Ganglioside GD1a is specifically formed by the addition of sialic acid to ganglioside GM1a by ST3 ${\beta}$-galactoside ${\alpha}$-2,3-sialyltransferase 2 (ST3GAL2). Above all, GD1a are known to be related with the functional regulation of several growth factor receptors, including activation and dimerization of epidermal growth factor receptor (EGFR) in tumor cells. The activity of EGF and EGFR is known to be a very important factor for meiotic and cytoplasmic maturation during in vitro maturation (IVM) of mammalian oocytes. However, the role of gangliosides GD1a for EGFR-related signaling pathways in porcine oocyte is not yet clearly understood. Here, we investigated that the effect of ST3GAL2 as synthesizing enzyme GD1a for EGFR activation and phosphorylation during meiotic maturation. To investigate the expression of ST3GAL2 according to the EGF treatment (0, 10 and 50 ng/ml), we observed the patterns of ST3GAL2 genes expression by immunofluorescence staining in denuded oocyte (DO) and cumulus cell-oocyte-complex (COC) during IVM process (22 and 44 h), respectively. Expression levels of ST3GAL2 significantly decreased (p<0.01) in an EGF concentration (10 and 50 ng/ml) dependent manner. And fluorescence expression of ST3GAL2 increased (p<0.01) in the matured COCs for 44 h. Under high EGF concentration (50 ng/ml), ST3GAL2 protein levels was decreased (p<0.01), and their shown opposite expression pattern of phosphorylation-EGFR in COCs of 44 h. Phosphorylation of EGFR significantly increased (p<0.01) in matured COCs treated with GD1a for 44 h. In addition, ST3GAL2 protein levels significantly decreased (p<0.01) in GD1a ($10{\mu}M$) treated COCs without reference to EGF pre-treatment. These results suggest that treatment of exogenous ganglioside GD1a may play an important role such as EGF in EGFR-related activation and phosphorylation in porcine oocyte maturation of in vitro.

We have developed a novel type of polymer nanogel loaded with anticancer drug based on bio-derived poly(${\gamma}$-glutamic acid) (${\gamma}$-PGA). ${\gamma}$-PGA is a highly anionic polymer that is synthesized naturally by microbial species, most prominently in various bacilli, and has been shown to have excellent biocompatibility. Thiolated ${\gamma}$-PGA was synthesized by covalent coupling between the carboxyl groups of ${\gamma}$-PGA and the primary amine group of cysteamine. Doxorubicin (Dox)-loaded ${\gamma}$-PGA nanogels were fabricated using the following steps: (1) an ionic nanocomplex was formed between thiolated ${\gamma}$-PGA as the negative charge component, and Dox as the positive charge component; (2) addition of poly(ethylene glycol) (PEG) induced hydrogen-bond interactions between thiol groups of thiolated ${\gamma}$-PGA and hydroxyl groups of PEG, resulting in the nanocomplex; and (3) disulfide crosslinked ${\gamma}$-PGA nanogels were fabricated by ultrasonication. The average size and surface charge of Dox-loaded disulfide cross-linked ${\gamma}$-PGA nanogels in aqueous solution were $136.3{\pm}37.6$ nm and $-32.5{\pm}5.3$ mV, respectively. The loading amount of Dox was approximately 38.7 ${\mu}g$ per mg of ${\gamma}$-PGA nanogel. The Dox-loaded disulfide cross-linked ${\gamma}$-PGA nanogels showed controlled drug release behavior in the presence of reducing agents, glutathione (GSH) (1-10 mM). Through fluorescence microscopy and FACS, the cellular uptake of ${\gamma}$-PGA nanogels into breast cancer cells (MCF-7) was analyzed. The cytotoxic effect was evaluated using the MTT assay and was determined to be dependent on both the concentration and treatment time of ${\gamma}$-PGA nanogels. The bio-derived ${\gamma}$-PGA nanogels are expected to be a well-designed delivery carrier for controlled drug delivery applications.
