Accelerating Effect of $TNF-{\alpha}$ on the Rhus verniciflua-induced Growth Inhibition and Apoptosis in Human Osteosarcoma Cells

  • Previously, a flavonoid fraction, which consisted mainly of protocatechuic acid, fustin, fisetin, sulfuretin, and butein, here named RCMF [${\underline{R}}hus$ verniciflua Stokes (RVS) ${\underline{c}}hloroform-{\underline{m}}ethanol\;{\underline{f}}raction$], was prepared from a crude acetone extract of RVS which is traditionally used as a food additive and as an herbal medicine. In the present study, we investigated the effects of $TNF-{\alpha}$ on RCMF-induced growth inhibition and apoptosis induction using human osteosarcoma (HOS) cells. The results from tritium uptake and MTT assays showed that $TNF-{\alpha}$ treatment itself (10 ng/ml) did not induce any cytotoxicity, but it actively accelerated RCMF-mediated cytotoxicity of HOS cells. RCMF-induced cytotoxicity and its facilitation by $TNF-{\alpha}$ was verified to be apoptotic, based on the increased DNA fragmentation and low fluorescence intensity in nuclei after propidium iodide (PI) staining of HOS cells. This speculation was further demonstrated by monitoring the Annexin V/PI double staining which could discriminate the difference between apoptotic and necrotic deaths. Collectively, our findings indicate that $TNF-{\alpha}$ accelerates RCMF-induced cytotoxicity in HOS cells.

Evaluation of Antioxidant and Antimicrobial Activities of Ethanol Extracts of Three Kinds of Strawberries

  • The antioxidant and antimicrobial activities of three kinds of strawberry ethanol extracts from Robus corchorifolius L. f. (RCL), Rubus parvifolius L. var. parvifolius (RPL), and Duchesnea chrysantha Miq. (DCM) were investigated. The RPL was highest (P<0.05) in phenolic, flavonoid, and anthocyanin contents. 2,2-Diphenyl-1-picrylhydrazyl and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) radical scavenging activities of RPL and DCM extracts were higher than that of RCL (P<0.05). Hydrogen peroxide scavenging activity of RPL was high compared to DCM and RCL (P<0.05). RCL exhibited a significant (P<0.05) potent antioxidant activity in nitric oxide radical inhibition. Inhibition diameter zone (nearest mm) of extracts against the test bacteria ranged from 11.5 in RCL to 12.5 in DCM against Staphylococcus aureus, from 10.5 in RCL to 13.5 in DCM against Streptococcus pneumoniae, from 8.5 in DCM to 10.5 in RCL against Escherichia coli, and the same inhibition of 10 mm in three of the extracts against Klebsiella pneumoniae. However, there was no inhibition against fungi Aspergillus niger and Candida albicans. Three of the extracts had the same minimum inhibitory concentration values of 12.50, 12.50, and $6.25{\mu}g/mL$ against S. aureus, K. pneumoniae, and S. pneumoniae, respectively. On the other hand, MIC values of 12.50, 12.50, and $6.50{\mu}g/mL$ were recorded for RPL, DCM, and RCL against E. coli, respectively. The result of present study revealed that extracts from three kinds of strawberries could be potential candidates as antioxidant and antimicrobial sources for functional food industries.

Bioavailability of plant pigment phytochemicals in Angelica keiskei in older adults: A pilot absorption kinetic study

  • BACKGROUND/OBJECTIVES: Angelica keiskei is a green leafy vegetable rich in plant pigment phytochemicals such as flavonoids and carotenoids. This study examined bioavailability of flavonoids and carotenoids in Angelica keiskei and the alteration of the antioxidant performance in vivo. SUBJECTS AND MATERIALS: Absorption kinetics of phytochemicals in Angelica keiskei were determined in healthy older adults (> 60 y, n = 5) and subjects with metabolic syndrome (n = 5). Subjects consumed 5 g dry Angelica keiskei powder encapsulated in gelatin capsules with a low flavonoid and carotenoid liquid meal. Plasma samples were collected at baseline, 0.5, 1, 2, 3, 4, 5, 6, 7, and 8 h. Samples were analyzed for flavonoids and carotenoids using HPLC systems with electrochemical and UV detection, respectively, and for total antioxidant performance by fluorometry. RESULTS: After ingestion of Angelica keiskei increases in plasma quercetin concentrations were observed at 1-3 and 6-8 hr in the healthy group and at all time points in the metabolic syndrome group compared to baseline (P < 0.05). Plasma lutein concentrations were significantly elevated in both the healthy and metabolic syndrome groups at 8 hr (P < 0.05). Significant increases in total antioxidant performance were also observed in both the healthy and the metabolic syndrome groups compared to baseline (P < 0.05). CONCLUSIONS: Findings of this study clearly demonstrate the bioavailability of phytonutrients of Angelica keiskei and their ability to increase antioxidant status in humans.

