전반사 형광 이미지 분석을 통한 세포 부착점의 운동 특성에 관한 연구 (On the Dynamic Characteristics of Cell Contact by Analyzing TIRE Images)

  • We carried out an image analysis of living cells forming their contacts at the bottom of the cell culturing substrate. In order to visualize the contact area selectively, we adopted total-internal-reflection-fluorescence (TIRF) method, which can illuminate the specimen volume within only several hundred nano-meters above the substrate. From the fluorescent intensity of the TRF image, we could calculate the distance of the cell surface from the substrate. As a result, we visualized the origin of cell contacts, their movements, and the change of cell-contact type from the close-contact into focal-contact with information of its vertical displacement representing the temporal evolution process of the three-dimensional cell-surface-profile near the contact area during this metamorphosis.

  • The microbolometer array sensor with fine pitch pixel array has been implemented to the released amorphous silicon layer supported by two contact pads. For the design of focal plane mirror with geometrical flatness, the simple beam test structures were fabricated and characterized. As the beam length decreased, the effect of beam width on the bending was minimized, Mirror deformation of focal plane in a real pixel showed downward curvature by residual stress of a-Si and Ti layer. The mirror tilting was caused by the mis-align effect of contact pad and confirmed by FEA simulation results. The properties of bolometer have been measured as such that the NETD 145 mK, the TCR -2 %/K, and thermal time constant 1.99 ms.

마이크로 유체렌즈를 이용한 마이크로 PIV 측정에 관한 실험적 연구 (Experimental Study on Micro PIV Measurement using a Micro Liquid Lens)

  • In the present study, we performed the velocity field measurement in a microchannel using a focal length variable micro liquid lens. The liquid lens is used as a beam expander in a micro-PIV system to acquire the scatter image of the seeded particle. A thin film-type micro liquid lens was made of PDMS material and it was attached on top of the 700-micron-wide working fluid supply channel trench. As a result, the focal length and contact angle of the liquid lens changed with variations in applied pressure.

  • Cell adhesion is a coordinated process involving initial binding of integrin receptors to extracellular matrix (ECM), recruitment of adhesion proteins, and focal adhesion assembly. The formation of mechanically stable focal adhesion assembly of cells within surrounding ECM is a key parameter to direct numerous cellular functions including cell migration, differentiation, and apotosis. With current cell adhesion assays, it is difficult to understand contributions of each coordinated event on evolution of cell adhesion strengthening since cells spontaneously spread upon their adhesion to the substrate, thus remodeling their cytoskeletal structure. In this presentation, novel approaches for analysis of cell adhesion strengthening process based on the combination of mechanical device, micro-patterned substrates, and molecular biological techniques will be discussed.

Adhesion of Human Osteoblasts Cell on CrN Thin Film Deposited by Cathodic Arc Plasma Deposition

  • Interaction between human osteoblast (hFOB 1.19) and CrN films was conducted in vitro. CrN films were produced by cathodic arc plasma deposition. The surface was characterized by atomic force microscopy (AFM). CrN films, glass substrates and TiN films were cultured with human osteoblasts for 48 and 72 hours. Actin stress fiber patterns and cell adhesion of osteoblasts were found less organized and weak on CrN films compared to those on the glass substrates and the TiN films. Human osteoblasts also showed less proliferation and less distributed microtubule on CrN films compared to those on glass substrates and TiN films. Focal contact adhesion was not observed in the cells cultured on CrN films, whereas focal contact adhesion was observed well in the cells cultured on glass substrates and TiN films. As a result, the CrN film is a potential candidate as a surface coating to be used for implantable devices which requires minimal cellular adhesion.