Eukaryotic translation termination is governed by eRF1 and eRF3. eRF1 recognizes the stop codons and then hydrolyzes peptidyl-tRNA. eRF3, which facilitates the termination process, belongs to the GTPase superfamily. In this study, the effect of the MC domain of eRF1a (eRF1aMC) on the GTPase activity of eRF3 was analyzed using fluorescence spectra and high-performance liquid chromatography. The results indicated eRF1aMC promotes the GTPase activity of eRF3, which is similar to the role of eRF1a. Furthermore, the increased affinity of eRF3 for GTP induced by eRF1aMC was dependent on the concentration of $Mg^{2+}$. Changes in the secondary structure of eRF3C after binding GTP/GDP were detected by CD spectroscopy. The results revealed changes of conformation during formation of the eRF3C GTP complex that were detected in the presence of eRF1a or eRF1aMC. The conformations of the eRF3C eRF1a GTP and eRF3C eRF1aMC GTP complexes were further altered upon the addition of $Mg^{2+}$. By contrast, there was no change in the conformation of GTP bound to free eRF3C or the eRF3C eRF1aN complex. These results suggest that alterations in the conformation of GTP bound to eRF3 is dependent on eRF1a and $Mg^{2+}$, whereas the MC domain of eRF1a is responsible for the change in the conformation of GTP bound to eRF3 in Euplotes octocarinatus.
3주간 정상적으로 생장한 닭의장풀을 Hoagland 용액 (대조구, 100 μM Cd/sup 2+/, 100 μM Cd/sup 2+/ + 100 μM ABA, 100 μM Cd/sup 2+/+50 mM KCl)에서 1주간 수경재배 한 후 생장, 엽록소 함량, 엽록소 형광, 수분 스트레스와 Cd/sup 2+/의 축적을 조사하여 Cd/sup 2+/ 축적과 식물의 생리적 반응과의 연관성을 찾고자 하였다. 식물의 생장에서 Cd/sup 2+/과 Cd/sup 2+/+ABA 처리구는 대조구에 비해 각각 71%와 81%생장이 감소하였고, Cd/sup 2+/+KCl 처리구는 Cd/sup 2+/ 처리구와 생장에 큰 차이가 없었다. 엽록소 함량은 Cd/sup 2+/, Cd/sup 2+/+ABA, Cd/sup 2+/+KCl 처리구에서 대조구에 비해 각각 32%, 41%, 29% 억제되었다. 엽록소 형광 측정의 경우 Cd/sup 2+/ 처리구는 대조구에 비해 모든 광도에서 Fv/Fm 비율이 14∼20% 감소하였다. Cd/sup 2+/+ABA 처리구는 Fv/Fm 비율이 23% 내외에서 감소하였으며, Cd/sup 2+/+KCl 처리구는 Cd/sup 2+/ 처리구와 Fv/Fm 비율에 큰 차이가 없었다. 수분 스트레스에 대한 Cd/sup 2+/과 ABA 처리 효과는 Cd/sup 2+/ 처리구에서 대조구에 비해 2.5배, Cd/sup 2+/+ABA 처리구에서 3배, Cd/sup 2+/+KCl 처리구에서 2.25배 각각 수분 스트레스가 증가하였다. Cd/sup 2+/ 수송에 대한 ABA의 효과를 살펴보면, Cd/sup 2+/+ABA 처리구에서 ABA는 Cd/sup 2+/ 수송을 억제하거나 촉진시키지는 않았지만 Cd/sup 2+/의 뿌리 축적을 약 13% 심화시켰다. 반면에 KCl은 뿌리 내 Cd/sup 2+/ 축적을 약 60% 억제시켰다. 따라서 대표적인 스트레스 내성 호르몬 ABA는 Cd/sup 2+/축적으로 인해 발생된 식물의 생리적 독성을 완화시켜 주지 못했으며. 오히려 독성을 증가시켰다.