The Effect of Antioxidant and Antimicrobial Activity on the Extracted Its Material and Aloe vera L. Callus Culture by the Natural Fruit Juice (천연과즙을 이용한 Aloe vera L.의 callus 배양과 이들 추출물의 항산화 및 항균활성 효과 검정)

  • In order to obtain functional materials from aloe callus, we cultured Aloe vera L. leaf on MS medium added 0.2 mg/L IAA, 0.3 mg/L kinetin and 100 mg/L grape or/and apple juice for 30 days. While a callus differentiation during callus culture did not show, the cultured leaves were uniquely released extracellular material into the agar plate. After cultivation for 18 days, the cultured leaf and agar were harvested for extraction a functional material. The materials extracted were measured on the amount of total phenols, flavonoids and polysaccharides and determined on the antioxidant and antimicrobial activity. In result, callus extracts of additive free (CT) and added apple juice (2T) had more amount of phenol compound ($659{\mu}g/mL$, $533{\mu}g/mL$) and flavonoid ($580{\mu}g/mL$, $501{\mu}g/mL$) than natural leaf (p: $525{\mu}g/mL$, f: $301{\mu}g/mL$). However, the extract of natural leaf had the better effect of lipid peroxidation and polysaccharide content than the culture extract. All samples extracted had same effect on the nitrite scavenging activity. On the other hand, only 2T extract showed excellent 72% nitric oxide scavenging activity. The agar extract was also confirmed to contain polyphenol compound and polysaccharide content that had antioxidant and antimicrobial activity partly.

  • Hwanggeumjackyak-tang (HJT) composed of Scatellaria baicalensis Georgi, Paeonia Lactiflora and Glycyrrhizae uralensis Fischer is a traditional Korean herbal medicine widely used for acute enteritis. In order to develop the tablet formulation of HJT, evaluation of the flow properties, thickness, diameter, hardness, friability and disintegration was carried out on four HJT granules according to mixed content of seven additives. Simultaneous analysis used HPLC method was performed of HJT tablet and was determined of the seven marker components; Albiflorin, Paeoniflorin, Liquiritin, Baicalin, Baicalein, Glycyrrhizic acid and Wogonin. The biological activities were examined the effect of HJT on anti-oxidation and pro-inflammation mediated by LPS-stimulation. We confirmed that both of HJT-Decoction (HJT-D) and HJT-Formulation (HJT-F) have the similar contents on total polyphenol and flavonoid and inhibited the secretion of nitro oxide (NO), interleukin $(IL)-1{\beta}$,interleukin (IL)-6, tumor necrosis factor $(TNF)-{\alpha}$ and the expression of nitric oxide synthase (iNOS), $IL-1{\beta}$, IL-6, $TNF-{\alpha}$. Therefore, the developed formulation for tablet of HJT would provide chemically and biologically the same effect compared with decoction of HJT.

Characterization of Purple-discolored, Uppermost Leaves of Soybean; QTL Mapping, HyperspectraI Imaging, and TEM Observation

  • Purple-discoloration of the uppermost leaves has been observed in some soybean cultivars in recent years. The purpose of this study was to characterize the novel phenotypic changes between the uppermost and middle leaves via multiple approaches. First, quantitative trait loci mapping was conducted to detect loci associated with the novel phenotype using 85 recombinant inbred lines (RILs) of the 'Daepung' × PI 96983 population. 180K SNP data, a major quantitative trait locus (QTL) was identified at around 60 cM of chromosome 6, which accounts for 56% of total phenotypic variance. The genomic interval is about ~700kb, and a list of annotated genes includes the T-gene which is known to control pubescence and seed coat color and is presumed to encode flavonoid 35-hydroxylase (F3'H). Based on Hyperspectral imaging, the reflectance at 528-554 nm wavelength band was extremely reduced in the uppermost leaves compared to the middle (green leaves), which is presumed die to the accumulation of anthocyanins. In addition, purple-discolored leaf tissues were observed and compared to normal leaves using a transmission electronic microscope (TEM). Base on observations of the cell organelles, the purple-discolored uppermost leaves had many pigments formed in the epidermal cells unlike the normal middle leaves, and the cell wall thickness was twice as thick in the discolored leaves. The thickness of the thylakoid layer in the chloroplast the number of starch grains, the size of starch all decreased in the discolored leaves, while the number of plastoglobule and mitochondria increased.

