Simultaneous Determination of 80 Unapproved Compounds using HPLC and LC-MS/MS in Dietary Supplements

  • We developed analytical methods using high performance chromatography (HPLC) and liquid chromatography tandem mass spectrometry (LC-MS/MS) for the simultaneous determination of 80 unapproved compounds in dietary supplements. The target compounds for analysis were unapproved ingredients (e.g., pharmaceuticals) that have potential adverse effects on consumers owing to accidental misuse, overuse, and interaction with other medication in dietary supplement. Two analytical methods were tested to identify the optimal validation results according to AOAC guideline. As a result, limit of quantification (LOQ) was 0.14-0.5 ㎍ mL-1; linearity (r2) was ≥ 0.99; accuracy (expressed as recovery) was 78.9-114%; precision (relative standard deviation) was ≤ 4.28% in the HPLC method. In the LC-MS/MS method, LOQ was 0.01-2 ng mL-1, linearity (r2) was ≥0.98, accuracy was 71.7-119%; precision was ≤ 12.5%. The developed methods were applied to 51 dietary supplements collected from 2019 to 2021 through MFDS alert system. Based on our previous monitoring study, major compounds were icariin, sibutramine, yohimbine, sildenafil, tadalafil, sennosides (A, B), cascarosides (A, B, C, D), and phenolphthalein. In this study, we re-analyzed samples of detected compounds, and evaluated the statistical difference using Bland-Altman analysis to compare two analytical approaches between HPLC and LC-MS/MS. These results showed a good agreement between two methods that can be used to monitor the unapproved ingredients in dietary supplements. The developed two methods are complementarily suitable for monitoring the adulteration of 80 unapproved compounds in dietary supplements.

Rapid Detection for Salmonella spp. by Ultrafast Real-time PCR Assay (Ultrafast Real-time PCR법을 이용한 살모넬라의 신속 검출)

  • Salmonella continue to be a major cause of food poisoning worldwide. The rapid detection method of food-borne Salmonella is an important food safety tool. A real-time polymerase chain reaction (PCR) has been used as a rapid method for the detection of pathogens. It has been recently reported that NBS LabChip real-time PCR is a novel, ultrafast, and chip-type-convenient real-time PCR system. We developed the assay method based on NBS LabChip real-time PCR for the rapid detection of Salmonella, which its reaction time was within 20 minutes. Two target genes (invA and stn) were selected to design target specific primers and probes. The new method was validated by checking specificity and sensitivity (limit of detection). This study included forty-two target and twenty-one non-target strains to assess the specificity. This assay was able to identify the 42 Salmonella strains correctly. The limit of detection (LOD) was $10^1copies/{\mu}L$ in Salmonella genomes DNA, while LOD incubated for 4 hr in the inoculated sausage sample ranged from $10^1CFU/g$ to $10^2CFU/g$ as an inoculated cell count. The assay developed in this study could be applied for the investigation of food poisoning pathogens.

  • Food safety accidents have been increasing by 2% over 5,000 cases every year since 2009. Most people know that the best method to prevent food safety accidents is a quick inspection, but there is a lack of inspection technology that can be used at the non-analytic level to food production and distribution sites. Among the recent on-site diagnostic technologies, the methods for testing gene-based food poisoning bacteria were introduced with the STA technology, which can range from sample to detection. If food safety information can be generated without forgery by directly inspecting food hazard factors by remote, unmanned, not human, pollution sources can be managed by predicting risks more accurately from current big-data and artificial intelligence technology. Since this information processing can be used on smartphones using the current cloud technology, it is judged that it can be used for food safety to small food businesses or catering services.

  • Dietary life of Korean population was greatly improved by the increased food production, enlarged food processing and betterment of food distribution system during the last three decades. On the other hand, food pollution issues have arisen from the increased use of chemicals and advancements in analytical techniques and toxicological information. The health risk of hazardous substances in Korean diet was estimated to be in the decreasing order of heavy metals>pesticide residues>food additives>mycotoxins>radionuclides>PCBs whereas consumers do not recognize it in the same order. It is needed to evaluate the food safety more accurately based on scientific evidences and to adopt reasonable regulatory measures in Korea for the benefits of people's health and national interest.