Mitochondrial calcium overload is a crucial event in determining the fate of neuronal cell survival and death, implicated in pathogenesis of neurodegenerative diseases. One of the driving forces of calcium influx into mitochondria is mitochondria membrane potential (${\Delta}{\psi}_m$). Therefore, pharmacological manipulation of ${\Delta}{\psi}_m$ can be a promising strategy to prevent neuronal cell death against brain insults. Based on these issues, we investigated here whether nobiletin, a Citrus polymethoxylated flavone, prevents neurotoxic neuronal calcium overload and cell death via regulating basal ${\Delta}{\psi}_m$ against neuronal insult in primary cortical neurons and pure brain mitochondria isolated from rat cortices. Results demonstrated that nobiletin treatment significantly increased cell viability against glutamate toxicity ($100{\mu}M$, 20 min) in primary cortical neurons. Real-time imaging-based fluorometry data reveal that nobiletin evokes partial mitochondrial depolarization in these neurons. Nobiletin markedly attenuated mitochondrial calcium overload and reactive oxygen species (ROS) generation in glutamate ($100{\mu}M$)-stimulated cortical neurons and isolated pure mitochondria exposed to high concentration of $Ca^{2+}$ ($5{\mu}M$). Nobiletin-induced partial mitochondrial depolarization in intact neurons was confirmed in isolated brain mitochondria using a fluorescence microplate reader. Nobiletin effects on basal ${\Delta}{\psi}_m$ were completely abolished in $K^+-free$ medium on pure isolated mitochondria. Taken together, results demonstrate that $K^+$ influx into mitochondria is critically involved in partial mitochondrial depolarization-related neuroprotective effect of nobiletin. Nobiletin-induced mitochondrial $K^+$ influx is probably mediated, at least in part, by activation of mitochondrial $K^+$ channels. However, further detailed studies should be conducted to determine exact molecular targets of nobiletin in mitochondria.
본 연구에서는 멸종위기식물인 큰바늘꽃(Epilobium hirsutum L.)의 효과적인 보전 및 복원을 위한 기초자료를 얻기 위해 토양의 수분함량과 유기물함량이 번식계절과 생리 반응에 어떠한 영향을 주는지에 대하여 알아보았다. 큰바늘꽃은 다년생식물이지만 모든 구배에서 한 해에 생식생장을 하였다. 꽃봉오리, 꽃 그리고 열매주머니는 수분구배와 영양소구배에서 각각 높은 수분 조건과 높은 유기물 조건에서 가장 이른 시기에 성숙하였다. 그리고 꽃 수와 열매주머니 수는 높은 수분 조건과 높은 유기물 조건에서 더 빨리 증가하였다. 엽록소 함량은 수분구배에서 높은 중간 수분 조건과 높은 수분 조건에서 가장 많았고, 영양소구배에서는 차이가 없었다. 최소엽록소형광 값은 수분구배와 영양소구배 모두 차이가 없었고, 최대엽록소형광 값은 높은 수분 조건과 높은 유기물 조건에서 가장 높았다. 광계 II의 광화학적 효율 값은 모든 수분구배에서 0.75로 차이가 없었고, 영양소구배에서의 경우 높은 유기물 조건에서 0.78로 가장 높았다. 큰바늘꽃은 수분이 증가할수록 엽록소 함량이 많아지고, 유기물이 증가할수록 Fv/Fm 값이 높아졌다. 이상의 연구결과는 토양의 충분한 수분과 유기물 함량은 큰바늘꽃의 번식계절을 앞당겨 주고 생식생장을 촉진한다는 것을 보여준다. 추후 멸종위기종인 큰바늘꽃의 개체군 유지와 서식지를 관리하는데 중요한 정보가 될 것으로 판단된다.
이 연구의 목적은 와동벽에서 다른 위치에서의 상아질 접착제의 두께를 평가하고, 이런 다양한 접착제의 두께와 미세 인장 강도 사이의 관계를 평가하기 위한 것이다. 여섯 개의 인간 대구치에 모든 상아질 면이 노출되도록 I급 와동을 형성하였다 3개의 치아는 filled adhesive ($Clearfil^{TM}$ SE bond)를 와동 내에 도포하였고, 다른 3개의 치아는 unfilled adhesives ($Scotchbond^{TM}$ Multi Purpose)를 도포하였다. 형광 현미경을 이용하여 접착층의 형태와 두께를 관찰하였다. 접착제의 두께는 수직 와동벽을 따라 와동 변연, 와동벽 1/2, 와동 내각의 세 지점에서 측정되었다. $Scotchbond^{TM}$ Multi Purpose와 $Clearfil^{TM}$ SE bond가 와동 변연과 와동벽 1/2, 와동 내각에서의 접착제의 두께를 재현하여 미세 인장 결합 강도를 측정하였다. 이 실험의 결과에서 두 가지 상아질 접착제 모두에서 와동 내각에서의 접착제의 두께가 와동 변연과 와동벽 1/2위치에서의 두께보다 두꺼웠으며, 와동 내각의 두꺼운 접착제의 미세 인장 결합 강도는 와동 변연과 와동벽 1/2에서의 얇은 접착제 두께의 미세 인장 결합 강도보다 유의성 있게 높게 나타났다.