A Set of Anthocyanin Biosynthetic Genes are Differentially Expressed in Strawberry (Fragaria x ananassa cv Maehyang) during the Fruit Development Process (매향 딸기로부터 anthocyanin 합성 유전자의 분리 및 과실발달 과정에서의 발현 분석)

  • Anthocyanin synthesis in strawberry (Fragaria x ananassa cv Maehyang) begins approximately 26 days postflowering and continued throughout fruit ripening. A set of cDNA clones encoding the anthocyanin biosynthetic enzymes were isolated from strawberry. A pair of primers were designed for polymerase chain reaction (PCR) through the comparison of the nucleotide sequences of homologous genes from diverse plants. Reverse transcriptase-PCRs were performed using cDNA synthesized from ripe fruit total RNA and the primers corresponding to each gene. Eight genes of the anthocyanin pathway were cloned and confirmed by sequencing to code for phenylalanine ammonia lyase (PAL), 4-cummarate CoA ligase (4CL), chalcone synthase (CHS), chalcone isomerase (CHI), flavanone-3-hydroxylase (F3H), dihydroflavonol 4-reductase (DFR), anthocyanidine synthase (ANS), UDP-glucose:flavonoid-3-O-glucosyl-transferase (UFGT). Northern analyses showed that the corresponding genes were differentially expressed during the fruit development process. All genes except PAL were predominantly expressed in fruit. Expression of PAL, DFR and ANS was detected 10 days postflowering at the early stage of fruit development, declined for a while and sharply increased 22 days postflowering then showed a peak 34 days postflowering. The other genes, however, were not expressed up to 22 or 30 days postflowering when the initial fruit ripening events occur at the time of initiation of anthocyanin accumulation. The onset of anthocyanin synthesis in ripening strawberry coincides with a coordinated induction of the anthocyanin pathway genes, suggesting the involvement of regulatory genes. We propose that at least two different regulatory mechanisms playa role in the biosynthesis of anthocyanin during color development of strawberry.

Comparison of ginsenoside contents and antioxidant activity according to the size of ginseng sprout has produced in a plant factory (식물공장에서 생산된 새싹인삼의 크기에 따른 진세노사이드 함량 및 항산화 활성 비교)

  • In this study, the ginseng sprout has produced through smart farm was classified according to its size and divided into above-ground (AG) and below-ground (BG) parts to compare ginsenoside contents and antioxidant activity. In the case of the AG part, the total phenolic contents were the highest at 5.16 mg/g in medium (M) size and the lowest at 2.23 mg/g in largest (L) size. The BG part also showed the highest content in the M size, but there was no significant difference. Also, the total flavonoid contents were also high in the M size in both the AG (5.16 mg/g) and BG (1.28 mg/g) parts. The major ginsenosides in the AG part were Re (20.33-24.15 mg/g) > Rd (11.36-27.42 mg/g) > Rg1 (4.48-5.54 mg/g) and the main ginsenosides in the BG part were Rb1 (5.09-8.61 mg/g) > Re (4.48-5.54 mg/g) > Rc (3.11-4.11 mg/g) in orders. In the case of M size, Re and Rd were approximately 4- and 19-folds higher at 24.15 mg/g and at 27.42 mg/g in the AG part and 5.20 mg/g and 1.43 mg in the BG part, respectively. In addition, F3 and Rh1 were detected in the AG part, but not in the BG part. 2,2-diphenyl-1-picrylhydrazyl (74.95%), 2,4,6-azino-bis (3-ethylbenzothiazoline-6-sulphnoic acid) diammonium salt (94.47%), and hydroxyl (70.39%) radical scavenging activities and FRAP (2.169) assay were the highest in M size than other sizes.

Antioxidative Properties of Asparagus cochinchinensis Root (천문동 건근의 항산화 효과에 관한 연구)

  • This study was performed to compare the antioxidative activities of methanol extracts from Asparagus cochinchinensis with whole root (W-AC), flesh (F-AC), and root bark (B-AC). To evaluate the antioxidative properties of their methanol extracts, 1,1-diphenyl-2-picrylhydrazyl radical, nitrite, hydroxyl radical, 2,2'-azino-bis(3-ethylbenz thiazoline-6-sulfonate) radical scavenging activities, and contents of total flavonoid and polyphenol contents were measured. B-AC extract showed the highest antioxidative activity, whereas F-AC extract showed the lowest. For B-AC extract, caffeic acid was isolated by preparative high-performance liquid chromatography and confirmed by liquid chromatography-mass spectrometry and absorption spectroscopy, which showed 1.6% of total polyphenol contents among all methanol extracts.

Nutritional composition and functionality of mixed cereals powder (혼합잡곡분말의 영양성 및 기능성)

  • Six types of mixed cereals powder were prepared using different mixing ratios of barley, proso millet, sorghum, and adlay, and their nutritional components and functionalities were analyzed. The mixed cereals powders contained high levels of crude protein. The free amino acid and mineral contents of Sample E were the highest among all the samples. Fructose, glucose, sucrose, and maltose were detected as the four major free sugars in all the mixed cereals powders, and Sample D had the highest amount of reducing sugar among all the samples. The total polyphenol and total flavonoid contents of Sample F were significantly higher than those of the other samples. Sample C showed the highest level of DPPH radical scavenging activity and reducing power. As for the ABTS radical scavenging activity, the $IC_{50}$ of Sample D was the lowest among all the samples, but did not significantly differ from that of Sample C. Therefore, we suggest 2:2:1:1 (in Sample D) as the best mixing ratio of barley, proso millet, sorghum, and adlay to produce a mixed cereals powder with excellent nutritional value and antioxidant activities.
