Monitoring of Changes in Molecular Weight Distribution and Fluorescence Properties of Dissolved Matter (DOM) in Water Treatment Processes (정수처리공정 중 자연유기물질의 분자량 분포 및 형광특성 변화 모니터링)

  • Monitoring of NOM characteristics is important for improving removal efficiency of natural organic matter (NOM) in water treatment processes. In this study, several NOM characteristics, which include specific UV absorbance (SUVA), total carbonate content, molecular weight distribution, and fluorescence properties, were measured using samples collected from a pilot-scale water treatment plant consisting of coagulation/flocculation (C/F), filtration, ozonation and granular activated carbon (GAC) processes. The highest removal of NOM was observed in C/F and filtration processes as demonstrated by the reduction of dissolved organic carbon (DOC) by 25% and 21%, respectively. Despite nearly no change in DOC, however, the lowest SUVA value and the highest total carbohydrate content were observed in the sample from ozonation process. This indicates that non-degradable aromatic compounds become depleted and biodegradable organic compounds are enriched during the process. Comparison of synchronous fluorescence spectra of the samples showed that ozoation process increased protein-like fluorescence while it decreased fulvic-like and terrestrial humic-like fluorescence. Consistently, a slight peak of protein-like fluorescence was observed in the sample from ozonation process. The greatest change in molecular weight distributions of the samples was observed in C/F process. Comparison of size exclusion chromatogram of the samples revealed that NOM fractions with the molecular weight greater than 2000 Da were reduced by over 90% after C/F process. SUVA values and total carbohydrate content of the samples were well correlated with a ratio of protein-like fluorescence and terrestrial humic-like fluorescence intensities with the correlation coefficients of 0.99 and 0.91, respectively. This suggests that synchronous fluorescence properties of NOM could be used as useful tolls for monitoring changes of some NOM characteristics during water treatment processes.

  • A tetradentate ligand of 2-phenyl-4,6-di(pyridin-2-yl)pyrimidine (L) has been synthesized and its complexes with $ZnI_2$ and CuI have been obtained by hydrothermal method. single crystal X-ray diffraction analysis indicates that ligand L coordinates with Zn(II) ions to form a simple four-coordinate di-nuclear complex, while the complexation of L with Cu(I) constructs a one-dimensional chain polymer. The existence of $I^-$ ion hampers the L to assemble grid-type complexes with Zn(II) and Cu(I). Fluorescence spectra show that the L emits blue fluorescence while its Cu(I) polymer decrease the fluorescence intensity and Zn(II) complex quenches the fluorescence.

  • Fluorescence quenching of norfloxacin (NOR) by Cu2+, Ni2+, Co2+ and Mn2+ was studied in water. The change in the fluorescence intensity and lifetime was measured as a function of quencher concentration at various temperatures. According to the Stern-Volmer plots, the NOR was quenched both by collisions and complex formation with the same quencher. However, the static quenching had a more important effect on the emission. Large static and dynamic quenching constants support significant ion-dipole and orbital-orbital interactions between NOR and cations. The both quenching constants by Cu2+ were the largest among quenchers. Also, quenching mechanism of Cu2+ was somewhat different. The change in the absorption spectra due to the quencher provided information on static quenching. The fluorescence of NOR was relatively insensitive to both the dynamic and static quenching compared with other quinolone antibiotics. This property can be explained by the twisted intramolecular charge transfer.

Dyeing of High Strength and High Molecular Weight Polyethylene Fiber Using Super Hydrophobic Red Fluorescence Dyes (고강도/고분자량 폴리에틸렌 섬유의 적색 초소수성 형광염료 염색)

  • Three super hydrophobic red fluorescence dyes were selected to dye high molecular weight polyethylene fiber. Their absorbance and emission spectra were obtained and Stokes' shift was measured. Fluorescence emission strength of the dyes on the fiber was investigated and therefore Fluoro Red 3 was determined as the best one among those three dyes in this experiment. Dyeing properties and fluorescence intensities were investigated using the Fluoro Red 3 on high molecular weight polyethylene fiber at various dyeing conditions. The optimum concentration of a dispersing agent was appeared at 10wt% in aqueous solution. The best dyeing was obtained at $125^{\circ}C$ for 1 hour. The color fastnesses to the washing and rubbing were as high as ratings 4~5, however, the fastness to light was exhibited ratings 2~3.