본 연구에서는 사용 후 태양광 셀을 HCl 용액 및 초음파세척기의 cavitation효과를 사용하여 셀 표면의 불순물(Al, Zn, Ag 등)을 제거하여 Si을 선택적으로 회수하기 위한 최적 공정 조건을 찾기 위한 연구를 진행하였다. 태양광 셀에서 Si을 선택적으로 회수 하기 위해 HCl 용액 및 초음파세척기를 사용하여 침출을 진행하였고, 반응이 끝난 태양광 셀은 증류수로 세척 후 건조 오븐에 건조를 실시하였고, 반응된 HCl 용액은 감압 여과 실시 후 여과된 용액은 ICP-Full Scan 분석을 실시하였다. 또한, 건조 된 태양광 셀은 유발을 사용하여 파쇄 후 XRD, XRF, 및 ICP-OES 분석을 실시하였으며, 이를 통해 Si의 순도 및 회수율을 알 수 있었다. 실험은 산용액 농도, 반응 온도, 반응 시간, 초음파 세기를 변수로 두고 실험을 진행하였다. 위 과정을 통해 최종적으로 태양광 셀로부터 Si을 선택적으로 회수하기 위한 최적 공정 조건은 산용액 농도 3M HCl, 반응 온도 60℃, 반응 시간 120min, 초음파 세기 150W인것을 알 수 있었고, 최종적으로 Si의 순도는 99.85%, 회수율은 99.24%로 측정되었다.
본 연구는 그람 음성균과 양성균인 E. coli와 L. monocytogenes를 인위적으로 오염시킨 음용수를 대상으로 UV-C 정수기의 살균 가능성과 미생물의 생육 저해에 따른 형태학적 특성을 조사하였다. UV-C를 이용한 음용수 살균 능력은 그람 음성균 E. coli와 양성균 L. monocytogenes의 저농도부터 고농도(E. coli 3.2×103-3.2×107 CFU/2.8 L; L. monocytogenes 8.4×103-8.4×107 CFU/2.8 L) 세균 모두의 생육을 저해할 수 있는 효과가 나타났다. 따라서 UV를 이용한 음용수 살균은 유속 3.4 L/min에서 UV-C 파장 254 nm, 조사선량 40 mJ/㎠로 조사할 경우 E. coli 3.2×107 CFU/2.8 L와 L. monocytogenes 8.4×107 CFU/2.8 L 이하 농도의 오염된 음용수의 살균이 가능한 것으로 나타났다. 정수기에 UV-C 살균장치를 추가하는 것이 물의 미생물 안전성에 효율적이라고 사료되며, 이 연구결과는 UV살균 장치 활용의 기초자료로 제공되어 향후 연구 수행에 도움을 줄 수 있을 것이다.
본 연구에서 N. commune으로부터 U937과 SK-HEP-1 세포에 대해 apoptosis를 유도하는 광과민성물질을 순수 분리하였다. N. commune을 dichloromethane : Methanol (1:1)로 추출하여 Hexane, EtOAc, MeOH로 분획하였다. 높은 광역학활성을 나타내는 EtOAc 분획물에서 NC-1을 분리하였으며, 분자량 870의 pheophytin a ($C_{55}H_{74}N_4O_5$)인 것으로 구조를 동정하였다. 광역학활성 조사는 NC-1을 $1.15{\mu}M$에서 $23.00{\mu}M$까지 5개의 농도를 처리하여 광을 조사하는 것과 조사하지 않는 조건으로 나누어 평가하였다. U937과 SK-HEP-1 두 세포에서 apoptotic body 형성, cell blebbing 등의 형태적 변화와 세포독성, caspase 활성, 세포핵의 응축, DNA 단편화 등이 광 의존적이며 농도 의존적으로 활성이 증가하였다. 추가적으로 유세포분석을 통한 apoptosis 유도의 정량적 분석 결과에서도 광조건에서 농도가 증가할수록 apoptosis 유도가 증가하는 것을 확인하였다. 이러한 결과를 통해 NC-1의 U937과 SK-HEP-1 세포에 대한 사멸은 광역학적 apoptosis에 의한 것임을 확인하였다. 본 연구 내용을 N. commune으로부터 광감각제 개발의 기초자료로 활용할 수 있을 것이다.

제 5 장 계약 해지 및 이용 제한

② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
