0.86, 0.79, 0.85, 0.82 and 0.78 respectively and standard error of cross-validation (SECV) of 1.89, 2.02, 2.79, 1.14, 1.47 and 0.46 % DM respectively. Results of this experiment showed the possibility of NIRS method to predict the chemical parameters of winter rye silages as routine analysis method in feeding value evaluation and for farmer advice.

  • 대관령 지역에서 Tall fescue 9개 품종을 공시하여 가을철 예취시기에 따른 2번초의 수량구성 요소와 개체중의 변화를 검토하여 이 지역의 환경조건에 적응성이 높은 품종을 선발하여고 하였으며, 얻어진 결과를 요약하면 다음과 같다. 1. 2번초에서 개체당 경수가 많은 품종(Barcel, Forager)이 개체중도 무거워서 이지역의 환경조건에 적응성이 높은 품종이라고 생각되었다. 2. 가을철 예취시기에 따른 2번초의 개체당 경수는 개체중과 0.1%의 유의한 정상관을 나타냈으나 1경중은 유의한 상관이 인정되지 않았다. 3. 2번초의 편균 개체중이 최대에 도달하는 가을철 예취시기는 $C_3$(10월28일)으로 추정되었다. 4. 2번초의 평균 개체중은 1번초의 평균 개체중과 반비례적인 관계를 나타내었다.

  • Near infrared reflectance spectroscopy (NIRS) has become increasingly used as a rapid and accurate method of evaluating some chemical compositions in forages and feedstuff. This study was carried out to explore the accuracy of near infrared spectroscopy (NIRS) for the prediction of chemical parameters of fresh whole crop barley silages. A representative population of 284 fresh whole crop barley silages was used as a database for studying the possibilities of NIRS to predict chemical composition. Samples of silage were scanned at 1 nm intervals over the wavelength range 680~2,500 nm and the optical data were recorded as log 1/Reflectance (log 1/R) and were scanned in fresh condition. NIRS calibrations were developed by means of partial least-squares (PLS) regression. Evaluation of Physically Effective Fiber for Distributing Total Mixed Rations in Korea

  • This study was carried out to evaluate the physical characteristics of total mixed rations (TMR) prepared in Korea for improving productive effectiveness of dairy and beef cattle. The 40 samples of commercial TMR for dairy and beef cattle in 6 municipals were collected and analyzed for chemical composition and physical properties. There were significant regional differences (p<0.05) in dry matter and crude protein contents of TMR for dairy and beef cattle. In roughage value index (RVI), neutral detergent fiber (NDF) content, and physically effective NDF (peNDF)$_{1.18}$ content, there were no regional differences in TMR for dairy cattle, but there were significant regional differences (p<0.05) in TMR for beef cattle. Thus the results from this study support that because RVI, NDF content, and peNDF content in TMR for dairy and beef cattle exceeded recommended ranges, a little adjustment in TMR for dairy cattle will be necessary to obtain optimal productivity.

품종과 보존제의 처리가 춘계수확 연맥의 특성과 사료가치에 미치는 영향 (Effect of Cultivar and Preservative on Characteristics and Quality of Spring Harvested Oat Hay)

  • An experiment was carried out to determine effect of cultivar and preservative on the characteristics and quality of baled oat(Avena sativa L.) hay after 60 days of storage. Ihe main plots consisted of the cultivars such as 'Foothill' and 'Swan', and the subplots consisted of preservatives such as commercial propionic acid(CA), lactic bacteria inoculant(LB) and control(C0). Bale temperature of 'Foothill' oat hay maintained higher than that of 'Swan' oat hay during storage. Bale temperature of oat hay treated with CA remained lowest during the earlier storage period and no significant difference was found among preservatives during the latter storage period. Visual assessment of bale of 'Swan' oat hay was better in the evaluation of mold, odor and color than that of 'Foothill' oat hay after storage. DM content of Foothill oat hay increased markedly than that of 'Swan' oat hay through the storage period. CFU(Colony fodng unit) of total viable micmrganism of oat hay bales was not quite different between preservatives, but that of lactic bacteria of 'Swan' oat hay and 'Foothill' was different by over 16 and lo4, respectively. Total nitrogen content of 'Swan' and 'Foothill' oat hay was 1.45 and 2.22%, respectively(P<0.05) and effect of preservatives on total N content was higher in the order of LB, CA, and CO.   • This study was investigated the herbage productivity and nutrient contents of corn (Suweon 19 and Kwanganok) and sweet sorghum (Ramiki sorgo and Silage sorgo) at the stage of maturity (10-DBS, silking, milky and dough stages) to identify the utility value of sweet sorghum as a substituting crop for corn. The silage materials (dough stage) were chopped and were storaged for 90 days in polyethlene bag with O.lmm thickness. Chemical composition, nitrogen content and in vitm dry matter digestibility were evaluated. Also, daily intake and palatability were checked in the feeding trial with four male sheep in average weight about 57kg. Both fresh and dry matter yield of corn and sweet sorghum were increased as the maturity processed (P< 0.05), and sweet sorghum showed higher total yield potential than corn. Crude protein content of the cultivars was decreased as the maturity was processed (P< 0.05). Neutral detergent fiber (NDF) content of the cultivars, in general, showed highest at silking stage, followed by 10 days before silking (10-DBS), and milky and dough stages (Pi0.05) in order. Acid detergent fiber (ADF) content was increased as the maturity processed, and it showed the highest value at milk stage. However, the contents of ADF in sorghum plant was decreased during the late maturity. Crude protein content of the silage was not significantly different among cultivars used, while NDF content was highest in Ramiki sorgo, followed by Silage sorgo, Kwanganok and Suweon 19. In v i m DM digestibility of the silage was highest in Kwanganok, followed by Suweon 19, Silage sorgo and Ramiki sorgo. Total N content of the silage was highest in Ramiki sorgo, followed by Suweon 19, Silage sorgo and Kwanganok. DM intake of the silage was highest in Ramiki sorgo, followed by Suweon 19, Silage sorgo and Kwanganok and it was closely related to the palatability value.

Evaluation of Exogenous Promoters for Use in Brachiaria brizantha Transformation

  • Brachiaria (Poaceae) is the most important forage genus for cattle production in Brazil. The genetic breeding of this genus is limited by the incompatibility among species, differences in ploidy level and the natural cloning of plants by apomixis (Valle and Miles 1992). However, plant regeneration via tissue culture methods and genetic engineering provide an opportunity to introduce new characteristics in plants of this genus. We have developed methods for the 'genetic modification of Brachiaria brizantha cv. Marandu via biolistic transformation. A higher number of shoots was obtained with 4 mg/L 2.4-diclorophenoxyacetic acid and 0.2 mg/L benzylaminopurine in calli induction medium and 0.1 mg/L naphtaleneacetic acid and 4.0 mg/L kinetin in shoot regeneration medium. A selection curve for mannose was determined to use phospho mannose isomerase (PMI) gene of Escherichia coli as a selection marker. Calli formation was inhibited from 5 g/L mannose, even in the presence of sucrose while calli that were formed in the presence of mannose failed to develop embryos showing that PMI gene can be used for selection of transformants of this grass. Different promoters were tested to evaluate the efficiency based on the detection of the GUS gene expression (Jefferson et al. 1987). The monocot promoters, act1-D and ubi-1, resulted in higher expression levels than dicot promoters, ubi-3 and act-2, or the CaMV35S and CVMV promoters.
