Toxicity of Fungicides in vitro to Cylindrocarpon destructans

  • As part of a study on the ability of fungicides to control disappearing root rot of ginseng (Panax quinquvdius) caused by Cylindruarpn destmtans, 15 fungicides were screened for toxicity to the fungus in vitro. Highly toxic fungicides were Benlate (benomyl), Thiram (thiram), and Orbit (propiconazole). EC5O values (mg a.i./L) were less than 1 and EC95 values were less than 10. Crown (carbathiin and thiabendazole), ASC-66835 (fluazinam), and UBI-2584 (tebuconazole) were moderately toxic, with EC5O values in the range 1-10 and EC95 values in the range 32-45. Weakly toxic fungicides (EC5O in the range 20-80, EC95 in the range 35-140) included UBI-2643 (thiabendazole), UBI-2565 (cyproconazole), and Vitaflo-280 (carbathiin and thiram). Anvil (hexaconazole), Vitaflo-250 (carbathiin), UBI-2383 (triadimenol), Daconil (chlorothalonil), CGA-173506 (fludioxonil), and CGA-169374 (difeno- conazole) were considered nontoxic to C. destmtan (EC5O 1.29->600, EC95>500). Relations between proportional inhibition of growth and concentration of fungicide were linear on arithmetic plots (Benlate, UBI-2643, UBI-2565, Vitaflo-280) or logarithmic plots (all other fungicides). Based on toxicity in vitro and formulation, it is recommended that Benlate, Orbit, and ASC-66835 be tested as soil drenches, and Benlate, Thiram, UBI-2584, and Crown be tested as seed treatments for controlling disappearing root rot.

Survey of Fungicide Resistance for Chemical Control of Botrytis cinerea on Paprika

  • Four hundred and sixty six isolates of B. cinerea were obtained from infected leaves, stems and fruits of paprika grown in greenhouses or plastic film houses in Gangwon province, Korea, between August and November in 2006 and 2007. These isolates were classified into five representative phenotypes of resistant (R) and sensitive (S) reactions as SSR, SRR, RSS, RRS and RSR according to the responses of isolates against benzimidazole, dicarboximide and N-phenyl-carbamate fungicide in order. The percentage of five phenotypes were 51.3, 2.4, 35.6, 8.1 and 2.6%, respectively. The SSR phenotype (51.3%) was the most common. Among the nineteen fungicide products evaluated to compare their efficacy against gray mold pathogen on the paprika fruit inoculated with fungal mycelia, the mixture of diethofencarb and carbendazim was the most effective followed by iprodione, boscalid, the mixture of iprodione and thiophanate-methyl, fludioxonil, polyoxin-B, fluazinam, the mixture of tebuconazole and tolyfluanid and procymidone; while in the assay methods inoculated with fungal spores, the mixture of tebuconazole and tolyfluanid was the most effective in controlling gray mold followed by boscalid, fludioxonil, the mixture of diethofencarb and carbendazim and the mixture of pyrimethanil and chlorothalonil.

Effects of Prochloraz and Tebuconazole on Control of Fusarium Bulb and Root Rot of Oriental Orchid, Cymbidium goeringii (Prochloraz와 Tebuconazole의 Fusarium oxysporum에 의한 춘란(Cymbidium goeringii) 구경썩음병 방제효과)

  • Eight fungicides including prochloraz, tebuconazole, benomyl, mancozeb, thiophanate-methyl, azoxystrobin, and fluazinam were examined for their control effects on Fusarium bulb and root rot of oriental orchid, Cymbidium goeringii. Among the chemicals, prochloraz and tebuconazole were the most effective on suppression of the causal pathogen, F. oxysporum in vitro and on control of the disease in vivo. Prochloraz and tebuconazole inhibited mycelial growth of the fungi 95~100% at 10ppm a.i. and microconidial germination 75~100% at 100ppm a.i. Prochloraz and tebuconazole showed 80~92% and 84~88% protective control value on the disease, respectively. However, curative effects on infected orchid were relatively low. Other chemicals showed no or lower than 20% curative and 50% protective control value. Results indicated that prochloraz and tebuconazole can be used for the control of the Fusarium bulb and root rot of oriental orchids. However, the chemicals need to be applied prior to the disease development to achieve successful control efficacy.