  • 물체의 형상검사를 위한 검사 방법 가운데 광삼각법을 이용한 레이저 삼각검사 방식은 원격 비접촉식의 고정밀, 고속 측정이 가능하다는 장점이 있다. 이러한 레이저 삼각측정 방식은 평면뿐만 아니라 경사면에 위치한 홀의 길이를 측정하는데 이용하고 있다. 그러나 기존의 방식으로는 렌즈의 초점 거리가 정해져 있었으므로 한정된 영역만을 측정할 수 있다는 단점이 있었다. 이러한 문제는 CCD 카메라와 측정 대상체 사이의 거리를 변화시키는 간단한 방법으로 해결할 수 있다. 이를 원리로 좀더 효율적인 방식을 채택한 것이 가변적인 초점 거리를 갖는 줌 렌즈를 사용하는 방법이다 주 논문에서는 가변적인 초점 거리를 갖는 줌렌즈를 사용함으로써 측정 물체의 크기에 따라 렌즈 배율을 최적화하여 개선된 분해능을 갖는 형상측정 시스템을 제안하였고 정밀도의 향상을 실험적으로 실증하였다.

Clostridium difficile Toxin A Inhibits Erythropoietin Receptor-Mediated Colonocyte Focal Adhesion Through Inactivation of Janus Kinase-2

  • Previously, we demonstrated that the erythropoietin receptor (EpoR) is present on fibroblasts, where it regulates focal contact. Here, we assessed whether this action of EpoR is involved in the reduced cell adhesion observed in colonocytes exposed to Clostridium difficile toxin A. EpoR was present and functionally active in cells of the human colonic epithelial cell line HT29 and epithelial cells of human colon tissues. Toxin A significantly decreased activating phosphorylations of EpoR and its downstream signaling molecules JAK-2 (Janus kinase 2) and STAT5 (signal transducer and activator of transcription 5). In vitro kinase assays confirmed that toxin A inhibited JAK 2 kinase activity. Pharmacological inhibition of JAK2 (with AG490) abrogated activating phosphorylations of EpoR and also decreased focal contacts in association with inactivation of paxillin, an essential focal adhesion molecule. In addition, AG490 treatment significantly decreased expression of occludin (a tight junction molecule) and tight junction levels. Taken together, these data suggest that inhibition of JAK2 by toxin A in colonocytes causes inactivation of EpoR, thereby enhancing the inhibition of focal contact formation and loss of tight junctions known to be associated with the enzymatic activity of toxin A.

고농축 표적을 이용한 비파괴검사용 $^{192}Ir$ 미세초점선원 개발 (Development of $^{192}Ir$ Small-Focal Source for Non-Destructive Testing Application by Using Enriched Target Material)

  • 하나로와 동위원소 생산시설을 활용하여 비파괴검사용 $^{192}Ir$ 미세초점선원의 생산기술을 개발하였고 현장적용시험을 통하여 개발된 제품의 성능을 확인하였다. 직경 0.5 mm ${\times}$ 높이 0.5 mm인 선원의 개발을 위해 알루미늄 캡슐 압착 방법 및 장치를 개발하여 표적을 제작하였으며, 하나로를 이용한 방사능 생성량을 평가한 결과 약 3.0 Ci 방사능의 선원 제조가 가능함을 확인하였다. 또한 컴퓨터 CPU 및 탄소강에 대한 비파괴검사를 실시하여 투과도계 감도 및 기하학적 불선명도가 비교 대상 선원에 비해 우수함을 확인하였다. 이 선원의 초점크기는 기존의 선원에 비해 매우 작기 때문에 근접 및 접촉촬영과 튜브-튜브시트 용접부의 비파괴검사에 있어 기하학적 불선명도를 최소화 할 수 있을 것이다.

  • The robot systems are widely used in the welding manufacturing. The essential tasks to operate the welding robot are the acquisition of the position and/or shape of the parent metal. For the seam tracking or the robot automation, many kinds of contact and non-contact of the system which monitors the shape of the welding part is described. This system uses the line-type structured laser diode and the visual sensor. It includes the correction of radial distortion which is often found in the image from the camera with short focal length. Direct Linear Transformation (DLT) is used for the camera calibration. The three dimensional shape of the parent metal is obtained after simple linear transformation. Therefore, the system operates in real time. Some experiments are carried out to evaluate the performance of the developed system.
