Pulsatility Estimation of a Pulsatile Decellularizing Device for the Fabrication of Organ Scaffold (생체장기용 지지체 제작을 위한 박동형 탈세포화 장치의 박동성 평가)

  • To identify a solution for the restricted availability of healthy lungs and the high risk of immune rejections following organ transplantation, tissue engineering techniques for culturing lungs have been studied by many research groups. The most promising method for culturing lungs is the utilization of a bio-scaffold that was prepared using harvested organs from human donors or other animals by removing their original cells. In this study, a pulsatile perfusion pump was used to alleviate the cell removal effect with the high fluid-dynamic power of the perfusion stream during the decellularization process, while other conventional studies focused on chemical methods to identify efficient detergents. The purpose of this study was to analyze the developed device by using energy equivalent pressure (EEP), which is an indicator of pulsatility, to understand the characteristics of pulsatile energy transmitted according to the load size by using the artificial model and compare it with the measured EEP. The pulsatility of the device can be estimated with the concept of fluid-dynamic energy during a particular constant time period or fluid-dynamic power represented as EEP and EEP increment. Because the measured EEP of perfusion flow during decellularization can be changed by the amount of fluid leakage and the degree of clogging in the capillary vessels, EEP should be measured to determine whether the decellularization is progressing without problems. The decrement of EEP caused by the high perfusion resistance was observed from some experimental results that were obtained with artificial models. EEP can be used to monitor the decellularization process after analyzing the varying EEP according to the amount of load. It was confirmed that the EEP was maintained at a high level in the experiment using the harvested lungs from 12-13-week-old rats. In addition, it was confirmed that the cell removal time was faster than when continuous perfusion was performed. In this study, pulsatile power delivered to the lungs was measured to monitor the process of cell removal, and it serve as the evidence for efficient decellularization.

  • Rotary disc ultrafiltration module(RDM) was developed for the separation of oil e$$\mu$sions. This module was devised to reduce the gel polarization phenomenon by decoupling the operation pressure and the surface velocity of solution in ultrafiltration(UF) processes. The rotary disc membrane consists of 3mm-thick ABS plate covered with UF membrane (UOP, U.S.A.). When the angular velocity($\omega$) was increased, the pure water flux was slightly decreased due to pressure drop caused by centrifugal force and slip flow at the surface of membrane. The pressure drop was proportional to the square of linear velocity(${\omega}r$). When the angular velocity was changed from 52.36rad/s to 2.62rad/s, the flux decline for 5% cutting oil in one-shaft RDM at $25^{\circ}C$ and 0.1MPa was 30.16%. In the lower concentrations, angular velocity tends to give less effect on the flux. Flux(J; $kg/m^{2} \cdot s$) in a rotating disc module is mainly a function of the bulk concentration($C_{B}$; %), the linear velocity(${\omega}r$; m/s) and the effective transmembrane pressure($\Delta P_{T}$ ; Pa). Using a modified resistance-in-series model, the flux data of cutting oil experiments were fitted to give the following equation.

Role of Tumor Necrosis Factor-Producing Mesenchymal Stem Cells on Apoptosis of Chronic B-lymphocytic Tumor Cells Resistant to Fludarabine-based Chemotherapy

  • Background: B-cell chronic lymphocytic leukemia B (B-CLL), the most common type of leukemia, may be caused by apoptosis deficiency in the body. Adipose tissue-derived mesenchymal stem cells (AD-MSCs) as providers of pro-apoptotic molecules such as tumor necrosis factor-related apoptosis-inducing ligand (TRAIL), can be considered as an effective anti-cancer therapy candidate. Therefore, in this study we assessed the role of tumor necrosis factor-producing mesenchymal stem cells oin apoptosis of B-CLL cells resistant to fludarabine-based chemotherapy. Materials and Methods: In this study, after isolation and culture of AD-MSCs, a lentiviral LeGO-iG2-TRAIL-GFP vector containing a gene producing the ligand pro-apoptotic with plasmid PsPAX2 and PMDG2 virus were transfected into cell-lines to generate T293HEK. Then, T293HEK cell supernatant containing the virus produced after 48 and 72 hours was collected, and these viruses were transduced to reprogram AD-MSCs. Apoptosis rates were separately studied in four groups: group 1, AD-MSCs-TRAIL; group 2, AD-MSCs-GFP; group 3, AD-MSCs; and group 4, CLL. Results: Observed apoptosis rates were: group 1, $42{\pm}1.04%$; group 2, $21{\pm}0.57%$; group 3, $19{\pm}2.6%$; and group 4, % $0.01{\pm}0.01$. The highest rate of apoptosis thus occurred ingroup 1 (transduced TRAIL encoding vector). In this group, the average medium-soluble TRAIL was 72.7pg/m and flow cytometry analysis showed a pro-apoptosis rate of $63{\pm}1.6%$, which was again higher than in other groups. Conclusions: In this study we have shown that tumor necrosis factor (TNF) secreted by AD-MSCs may play an effective role in inducing B-CLL cell apoptosis.