Review of Researches on Clubroot Disease of Chinese Cabbage in Korea and Future Tasks for Its Management (우리나라 배추 뿌리혹병 연구 현홍과 향후과제)

  • Clubroot disease of curcifer crops caused by Plasmodiophora brassicae had been first reported in 1928 in Korea, and maintained mild occurrence until 1980s. Since 1990s the disease has become severe in alpine areas of Kyonggi and Kangwon, gradually spread to plain fields throughout the country, and remains as the great-est limiting factor for its production. Researches on the disease has begun in late 1990s after experiencing severe epidemics. Survey of occurrence and etiological studies have been carried out, particularly, on the pathogen physiology, race identification, quantification of soil pathogen population, and host spectrum of the pathogen. Ecology of gall formation and its decay, yield loss assessment associated with time of infection, and relationships between crop rotation and the disease incidence was also studied during late 1990s. In studies of its control, more than 200 crucifer cultivars were evaluated for their resistance to the disease. Lime applica-tion to field soil was also attempted to reduce the disease incidence. Resistant radish and welsh onion were recommended as rotation crops with crucifers after 3-year field experiments. However, so for, most studies on clubroot disease in Korea have been focused on chemical control. Two fungicides, fluazinam and flusulfamide, were selected and extensively studied on their application technologies and combination effects with lime application or other soil treatment. To develop environmentally-friendly control methods, solar-disinfection of soil, phosphoric acid as a nontoxic compound, and root-parasiting endophytes as biocontrol agents were examined for their effects on the disease in fields. In the future, more researches are needed to be done on development of resistant varieties effective to several races of the pathogen, establishment of economically-sound crop rotation system, and improvement of soil-disinfection technique applicable to Korean field condi-tion, and development of methodology of pretreatment of fungicides onto seeds and seedbeds.

Fungicide selections for control of chili pepper stem rot caused by Sclerotium rolfsii using an agar dilution method

  • Sclerotium rolfsii causing southern blight on numerous vegetable and fruit crops was isolated from stems of chili peppers showing wilting symptoms. The pathogen was identified by morphological observation and DNA sequencing analysis of ITS region. To select an effective fungicide for control of southern blight, we investigated the inhibition efficacy of thirty fungicides included in nine groups of fungicides with different mechanisms of action. A fungal growth inhibition assay was conducted through an agar dilution method by using mycelial discs and sclerotia of the pathogen as inoculum, respectively. When mycelial discs were used as an inoculum, several fungicides showed good inhibitory activity against the mycelial growth of S. rolfsii 12-6. All DMI fungicides tested had a good inhibition except for prochloraz which had low inhibitory effect. All strobilurin fungicides tested except for kresoxim-methyl and all SDHI fungicides tested except for boscalid and fluopyram, had a good inhibition. Also, fludioxonil, a protective fungicide and fluazinam had a good inhibitory effect. Interestingly, when sclerotia were used as an inoculum, inhibition efficacy was increased for fluopyram, a SDHI fungicide, and for some protective fungicides such as propineb, chlorothalonil, dithianon, and folpet. All the fungicides selected in this study should be tested in the field for their control activities against stem rot for practical use in chili pepper cultivation.

Development of assay method for the activities of new compounds, and the effect of several fungicides against spore germination, adhesion, and myceial growth of Colletotrichum sp. causing red pepper anthracnose (고추 탄저병균의 포자 발아와 부착, 균사 생장에 미치는 화합물의 활성 검정법 확립 및 살균제의 효과)

  • With microtiter plate, the assay method was developed for detecting the fungicidal activity of new compounds against spore germination, spore adhesion and mycelial growth of Colletotrichum sp. JC24 cal1Sing red pepper anthracnose. Also, the effects of some commercialized fungicides on fungal development like above mentioned were investigated by measuring the optical density of mycelia grown into wells of microtiter plate. For the standardization of assay method, some factors, such as the treatment of MTT and/or propanol, inodulum density and incubation period, affecting on mycelial optical density were investigated. For obtaining precise and consistent mycelial optical density, it was necessary the treatment of MTT for 12 hrs and propanol for 1 hr. inoculum density adjusted to $1\times10^5$ spores/mL and incubation period for 36 hrs at $25^{\circ}C$. For fungicidal activities, 6 protective fungicides, 6 ones inhibiting sterol biosynthesis, and one inhibiting respiration were used in this study. While mancozeb, chlorothalonil and dithianon among 6 protective fungicides inhibited strongly spore germination, adhesion, and mycelial growth at $6.25{\mu}g/mL$, propineb, iminoctadine and fluazinam inhibited intermediately spore germination and mycelial growth at $100{\mu}g/mL$. Washing above 3 fungicides with new PD broth, their activity against spore adhesion decreased. With hexaconazole, tebuconazole and myclobutanil, the tendency of the activity against fungal differentiation of the early infection stage was similar to the latter group of protective fungicides, showing the decrease of the inhibitory activity against spore adhesion by washing 2 hrs after incubation. However, kresoxim-methyl inhibited spore adhesion distinctly, depending on the applied concentrations. Based on these results, it might be able to assess the fungicidal activity of many compounds against spore germination, adhesion and mycelial growth by the use of microtiter plate in vitro. Using the assay developed in this report, it was possible to investigate the inhibitory activity of some commercialized fungicides, too.