Determination of Adulteration of Chicken Meat into Minced Beef Mixtures using Front Face Fluorescence Spectroscopy Coupled with Chemometric

  • The objective of this study was to explore the potential of front face fluorescence spectroscopy (FFFS) as rapid, non-destructive and inclusive technique along with multi-variate analysis for predicting meat adulteration. For this purpose (FFFS) was used to discriminate pure minced beef meat and adulterated minced beef meat containing (1%, 2%, 3%, 4%, 5%, 10%, 20%, 30%, 40%, 50%, 60%, 70%, 80%, 90%, and 100%) of chicken meat as an adulterant in uncooked beef meat samples. Fixed excitation (290 nm, 322 nm, and 340 nm) and fixed emission (410 nm) wavelengths were used for performing analysis. Fluorescence spectra were acquired from pure and adulterated meat samples to differentiate pure and binary mixtures of meat samples. Principle component analysis, partial least square regression and hierarchical cluster analysis were used as chemometric tools to find out the information from spectral data. These chemometric tools predict adulteration in minced beef meat up to 10% chicken meat but are not good in distinguishing adulteration level from 1% to 5%. The results of this research provide baseline for future work for generating spectral libraries using larger datasets for on-line detection of meat authenticity by using fluorescence spectroscopy.

  • Polysaccharides have attracted great attention for their wide range of applications in biological and medical fields. In this paper, the interaction of polysaccharides with human serum albumin (HSA) was systematically investigated by fluorescence (FL) spectroscopy and circular dichroism (CD) spectra under different conditions. The Stern-Volmer quenching constants ($K_a$) at different ionic strength and pH were calculated, and information of the structural features of HSA was discussed. FL and CD results indicate that both hydrophobic and electrostatic interactions play important roles during the binding process. The quenching of the fluorescence resulting the binding of polysaccharides and HSA is static.

  • A rapid and simple method for detecting chemical preservatives was attempted on the basis of emitted fluorescence at the illumination of UVSL-25 mineral light. Absorption and fluorescence spectra of powdered samples dispersed in liquid paraffin revealed characteristic patterns depending on chemical preservatives. Detection of chemical preservatives was more readily accomplished by simultaneous comparison of spectral characteristics at long and short wave ranges of the exciting light.

  • The steady-state emission spectra of dicoumarol (DC) in ethanol and EPA have been examined at various temperatures (77-298 K). At room temperature, a fluorescence spectrum of DC in ethanol shows a emission maximum at 350 nm. In EPA a Stokes-shifted emission band appears around 470 nm in addition to the 350 nm emission, and its intensity is enhanced as temperature decreases. This emission is attributed to a zwitterionic tautomer of DC formed by a single excited-state intramolecular proton transfer (ESIPT) along the internal hydrogen-bonding. The fluorescence lifetimes have been measured at 350 and 450 nm as a function of temperature. The fluorescence decay at 350 nm is single exponential at any temperature, whereas the one at 450 nm becomes biexponential at temperatures below 250 K. These results are discussed in terms of a conformational change followed by the ESIPT. The activation energy barrier for the conformational change has been determined to be 3.7 $\pm$ 0.2 kJ/mole.

  • Binding of six cephalosporins (cefotaxime, cefuroxime, cefazoline, cephalothin, cephaloridine, cephacetrile) to bovine serum albumin was studied. Fluorescence probe technique and difference spectrophotometry were employed to evaluate the nature and degree of association of cephalosporin albumin complex. 1-Anilinonaphthalene-8-sulfonate (ANS) was used as the fluorescence probe. 2-(4'-hydroxybenzeneazo) benzoic acid(HBAB) was employed as the UV spectrophotometric probe. Compentitive bindings between cephalosporins and probes were observed. The number of binding sites of bovine serum albumin for each cephalcsporin is 2. Among six cephaloporins, cefotaxime has the highest binding constant followed by cafazoline, cefuroxime, cephalothin, cephaloridine and cephacetrile.

  • A series of $Er^{3+}/Yb^{3+}$ co-doped fluorophosphate glasses with varying $YbF_3$ concentration were prepared by a high temperature melt quenching technique. Absorption and emission cross-sections were determined by using the McCumber theory. The larger emission cross-section ($9.86{\times}10^{-21}cm^2$) and longer fluorescence lifetime (12.37 ms) were obtained for the $^4I_{13/2}{\rightarrow}^4I_{15/2}$ transition of ABS3Er4Yb glass. The sensitivity and temperature of the maximum sensitivity were evaluated by the fluorescence intensity ratio method from the measured upconversion spectra. The results were discussed and compared to the other reported glasses.