Autothermal Reforming of Propane over Ni/CexZr1-xO2 Catalysts (Ni 담지 CexZr1-xO2 촉매상에서 프로판의 자열개질반응)

  • In this study, the catalytic performance and characterization of $Ni/Ce_xZr_{1-x}O_2$ were investigated using an autothermal reforming (ATR) process for hydrogen production. The $Ni/Ce_xZr_{1-x}O_2$ catalysts were prepared using the following methods: the water method (CZ-W), urea water method (CZ-UW) and urea, water and ethanol method (CZ-UWA). The performance of $Ni/Ce_xZr_{1-x}O_2$ catalysts in autothermal reforming of propane for hydrogen production was studied in a fixed-bed flow reactor. Reaction tests were conducted by using a feed of $H_2O/C_3H_8/O_2$=3/1/0.37 and $300{\sim}700^{\circ}C$. The CZ-UW and CZ-UWA catalysts showed higher propane conversion and hydrogen yield than the CZ-W catalyst. The activity test confirmed that the improvement in the water-ethanol catalyst was due to the low level of carbon deposition. SEM showed that the surface carbon consisted of clusters on the used CZ-UW catalyst, which is incontrast to the nano-fiber morphology observed on the used CZ-UWA catalyst. It was found that the amount of carbon deposition depends on the preparation method. Especially the $Ni/Ce_{0.75}Zr_{0.25}O_2$ was showed higher propane conversion and hydrogen yield than the other catalysts. Also TGA showed that the resistance of carbon deposition increase to Co addition.

  • Telemetering may be described as the art of metering at a considerable distance those quantities which are ordinarily encountered in industry, and in the generation of electric power. It is in the production of electric power that telemetering is particularly important, for it permits the system operator, or load dispatcher, to have before him at all times a continuous graphic record of the power output of each individual generating station together with an automatically made continuous graphic record of the total system output. There desired individual graphic records may be obtained showing power flow in or out, on important tie lines, etc. Such arrangements have the very great advantage that loads may be assigned to each generating station so that the best over-all system economy may be obtained and the system operator at all times may see with his own eyes that loads scheduled are actually held at the various stations. Moreover, with such equipment, in the event of system or station trouble the load dispatcher can see exactly what station, or stations, are affected and to what extent, without having to get in touch with anyone by telephone. Decisions can, therefore, be quickly reached for rescheduling the load. One of the most accurate and reliable telemetering systems is based on the use of potentiometric circuits, the fundamentals of which are discussed below. A member of such telemetering systems have been installed for the Boston Edicon Co., Boston, Massachusetts, the Consolidated Edison Co. of New York City. The Public Service Gas & Electric Co. of Newark, new Jersey. The Philadelphia Electric Co. for Philadelphia, Pennsylvania, and the Pennsylvania Railroad Co. for their electrified zone between New York and Washington a distance of over 200 miles. The scale of the totalizing recorder for the New York area is 3,000,00 KW. That of the totalizing recorder for the Philadelphia area is 2,000,000 KW. The initial installation using this type of equipment described was placed in service for the Philadelphia Electric Co. in 1923. All of the original recording instruments are still in service, later instruments have been added to take care of additions to the power system and naturally these later recorders have incorporated in those refinements in design made since the earlier ones were manufactured. Many other installations of similar equipment have been made in the United States in various locations such as at St. Louis, on the West Coast, at Baltimore and in Washington, D.C. While the use of these basic potentiometric circuits involves the use of continuous metallic circuits of good insulation resistance and free of grounds, nevertheless, intermediate transmission links, involving and impulse method suitable for use on telephone Morse carrier channels is available. This same method may be employed on power line carrier systems and is also suitable for use on beam type microwave transmission. Many impulse type units are also used as a link in these potantiometric methods. For the sake of brevity a description is given only of these basic potentiometric circuits. If there is sufficient interest in Korea, a further paper can be given covering those impulse circuits also.

  • Various researches on the application of polymer dispersions to the cement mortar and concrete have been practised in many countries like America, Japan and Germany and so on because of high performance and good modification effect of these. In this study, SBR, Polymer dispersion that widely used in situ is employed that the self-flowability may be induced in the cemen mortar. In order to comprehend and investigate the modification of cement mortar with self-flowability by SBR and properties and fracture mode of adhesive strength in tension of that, experimental parameter was set as SBR solid-Cement ratio(S/C) and Cement:Fine aggregate(C:F) and the experiments such as Unit weight, Flow, Consistency change, Crack resistance and Segregation that inform on the general properties have been done. In addition of that, Adhesion in tension is measured with a view to comprehending the properties and fracture mode in tensile load. Consistency change of cement mortar modified by SBR did grow better as the ratio of SBR solid-Cement increased and was much superior to that of resin based flooring such as polyurethane and epoxy which recorded the loss of consistency in 90min. after mixing. Adhesive strength in tension increased with continuity in the curing age and showed the maximum in case of C:F=1:1 and S/C=20%. As the increase of curing age, the fracture mainly happened in the concrete substrate and the interface between the specimen and concrete substrate.