Plant Diseases of Safflower (Carthamus tinctorius) and Their Chemical Control (잇꽃의 식물병 발생양상과 주요 식물병의 약제방제)

  • This study were carried out to identify pathogens and determine the seasonal occurrence and chemical control of safflower (Carthamus tinctorius) diseases from 2000 to 2002 in Gyengbuk province, Korea. Major diseases of safflower were, anthracnose caused by Colletotricum acutatum in open field, and gray mold by Botrytis cinerea in rain sheltered plastic house. Other diseases occurred were powdery mildew caused by Sphaerotheca fuliginea, collar rot by Sclerotium rolfsii, leaf spot by Alternaria carthami and A. alternata, rust by Puccinia carthami, root-rot and stem-rot by Phytophthora cactorum, root-rot and wilt by Fusarium oxysporum and damping-off by Pythium ultimum. Seasonal occurrence of anthracnose on safflower has begun from late April, and increased until harvesting, especially rapid increased after rainfall during stem elongation season that is from May to June. In open fields, maximum incidence of anthracnose was 67 % in late July. But in rain-sheltered plastic house, it was very low, about 5% in July. Gray mold caused by Botrytis cinerea was most important disease in rain-sheltered plastic house cultivation. Maximum incidence of gray mold on floral head was 27.4%, whereas other diseases occurred below 1 %. In the test of the chemical control of the safflower anthracnose, metiram WP, carbendazim$.$kasugamycin WP and iminoctadintris$.$thiram WP were the highest controlling chemicals. In chemical control of gray mold, iminoctadintris$.$thiram WP, fluazinam WP and iprodion WP showed highest controlling effects.

Biocontrol Efficacy of Endophytic Bacteria Flavobacterium hercynim EPB-C313 for Control of Chinese Cabbage Clubroot (Flavobacterium hercynium EPB-C313 균주를 이용한 배추 뿌리혹병 생물적 방제)

  • Clubroot of Chinese cabbage by Plasmodiophora brassicae, was found to be high virulent to the Chinese cabbage, turnips and cabbage. It this study, the endophytic bacteria Flavobacterium hercynium EPB-C313, which was isolated from tissues of Chinese cabbage, was investigated the antimicrobial activity on the inactivation of resting spores and its control effect on clubroot disease by bioassay. The bacterial cells, culture solutions, and culture filtrates of F. hercynium EPB-C313 inactivate the resting spores of P. brassicae with 90.4, 36.8, and 26.0%, respectively. The clubroot was inhibited with 100% by dipping the seedlings of Chinese cabbage in culture solutions of F. hercynium EPB-C313 before planting, however the chemmical 'fluazinam' was 91.7% in pot tests. Complex treatment were highly enhanced control efficacy with 63.7% at field of 50% diseased plants by soil incorporation with the pellet contains organic matter and F. hercynium EPB-C313, seedling drench of culture solution of F. hercynium EPB-C313 and soil drench with 10 days after planting. These results imply that the F. hercynium EPB-C313 is a very useful biological control agent of clubroot disease of Chinese cabbage.

Some Environmental Factors Affecting Germination and Survival of Resting Spores of Plasmodioprora brassicae (배추무사마귀병균 휴면포자의 발아 및 생존에 미치는 몇가지 환경요인)

  • Effect of temperature on resting spore germination of Plasmodioprora brassicae was indirectly estimated based on examining temporal change of number of inactive resting spores. Resting spore germination was the highest at $28^{\circ}C$ reaching 55.6% and 82.5%, 24hr and 132hr after treatment, respectively. Optimum pH for resting spore germination was pH6, following pH7 and pH8, and the germination was inhibited at pH 4, and pH9. termination of resting spores was stimulated by root extracts of radish, Chinese cabbage and kidney bean, but inhibited by that of lettuce. Number of inactive resting spores was increased as temperature increases and time prolongs after temperature treatment. However, degree of inactivation of resting spores after 1hr at $40{\sim}65^{\circ}C$ was similar with $40{\sim}60%$, but rapidly increased to 91.5% at $70^{\circ}C$. When root galls were submerged in water, density of inactive resting spores was increased rapidly and reached 60.3% 9 days after treatment. Flooding of infested soil resulted in 30% reduction of survived resting spores 5 months later. Among the two registered fungicides, fluazinam was better for inactivation of resting spores than flusulfamide, but both fungicides were inferior to phosphoric acid.

Investigation of Various Pesticide Residues in Commercial Bee Pollen Products Sold in South Korea (한국에서 유통되는 화분식품의 잔류농약 함량 분석)

  • To analyze the pesticide residues in commercial bee pollen products in South Korea, 61 samples were collected and screened for 339 pesticides. Results revealed that approximately 34% (>LOQ) of samples were contaminated with at least one pesticide. The pesticide residue detection rates of domestic and imported samples were 31% and 44%, respectively. Furthermore, the pesticide residue detection rate of online distribution (60%) was higher than that of offline distribution (27%). Fifteen pesticides were discovered in bee pollen, and pendimethalin, chlorfenvinphos, chlorpyrifos, and fluazinam were detected in 7, 6, 3, and 2 order of frequency, respectively. Even though its concentration was low, chlorfenvinphos which is banned in food crops in the United States, European Union, and Korea, was detected in bee pollen samples commonly. Therefore, continuous investigation of pesticide residues in bee pollen products and their acceptance criteria is required for safety.