  • The demand for food safety has emerged as a major social issue due to changes in diet patterns and consumers' perceptions, along with the advancements in society and the development of the food industry. Consumers are demanding more information about the food they consume, and are sensitive to food scandals. With such interest in food safety, blockchain technology is attracting attention as a means of effectively responding to poor food management resulting in food fraud or unsafe distribution. By ensuring the accuracy of, and trust in, traceability in the food supply chain, it is possible to build trust between traders and to ensure safe food distribution. This paper proposes a next-generation agri-food distribution system that can provide and manage (for suppliers, consumers, and distribution officials) a variety of agri-food information, such as the history, distribution, safety, quality, and freshness of food. Information on product status and distribution status in all processes, including production, processing, distribution, sales, and consumption, can be monitored and controlled in real time (anytime, anywhere), and users can check the safety level of each type of food in real time through an app.

  • The meanings, classification, and regulatory status of 'natural food colorants' are discussed. Nowadays the technological advances for processing Cochineal color is flourishing here and aboard, makes highly concentrated products (e.g., 95% Carminic acid) is comercially available. Application of carminic acid for a food system needs target-specific formulation, mixing with alumn, tartaric acid, and other salts to stabilize the color hue in food system. On the other hand, carmine, the aluminium-lake of carminic acid, is more convenient product to be used and would give more strong color. Of these reasons Carmine is much popular product than Cochineal extract in Western and European countries. We insist that carmine should be also certified as natural colorant together with Cochineal extract in Korea. Authors expect that several kinds of anthraquinone colorant (Lac and Kermes) would be introduced in Korean market in near furture.

Safety Assessment of Foods Produced Using Recombinant DNA Techniques

  • The introduction of genetically modified crops has raised concerns regarding safety issues over the insertion of foreign genes into plant genomes using recombinant DNA technology. Since 1991 in Japan, 29 foods and 6 food additives have been evaluated, based on the "Guideline for Safety Assessment", before these foods were marketed. The MHW, however, decided that safety assessment of such foods and food additives should be legally imposed. because soon such foods and food additives are expected to circulate globally and a new system for assessing safety of such foods and food additives at a pre-market stage is necessary, in order to avoid the distribution of any genetically modified foods that have had no safety assessment. The MHW published relevant announcements to amend existing regulations on 1 May 2000. "Standards for safety assessment of seed plant" is established based on a concept of substantial equivalence, and applicable to the products which are regarded as equivalent to the existing products used as foods and food additives. The characterization of the food products entails consideration of the molecular characterization. phenotypic and compositional characteristics, key nutrients and toxicants, and toxicity and allergenicity of the introduced proteins, and if there are indications of unintended effects of the modification, whether further safety testing (animal studies etc.) is needed should be considered. Safety and wholesomeness studies with whole foods should be care fully designed in order to avoid nutritional imbalances causing artifacts and uninterpretable results as was the case of Dr. Pusztaiis report. A case study of genetically modified soybeans (glyphosate-tolerant soybeans) on the immune system of rats and mice is shown. Chemical compositions were also compared with those of the non-GM soybeans. The studies failed to detect any differences in immuno-toxic activity.muno-toxic activity.

  • In the 4th industrial revolution, Artificial Intelligence (AI), big data, Internet of Things (IoT) are already around us, making our society hyper-connected and blurring the lines between the digital and biological spheres. We witness drastic changes not only in the food industry, but also in economy, society and our life as a whole. Technologies bring industrial reorganization and greater changes at the system level and the food industry is not exceptional. Human demand for foods continues to grow and the very nature of the food industry remains unchanged, but its production, distribution and marketing face unprecedent innovations. Passing through the global pandemic, the food industry has been evolved into 'contact-free', as the safety become our top priority. Amid the gradual shift to technology-oriented society, the smarter food safety management skills and tools are being adopted in many countries exerting greater efforts to enhance traceability and to upgrade AI-powered safety management system.