Gender Differences in Physiological Effects of a Transient Exposure to Experimental Noise

  • The physiological responses following stress are different in individual or personality. We performed this study to clarify gender differences in influences of noise stress on physiological factors. 70 healthy subjects, which was divided man (n=30) and woman (n=40) groups, were exposed to 85 decibels of excavator noise for 15 minutes. Cardiac factors such as heart rate (HR), systolic and diastolic blood pressures (SBP and DBP, respectively), and heart rate-systolic pressure product (RPP) were determined. Transcranial Doppler ultrasound (TCD) was used to measure mean blood flow velocity (Vm), pulsatility index (PI), and resistance index (RI) in the middle, anterior and posterior cerebral arteries (MCA, ACA and PCA, respectively) before and during noise exposure. Cortisol level and hematological variables were also measured before (baseline) and immediately after the end of noise exposure. In the both groups HR, SBP, and RPP significantly decreased during noise exposure (P<0.05) but not significantly different between two groups (P>0.05). Vms of three cerebral arteries in man group decreased, whereas Vm of PCA in woman group fell during noise exposure (P<0.05). Vm, PI and RI in MCA and ACA during noise exposure were low in man group compared with woman group (P<0.05). Vm of PCA was low, whereas PI and RI of PCA were high in man group compared with woman group during noise exposure (P<0.05). Total leukocyte and red blood cell (RBC) counts slightly decreased during noise exposure but not significant (P>0.05). Levels in hematological variables decreased but not significant changed following noise exposure. Decreased rate of total leukocyte in man group was higher (P<0.05). Cortisol levels in the both groups decreased immediately after the end of noise exposure, while the decreased rate in man group was greater than that in woman group (p<0.05). These findings indicate that a transient exposure to experimental excavator noise may cause decreased changes in cardiac factors, cerebral hemodynamics and cortisol levels and the changes may be greater in men than in women.

Variation of Soil Properties by Permeating Injection of Chemical Grouts (약액(藥液)의 침투주입(浸透注入)에 의한 토질성상변화(土質性狀變化))

  • Variation of soil properties is studied by permeating injection of chemical grouts, such as cement type, water-glass type and acrylamide type, to the same soil samples with different densities. Moreover, injection tests using specially prepared equipments of 1.0 shot system and 1. 5 shot system are attempted to investigate permeating injection effects in highly compacted soil and in the presence of ground water. The main factor which causes the improvement of cut-off effect and shearing strength is the cohesion of soil. The strength in the loose state is fundamentally governed by the membrane cohesion, meanwhile, in the loose state is governed by the structural cohesion. Injection effects under the ground water flow is considerably decreased, and effective gelling ratio of approximate 45~80% is observed by variation of velocity and gel time, besides grading of injection materials has high relation with permeation and traveling length but has little relation with effective gelling ratio. Permeating injection effects, such as gelling scope, gelling strength in highly compaoted soil conditions can be confirmed by penetration resistance diagram and iso-strength curve.

  • In this study, the energy conversion equipment from the radiation energy to mechanical energy by using n­pentane as the operating fluid was constructed and the performance to pump the water was tested for the utilization of solar powered water pump. The equipment was designed optimally, after the theoretical analyses of the water pumping head and water quantity per cycle were done. The pentane vapour temperature in the condenser and the temperature of the outlet water from the condenser became lowered and the heat transfer rate became higher with decreasing the water inlet level to the condenser. The temperature difference between the condenser and the water tank was significant. Therefore, the distance between the water tank and condenser was recommended to be shorten and the diameter of their connecting pipe was recommended to be narrow in order to reduce the resistance of the fluid passage and improve the heat transfer rate. The amount of water pumped was 1.6­2.4 liters. Mass flow rate of the cooling water became lowered when the cooling water pipe was prolonged from the condenser to improve the heat transfer rate.

Time - and Concentration - Dependent Effects of Resveratrol on miR 15a and miR16-1 Expression and Apoptosis in the CCRF-CEM Acute Lymphoblastic Leukemia Cell Line

  • Background: Chemotherapy is one of the common approaches in treatment of cancers, especially leukemia. However, drug resistance phenomena reduce the likelihood of treatment success. Resveratrol is a herbal compound which through complicated processes makes some selected cells sensitive to treatment and induction of apoptosis. In the present study, the effects of resveratrol on the expression of miR 15a and miR16-1 and apoptosis in the CCRF-CEM cell line were investigated. Materials and Methods: The CCRF-CEM cell line was cultured under standard conditions and changes in miR 15a and miR 16-1 expression were analyzed by real time-PCR technique, with attention to reveratrol dose and time dependence. Also, apoptosis is evaluated by flow cytometry using annexin V and PI. Results: CCRF-CEM cells underwent dose-dependent apoptotic cell death in response to resveratrol. MiR 15a and miR 16-1 expression was up-regulated after 24 and 48 hours resveratrol treatment (p<0.05). Conclusions: The results of our study indicate that resveratrol induces apoptosis in a time and dose-dependent manner in CCRF-CEM cells. Also, increased expression level of miR 16-1 and miR 15a by means of resveratrol in CCRF-CEM cells might have a role in apoptosis induction and predisposition. According to our results resveratrol can be regarded as a dietary supplement to improve efficacy of anti-leukemia therapies.
